Protective effect of glial cell line-derived neurotrophic factor on neural function in rats with cerebral hemorrhage

Abstract:Aim: To investigate the protective effect of glial cell line-derived neurotrophic factor (GDNF) on neural function in rats with cerebral hemorrhage and study the genesis mechanism. Methods: The total sequence of rat GDNF gene was cloned to construct pCDNA3-GDNF-GFP plasmid which was injected into the region of hemorrrhage in rates. Adult male SD rats were selected to establish the model of cerebral hemorrhage. All rates were divided into 9 groups and each group 20 rats. Praxiology, the levels of water, the changes of sodium and potassium in brain were observed in groups I of artificial operation, cerebral hemorrhage and intervention. The changes of blood brain barrier in groups II of artificial operation, cerebral hemorrhage and intervention were examined. Histological and the express of GDNF positive cells after hemorrhage during different times respectively were observed. Results: In the cerebral hemorrhage group and intervention group, the markers of neurologic defect 24 hours after hemorrhage were 8.53 ± 1.44 and 8.34 ± 1.28 respectively (P > 0. 05), and after 72 hours, 7 days, 14 days were 7.58 ± 1.08, 5.46 ± 0.74, 3.06 0.43, and 4.21 ± 0.76, 3.25 ± 0.52, 1.92 ± 0.26 (P < 0. 01). The contents of water in brain at 72 hours, 7 days were (84.26 ± 0.64)%, (80.38 ± 0.86)% and (78.26 ± 0.42)%, (77.51 ± 0.33)% (P < 0.01). There was significant difference between the two groups of sodium and potassium in brain at 72 hours, 7 days (P < 0.01). In intervention groups, necrotic volume of cerebral constitution decreased and the positive expression of GDNF increased significantly as compared with the cerebral hemorrhage group (P < 0.01). Conclusion: GDNF can ameliorate the praxiology, relieve the cerebral edema, benefit to the recovery of neural function and increase ability of antihurts in brain, and have therapeutic significance.
