Abstract:The effects on catalase (CAT), glutathione -S-transferase(GST) activities and content of malondialdehyde (MDA), reduced glutathione (GSH) in the gill of Meretrix meretrix were evaluated, after it had exposed to tributyltin (TBT) at environmental levels (0.1, 1, 10 ng·L -1 as Sn) for 2, 8, 20 days and recovered for 7, 20 days, in experimental condition. The results showed that the activities of GST and CAT and GSH content in Meretrix meretrix exposed to TBT were principally induced, except that were mainly inhibited in the clams exposed to 10 ng·L -1 for 2 days. MDA content were significantly induced after exposure for 8d. After the clams had been transferred to clean recovery tanks for 7 days, the indexes were recovered to the level corresponding to the control group, except MDA content in the clams exposed to 10 ng·L -1 was sequentially induced. These results in the present work showed that oxidative stress in Meretrix meretrix would be produced by exposure to TBT at realistic levels in the environment. The antioxidant defense systems could act as biomarkers to monitor marine contamination of organotins.

Abstract:The effect of TBT exposure on hard clam Meretrix meretrix,which is a commercially important(fisheries and aquaculture) mollusc species along the coast in China was investigate.Meretrix meretrix were exposed to tributyltin(TBT) at environmental levels(0.1,1,10 ng/L as Sn) for 2,8,20 days respectively and recovered for 7,20 days,at experimental condition.The effects on catalase(CAT),superoxide dismutase(SOD) activities and content of malondialdehyde(MDA) in the digestive gland were evaluated.The results showed that the activity of CAT was inhibited in the early period and then unaffected by TBT exposure.The SOD activity and MDA content were induced after exposure,SOD were significantly induced after exposure to 10 ng/L TBT for 2,8 and 20 d.The MDA contents were significantly increased after exposure to the three concentrations of TBT for 8,20 d compared with that of the control group.The SOD activities and MDA contents in all exposure groups,transferred to clean recovery tanks for 20 days,were recovered to the level corresponding to the control group.These results in the present work show that oxidative stress in Meretrix meretrix would be produced by exposure to TBT at environmentally realistic levels.These results suggested that the SOD activity and MDA content could be regarded as potential biomarkers to monitor marine contamination of organtins.

Abstract:Organotin compounds, such as tributyltin (TBT) used as an antifouling biocide, can induce masculinization in female mollusks. However, few studies addressing the effect of TBT in fish have been reported. This study was conducted to investigate effects of TBT at environmental levels (1, 10, 100ng/L) on the development of ovary in female cuvier. TBT exposure elevated testosterone levels, increased the ratio of testosterone to 17β-estradiol and decreased 17β-estradiol levels in ovaries after 50 days compared to the control. Three stages of follicles (primary growth stage, yolk vesicle stage, vitellogenic stage) were observed in the ovaries of cuvier at the control and 1ng/L TBT group. The ovaries at the 10ng/L TBT group were characterized by the lack of vitellogenic stage follicles and instead had higher proportions of primary growth stage follicles. 100ng/L TBT resulted in follicles that were entirely at the earliest (primary growth stage) stages of development. There was a significant increase in apoptotic ovarian follicular cells judged by TUNEL-positive cell at the 10ng/L TBT group. The TUNEL-positive follicles were observed at the 100ng/L TBT group. The result in the present study showed that TBT at environmentally realistic concentrations can inhibit the ovarian development in fish. Besides the changes of sex hormone induced by TBT, apoptosis appears to be one mechanism affecting ovarian development.

Abstract:The effects on ethoxyresorufin O-deethylase (EROD), NADPH-cytochrome c reductase and NADH cytochrome b5 reductase activities of digestive gland in Meretrix meretrix exposed to tributyltin (TBT) at environmental levels (0.1, 1.0, 10.0 ng/dm(3) as stannum concentration), in experimental condition, were evaluated. The EROD, NADH cytochrome b5 reductase activities were significantly inhibited after exposure to 10.0 ng/dm(3) TBT for 8 and 20 d, the NADPH cytochrome c reductase activities were significantly inhibited after exposure to 0.1, 1.0 and 10.0 ng/dm(3) TBT for 8 d and to 1.0 and 10.0 ng/dm(3) for 20 d, as compared with the matched control, while NADH cytochrome b5 reductases and NADPH cytochrome c reductase activities were induced after exposure to 10. 0 ng/dm(3) TBT for 2 d. The EROD activity in the 10 ng/dm(3) group, and the NADH cytochrome b5 reductases activities in 1.0 and 10.0 ng/dm(3) groups, were significantly induced when transfer-red to clean recovery tanks for 7 d. The three enzymic activities in the clams exposed to TBT were recovered to the level corresponding to that of the control group after transfer to clean recovery tanks for 20 d. NADPH cytochrome c reductase activity in Meretrix meretrix seems to be more sensitive to exposure of TBT than that of the EROD and NADH cytochrome b5 reductases. The results suggest that induction and inhibition by TBT to the monooxygenase system enzymic activity in Meretrix meretrix would simultaneously exist. The enzymic activities were inhibited by exposure for a long time. The results suggest that inhibition of the monooxygenase system should be an indication of the exposure to environmentally relevant concentrations of TBT for a long time.

