Abstract:Allelopathic inhibition activities of four submerged macrophytes (Potamogeton maackianus, Vallisneria spiralis, Elodea nuttalli Ceratophyllum demersum) on Microcystis aeruginosa are investigated by a series of experiments in a short-term bioassay. Individual algal test reveals culture solution of P. maackianus and C. demersum have better effects on algal growth inhibition. Furthermore, continuous addition assays show strong growth inhibition by culture solution. Inhibitory activity is different in aquatic extracts from different section of P. maackianus. Removing the disturbance of microorganism by autoclaving influences algal assay. These results provide that unstable and excreting continuously allelopathic compounds by submerged macrophytes are the primary cause of algal growth inhibition. Interference with algal growth experiments was investigated in microorganism and nutrient elements of nitrogen and phosphorus.

Abstract:For revealing the mechanism of allelopathic influence on phytoplankton by aquatic macrophytes, the growth and photosynthetic activities of cyanobacteria Microcystis aeruginosa and the chlorophyte Selenastrum capricornutum were investigated when they coexisted with submerged macrophyte Myriophyllum spicatum and were exposed to allelopathic polyphenols: pyrogallic acid (PA), gallic acid (GA), ellagic acid (EA) and (+)-catechin (CA). According to the results of coexistence assays, the non-photochemical quenching (NPQ) and effective quantum efficiency (YII) of M. aeruginosa were affected earlier and more rapidly than the cell density. However, the influence of M. spicatum on S. capricornutum was not found. When the Toxicity Index (TI) was applied to evaluate the combined effects of binary and multiple mixtures of polyphenols, it was found that the four tested polyphenols with the proportion identified in the M. spicatum-cultured solution were observed to present synergistic effect (0.36-0.49) according to the cell density, NPQ and YII of M. aeruginosa. With the combined effects of polyphenols on S. capricornutum, only additive action (0.52-1.62) was found. On the other hand, PA (2.97mgL(-1)), GA (2.65mgL(-1)) caused significant reductions of photosystem II (PSII) and whole electron transport chain activities of M. aeruginosa by 71.43 and 18.37%, 70.95 and 40.77% (P<0.05), respectively, after 24-h exposure, but no inhibition effect was found in S. capricornutum. The dark respiration and photosystem I (PSI) activities of M. aeruginosa were significantly increased by exposure to PA and GA (P<0.05). Nevertheless, EA and CA had no influence on the electron transport activities of the tested organisms. These results indicate that the reduction in photosynthetic activity of M. aeruginosa and the synergistic effect of allelochemicals may be two important causes for the inhibition of undesired phytoplankton by submersed macrophytes in natural aquatic ecosystems, and PSII in cyanobacteria is considered to be one of the target sites attacked by allelopathic polyphenols.

Abstract:This work aims to identify adequate species, tissues, pretreatments and allelochemicals to inhibit algal growth. Physiological indexes were investigated to explore the mechanism underlying allelopathic effect. Results demonstrated that Microcystis aeruginosa growth could be suppressed by the four tested aquatic macrophytes, namely emergent Oenanthe javanica, Iris pseudacorus, Canna indica, and submerged Potamogeton crisp us. The leaves were responsible for releasing anti-algal inhibitors. Leaf extracts of I pseudacorus inhibited M aeruginosa by 76.60% after 11 d. Pretreatments influenced the release of inhibitors. The inhibition rate of I pseudacorus leaf extracts collected by water bath heating was 55.20% higher than that of fresh leaves. Furthermore, rare algal growth was observed after 5 d of exposure to I. pseudacorus and C. indica culture water. The allelochemicals in tissue extracts could inhibit the synthesis of chlorophyll a (Chl-a) and enhance the activity of superoxide dismutase (SOD), thus affecting algal growth. The highest removal rates of Chl-a was 78.83%. SOD activities were triggered 2 times by leaf extracts than the control. Besides, SOD activity may be impaired by the excessive algal damage. The results suggest that the major allelochemicals in I. pseudacorus culture water were identified to be aromatic compounds, including dimethyl phthalate, ethyl phthalate, phthalic acid, dibutyl phthalate, and 3-nitrophthalic acid.

