Abstract:For revealing the mechanism of allelopathic influence on phytoplankton by aquatic macrophytes, the growth and photosynthetic activities of cyanobacteria Microcystis aeruginosa and the chlorophyte Selenastrum capricornutum were investigated when they coexisted with submerged macrophyte Myriophyllum spicatum and were exposed to allelopathic polyphenols: pyrogallic acid (PA), gallic acid (GA), ellagic acid (EA) and (+)-catechin (CA). According to the results of coexistence assays, the non-photochemical quenching (NPQ) and effective quantum efficiency (YII) of M. aeruginosa were affected earlier and more rapidly than the cell density. However, the influence of M. spicatum on S. capricornutum was not found. When the Toxicity Index (TI) was applied to evaluate the combined effects of binary and multiple mixtures of polyphenols, it was found that the four tested polyphenols with the proportion identified in the M. spicatum-cultured solution were observed to present synergistic effect (0.36-0.49) according to the cell density, NPQ and YII of M. aeruginosa. With the combined effects of polyphenols on S. capricornutum, only additive action (0.52-1.62) was found. On the other hand, PA (2.97mgL(-1)), GA (2.65mgL(-1)) caused significant reductions of photosystem II (PSII) and whole electron transport chain activities of M. aeruginosa by 71.43 and 18.37%, 70.95 and 40.77% (P<0.05), respectively, after 24-h exposure, but no inhibition effect was found in S. capricornutum. The dark respiration and photosystem I (PSI) activities of M. aeruginosa were significantly increased by exposure to PA and GA (P<0.05). Nevertheless, EA and CA had no influence on the electron transport activities of the tested organisms. These results indicate that the reduction in photosynthetic activity of M. aeruginosa and the synergistic effect of allelochemicals may be two important causes for the inhibition of undesired phytoplankton by submersed macrophytes in natural aquatic ecosystems, and PSII in cyanobacteria is considered to be one of the target sites attacked by allelopathic polyphenols.
Interspecific Competition Between Microcystis Aeruginosa and Pseudanadaena and Their Production of T&O Compounds.

Abstract:Microcystis aeruginosa and Pseudanabaena are two common cyanobacterial species/genus and they can occur coincidently in many eutrophic lakes globally. These two cyanobacteria could produce Taste & Odor (T&O) compounds, and their production of T&O compounds might be changed when they are present coincidently. The amounts of T&O compounds and their producers may influence the effectiveness of water treatment processes. Therefore, the mutual interactions between Microcystis aeruginosa (FACHB-905, M) and Pseudanabaena sp. (FACHB-1277, P) on T&O compounds in co-cultures were evaluated in this study. Different initial cell concentrations of M and P, with ratios of M:P = 1:1, M:P = 1:2 and M:P = 2:1 were applied in the co-cultures. The growth of M was enhanced under all of the cyanobacterial cell ratios. The growth of P was enhanced under the ratio of M:P = 1:1, while it was inhibited under the ratios of M:P = 1:2 and M: P = 2:1. In addition, the growth of the two cyanobacteria and their production of β-cyclocitral and 2-methylisoborneol (2-MIB) in the filtrate of P were higher than those in the filtrate of M, which may be attributed to their associated secondary metabolites. The cell integrity and photosynthetic capacity of the two studied cyanobacteria are greatly affected by exposure to β-cyclocitral and 2-MIB. The results showed that β-cyclocitral and 2-MIB had the allelopathic effects on the two cyanobacteria species which might influence the composition of co-existing cyanobacteria and their production of T&O compounds.