Abstract:The effects on alkaline phosphatase, aaciphosphatase and adenosine triphosphatase activities in mantle of Meretrix meretrix exposed to tributyltin (TBT) at environmental levels (0. 1,1, 10ng/dm3 as Sn) for 2, 8, 20 days and recovered for 7 and 20 days in experimental condition were evaluated. The results showed that the activity of alkalinephosphatase, aaciphosphatase and Na+ , K+ -ATPase were not significantly differences at earlier period (2days) exposure to tributyltin com-paired to control,and the activity of alkalinephosphatase in 1,10 ng/dm3 groups was induced with prolonged exposure. The activity of aaciphosphatase, Na + , K+-ATP ase were inhibited after exposure to TBT. The results suggested that Na+, K+ -ATPase activity would be a potential biomarker to monitor TBT pollution.
Effects of Tributyltin and Benzo[a]pyrene on the Immune-Associated Activities of Hemocytes and Recovery Responses in the Gastropod Abalone, Haliotis Diversicolor.

Abstract:Our previous study reports that short-term exposure to sublethal concentrations of benzo[a]pyrene (BaP) induces immunomodulation in the gastropod abalone, Haliotis diversicolor. In the present study, it was further observed that long-term chronic exposure to sublethal concentrations of BaP modulated the immunocompetence of abalones in terms of the change in activity of the antioxidant and immune associated parameters tested. In addition, the effect of tributyltin (TBT), another important genotoxicant in the aquatic environment, was investigated. Exposure of abalones to sublethal concentrations of TBT and BaP for 21 days resulted in significant decrease of total hemocyte count, phagocytosis, membrane stability and lysozyme activity. Conversely induction of extra and intra cellular superoxide generation, nitric oxide, nitric oxide synthase and myeloperoxidase activity was present when the abalones were exposed to TBT and BaP. Most of the immune associated parameters tested showed clear time dependent response to both toxicants. Within 14 days after the 21 day exposure to BaP, recovery was observed as evidenced by most of the parameters returning to their normal level. However, no recovery was observed within 14 days after the 21 day exposure to TBT as evidenced by continued elevation of intra cellular superoxide and nitrite production and decrease in THC, membrane stability and lysozyme activity. This suggested a prolonged TBT-induced impact on the immune reaction and possibly more damage than that caused by BaP. Overall the results suggest that chronic exposure to sublethal concentrations of TBT or BaP causes modulations in the immunocompetence of abalones with most of the immune associated parameters tested being stimulated, and this might be harmful to the host.
Transcriptomic analysis reveals the endocrine toxicity of tributyltin and triphenyltin on the whelk Reishia clavigera and mechanisms of imposex formation

Abstract:Organotin compounds (OTs) are endocrine disruptors that induce imposex in hundreds of gastropods, but little is known about their underlying molecular mechanisms. This study aimed to investigate the endocrine toxicity and molecular responses to tributyltin (TBT) and triphenyltin (TPT) exposure in the whelk Reishia clavigera, which often serves as a biomonitor for OT contamination. Over a 120-day exposure to environmentally relevant concentrations of TBT (1000 ng L-1) and TPT (500 ng L-1), we observed a significant increase in penis length in both male and female whelks. Notably, TPT exhibited a stronger potency in inducing pseudo-penis development and female sterility, even at a half dose of TBT. Bioaccumulation analysis also revealed higher persistence and accumulation of TPT in whelk tissues compared to TBT. Differential expression analysis identified a substantial number of differentially expressed genes (DEGs), with TPT exposure eliciting more DEGs than TBT. Our results demonstrated that OTs induced xenobiotic metabolism and metabolic dysregulation in the digestive gland, impaired multiple cellular functions and triggered neurotoxicity in the nervous system, and disrupted lipid homeostasis and oxidative stress in the gonads. Furthermore, imposex was possibly associated with disturbances in retinoic acid metabolism, nuclear receptor signaling, and neuropeptide activity. When compared to TBT, TPT exhibited a more pronounced endocrine-disrupting effect, attributable to its higher bioaccumulation and substantial interruption of transcriptional regulation, OT detoxification, and biosynthesis of retinoic acids in R. clavigera. Our results, therefore, highlight the importance of considering the differences in bioaccumulation and molecular toxicity between TBT and TPT in future risk assessments of these contaminants. Overall, our study provided molecular insights into the toxicity and transcriptome profiles in R. clavigera exposed to TBT and TPT, shedding light on the endocrine-disrupting effects and reproductive impairment in female gastropods.

