Abstract:It has been reported that there is a metabolic interaction between tributyltin (TBT), an organometal used as an antifouling biocide, and benzo[a]pyrene (BaP), a widespread carcinogenic polycyclic aromatic hydrocarbon. This study was therefore designed to examine the potential in vivo influence of TBT, BaP, and their mixture on hepatic antioxidant defense systems of Sebastiscus marmoratus, which were given a single ip injection of TBT (0.5, 1, 5, and 10mg/kg), BaP (0.5, 1, 5, and 10mg/kg), or both in combination (0.5, 1, 5, and 10mg/kg); control fish received olive oil vehicle only. Samples were collected for biochemical analysis after injection for 7 days. Cotreatment with BaP caused a significant inhibition of TBT-mediated malondialdehyde contents elevation. Cotreatment with TBT decreased BaP-mediated glutathione peroxidase activity induction. Cotreatment with TBT and BaP did not significantly alter the reduced glutathione levels, which were significantly induced by TBT or BaP alone. TBT-induced suppression of BaP bioactivation or BaP-induced stimulation of the phase II metabolism of TBT and its biliary excretion, both of which have been reported previously, could explain the observed antagonism. The results suggest that combined exposure of TBT and BaP increases the vulnerability of the fish to oxidative stress. BaP cotreatment decreased the induction of glutathione S-transferase (GST) activity by the lower dose of TBT, while cotreatment with TBT and BaP at the highest dose (10mg/kg) resulted in inhibition of the GST activity by 4.8-fold. The results suggest that these biomarkers should be interpreted with caution in biomonitoring studies. Combined effects of TBT and BaP exposure at environmental levels on these biomarkers should be further researched.

Abstract:The aim of our experiment was to study the effect of phenoloxidase of kidney and spleen on the activities of in Sebastiscus marmoratus.The abdominal injections of benzo(a)pyrene,tributyltin and their mixture were weekly done.The results showed that the following dose gradients of 0.5×10-6 、1.5×10-6 and 10×10-6 of body weight, the activities of phenoloxidase were induced during prophasic exposure,while reduced as the exposure continues.What's more,benzo(a)pyrene and tributyltin might corporate each other in mixture,enhance their immune toxicity.

Abstract:The impacts of environmental pollutants on marine organisms can be determined by the routes of exposure. Various routes of exposure, including dietary exposure and waterborne exposure with or without feeding, were applied to study the cytogenetic responses in marine mussels Mytilus galloprovincials to typical pollutants, BaP (53.74 +/- 19.79 mu g/L) and Cu (47.38 +/- 3.10 mu g/L). The increased DNA strand breaks and micronucleus formation were found in haemocytes of mussels via the dietary exposure, indicating the vital role of trophic transfer in toxicity induction. The deeper exploration to relate BaP induced cytogenetic alterations with key antioxidant defense factors, SOD and GST, was performed under different exposure routes. The results revealed the significantly inhibited SOD activity via the trophic transfer, suggesting more direct or prompt role of SOD in antioxidant defense. On contrary, gene expressions of both sod and gst were up-regulated upon all routes of exposures, and showed negative correlation with enzyme activities. The results suggested the asynchronous regulation of studied antioxidant factors at transcriptional and enzyme functional level in mussels upon the change of exposure routes. The study brings out the first observation of trophic transfer influenced cytogenetic and antioxidant responses to pollutants and their alterative risk to marine organisms.

Abstract:This study was designed to examine in vivo influence of tributyltin(TBT)at environmentally relevant concentrations on hepatic biomarkers in Sebastiscus marmoratus,which were exposed through water column to environmental concentrations of TBT(1,10,100 ng/L as Sn).Elevation of malondialdehyde(MDA)contents after exposure for 50 days indicates that TBT exposure at environmental concentrations could result in oxidative stress on liver.Glutathione peroxidase(GPx),superoxide dismutase(SOD)and catalase(CAT)activities were significantly induced by TBT after 50 days exposure.Reduced glutathione(GSH)contents in Sebastiscus marmoratus exposed to TBT for 7 days were induced and then inhibited in a dose-dependent manner for 50 days exposure.Glutathione S-transferase(GST)activities in Sebastiscus marmoratus exposed to 10 and 100 ng/L of TBT for 7 days were induced and then inhibited by TBT exposure for 25 days.These indices in all exposure groups,transferred to clean recovery tanks for 7 and 20 days,recovered to the level corresponding to that of the vehicle group.The results in the present work suggest that TBT exposure at environmental concentrations could result in oxidative stress on the fish.These biomarkers would indicate marine pollution of TBT at environmental concentrations.
BaP-metals Co-Exposure Induced Tissue-Specific Antioxidant Defense in Marine Mussels Mytilus Coruscus.