Abstract:Algae-induced odor problems in water have been repeatedly occurred concerns for drinking water quality. However, present researches mostly focus on the odor-producing pattern of algae in normal growth, and there is scarce discussion on those under stress. Microplastics (MPs) pollution have been global concern for their negative ecological impacts and frequently co-occurs with odor-producing algal bloom in freshwaters. Thus, this study aimed to elucidate the effects and mechanisms of MPs as an environmental stress on algal odorant production for good illustration of odor-producing response pattern under stress. Variation in MP size (polystyrene microspheres; 100 nm, 1000 nm and 10 μm) had significant effects on odorant formation (β-cycloidal, 2-methylisopropanol, 2,4-heptandienal and 2,4-decadienal) by four freshwater algae (Microcystis aeruginosa, Pseudanabaena sp., Cyclotella meneghiniana and Melosira varians). The size ratio of MPs over cells (SRMC) was proposed to categorize the size-ratio dependent effects on the algal odorant production. Interestingly, when SRMC was in the range of 0.1-1, there were always promoting effects; when SRMC < 0.1 or SRMC > 1, there exhibited inhibiting effects, and the inhibiting effects of SRMC < 0.1 were far more severe than those of SRMC > 1. The promotion on odorant production in the SRMC range of 0.1-1 was mainly attributed to the increase in cellular yield, which was related to the increased odorant precursors derived from the oxidation products of reactive oxygen species (ROS). Alternatively, the inhibition of odorant production caused by MPs with SRMC < 0.1 was the results of simultaneously inhibiting cellular density and cellular yield, which might be attributed to the cellular internalization of MPs, inducing the extensive toxic effects. This study illustrated the possibilities of MPs in impairing the esthetics of the source water and provided guidance for the future algal odor issues under stress.

Abstract:2-methylisoborneol (2-MIB), a secondary metabolite produced by cyanobacteria, often causes a musty odour in water, threatening the safety of drinking water supplies. This study investigated the effects of the growth phases on the production of 2-MIB by Pseudanabaena. The effects of cell characteristics on the production and release of 2-MIB were also explored. The total 2-MIB concentration increased during the exponential phase and decreased during the declining phase, which was consistent with the changes in cell density. However, the total 2-MIB yield (1.12-1.27 fg cell-1) of Pseudanabaena did not significantly differ throughout the growth cycle (p > 0.05). Meanwhile, the extracellular 2-MIB yield increased significantly from 0.31 fg cell-1 in the exponential phase to 0.76 fg cell-1 in the declining phase (p < 0.05), and the corresponding proportion of extracellular 2-MIB improved from 25.13% to 59.16% (p < 0.05). The surge in extracellular 2-MIB during the declining phase could be attributed to the breaking of the Pseudanabaena filament, as indicated by the decrease in Dmean during cell ageing. The findings of this study contribute to a more inclusive comprehension and management of musty odour issues resulting from cyanobacteria in the water supply.

Phenolic Compounds Exuded from Two Submerged Freshwater Macrophytes and Their Allelopathic Effects on Microcystis aeruginosa

Abstract:Culture solutions of the submerged freshwater macrophytes Hydrilla verticillata and Vallisneria spiralis, separately incubated at ID g of fresh weight (H) per liter for three days, were extracted by solid phase extraction (SPE) followed by liquid liquid extraction (LLE), and the acquired ethyl acetate fractions were analyzed by gas chromatograph-mass spectrometry (GC-MS) to identify potential allelochemicals exuded from these plant species. Freeze-dried plant tissues were separately methanol-extracted with a similar LLE-GC-MS procedure to compare allelochemical production and exudation. Four phenolic compounds were identified for both species: vanillic acid (VA), protocatechuic acid (PA), ferulic acid (FA), and caffeic acid (CA). H.verticillata produced 179 times the amount of phenolic compounds of V. spiralis. The phenolic contents in the culture solution were lower than 10% of those in the plant tissues for both submerged species in six of the eight cases. When evaluating the joint effects of the compounds using the Toxicity Index (TI) model, it was observed that the four phenolic compounds exerted additive and synergistic inhibition effects on the growth of Microcystis aeruginosa depending on the mixing ratios. These results indicate that H. verticillata and I! spiralis could release some phenolic allelochemicals to inhibit the growth of M. aeruginosa, and the joint action of multiple allelochemicals may be an important allelopathic pattern of submerged macrophytes to inhibit the growth of noxious cyanobacteria in natural aquatic ecosystems.