Abstract:Organotin compounds, such as tributyltin (TBT), that have been used as antifouling biocides can induce masculinization in female mollusks. However, few studies addressing the effects of TBT on fishes have been reported. The present study was conducted to investigate the effects of TBT at environmentally relevant concentrations (1, 10, and 100 ng/L) on testicular development in Sebastiscus marmoratus and to gain insight into its mechanism of action. After exposure for 48 d, the gonadosomatic index had decreased in a dose-dependent manner. Although the testosterone levels in the testes were elevated and the 17 beta-estradiol levels were decreased, spermatogenesis was suppressed. Moreover, gamma-glutamyl transpeptidase activity (which is used as a Sertoli cell marker) was decreased in a dose-dependent manner after TBT exposure, and serious interstitial fibrosis was observed in the interlobular septa of the testes in the 100 ng/L TBT test group. Increases in the retinoid X receptors and peroxisome proliferator activated receptor gamma expression and the progressive enlargement of lipid droplets in the testes were observed after TBT exposure. Estrogen receptor alpha levels in the testes of the fish exposed to TBT decreased in a dose-dependent manner. The reduction of estrogen receptor alpha mRNA resulted from the decrease of 17 beta-estradiol levels, and the progressive enlargement of lipid droplets may have contributed to the dysfunction of the Sertoli cells, which then disrupted spermatogenesis.

Na~+,K~+-ATPase could be a potential biomarkers to monitor the organotins pollution.
Developmental Exposure to the Organophosphorus Flame Retardant Tris(1,3-Dichloro-2-propyl) Phosphate: Estrogenic Activity, Endocrine Disruption and Reproductive Effects on Zebrafish

Abstract:Tris(1,3-dichloro-2-propyl) phosphate (TDCPP) is an organophosphate flame retardant that is detectable in the environment and biota, prompting concern over its risk to wildlife and human health. Our objective was to investigate whether long-term exposure to low concentrations of TDCPP can affect fish reproduction. Zebrafish embryos were exposed to low concentrations (0, 4, 20 and 100 mu g/L) of TDCPP from 2 h post-fertilization until sexual maturation. Exposure to TDCPP significantly increased plasma estradiol and testosterone levels in females, but had no effect in males. TDCPP exposure also caused a significant reduction in fecundity as indicated by decreased egg production. Real-time PCR was performed to examine selected genes in the hypothalamic-pituitary-gonadal (HPG) axis and liver. Principle component analysis (PCA) showed that sex hormone levels and fecundity were related to the mRNA level of several genes in the HPG axis. Furthermore, hepatic vitellogenin (vtg1 and vtg3) expression was upregulated in both females and males, suggesting TDCPP has estrogenic activity. Histological examination revealed promotion of oocyte maturation in the females, but retardation of spermiation in males. Reduced egg quality (e.g., egg diameter) and increased malformation rates were observed in the F1 generation. Chemical analysis showed significant levels of TDCPP and its metabolite bis(1,3-dichloro-2-propyl) phosphate in the gonads of males and females. Abstract:Introduction: Butachlor (BTL) and triadimefon (TDF), the widely used herbicide and fungicide, are unavoidable enter into the aquatic environment. However, there were limited study regarding to the joint toxicity of these two pesticides on fish at present.Aim: To evaluate the potential thyroid-disrupting toxicity and exposed to different concentrations of BTL mixed with TDF.Materials and methods: Zebrafish embryo (n=3) were exposed to 0.01 and 0.05 fold of LC50 from the acute joint toxicity test, of which 0.32 mg/L (BTL) and 9.41 mg/L (TDF) for single or mixture agents (BTL: 0.0064 mg/L, 0.032 mg/L; TDF: 0.1882 mg/L, 0.9410 mg/L; co-exposure: 0.0032 mg/L BTL + 0.0941 mg/L TDF, 0.016 mg/l BTL + 0.4705 mg/L TDF) after 10-day post-fertilization. Hatching, malformation, survival rates and thyroid hormones (THs), genes expression involved in HPT-axis of embryos were measured and detected in control and separately/co-exposure treatments. THs contents were evaluated by ELISA kit and the expression levels of genes were determined by RT-PCR.Results: Hatching, malformation and survival rates of embryos exposed to single BTL exhibited no statistically significant difference from the control besides decreased of high concentration in survival rates. Exposure to TDF reduced hatching, survival rate and increased malformation. The combined exposure to BTL and TDF resulted in greater adverse effects on embryonic development. BTL exposure significantly increased free T-3 and T-4 contents. Elevated free T-3 content was also observed in the larvae exposed with single BTL. Co-exposure of the two pesticides caused greater enhanced of T-3 and T-4 levels. Furthermore, gene data showed BTL up-regulated the mRNA expression of tpo, tsh beta, tg, ttr, dio2, TDF up regulated the mRNA expression of tpo, tr alpha, ttr, dio2 and down-regulated tr beta gene. The mixture of the two pesticides caused up-regulation mRNA expression of tra, tr beta, tg, ttr, dio2.Conclusion: BTL and TDF resulted in adverse effects on zebrafish embryonic development and caused thyroid endocrine disruption, BTL and TDF have a synergistic effect on development and thyroid endocrine by enhanced level of thyroid hormone. Gender Differences in TBT Accumulation and Transformation in Thais Clavigera after Aqueous and Dietary Exposure.