Abstract:Microcystis aeruginosa and Microcystis wesenbergii are two cyanobacteria commonly found in eutrophic shallow lakes. Previous studies reported that microcystin-producing M. aeruginosa could have an increased competitive potential on other algae and aquatic plants, and microcystin-LR (MC-LR) was regarded as an allelochemical. Based on this hypothesis, the allelopathic interaction between these two cyanobacteria was studied for the first time under laboratory conditions, and potential allelochemicals were screened. Cyanobacteria biomass and microcystin-LR (MC-LR) concentration were monitored under different culture conditions. The potential allelochemicals from M. aeruginosa were investigated by extract fractionation and GC(LC)/MS analysis. The growth of M. wesenbergii was inhibited by the addition of cell-free filtrates of M. aeruginosa whereas M. aeruginosa was promoted by the addition of cell-free filtrates of M. wesenbergii . The higher polarity the extract of M. aeruginosa is, the stronger the inhibition effect of the extract on M. wesenbergii will be. According to our results, M. aeruginosa has a significant allelopathic inhibition effect on M. wesenbergii . Allelopathic compounds from M. aeruginosa have synergistic effects on inhibition of M. wesenbergii . Besides microcystin, there may be other allelopathic compounds in M. aeruginosa .

Abstract:Aqueous extracts of three submerged macrophytes (Ceratophyllum demersum, Vallisneria spiralis and Hydrilla verticillata) and their ethyl acetate fractions were bioassayed using Microcystis aeruginosa. C. demersum showed stronger growth inhibition of M. aeruginosa than others. Ethyl acetate fraction contained succinic acid, azelaic acid, cis-aconitic acid, ferulic acid, 4-hydroxybenzoic acid, lactic acid and citric aicd. The potential allelopathic activities of these compounds on algal growth were discussed. Phenolic acid allelochemicals contributed more to the allelopathic effects of submerged macrophytes.

Abstract:Study was made on the allelopathic effect on Microcystis aeruginosa caused by filtered culturing water and extracts from different parts of Eichhornia crassipes. The results showed that filtered culturing water and extracts from different parts of Eichhornia crassipes had inhibition to Microcystis aeruginosa and reduced algal yield in stabili-zation period. When the concerntration of filtered culturing water was 10% ,50% ,100% ,the inhibition rates was 81. 06% ,89. 23% and 89. 78% respectively. The methanol and acetone extract of different part of Eichhornia crassipes inhibited Microcystis aeruginosa to various extent,of which the methanol abstract of Eichhornia crassipes root had the greatest inhibition. It was indicated that Eichhornia crassipes is one potential algaecide in the future.

Abstract:In natural environment, submergsed macrophytes have allelopathic effects on algaes, while algaes also have antagonistic allelopathic effects on submersed macrophytes. With static experiments, the authors investigated the biomass change of Ceratophyllum demersum and Microcystis aeruginosa cultured in Hoagland’s solution with different concentration levels of Microcystis aeruginosa. The results showed that the culture solution with higher concentration of Microcysuis aeruginosa (6.26×106 cells/mL) distinctly inhibited the growth of Ceratophyllum demersum, while the culture solution with lower concentration of Microcystis aeruginosa (0.63×106-3.13×106 cells/mL) was a stress to Ceratophyllum demersum, which stimulated Ceratophyllum demersum to inhibit algal growth. Allelochemicals in the culture solution were determined with SPE-HPLC.In the coexistence experiment the presence of algae might preferably stimulate the release of some potential allelochemicals of less polarity from macrophytes to inhibit the algal growth. Fifteen compounds detected from Ceratophyllum demersum were detected and identified by GC/MS. They were different from those extracted from Microcystis aeruginosa, and includeing N-phenyl-2-naphthylamine, methyl-dehydroabietate and ethyl-dehydroabietate with which have shown allelopathic activities confirmed in previous researches.
[Growth of Microcystis Aeruginosa Affected by Allelochemicals of Arundo Donax Linn. Extracted with Different Solvents].

Abstract:To study the effects of allelochemicals of Arundo donax Linn. on the growth of Microcystis aeruginosa, the allelochemicals were extracted with three solvents (methanol, ethyl acetate and hexane), respectively. Based on the observation of algal morphology and the measurement of algal density and cell size, the results showed the allelochemicals extracted with all the three solvents had inhibition on M. aeruginosa. The appearance time of allelochemical inhibition increased as follows, allelochemicals extracted with methanol < ethyl acetate < hexane. As treatment time extended, M. aeruginosa regrew at low doses of allelochemicals extracted with methanol. Their inhibitory effects at high doses were lower than those extracted with ethyl acetate and hexane. After 2 d and 4 d of treatment, the inhibition ratios of allelochemicals extracted with ethyl acetate and hexane reached almost 100%, respectively. The 50% effective concentrations (EC50, 6 d) of allelochemicals extracted with methanol, ethyl acetate and hexane were 0.17 g x L(-1), 0.05 g x L(-1) and 0.08 g x L(-1), respectively. The allelochemicals extracted with methanol caused cavities in cells, those with ethyl acetate caused cells into cavities, pieces and conglomeration, and those with hexane caused goffers on cells initially with conglomeration later. The allelochemicals extracted with all the three solvents decreased the cell size of M. aeruginosa, among which those extracted with ethyl acetate had the strongest action.
Growth inhibition and oxidative damage of Microcystis aeruginosa induced by aqueous extract of different submerged macrophytes