Abstract:Allelopathic inhibition activities of four submerged macrophytes (Potamogeton maackianus, Vallisneria spiralis, Elodea nuttalli Ceratophyllum demersum) on Microcystis aeruginosa are investigated by a series of experiments in a short-term bioassay. Individual algal test reveals culture solution of P. maackianus and C. demersum have better effects on algal growth inhibition. Furthermore, continuous addition assays show strong growth inhibition by culture solution. Inhibitory activity is different in aquatic extracts from different section of P. maackianus. Removing the disturbance of microorganism by autoclaving influences algal assay. These results provide that unstable and excreting continuously allelopathic compounds by submerged macrophytes are the primary cause of algal growth inhibition. Interference with algal growth experiments was investigated in microorganism and nutrient elements of nitrogen and phosphorus.

Abstract:This work aims to identify adequate species, tissues, pretreatments and allelochemicals to inhibit algal growth. Physiological indexes were investigated to explore the mechanism underlying allelopathic effect. Results demonstrated that Microcystis aeruginosa growth could be suppressed by the four tested aquatic macrophytes, namely emergent Oenanthe javanica, Iris pseudacorus, Canna indica, and submerged Potamogeton crisp us. The leaves were responsible for releasing anti-algal inhibitors. Leaf extracts of I pseudacorus inhibited M aeruginosa by 76.60% after 11 d. Pretreatments influenced the release of inhibitors. The inhibition rate of I pseudacorus leaf extracts collected by water bath heating was 55.20% higher than that of fresh leaves. Furthermore, rare algal growth was observed after 5 d of exposure to I. pseudacorus and C. indica culture water. The allelochemicals in tissue extracts could inhibit the synthesis of chlorophyll a (Chl-a) and enhance the activity of superoxide dismutase (SOD), thus affecting algal growth. The highest removal rates of Chl-a was 78.83%. SOD activities were triggered 2 times by leaf extracts than the control. Besides, SOD activity may be impaired by the excessive algal damage. The results suggest that the major allelochemicals in I. pseudacorus culture water were identified to be aromatic compounds, including dimethyl phthalate, ethyl phthalate, phthalic acid, dibutyl phthalate, and 3-nitrophthalic acid.
Growth inhibition and oxidative damage of Microcystis aeruginosa induced by aqueous extract of different submerged macrophytes

Abstract:With the increasing eutrophication of the aquatic environments, cyanobacteria blooms caused certain damage to the animals and plants in the aquatic environments. In this experiment, two species were selected from six species of submerged macrophytes, the experimental conditions were changed to achieve the best inhibitory effect on Microcystis aeruginosa, and oxidative damage analysis was carried out. The experiment results demonstrated that the inhibition rate of Vallisneria natans and Ceratophyllum demersum was nearly 100% at the concentration of 3 g/L after 15 days of anaerobic soaking extract. In addition, the longer the soaking time of the two submerged macrophytes, the weaker the photosynthesis effect, and the lower the chlorophyll fluorescence parameters, the more obvious the inhibition effect on M. aeruginosa. Lipid peroxidation injury of M. aeruginosa could be reflected by malondialdehyde (MDA) concentration. The MDA concentration in the experimental group was significantly higher than the control group. Results showed that V. natans and C. demersum could induce oxidative damage in M. aeruginosa. It was also observed that the secondary metabolites produced by V. natans were mainly fatty acids (e.g., the oxidative acid was 6.92 w/%, and the successful acid was 9.85 w/%) which inhibited M. aeruginosa in gas chromatography-mass spectrometry (GC-MS). The main secondary metabolites in C. demersum were hydroxyl acids (e.g., the 4-hydroxy-3-methoxyphenylacetic acid was 24.33 w/%), which could inhibit the algae through allelopathy. This study provided reference for submerged macrophytes to inhibit M. aeruginosa under different conditions.

Effects of Microcystin-Lr on the Tissue Growth and Physiological Responses of the Aquatic Plant Iris Pseudacorus L.

