Abstract:Lagotis yunnanensis is a folkloric medicine in China. In order to investigate its effective components, The 95% ethanol extract of whole plant of L. yunnanensis was separated and purified with chromatographic method. Eight compounds were obtained and identified as luteolin (1), chrysoeriol (2), apigenin-7-methyl ether (3), 3, 4-dimethoxycinnamyl-D-glucopyranoside (4), verbascoside (5), cistanoside D (6), 2-O--D-glucopyranospylcucurbitacin D (7) and arvenin I (8) by spectroscopic methods. Compounds 1~3 are flavonoids, compounds 4~6 are phenylpropanoids and compounds 7 and 8 are triterpenoids. Compounds 4~8 have been obtained from this species for the first time.

Abstract:Objective:To study the chemical components of the overground parts of Lamiophlomis rotata (Benth.) Kudo.Methods: Chemical components of Lamiophlomis rotata Kudo were isolated by chromatography methods and the products were identified by physiochemical and spectral analysis.Results: Three flavonoids and 3 phenylethanoid glycosides were isolated and identified: luteolin-7-O-β-D-glucopyranoside (Ⅰ),apigenin-7-O-β-D-glucopyranoside (Ⅱ),luteolin-7- O-[β-D-apiofuranosyl(1→6)-β-D-glucopyranoside (Ⅲ),forsythoside B(Ⅳ),verbascoside(Ⅴ),betonyosides A(Ⅵ).Conclusion: Compounds Ⅲ-Ⅵ have been isolated from Lamiophlomis rotata kudo for the first time.

Abstract:Lamiophlomis rotata (Lamiaceae), a perennial medicinal herb, is endemic to the Qinghai-Tibet Plateau. A total of 188 individuals from eight natural populations of L. rotata in Qinghai-Tibet Plateau (four from Tibet, two from Yunnan, and two from Qinghai) were analyzed using intersimple sequence repeats (ISSR) and randomly amplified polymorphic DNA (RAPD) techniques. Our results revealed that the level of genetic variation in L. rotata was relatively high (P = 94.85%, I = 0.440 +/- 0.220, H(T) = 0.289 +/- 0.028). Three genetic groups corresponding to the three geographic regions were detected, suggesting significant geographic structure. Our results suggest that the highly structured geographic pattern found in L. rotata might represent diverging evolutionary processes associated with the uplifting of the Qinghai-Tibet Plateau and Quaternary climatic oscillations. These findings imply that as many populations as possible should be preserved in situ for the conservation of this species. Given their genetic variability and peripheral distribution, Qinghai and Yunnan populations should be assigned priority for conservation. Optimal harvesting strategies, domestication and tissue culture of L. rotata should be developed as soon as possible to guarantee its sustainable use.
Target and non-target identification of chemical components in Lamiophlomis rotata by liquid chromatography/quadrupole time-of-flight mass spectrometry using a three-step protocol

Abstract:Rationale: As a herbal plant used in traditional Chinese medicine, Lamiophlomis rotata (Benth.) Kudo mainly displays its pharmacological effect by promoting blood circulation and hemostasis, dispelling wind, and acting as an analgesic. To identify the components contained in L. rotata, global detection and structural elucidation of both target and non-target components in the medicinal material was performed. Methods: L. rotata was ultrasonically extracted with methanol. Separation and analysis were achieved using liquid chromatography/quadrupole time-of-flight mass spectrometry (LC/QTOF-MS). A three-step protocol which included (1) potential components screening, (2) collection of qualitative information, and (3) database searching and structural elucidation was used for target and non-target identification. Results: A total of 42 components were tentatively identified, which included 12 iridoids (2 aglycones and 10 glucosides), 11 flavonoids (4 aglycones and 7 glucosides), and 13 phenylethanoid glycosides. Moreover, components of L. rotata extract belonging to the three main structural categories could be well separated in a 3D point plot according to their retention times, mass defects and degrees of unsaturation, facilitating the structural classification and identification in the subsequent studies. Conclusions: The results provide a reasonable picture of the components contained in L. rotata extract and promote the further pharmacodynamic and/or pharmacokinetic characterization of this medical material, meanwhile demonstrating the utility of a universal methodology for the systematical study of herbal medicines. Flavonoid Synthesis in Lamiophlomis Rotata from Qinghai-Tibet Plateau is Influenced by Soil Properties, Microbial Community, and Gene Expression