Abstract:Both benzo(α)pyrene (BaP) and metals are frequently found in marine ecosystem and can cause detrimental effects in marine organism, especially the filter feeder-marine mussels. Although the biological responses in mussels have been well-studied upon the single metal or BaP exposure, the information about antioxidant defense, especially in different tissues of mussels, are still limited. Considering the variety of contaminants existing in the actual marine environment, single BaP (56 μg/L) and the co-exposure with Cu, Cd and Pb (50 μg/L, 50 μg/L and 3 mg/L respectively) were applied in a 6 days exposure followed by 6 days depuration experiment. The alterations of superoxide dismutase (SOD), catalase (CAT) activities and total antioxidant capacity (TAC) level were assessed in haemolymph, gills and digestive glands of marine mussels, Mytilus coruscus. An unparalleled change in antioxidant biomarkers was observed in all cells/tissues, with the SOD activity showing higher sensitivity to exposure. A tissue-specific response showing unique alteration in gill was investigated, indicating the different function of tissues during stress responses. Depressed antioxidant effects were induced by BaP-metals co-exposure, indicating the interaction may alter the intact properties of BaP. To our knowledge, this is the first research to explore the antioxidant defense induced by combined exposure of BaP-metals regarding to tissue-specific responses in marine mussels. The results and experimental model will provide valuable information and can be utilized in the investigation of stress response mechanisms, especially in relation to tissue functions in marine organism in the future.

Abstract:The effects of benzo(a)pyrene(BaP),tributyltin(TBT) and their mixture exposure on lysozyme activity of kidney and spleen in Sebastiscus marmoratus were studied with doses of 0.5,1,5,10 mg/kg of body weight for intraperitoneal injected in experimental condition.The results showed that the lysozyme activities were induced during single exposure,but were inhibited by BaP and TBT cotreatment after 8 weeks.It suggests that combined exposure of the two chemicals for long time will cause detrimental effects on the nonspecific activity in Sebastiscus marmoratus.

Abstract:Tributyltin(TBT), an organometal used as an antifouling biocide, has been reported to induce masculinization of fish. Benzo [a]pyrene (BaP), a widespread carcinogenic polycyclic aromatic hydrocarbon, has been reported that its microsomal metabolites can produce an estrogenic response when tested in vitro. This study was therefore designed to examine the potential in vivo influence of TBT, BaP and their mixture on sex hormone levels in serum of Sebastiscus marmoratus, which were given 2 separate intraperitoneally (ip) injections(a single injection every 7 d) of TBT(0.5, 1, 5 and 10 mg/kg), BaP(0.5, 1, 5 and 10 mg/kg), or both in combination (0.5, 1, 5 and 10 mg/kg); control fish received olive oil vehicle only. Six days after the 2nd injection, serum samples were collected and analyzed for sex hormone levels and alkali labile protein phosphorus (ALPP), which is related to the yolk precursor protein vitellogenin. The pollutants at all doses significantly reduced serum testosterone, estradiol and ALPP content after 2 injections compared with the corresponding controls. The reduction of the estradiol levels should be response for the decrease of the vitellogenin levels. The results in the present study suggested that aromatase seems not the major target acted by TBT and BaP in fish. This study demonstrated that TBT or BaP exposure both inhibit the reproductive potential in female Sebastiscus marmoratus. Combined effect of TBT and BaP on the serum testosterone, estradiol and ALPP was not antagonism from the anticipation.

Abstract:Hepatic DNA strand break in Sebastiscus marmoratus exposed to tributyltin(TBT)was measured by alkaline unwinding assay.The results showed that DNA damage of the liver increased as increasing TBT doses,after intraperitoneally injected with 0.5,1,5,10 mg/kg(Bm)TBT.There is a dose-dependent relation.In general,the hepatic DNA damage in fish exposed to waterborne TBT(1,10,100 ng/L as Sn)increased as extension of exposure time and increasing dose.The results suggest that hepatic DNA integrity would be a potential biomarker to monitor TBT pollution.