Abstract:In this study, female and male Thais clavigera whelks were exposed to aqueous and dietary (using oysters as the prey) tributyltin (TBT) for up to 45 days, followed by a 30-day depuration, in order to examine the gender differences in TBT accumulation and transformation. The metabolites of TBT [dibutyltin (DBT) and monobutyltin (MBT)] were also measured in different tissues of the whelks (digestive, reproductive and remaining organs) during the exposure and depuration periods. By the end of the exposure period, all of the female whelks developed imposex after TBT exposure, and both the relative penis size index and the vas deferens sequence index were positively correlated with the tissue burden of TBT. However, biomagnification of TBT did not occur in the whelks. TBT was rapidly accumulated in their digestive and reproductive organs from both routes of exposure, and both elimination and biotransformation of TBT were also rapid. The redistribution of TBT among tissues was obvious during the exposure period but negligible during depuration. MBT was generally the major metabolite in each tissue, indicating a significant metabolism of TBT by the whelks. Accumulation, transformation, as well as elimination, were more significant following dietary exposure than following aqueous exposure. In particular, we observed gender-related differences in the biokinetics of TBT. Rapid biotransformation and elimination of TBT were detected in the male whelks, while the female whelks had higher bioaccumulation but lower elimination of TBT in their reproductive organs. Internal remobilization of TBT from digestive to reproductive organs was also more obvious in the females, indicating that the reproductive organs of females were the main targets of TBT accumulation.
Comparative Toxicogenomics of Benzotriazole Ultraviolet Stabilizers at Environmental Concentrations in Asian Clam (corbicula Fluminea): Insight into Molecular Networks and Behavior

Abstract:Benzotriazole ultraviolet stabilizers (BUVSs) are widespread emerging pollutants, which can pose exposure risks to benthic organisms. However, the toxicity and mechanisms of BUVSs congeners in benthic clams are far from elucidated. In this study, Asian clams (Corbicula fluminea) were exposed to one of UV-234, UV-326, UV-329, or UV-P at environmentally relevant levels (0.1, 1, and 10 μg/L) for 21 days. Filtration rate (FR) was increased in clams exposed to all BUVSs and there were notable histopathologic changes, including irregular digestive lumen, lipid droplet vacuolation, and degraded epithelial cells. To determine the molecular underpinnings following BUVSs exposure, the transcriptome responses in digestive glands were compared. Differentially expressed genes shared among BUVSs treatments were associated with focal adhesion, TNF-α/NF-κB proinflammatory pathways, and apoptosis. Following this, biochemical analysis of biomarkers related to apoptosis were conducted to further validate response. Exposure to BUVSs inhibited anti-oxidant enzyme activity and induced oxidative stress. Heat shock proteins were also triggered with exposure, and there was an induction of caspase-3 and caspase-9 activity. Molecular responses were not identical in the digestive gland of C. fluminea when comparing responses to BUVSs; nevertheless conserved mechanism (impairment of the oxidative defense system, immune system disruption, and induction of apoptosis) among BUVSs congeners was noted. This study provides novel insight into the toxicity and hazards of BUVSs in benthic organisms.