Abstract:Lamiophlomis rotata is a medicinal plant in Qinghai-Tibet Plateau, in which flavonoid compounds are the major medicinal components. However, it remains unclear how flavonoid metabolism of L. rotata is influenced by soil properties and microbial community. In this study, we collected L. rotata seedlings and rhizosphere soils from five habitats ranging from 3750 to 4270 m of altitude and analyzed the effects of habitat conditions on flavonoid metabolism. The activities of peroxidase, cellulase, and urease were increased with altitude, while those of alkaline phosphatase, alkaline protease, and sucrase were decreased with altitude. Analysis of OTUs showed that the total number of bacterial genera was higher than that of fungal genera. The highest number of fungal genera was 132, and that of bacterial genera was 33 in Batang (BT) town in Yushu County at an altitude of 3880 m, suggesting that the fungal communities may play a critical role in L. rotata rhizosphere soils. Flavonoids in leaves and roots of L. rotata shared a similar pattern, with a trend of increasing levels with altitude. The highest flavonoid content measured, 12.94 mg/g in leaves and 11.43 mg/g in roots, was from Zaduo (ZD) County at an altitude of 4208 m. Soil peroxidases affected quercetin content in leaves of L. rotata, while the fungus Sebacina affected flavonoid content in leaves and roots of L. rotata. The expression of PAL, F3′H, FLS, and FNS genes showed a declining trend in leaves with altitude, while F3H showed an increasing trend in both leaves and roots. Overall, soil physicochemical properties and microbial community affect flavonoid metabolism in L. rotata in Qinghai-Tibet Plateau. The variations in flavonoid content and gene expression as well as their associations with soil factors revealed the complexity of the growth conditions and genetic makeup in L. rotata habitats of Qinghai-Tibet Plateau.

Kudo, the compounds were isolated by silica gel, MCI chromatography and preparative HPLC.Their structures were then elucidated on the basis of chemical and spectroscopic measurements.The results indicated that eleven compounds were isolated from Lamiophlomis rotata ( Benth.) Abstract:Lamiophlomis rotata (Labiatae) is a Chinese folk medicinal plant in Xi-zang (Tibet), which has effects of promoting blood circulation to remove blood stasis, subduing swelling and alleviating pain. Four iridoid glucosides were isolated from the root of the species. On the basis of IR, UV, NMR, MS spectral data and chemical methods, their structures were identified as: 8-O-acetylshanzhiside methyl ester(I), 6-O-acetylshanzhiside methyl ester(II), penstemoside(III) and 7, 8-dehydropenstemoside(IV). The last one is a new compound.

Abstract:Flavonoids and volatiles in a traditional herbal medicine Chrysanthemum morifolium Ramat flower were determined by HPLC and GC/MS, respectively. Eight flavonoids and fifty eight volatiles were identified. Luteolin-7-glucoside and quercitrin were the most abundant flavonoids and they amounted for 85.7% of the total detected flavonoids. -Humulene was the most abundant volatile and ledene oxide-(I) the next abundant one and the two volatiles were 16.3 and 9.0% of total volatiles, respectively. The bioactivities of the partial components in C. morifolium Ramat flower were discussed. It is considered that the health benefits of C. morifolium Ramat might be related to its abundant flavonoids and volatiles.