Abstract:Tributyltin (TBT) and triphenyltin (TPT) coexist in the aquatic environment. However, the effects of TBT, TPT and a mixture of the two on DNA damage in marine fish livers and the mechanism involved remain to be elucidated. In the present study, we assessed their ability to cause hepatic DNA damage in Sebastiscus marmoratus liver and we investigated the related mechanism. The results showed that TBT, TPT and the mixture significantly decreased liver DNA integrity in a dose-dependent manner. Using Pearson correlation coefficient analysis, we identified that the hepatic total tin concentration was significantly correlated with hepatic DNA integrity after exposure to TBT alone and combined exposure to TBT and TPT. In order to clarify the mechanism which influences DNA repair, the mRNA levels of nucleotide excision repair (NER) genes were determined using real-time PCR analysis. The results showed that NER gene expression levels were disturbed. The expression of XPB, ERCC1, and DNA Pol ɛ was significantly decreased after treatment with TBT, TPT and the mixture, while the expression of PCNA, HR23B, XPG, and DNA lig III was significantly increased in treated-groups compared to the control. Based on this, we proposed that TBT, TPT and a mixture of the two induced DNA damage in marine fish livers by altering the transcription levels of the NER genes.

Abstract:This study investigated the influence of titanium dioxide nanoparticles (n-TiO2) on the bioavailability, metabolism, and toxicity of pentachlorophenol (PCP) in fish. Zebrafish (Danio rerio) embryos or larvae (2-h post-fertilization) were exposed to PCP (0, 3, 10, and 30 μg/L) alone or in combination with n-TiO2 (0.1mg/L) until 6 days post-fertilization. Results showed that n-TiO2 treatment alone did not induce lipid peroxidation, DNA damage, as well as the generation of reactive oxygen species (ROS) in the larvae. As compared with PCP treatment, the co-exposure of PCP and n-TiO2 enhanced the induction of ROS generation, eventually leading to lipid peroxidation and DNA damage. The nuclear factor erythroid 2-related factor 2 gene transcriptions were significantly upregulated in both PCP treatment alone and in combination with n-TiO2. Chemical analysis and histological examination showed that n-TiO2 adsorb PCP, and n-TiO2 are taken up by developing zebrafish larvae; however, PCP content was not enhanced in the presence of n-TiO2, but the metabolism of PCP to tetrachlorohydroquinone was enhanced in larvae. The results indicate that n-TiO2 enhanced the metabolism of PCP and caused oxidative damage and developmental toxicity, suggesting that NPs can influence the fate and toxicity of associated organic pollutants in the aquatic environment.

Abstract:Our previous study reports that short-term exposure to sublethal concentrations of benzo[a]pyrene (BaP) induces immunomodulation in the gastropod abalone, Haliotis diversicolor. In the present study, it was further observed that long-term chronic exposure to sublethal concentrations of BaP modulated the immunocompetence of abalones in terms of the change in activity of the antioxidant and immune associated parameters tested. In addition, the effect of tributyltin (TBT), another important genotoxicant in the aquatic environment, was investigated. Exposure of abalones to sublethal concentrations of TBT and BaP for 21 days resulted in significant decrease of total hemocyte count, phagocytosis, membrane stability and lysozyme activity. Conversely induction of extra and intra cellular superoxide generation, nitric oxide, nitric oxide synthase and myeloperoxidase activity was present when the abalones were exposed to TBT and BaP. Most of the immune associated parameters tested showed clear time dependent response to both toxicants. Within 14 days after the 21 day exposure to BaP, recovery was observed as evidenced by most of the parameters returning to their normal level. However, no recovery was observed within 14 days after the 21 day exposure to TBT as evidenced by continued elevation of intra cellular superoxide and nitrite production and decrease in THC, membrane stability and lysozyme activity. This suggested a prolonged TBT-induced impact on the immune reaction and possibly more damage than that caused by BaP. Overall the results suggest that chronic exposure to sublethal concentrations of TBT or BaP causes modulations in the immunocompetence of abalones with most of the immune associated parameters tested being stimulated, and this might be harmful to the host.