Abstract:The release of cyanobacterial toxins during algal bloom has adverse effects on aquatic plants and animals. This study aimed to understand the toxic effects and mechanism of microcystin-LR (MC-LR) on the seedling growth and physiological responses of Iris pseudacorus L. (calamus). After a one-month exposure experiment, the growth and development of the calamus leaves were significantly inhibited, and this inhibitory effect was verified to be concentration dependent. Furthermore, the cell membrane system was damaged, and the photosynthesis was also adversely affected by MC-LR. The relative conductivity of the leaves increased from 10.96% to 97.51%, and the total chlorophyll content decreased from 0.89 mg/g to 0.09 mg/g. Notably, the behavior of the roots in the presence of MC-LR was different from that of the leaves. The seedlings needed to absorb more nutrients to maintain the normal growth at low-toxin concentrations, but the high concentration of (over 250 mu g/L) MC-LR exceeded the tolerance of plants and inhibited the growth of roots. In addition, MC-LR led to an excessive accumulation of H2O2, and the seedlings enhanced the activities of catalase, peroxidase, and superoxide dismutase to resist oxidative stress. The presence of MC-LR also affected the capacity of the plants to absorb nitrogen and phosphorus. The removal efficiency of NO3--N, the main source of nitrogen, was 63.53% in the presence of 100 mu g/L MC-LR. As a result, the pH increased, and the growth of plants was indirectly inhibited. Therefore, the presence of MC-LR could affect the purification efficiency of calamus in eutrophic water. This study provides theoretical support for the selection of plants in the eutrophic water.
Growth Inhibition of Blue-green Algae by Allelopathic Effects of Macrophytes

Abstract:Inhibitory effects of macrophytes on the growth of blue-green algae (Microcystis aeruginosa, Anabaena flos-aquae, Phormidium tenue) were assayed in a mixed culture system with various concentrations of macrophytes, (i.e., coexistence assay) . Egeria densa, Cabomba caroliniana, Myriophyllum spicatum, Ceratophyllum demersum, Eleocharis acicularis, Potamogeton oxyphyllus, Potamogeton crispus, Limnophila sessiliflora and Vallisneria denseserrulata were used as macrophytes. The coexistence assay showed the inhibitory effects of the macrophytes on algal growth and that, among the 9 macrophytes, C. caroliniana and M. spicatum were the only species to inhibit growth of all the species of blue green algae. The inhibitory effects of M. spicatum were stronger than those of C. caroliniana.Due to its strong inhibition, we used the cultured solution of M. spicatum for an algal assay to confirm that the inhibitory effect of M. spicatum is caused by allelopathic compounds secreted by M. spicatum. The growth of M. aeruginosa was significantly inhibited by the quasi-continuous addition of the cultured solution of M. spicatum, whereas the initial addition did not inhibit their growth. Such results provide the evidence of the allelopathic compounds, which must be continuously secreted by M. spicatum.

Abstract:Many studies have attempted to find measures for control of cyanobacterial harmful algal blooms (cyanoHABs) caused by Microcystis, Anabaena, and other bloom-forming plankton species. We have investigated allelopathic inhibition of the submerged macrophyte Myriophyllum spicatum on four phytoplankton species of two taxonomic groups: Chlorophyta Selenastrum capricornutum, Scenedesmus obliquus, and cyanobacteria Microcystis aeruginosa (different strains for toxic, non-toxic, the North Han River originated (NHR) and colonies) and Anabaena circinalis. Inhibitions of unicellular cyanobacteria M. aeruginosa were over 50% for three consecutive days from the 3rd to the 5th day of the coexistence. M. spicatum even inhibited M. aeruginosa at a high initial concentration (1.1 mg L−1 Chl-a). Moreover, M. aeruginosa in a mixture of four phytoplankton species (S. capricornutum, S. obliquus, M. aeruginosa and A. circinalis) was selectively inhibited by M. spicatum. The inhibition of toxic, non-toxic, and NHR of Microcystis by M. spicatum were not significantly different. Colonial cyanobacteria strains were mostly not inhibited by M. spicatum.