Abstract:Tributyltin, an organotin, is ubiquitous in estuaries and freshwater systems. Previous reports suggest that tributyltin is an endocrine disruptor in many wildlife species and it inhibits aromatase in mammalian placental and granulosa-like tumor cell lines. However, no evidence showing the effects of tributyltin on oocytes or preimplantation embryonic developmental competence exists. Therefore, we investigated the role of tributyltin chloride (TBTCl) in the development of female oocytes and preimplantation embryos. Briefly, female ICR mice were gavaged with 0 (vehicle), 4, and 8 mg/kg of TBTCl each day for 18 days. The fluorescence intensity analysis showed that the 5-methylcytosine level decreased after TBTCl treatment, indicating that the general DNA methylation level decreased in the treated oocytes. Our results demonstrate that TBTCl treatment results in decreased mRNA levels of imprinted genes H19, Igf2r, and Peg3 during oocyte growth. The TBTCl-treated oocytes showed a significant increase in reactive oxygen species levels in germinal vesicle oocytes. In TBTCl-treated oocytes, there was no difference in GPx and Sod1 expression, but a decreased mRNA level of Cat occurred when compared with control. Moreover, the blastocysts with TBTCl exposure displayed higher apoptotic signals. These results suggest that TBTCl induces developmental defects in oocytes and preimplantation embryos.

Inhibition of Thyroidal Status Related to Depression of Testicular Development in Sebastiscus Marmoratus Exposed to Tributyltin

Abstract:Organotin compounds, such as tributyltin (TBT), which have been used as antifouling biocides, are still of concern with regard to their effects on marine organisms, and especially their reproductive systems. We investigated the toxicity of TBT at environmentally relevant concentrations (1, 10, and 100 ng/L) on testicular development in a marine fish, Sebastiscus marmoratus. After exposure for 50 days, the gonadosomatic index had decreased in a dose-dependent manner, and there was a reduced number of mature sperm and an abundance of the late stages of spermatocysts in the testes of S. marmoratus. Exposure has also caused serious histological damage to the testes including interstitial fibrosis and pyknotic nuclei. Analysis of the thyroid status revealed severe damage to the thyroid gland, decreased triiodothyronine and thyroxine in the serum and low expression of thyroid hormone receptor alpha in the testes at this time. Pearson correlation analysis indicated that the levels of thyroxine in the serum were significantly correlated with the gonadosomatic index. These results suggest that inhibition of thyroidal status induced by TBT might be one of the mechanisms affecting testicular development.

Abstract:The effects on reduced glutathione (GSH) levels, glutathione-S-transferases (GST) and glutathione peroxidase (GPx) activities of digestive gland in Meretrix meretrix exposed to tributyltin (TBT) at environmental levels (0.1, 1, 10ng/L as Sn), in experimental condition, were evaluated. The GST activities in 0.1ng/L groups were significantly elevated after exposure for 2 days, and were significantly inhibited after exposure to 10ng/L TBT for 8 and 20 days. The GPx activities were mainly induced by TBT exposure, except the GPx activities in 10ng/L groups were significantly inhibited after exposure for 2 days. The GSH content was significantly decreased with prolonged exposure. The GSH content, GST and GPx activities in all exposure groups, transferred to clean recovery tanks for 20 days, were recovered to the level corresponding to that of the control group. Taken together, our present studies indicate that exposure to TBT may induce strong production of reactive oxygen species in the clams.

Abstract:The reported adverse effects of Polybrominated diphenyl ether (PBDE) congeners on gonadogenesis in fish may also occur in marine bivalves especially the burrowing species such as manila clam Ruditapes philippinarum. In this study, clams were exposed to BDE-47 for 25days at 0, 0.1 and 1μg/L. By using the water temperature control method, gonadal maturation from resting to ripe stage were observed successively in both the control and the treatment groups during 25days. The results showed that exposure to BDE-47 at concentration below 1μg/L did not delay the gonadogenesis process of the clam R. philippinarum, and no evidence of adverse effects of BDE-47 on clam gonadal histology was observed. However, exposure to 1μg/L BDE-47 caused significant decreases of haemolymph testosterone levels in both female and male clams at day 5 and day 15. The mRNA expression of 3β-HSD in females exposed to BDE-47 was significantly decreased at day 5, while mRNA expression of 17β-HSD and CYP17 was not significantly changed in either sex. Exposure to BDE-47 also resulted in up-regulation of the mRNA expression of vitellogenin (Vtg) in both sexes and spermatogenesis associated protein 4-homolog (SAP4) in males. These results suggest a potential contribution of BDE-47 to reproductive disruption in the manila clams, especially in males. This study demonstrates the promising utility of water temperature control method in conjunction with histological endpoints and biomarkers such as mRNA levels of Vtg in determining the reproductive disturbances caused by EDCs on bivalves.