Abstract:The effects of tributyltin (TBT) on the activities of alkaline phosphatase (AKP) of liver,spleen and kidney in Sebastiscus marmoratus were studied at injected intraperitoneally with single doses of 1.9,9.6,19.3,193 μg /kg.After the injection 2,4 and 7 days,six fishes per dose group were killed and the liver,spleen and kidney were frozen in liquid N_2 immediately after collection and stored at -80℃ until analyzed.Each liver was measured solely,while 2 spleens or kidneys were got into a group for measure of enzymes activity,AKP activity was assayed using p-nitrophenylphosphate (pNPP) as the substrate.The results showed that the AKP activities were primarily induced in the fish exposed to TBT at low concentrations.The dose-response curve was a bell shape.AKP is participated in phosphoric metabolism directly,and participated in the metabolism of DNA,RNA,protein,lipid and so on.The results suggested that metabolism in the fish would be interfered by TBT exposure.The spleen and kidney are the immune and hemopoietic organs in fish.Effect of TBT on the activity of AKP in the spleen and kidney would affect the immune function in the fish.
Effects of Tributyltin on Antioxidant Enzymes Activity and Lipid Peroxidation in the Gill of Hard Clam Meretrix Meretrix

Abstract:The effects on catalase (CAT), glutathione -S-transferase(GST) activities and content of malondialdehyde (MDA), reduced glutathione (GSH) in the gill of Meretrix meretrix were evaluated, after it had exposed to tributyltin (TBT) at environmental levels (0.1, 1, 10 ng·L -1 as Sn) for 2, 8, 20 days and recovered for 7, 20 days, in experimental condition. The results showed that the activities of GST and CAT and GSH content in Meretrix meretrix exposed to TBT were principally induced, except that were mainly inhibited in the clams exposed to 10 ng·L -1 for 2 days. MDA content were significantly induced after exposure for 8d. After the clams had been transferred to clean recovery tanks for 7 days, the indexes were recovered to the level corresponding to the control group, except MDA content in the clams exposed to 10 ng·L -1 was sequentially induced. These results in the present work showed that oxidative stress in Meretrix meretrix would be produced by exposure to TBT at realistic levels in the environment. The antioxidant defense systems could act as biomarkers to monitor marine contamination of organotins.

Abstract:Organotin compounds, such as tributyltin (TBT), that have been used as antifouling biocides can induce masculinization in female mollusks. However, few studies addressing the effects of TBT on fishes have been reported. The present study was conducted to investigate the effects of TBT at environmentally relevant concentrations (1, 10, and 100 ng/L) on testicular development in Sebastiscus marmoratus and to gain insight into its mechanism of action. After exposure for 48 d, the gonadosomatic index had decreased in a dose-dependent manner. Although the testosterone levels in the testes were elevated and the 17 beta-estradiol levels were decreased, spermatogenesis was suppressed. Moreover, gamma-glutamyl transpeptidase activity (which is used as a Sertoli cell marker) was decreased in a dose-dependent manner after TBT exposure, and serious interstitial fibrosis was observed in the interlobular septa of the testes in the 100 ng/L TBT test group. Increases in the retinoid X receptors and peroxisome proliferator activated receptor gamma expression and the progressive enlargement of lipid droplets in the testes were observed after TBT exposure. Estrogen receptor alpha levels in the testes of the fish exposed to TBT decreased in a dose-dependent manner. The reduction of estrogen receptor alpha mRNA resulted from the decrease of 17 beta-estradiol levels, and the progressive enlargement of lipid droplets may have contributed to the dysfunction of the Sertoli cells, which then disrupted spermatogenesis.

Abstract:The aim of this study was to investigate the harmful effects of TBT and TPT using sea urchinStrongylocentyotus intermedius. At 2, 4, 6, 8, 10 days after TBT and TPT exposure, the examinations were taken. The results revealed that administration of TBT and TPT reduced the activities of antioxidants CAT and GPx, while MDA content increased. It was suggested that TBT and TPT were toxin with a variety of deleterious effects on sea urchin, indicating that oxidative injury and antioxidant defense system damage were induced by TBT and TPT.