Diversity and interaction of bacterial and microeukaryotic communities in sediments planted with different submerged macrophytes: Responses to decabromodiphenyl ether

Abstract:Although various persistent organic pollutants (POPs) can affect microbial communities and functions in aquatic ecosystems, little is known about how bacteria and microeukaryotes respond to the POPs in sediments planted with different submerged macrophytes. Here, a 60-day microcosm experiment was carried out to investigate the changes in the diversity and interaction of bacterial and microeukaryotic communities in sediments collected from Taihu lake, either with decabromodiphenyl ether (BDE-209) own or combined with two common submerged macrophyte species (Vallisneria natans and Hydrilla verticillate). The results showed that BDE-209 significantly decreased the bacterial α-diversity but increased the microeukaryotic one. In sediments planted with submerged macrophytes, the negative effect of BDE-209 on bacterial diversity was weakened, and its positive effect on microeukaryotic one was strengthened. Co-occurrence network analysis revealed that the negative relationship was dominant in bacterial and microeukaryotic communities, while the cooperative relationship between microbial species was increased in planted sediments. Among nine keystone species, one belonging to bacterial family Thermoanaerobaculaceae was enriched by BDE-209, and others were inhibited. Notably, such inhibition was weakened, and the stimulation was enhanced in planted sediments. Together, these observations indicate that the responses of bacteria and microeukaryotes to BDE-209 are different, and their communities under BDE-209 contamination are more stable in sediments planted with submerged macrophytes. Moreover, the effects of plant species on the microbial responses to BDE-209 need to be explored by more specific field studies in the future.

Abstract:Outbreaks of Microcystis blooms can affect growth of submerged plants, which in turn can inhibit cyanobacterial growth. Microcystin (MC)-producing and non-MC-producing Microcystis strains typically coexist in Microcystis -dominated blooms. However, the interaction between submerged plants and Microcystis at strain level is not clear. This study was aimed at assessing the effects of a submerged macrophyte Myriophyllum spicatum on one MC-producing versus one non-MC-producing strains of the cyanobacterium Microcystis using plant- Microcystis co-culture experiments. The impacts of Microcystis on M. spicatum were also examined. It showed that the MC-producing Microcystis strain had a higher resistance to negative impacts by the cocultured submerged plant M. spicatum than the non-MC-producing strain. By contrast, the plant M. spicatum was impacted more by the MC-producing Microcystis than the non-MC-producer. The associated bacterioplankton community was affected more by the MC-producing Microcystis than the cocultured M. spicatum . The MC cell quotas were significantly higher in the coculture treatment (the PM + treatment, p < 0.05), indicating that the production and release of MCs might be a key factor responsible for the reduced impact of M. spicatum . The higher concentrations of dissolved organic and reducing inorganic compounds might eventually exacerbate the recovering capacity of coexisting submerged plants. Overall, this study indicated that the capacity to produce MCs, as well as the density of Microcystis , should be taken into account when attempting to reestablish submerged vegetation to undertake remediation works.

Abstract:Aqueous extracts of three submerged macrophytes (Ceratophyllum demersum, Vallisneria spiralis and Hydrilla verticillata) and their ethyl acetate fractions were bioassayed using Microcystis aeruginosa. C. demersum showed stronger growth inhibition of M. aeruginosa than others. Ethyl acetate fraction contained succinic acid, azelaic acid, cis-aconitic acid, ferulic acid, 4-hydroxybenzoic acid, lactic acid and citric aicd. The potential allelopathic activities of these compounds on algal growth were discussed. Phenolic acid allelochemicals contributed more to the allelopathic effects of submerged macrophytes.