Abstract:With the increasing eutrophication of the aquatic environments, cyanobacteria blooms caused certain damage to the animals and plants in the aquatic environments. In this experiment, two species were selected from six species of submerged macrophytes, the experimental conditions were changed to achieve the best inhibitory effect on Microcystis aeruginosa, and oxidative damage analysis was carried out. The experiment results demonstrated that the inhibition rate of Vallisneria natans and Ceratophyllum demersum was nearly 100% at the concentration of 3 g/L after 15 days of anaerobic soaking extract. In addition, the longer the soaking time of the two submerged macrophytes, the weaker the photosynthesis effect, and the lower the chlorophyll fluorescence parameters, the more obvious the inhibition effect on M. aeruginosa. Lipid peroxidation injury of M. aeruginosa could be reflected by malondialdehyde (MDA) concentration. The MDA concentration in the experimental group was significantly higher than the control group. Results showed that V. natans and C. demersum could induce oxidative damage in M. aeruginosa. It was also observed that the secondary metabolites produced by V. natans were mainly fatty acids (e.g., the oxidative acid was 6.92 w/%, and the successful acid was 9.85 w/%) which inhibited M. aeruginosa in gas chromatography-mass spectrometry (GC-MS). The main secondary metabolites in C. demersum were hydroxyl acids (e.g., the 4-hydroxy-3-methoxyphenylacetic acid was 24.33 w/%), which could inhibit the algae through allelopathy. This study provided reference for submerged macrophytes to inhibit M. aeruginosa under different conditions.

Phenolic Compounds Exuded from Two Submerged Freshwater Macrophytes and Their Allelopathic Effects on Microcystis aeruginosa

Abstract:Culture solutions of the submerged freshwater macrophytes Hydrilla verticillata and Vallisneria spiralis, separately incubated at ID g of fresh weight (H) per liter for three days, were extracted by solid phase extraction (SPE) followed by liquid liquid extraction (LLE), and the acquired ethyl acetate fractions were analyzed by gas chromatograph-mass spectrometry (GC-MS) to identify potential allelochemicals exuded from these plant species. Freeze-dried plant tissues were separately methanol-extracted with a similar LLE-GC-MS procedure to compare allelochemical production and exudation. Four phenolic compounds were identified for both species: vanillic acid (VA), protocatechuic acid (PA), ferulic acid (FA), and caffeic acid (CA). H.verticillata produced 179 times the amount of phenolic compounds of V. spiralis. The phenolic contents in the culture solution were lower than 10% of those in the plant tissues for both submerged species in six of the eight cases. When evaluating the joint effects of the compounds using the Toxicity Index (TI) model, it was observed that the four phenolic compounds exerted additive and synergistic inhibition effects on the growth of Microcystis aeruginosa depending on the mixing ratios. These results indicate that H. verticillata and I! spiralis could release some phenolic allelochemicals to inhibit the growth of M. aeruginosa, and the joint action of multiple allelochemicals may be an important allelopathic pattern of submerged macrophytes to inhibit the growth of noxious cyanobacteria in natural aquatic ecosystems.

Growth Inhibition of Blue-green Algae by Allelopathic Effects of Macrophytes

Abstract:Inhibitory effects of macrophytes on the growth of blue-green algae (Microcystis aeruginosa, Anabaena flos-aquae, Phormidium tenue) were assayed in a mixed culture system with various concentrations of macrophytes, (i.e., coexistence assay) . Egeria densa, Cabomba caroliniana, Myriophyllum spicatum, Ceratophyllum demersum, Eleocharis acicularis, Potamogeton oxyphyllus, Potamogeton crispus, Limnophila sessiliflora and Vallisneria denseserrulata were used as macrophytes. The coexistence assay showed the inhibitory effects of the macrophytes on algal growth and that, among the 9 macrophytes, C. caroliniana and M. spicatum were the only species to inhibit growth of all the species of blue green algae. The inhibitory effects of M. spicatum were stronger than those of C. caroliniana.Due to its strong inhibition, we used the cultured solution of M. spicatum for an algal assay to confirm that the inhibitory effect of M. spicatum is caused by allelopathic compounds secreted by M. spicatum. The growth of M. aeruginosa was significantly inhibited by the quasi-continuous addition of the cultured solution of M. spicatum, whereas the initial addition did not inhibit their growth. Such results provide the evidence of the allelopathic compounds, which must be continuously secreted by M. spicatum.