A Transcriptomic and Metabolomic Study on the Biosynthesis of Iridoids in Phlomoides rotata from the Qinghai–Tibet Plateau

Abstract:Phlomoides rotata is a traditional Chinese herbal medicine that grows in the Qinghai–Tibet Plateau region at a 3100–5000 m altitude. Iridoid compounds are the main active compounds of the P. rotata used as medical ingredients and display anti-inflammatory, analgesic, and hepatoprotective properties. To better understand the biological mechanisms of iridoid compounds in this species, we performed a comprehensive analysis of the transcriptome and metabolome of P. rotata leaves from four different regions (3540–4270 m). Global metabolome profiling detected 575 metabolites, and 455 differentially accumulated metabolites (DAMs) were detected in P. rotata leaves from the four regions. Eight major DAMs related to iridoid metabolism in P. rotata leaves were investigated: shanzhiside methyl ester, 8-epideoxyloganic acid, barlerin, shanzhiside, geniposide, agnuside, feretoside, and catalpin. In addition, five soil physical and chemical indicators in P. rotata rhizosphere soils were analyzed. Four significant positive correlations were observed between alkaline nitrogen and geniposide, exchangeable calcium and geniposide, available potassium and shanzhiside, and available phosphorus and shanzhiside methyl ester. The transcriptome data showed 12 P. rotata cDNA libraries with 74.46 Gb of clean data, which formed 29,833 unigenes. Moreover, 78.91% of the unigenes were annotated using the eight public databases. Forty-one candidate genes representing 23 enzymes involved in the biosynthesis of iridoid compounds were identified in P. rotata leaves. Moreover, the DXS1, IDI1, 8-HGO1, and G10H2 genes associated with iridoid biosynthesis were specifically expressed in P. rotata. The integration of transcriptome and metabolome analyses highlights the crucial role of soil physical and chemical indicators and major gene expression related to iridoid metabolism pathways in P. rotata from different areas. Our findings provide a theoretical foundation for exploring the molecular mechanisms underlying iridoid compound accumulation in P. rotata.

Abstract:An ultra-high performance liquid chromatography coupled with time-of-flight mass spectrometry technique and a subsequent LC-MS/MS method were developed for metabolite profile study of Lamiophlomis rotata extract after its oral administration. Metabolite profile analysis for plasma, bile, urine and feces were performed on a sub-2-mm analytical column, and showed that 39 prototypes and 47 metabolites were identified or tentatively characterized. More than half the metabolites underwent phase II reaction, including sulfate, glucuronide, taurine, glycine, and glutathione conjugation. The targeted and valid LC-MS/MS method, using one-step protein precipitation for rat plasma, was applied for the pharmacokinetic study of shanzhiside methyl ester, 8-O-acetylshanzhiside methyl ester, lamalbid, verbascoside, forsythoside B, and luteolin-7-O-beta-D-glucopyranoside. Pharmacokinetics results showed that six bioactive components did not vary significantly in rats after administration of the aerial parts and the whole plant, which was consistent with the metabolite profile comparison between two groups. This article contributes to the understanding of the metabolism of L. rotata, and provides detailed performance analysis and scientific data to support the medicine parts alteration in the Chinese Pharmacopoeia.

Abstract:OBJECTIVE:To investigate the chemical constituents from Lamiophlomis rotata and the bioactivities of 8-epideoxyloganic acid.METHOD:The constituents were isolated by using a combination of various chromatographic techniques including column chromatography over ployamide, silica gel and Sephadex LH-20. Structures of the isolates were identified by spectroscopic data analysis. Bioactivities were screened by using models in vivo.RESULT:Five constituents were isolated. 8-epideoxyloganic acid was isolated for the first time in L. rotata and also in lamioplomis genus. 8-epideoxyloganic acid could significantly inhibit aectic acid-induced twisting times and slower the time of homeostatsis, also inhibit xylene-induced ear edema in mice.CONCLUSION:8-epideoxyloganic acid possesses bioactivities of analgesia, homeostasis and anti-inflammatory.