Abstract:Introduction: Cyanobacterial blooms in tropical water bodies are increasingly common, because of eutrophication and rising temperatures. Consequently, many freshwater systems are affected, by reducing water quality, biodiversity, and ecosystem services. With the increased frequency of harmful algal blooms, the development of biological tools to improve water quality is an urgent issue. Objective: To evaluate the effects of a submerged macrophyte and a cladoceran on the microcystin-producing cyanobacteria Microcystis aeruginosa (NPLJ-4) and the chlorophyte Raphidocelis subcapitata (BMIUFRPE-02) in mixed cultures. Methods: Two parallel experiments were carried out for ten days to evaluate the effects of the submerged macrophyte Ceratophyllum demersum and the cladoceran Moina micrura on microalgal interactions. Microalgal strains were cultivated in the ASM1 culture medium, under controlled laboratory conditions. The first experiment presented four treatments: M (C. demersum), Z (M. micrura), MZ (C. demersum and M. micrura), and C (control). Meanwhile, the second experiment consisted of five treatments, in which the microalgae were cultivated together at different Microcystis:Raphidocelis ratios: 1:0, 3:1, 1:1, 1:3, and 0:1. Biomass and growth rates of the strains were evaluated every two days, which were statistically treated with three-way or two-way repeated-measures ANOVA. Results: In the first experiment, M. aeruginosa was significantly inhibited in M and MZ treatments from the second day, and Z from the fourth, while R. subcapitata showed no reduction in its biomass in any treatment. On the other hand, R. subcapitata was stimulated from the eighth and tenth days in M treatment and only on the eighth day in Z treatment. In the second experiment, M. aeruginosa was significantly inhibited when cultivated with R. subcapitata in low ratios (Microcystis:Raphidocelis ratio of 1:3) throughout the experiment, while the chlorophyte was stimulated in that treatment. Conclusions: The coexistence of a cyanobacterium with a green alga did not alter the main negative response of M. aeruginosa to the submerged macrophyte and zooplankton but stimulated the green alga. Accordingly, the introduction of submerged macrophytes and cladocerans already adapted to eutrophic conditions, both isolated and combined, proved to be a good method to control cyanobacterial blooms without negatively affecting other coexisting phytoplankton species.
MIB-producing cyanobacteria (Planktothrix sp.) in a drinking water reservoir: distribution and odor producing potential

Abstract:The production of odorant 2-methylisoborneol (MIB) in water bodies by Planktothrix sp. have not been understood very well. Through a four-year investigation in Miyun Reservoir, a huge mesotrophic drinking water reservoir known to have the MIB episodes, we found that the Planktothrix sp. bloomed during September and October causing the high levels of MIB in the reservoir. The concentration of MIB and the biomass of MIB-producing cyanobacteria Planktothrix were measured (n = 887) at different sites and depths during different seasons. The results indicated that the shallow region of the reservoir is the major habitat for Planktothrix sp. due to that the light is able to penetrate down to the relatively high concentrations of nutrients close to the sediments. Quantile regression analysis between Planktothrix biomass and MIB concentration shows that the risk of MIB exceeding the odor threshold (15 ng L⁻¹) in water was as high as 90% when the Planktothrix density was more than 4.0 × 10⁵ cells L⁻¹, while the risk was reduced to 10% when the Planktothrix density remained below 1.6 × 10⁴ cells L⁻¹. This study will improve the understanding of the environmental behaviors of Planktothrix sp., and can provide useful information for better management of drinking water lakes/reservoirs experiencing the taste and odor (T&O) problems caused by deep living cyanobacterial species.
Maintenance of cell integrity during hydroxyl radical rapid inactivation of Pseudanabaena sp. and simultaneous mineralization of odor compound 2-methylisoborneol

Abstract:Pseudanabaena sp. and the odor compound it produces, 2-methylisoborneol (2-MIB), has been reportedly responsible for off-flavor pollution worldwide, leading to substandard drinking water sensory indicators and serious water supply crises. In this paper, the hydroxyl radical (•OH) produced by the synergistic effect of strong ionization discharge and hydrodynamic cavitation rapidly inactivated Pseudanabaena sp. and simultaneously mineralized 2-MIB to a concentration of 2.57 ng/L, which is below the odor threshold of 10 ng/L for a total reactive oxidants (TRO) concentration of 1.2 mg/L within 12 s. Crucially, the intracellular 2-MIB level was maintained in approximately 155.26- 162.29 ng/L range, indicating that 2-MIB was not released from the cells. Based on the scanning electron microscopy (SEM) results, the integrity of Pseudanabaena sp. cells was maintained with intact membranes and no intracellular organic matters (IOM) released during •OH inactivation. In contrast, ClO2 caused severe membrane rupture and massive IOM release. Based on the gas chromatograph/mass spectrometer (GC/MS) analyses and mass spectral database, the chromatogram fitted the baseline with a TRO concentration of 4 mg/L and no peaks corresponding to intermediates were detected. Moreover, •OH could mineralize 2-MIB by opening the ring structures of 1,2,3,3-tetramethyl-4-cyclopentenone, neomenthol, and 2-methylcyclohexene-1-aldehyde to produce small-molecule compounds, finally leading to CO2 and H2O formation via three reaction pathways. Therefore, the •OH not only maintained the cell integrity of Pseudanabaena sp. during inactivation but also mineralized 2-MIB simultaneously.