Abstract:Eutrophication with a large number of Microcystis aeruginosa commonly occurs worldwide, thereby threatening the aquatic ecosystem and human health. In this study, four kinds of algicides were tested to explore their influence on cell density and chlorophyll-a of M. aeruginosa. Results showed that aluminum silicate agent, which inhibited more than 90% cell growth compared with the control group, demonstrated the strongest inhibition effect immediately on M. aeruginosa growth. Furthermore, the production and release of microcystin (MC)-LR were investigated. Aluminum silicate, CuSO4, and Emma-11 were more effective than pyrogallic acid in disrupting the cells of M. aeruginosa, thereby increasing the extracellular MC-LR concentration. Aluminum silicate caused the highest extracellular MC-LR concentration of more than 45 mg·L–1. Biotoxicity was also detected to evaluate the environmental risks of MC-LR release, which were related to the usages of different algicides. Extracellular MC-LR concentration mostly increased when the biotoxicity of algae solution increased. The experiments were also designed to reveal the effects of physical conditions in riverways, such as natural sunlight, aeration and benthal sludge, on MC-LR degradation. These findings indicated that UV rays in sunlight, which can achieve a MC-LR removal efficiency of more than 15%, played an important role in MC-LR degradation. Among all the physical pathways of MC-LR removal, benthal sludge adsorption presented the optimal efficiency at 20%.

Abstract:This study investigated the allelopathic effects of Microcystis aeruginosa (M. aeruginosa) extracts and exudates on the physiological responses, photosynthetic activity, and microbial structure of biofilms on leaves of Vallisneria natans (V. natans). By measuring physiological and photosynthetic indices, the results showed that M. aeruginosa allelochemicals inhibited photosynthesis, oxidative stress and antioxidant system stress response in the biofilms of V. natans leaves. Multifractal analysis found that the surface topography of V. natans leaves was altered due to the allelochemicals found in M. aeruginosa. Microbial diversity on the leaves was analyzed using high-throughput sequencing, and the results showed that M. aeruginosa exudates had a stronger effects on the microbial community structure of biofilms compared to extracts. These findings highlight how cyanobacterial allelochemicals induce negative effects on submerged macrophytes.
Nutrient conditions influence allelopathic capabilities of Ludwigia adscendens and other tropical macrophytes against Microcystis aeruginosa

Abstract:Macrophytes may release allelochemicals that suppress competing phytoplankton, although it is unclear how their effectiveness could be influenced by environmental nutrient conditions. To approach this, we conducted a series of bioassays against the bloom‐forming cyanobacterium Microcystis aeruginosa using extracts and exudates from potentially allelopathic macrophytes under varying nutrient conditions. We screened six different tropical macrophytes and identified antialgal effects in both extracts and exudates of Ludwigia adscendens, Persicaria barbata, Pontederia crassipes and Vallisneria spiralis. Growth assays indicated that extract inhibitory effectiveness decreased as nutrient concentration increased from 5% to 50% strength. Focusing on the potent antialgal extract of L. adscendens, we determined that its inhibitory effects were enhanced by specifically reducing the availability of either micronutrient or bicarbonate availability, indicating a role of these nutrients in protecting algal cells from allelochemicals. Bioactivity‐guided fractionation and ultra‐performance liquid chromatography – mass spectrometry analyses of L. adscendens extract indicated that ellagitannins were the major constituent of bioactive extract fractions, suggesting an antialgal mechanism common to other tannic compounds. Novel allelopathic alkaloids and terpenoid compounds also were described, although their mechanisms of action are unknown. Our findings provide evidence that the availability of carbon and micronutrients to cyanobacteria could influence the effects of macrophyte allelochemicals. This interaction between the nutrient environment and allelochemical sensitivity of cyanobacteria can be substantial, and studies or applications involving allelopathic interactions between macrophytes and phytoplankton should consider this.