Abstract:Objective To analyze the chemical constituents of the volatile oil from flower of Stellera chamaejasma Linn. Methods The volatile oil from flower of Stellera chamaejasma Linn was extracted by steam distillation method,and was analyzed by gas chromatography-mass spectrometry(GC-MS). Results 22 compounds were identified,which represented 97.59%of the total GC peak area of the volatile oil.The main components were benzyl benzoate(24.216%)、4aH-cycloprop[e]azulen-4a-ol,decahydro-1.1.4.7-Tetramethyl-(14.092%),eugenol(9.313%),lilac alcohol C(6.166%),nerolidol 2(6.347%),alpha-bisabolol(2.98%),tricosane(8.769%),heneicosane(4.755%),benzoic acid,2-hydroxy-,phenylmethyl ester(2.571%)、pentacosane(2.522%),etc. Conclusion The present study provided scientific bases for Stellera chamaejasma Linn exploitation in reason.

Abstract:The composition of the volatile oils obtained from the leaves and flowers of Salvia przewalskii Maxim. grown in Tibet was analysed by gas chromatography–mass spectrometry (GC–MS) for the first time. Totals of 43 compounds were identified in the leaf oil and 69 compounds in the flower oil. A comparative study of the oil composition from the leaves and flowers found 22 common constituents of these oils, viz. α-thujene, β-phellandrene, linalool, terpinene-4-ol, diisopropenyl methyl vinyl cyclohexane, caryophyllene, germacrene D, borneol, caryophyllene oxide, spathulenol, germacrene B, δ-cadinene, humulene, farnesane, α-cadinol, β-caryophyllene, elemene, sabinene hydrate, γ-terpinene, p-cymene, camphene and α-pinene. The percentages of the most components were higher in the leaf oil than in the flower oil, especially those of β-phellandrene and caryophyllene (26.08% and 11.11%, respectively). Farnesene, chamigrene, n-hexadecane, epicedrol, cyclotetradecane, n-octadecane, 9-octadecen-1-ol, 1-heneicosyl formate, Z-8-hexadecen-1-ol acetate, n-heneicosane and n-pentacosane were the specific constituents found only in the flower oil, while β-cadinol was the specific constituent found only in the leaf oil. Abstract:Phlomoides rotata is a traditional medical plant at 3100-5200 m altitude in the Tibet Plateau. In this study, flavonoid metabolites were investigated in P. rotata from Henan County (HN), Guoluo County (GL), Yushu County (YS), and Chengduo County (CD) habitats in Qinghai. The level of kaempferol 3-neohesperidoside, sakuranetin, and biochanin A was high in HN. The content of limocitrin and isoquercetin was high in YS. The levels of ikarisoside A and chrysosplenol D in GL were high. Schaftoside, miquelianin, malvidin chloride, and glabrene in CD exhibited high levels. The results showed a significant correlation between 59 flavonoids and 29 DEGs. Eleven flavonoids increased with altitude. PAL2, UFGT6, COMT1, HCT2, 4CL4, and HCT3 genes were crucial in regulating flavonoid biosynthesis. Three enzymes CHS, 4CL, and UFGT, were crucial in regulating flavonoid biosynthesis. This study provided biological and chemical evidence for the different uses of various regional plants of P. rotata.

Abstract:Hydrodistilled leaf oils of Pistacia chinensis Bunge from five locations in China were analyzed using GC/MS. A total of 58 compounds was identified in the oils, and a relatively high variation in their contents was found. The major compounds include β-phellandrene (0.54–53.86%), α-pinene (4.74–54.44%), β-pinene (0.49–42.90%), caryophyllene (5.64–20.01%), cis -ocimene (tr−43.93%), eudesmadiene (0–15.06%), and camphene (tr−20.57%). Cluster analysis classified the leaf oils into two chemotypes: one rich in α-pinene and β-pinene, and the other rich in β-phellandrene.