Abstract:The purpose of this study was to evaluate the permeability characteristics of a previously reported in vitro corneal model that utilizes SIRC rabbbit corneal cells and to investigate the permeability of three novel esters of phenylephrone chemical delivery systems (CDS) under different pH conditions using this in vitro model. The SIRC rabbit corneal cell line was grown on transwell polycarbonate membranes, and the barrier properties were assessed by measuring transepithelial electrical resistance (TEER) using a voltohmmeter. The permeabilities of esters of phenylephrone CDS across the SIRC cell layers were measured over a pH range 4.0-7. 4. The esters tested include phenylacetyl (1), isovaleryl (2), and pivalyl (3). The SIRC rabbit corneal cell line, when grown on permeable filters, formed tight monolayers of high electrical resistance with TEER values increasing from 71.6 +/- 20.8 Omega.cm2 at day 3 in culture to 2233.42 +/- 15.2 Omega.cm2 at day 8 in culture and remained constant through day 14 in culture. The transepithelial permeability coefficients (Papp) at pH 7.4 ranged from 0.58 x 10(-6) cm/s for the hydrophilic marker, mannitol, to 43. 5 x 10(-6) cm/s for the most lipophilic molecule, testosterone. The Papp at pH 7.4 for phenylephrine was 4.21 x 10(-6) cm/s. The Papp values and the lag times of the three esters of phenylephrone were pH dependent. The Papp for 1, 2, and 3 at pH 7.4 were 14.76 x 10(-6), 13.19 x 10(-6), and 12.86 x 10(-6) cm/s, respectively and the permeabilities decreased at conditions below pH 7.4. The lag times at pH 7.4 were 0.10, 0.17, and 0.12 h for 1, 2, and 3, respectively, and the values increased at lower pH conditions. The TEER values of SIRC cell line observed at day 8 to day 14 in the present investigation are similar to the resistance value reported for rabbit cornea (2 kOmega.cm2). All the esters showed significantly (p < 0.05) higher permeabilities than phenylephrine at pH 7.4. The rate and extent of transport of the drugs across the cell layers were influenced by the fraction of ionized and un-ionized species and the intrinsic partition coefficient of the drug. The results indicate that the permeability of ophthalmic drugs through ocular membranes may be predicted by measuring the permeability through the new in vitro cell culture model.
Algal control ability of allelopathically active submerged macrophytes: a review]

Abstract:The inhibitory effect of allelochemicals released by submerged macrophytes on phytoplankton is considered as one of the mechanisms that contribute to the stabilization of clear-water status in shallow lakes. This paper reviewed the research progress in the allelopathy of submerged macrophytes on algae from the aspects of the occurrence frequency and coverage of allelopathically active submerged macrophytes in lakes, and the kinds and allelopathical effects of the allelochemicals released from the macrophytes. The previous researches indicated that allelopathically active submerged macrophyte species such as Myriophyllum, Ceratophyllum, and Elodea were efficient to control phytoplankton, especially when their biomass was high enough, and the dominant algae were sensitive species. The allelochemicals such as hydroxybenzene released by the submerged macrophytes could inhibit the growth of algae. Different phytoplankton species exhibited different sensitivity against allelochemicals, e.g., cyanobacteria and diatom were more sensitive than green algae, while epiphytic species were less sensitive than phytoplankton. Environmental factors such as light, temperature, and nutrients could significantly affect the allelopathical effect of submerged macrophytes. The research of the allelopathy of submerged macrophytes is still at its beginning, and further researches are needed on the effects of environmental factors on the allelopathy, extraction and identification of allelochemicals, selective algal control mechanisms, and metabolism of the allelochmicals.