Abstract:In the present study, a microcosm test was used to explore the effects of the zoobenthos on macrophyte allelopathy. Four representative zoobenthos showed low algal inhibition ranging within 0.05–0.16 in separate co-culture. When zoobenthos and tested microalgae were mixed completely, the inhibition rate was enhanced by 0.68. The inhibition potential followed the order: Corbicula fluminea > Palaemonetes sinensis > Chironomus plumosus > Limnodrilus hoffmeisteri. Furthermore, the benthic fauna significantly enhanced algal inhibition of the emergent plant Typha angustifolia and the submerged plant Potamogeton crispus by 0.43 and 0.32, respectively. Meanwhile, there was a significant difference in algal inhibition between five growth phases of macrophytes combined with zoobenthos community. In addition, a significant positive relationship occurred between the algal inhibition of allelopathic macrophytes and the mean individual biomass of the zoobenthos. Through the determination of physiological and biochemical traits of macrophytes, it was concluded that the zoobenthos would be a significant disturbance factor and induce strong stress-resistance response in macrophytes. Subsequently, the specific response will facilitate the algal inhibition of allelopathic macrophytes. Therefore, to keep a reasonable biodiversity will fully display strong function of the aquatic ecosystem and efficiently control harmful algal bloom.

Abstract:A comparative study was made on the two exotic submerged macrophyte species(Elodea nuttallii and Myriophyllum aquaticum) and their native congeners(Hydrilla verticillata and M.spicatum,with the differences in the community composition and structural characteristics of the epiphytic algae living on the two native species analyzed.Then,two concentrations(0.5 and 2 mg·mL-1) of the leaf extracts from the exotic and native submerged macrophytes were used to explore how the extracts affect the growth and community composition of epiphytic algae living on the native macrophytes.The results showed that the extracts from the two exotic submerged macrophytes showed stronger algal inhibiting effect than those from the native ones,being more significant in high concentrations of the extracts.The extracts from the four submerged macrophytes all reduced the total number of the epiphytic algae living on the native macrophytes,and showed the trend that low concentration extracts could promote the species richness and diversity of the epiphytic algae,while high concentration extracts showed inhibiting effect.The extracts from exotic species had more significant negative effects,and made the epiphytic algae community composition tend to be simple.The extracts from fresh plant materials showed stronger allelopathic algal inhibiting effect than those from dry ones.

Abstract:Many studies have attempted to find measures for control of cyanobacterial harmful algal blooms (cyanoHABs) caused by Microcystis, Anabaena, and other bloom-forming plankton species. We have investigated allelopathic inhibition of the submerged macrophyte Myriophyllum spicatum on four phytoplankton species of two taxonomic groups: Chlorophyta Selenastrum capricornutum, Scenedesmus obliquus, and cyanobacteria Microcystis aeruginosa (different strains for toxic, non-toxic, the North Han River originated (NHR) and colonies) and Anabaena circinalis. Inhibitions of unicellular cyanobacteria M. aeruginosa were over 50% for three consecutive days from the 3rd to the 5th day of the coexistence. M. spicatum even inhibited M. aeruginosa at a high initial concentration (1.1 mg L−1 Chl-a). Moreover, M. aeruginosa in a mixture of four phytoplankton species (S. capricornutum, S. obliquus, M. aeruginosa and A. circinalis) was selectively inhibited by M. spicatum. The inhibition of toxic, non-toxic, and NHR of Microcystis by M. spicatum were not significantly different. Colonial cyanobacteria strains were mostly not inhibited by M. spicatum.

Abstract:The present work describes the co-cultivation of Microcystis aeruginosa and Ceratophyllum demersum and the mechanism of their interactions. The alga density, chlorophyll-a content, phycobiliprotein content, SOD activity and MDA content were determined to study the inhibition of M. aeruginosa by C. demersum. Results showed that the growth of M. aeruginosa was substantially inhibited by C. demersum and the alga died out by 96 h. The inhibition of M. aeruginosa was due to allelopathy rather than competition between them for light and nutrients Chlorophyll-a and phycobiliprotein (including PC, APC and PE) were damaged during the co-cultivation, especially PC. SOD activity and MDA accumulation initially increased, then decreased. Although allelopathic inhibition of M. aeruginosa on C. demersum occurred in co-cultivation, M. aeruginosa also hindered the growth of C. demersum resulting in a decrease of the content of chlorophyll-a and carotenoids.