Abstract:The effects of Benzo(a)pyrene and benzo(k)flouranthene mixture on EROD, GST, antioxidant enzymes activities and LPO levels of digestive gland and gills of scallop Chlamys Ferrari were studied. The results indicated that there is no significant effect on EROD activity in digestive gland at low concentrations (0.5 μg·L~ -1 and 1.0 μg·L~ -1 ) of PAHs, whereas it has induced the GST activity, that while in high concentration (10.0 μg·L~ -1 and 50.0 μg·L~ -1 ) of PAHs, EROD activity increased significantly, and the GST activity showed peak changes. Three antioxidant enzymes (SOD、CAT and GPx) all showed great changes and were restrained in high PAHs concentrations. Furthermore the gill was more sensitive to BaP than digestive gland, and its detoxification function was less strong than digestive gland, which indicated that digestive gland was the main tissue to metabolize oxyradicals. The contents of MDA increased with the treating time, and a positive correlation (concentration-effect) was shown between the MDA contents and the concentrations of the PAHs exposure. According to the studies, the increase of EROD, the decrease of GST and the changes of antioxidant enzymes can reflect the toxicity of PAHs. Therefore the LPO levels showed as a direct biomarker reflecting the oxidant stress. Furthermore, an important relationship in detoxification of these biomarkers can be seen in the results.

Abstract:The effects on ethoxyresorufin O-deethylase (EROD), NADPH-cytochrome c reductase and NADH cytochrome b5 reductase activities of digestive gland in Meretrix meretrix exposed to tributyltin (TBT) at environmental levels (0.1, 1.0, 10.0 ng/dm(3) as stannum concentration), in experimental condition, were evaluated. The EROD, NADH cytochrome b5 reductase activities were significantly inhibited after exposure to 10.0 ng/dm(3) TBT for 8 and 20 d, the NADPH cytochrome c reductase activities were significantly inhibited after exposure to 0.1, 1.0 and 10.0 ng/dm(3) TBT for 8 d and to 1.0 and 10.0 ng/dm(3) for 20 d, as compared with the matched control, while NADH cytochrome b5 reductases and NADPH cytochrome c reductase activities were induced after exposure to 10. 0 ng/dm(3) TBT for 2 d. The EROD activity in the 10 ng/dm(3) group, and the NADH cytochrome b5 reductases activities in 1.0 and 10.0 ng/dm(3) groups, were significantly induced when transfer-red to clean recovery tanks for 7 d. The three enzymic activities in the clams exposed to TBT were recovered to the level corresponding to that of the control group after transfer to clean recovery tanks for 20 d. NADPH cytochrome c reductase activity in Meretrix meretrix seems to be more sensitive to exposure of TBT than that of the EROD and NADH cytochrome b5 reductases. The results suggest that induction and inhibition by TBT to the monooxygenase system enzymic activity in Meretrix meretrix would simultaneously exist. The enzymic activities were inhibited by exposure for a long time. The results suggest that inhibition of the monooxygenase system should be an indication of the exposure to environmentally relevant concentrations of TBT for a long time.

Abstract:There is a growing recognition that the toxic effects of chemical mixtures are been an important issue in toxicological sciences. Tributyltin (TBT) and benzo[a]pyrene (BaP) are widespread pollutants that occur simultaneously in the aquatic environments. This study was designed to examine comprehensively the combined effects of TBT and BaP on zebrafish (Danio rerio) embryos using toxicogenomic approach combined with biochemical detection and morphological analysis, and tried to gain insight into the mechanisms underlying the combined effects of TBT and BaP. The results of toxicogenomic data indicated that: (1) TBT cotreatment rescued the embryos from decreased hatching ratio caused by BaP alone, while the alteration of gene expression (in this article the phrase gene expression is used as a synonym to gene transcription, although in is acknowledged that gene expression can also be regulated by, e.g., translation and mRNA or protein stability) relative to zebrafish hatching in the BaP groups was resumed by the cotreatment with TBT; (2) BaP cotreatment decreased TBT-mediated dorsal curvature, and alleviated the perturbation of Notch pathway caused by TBT alone; (3) cotreatment with TBT decreased BaP-mediated bradycardia, which might be due to that TBT cotreatment alleviated the perturbation in expression of genes related to cardiac muscle cell development and calcium handling caused by BaP alone; 4) TBT cotreatment brought an antagonistic effect on the BaP-mediated oxidative stress and DNA damage. These results suggested that toxicogenomic approach was available for analyzing combined toxicity with high sensitivity and accuracy, which might improve our understanding and predictability for the combined effects of chemicals.
