Abstract:Polymerase chain reactions (PCR), thermally activated chemical reactions which are widely used for nucleic acid amplification, have recently received much attention in microelectromechanical systems and micro total analysis systems because a wide variety of DNA/RNA molecules can be enriched by PCR for further analyses. In the present work, a droplet-based micro oscillating-flow PCR chip was designed and fabricated by the silicon microfabrication technique. Three different temperature zones, which were stable at denaturation, extension and annealing temperatures and isolated from each other by a thin-wall linkage, were integrated with a single, simple and straight microchannel to form the chip's basic functional structure. The PCR mixture was injected into the chip as a single droplet and flowed through the three temperature zones in the main microchannel in an oscillating manner to achieve the temperature maintenance and transitions. The chip's thermal performance was theoretically analyzed and numerically simulated. The results indicated that the time needed for the temperature of the droplet to change to the target value is less than 1 s, and the root mean square error of temperature is less than 0.2 °C. A droplet of 1 µl PCR mixture with standard HPV (Human Papilloma Virus)-DNA sample inside was amplified by the present chip and the results were analyzed by slab gel electrophoresis with separation of DNA markers in parallel. The electrophoresis results demonstrated that the micro oscillating-flow PCR chip successfully amplified the HPV-DNA, with a processing time of about 15 min which is significantly reduced compared to that for the conventional PCR instrument.

Abstract:An innovative micro polymerase chain reaction (PCR) chip, micro oscillating flow-through PCR chip, was designed and fabricated by using silicon microfabrication technique with double-sided lithography. Three different temperature zones, which are stable at 93/spl deg/C, 72/spl deg/C and 50/spl deg/C and isolated from each other by deep groove, are integrated with a straight microchannel in the chip to form the basic functional structure. It is indicated numerically that the time needed for the temperature varying to the target value is less than 1 second, and the temperature difference in the reaction zone is less than 0.2/spl deg/C at the same time. A quasi multi-systems lumped heat capacity method has been advanced to analyze the chip's transient thermal performance, which was validated by numerical simulation. A microliter-sized droplet of reagent with HPV-DNA inside was injected into the chip and forced to flow through the three temperature zones in an oscillating manner to realize DNA amplification. The amplification results of the chip are analyzed by slab gel electrophoresis with separation of DNA markers in parallel.
Oscillatory-Flow PCR Microfluidic Chip Driven by Low Speed Biaxial Centrifugation.

Abstract:PCR is indispensable in basic science and biotechnology for in-orbit life science research. However, manpower and resources are limited in space. To address the constraints of in-orbit PCR, we proposed an oscillatory-flow PCR technique based on biaxial centrifugation. Oscillatory-flow PCR remarkably reduces the power requirements of the PCR process and has a relatively high ramp rate. A microfluidic chip that could perform dispensing, volume correction, and oscillatory-flow PCR of four samples simultaneously using biaxial centrifugation was designed. An automatic biaxial centrifugation device was designed and assembled to validate the biaxial centrifugation oscillatory-flow PCR. Simulation analysis and experimental tests indicated that the device could perform fully automated PCR amplification of four samples in one hour, with a ramp rate of 4.4 ∘C/s and average power consumption of less than 30 W. The PCR results were consistent with those obtained using conventional PCR equipment. Air bubbles generated during amplification were removed by oscillation. The chip and device realized a low-power, miniaturized, and fast PCR method under microgravity conditions, indicating good space application prospects and potential for higher throughput and extension to qPCR.

Abstract:The microfluidic polymerase chain reaction (PCR) chips have undergone extensive development and nowadays have become an important domain of miniaturization technology application. Here, we review the advances of microfluidic PCR chips over the past years, from the first single chamber stationary PCR chip to the new SlipChip PCR. First, the three distinct types of microfluidic PCR chips are discussed, including chamber stationary PCR chips, flow-through PCR chips and convection PCR chips. Then we focus on droplet PCR chips and SlipChip PCR. Although they are at an early stage, they show the great potential for high-throughput PCR and robust chip. Finally, general discussions on integrated chips are given. The low cost, portable, high-throughout integrated PCR chips will certainly be further developed in spite of many challenges.

Abstract:The combination of multiplex polymerase chain reaction (mPCR) and microfluidic technologies demonstrates great significance in biomedical applications. However, current microfluidics-based molecular diagnostics face challenges in multi-target detection due to their limited fluorescence channels, complicated fabrication process, and high cost. In this research, we proposed a cost-effective sandblasting method for manufacturing silicon microchips and a chip-based microdevice for field mPCR detection. The atomic force microscopy (AFM) images showed a rough surface of the sandblasted microchips, leading to poor biocompatibility. To relieve the inhibitory effect, we dip-coated a layer of bovine serum albumin (BSA) on the irregular substrate. The optimized coating condition was determined by scanning electron microscope (SEM) and energy-dispersive X-ray spectroscopy (EDS) (65 °C for 60 min). After sufficient coating, we performed on-chip PCR tests with 500 copies/mL Coronavirus Disease 2019 (COVID-19) standard sample within 20 min, and the sandblasted microchip displayed a higher amplification rate compared to dry etching chips. Finally, we achieved a 50 min mPCR for screening five resistance genes of the endophthalmitis pathogens on our microdevices, with strong specificity and reliability. Thus, this sandblasted microchip-based platform not only provides a rapid, accessible, and effective solution for multiplex molecular detection but also enables large-scale microfabrication in a low-cost and convenient way.

Abstract:The chip-based digital polymerase chain reaction (PCR) is an indispensable technique for amplifying and quantifying nucleic acids, which has been widely employed in molecular diagnostics at both fundamental and clinical levels. However, the previous designs have yet to achieve widespread application due to limitations in complex chip fabrication, pretreatment procedures, special surface properties, and low throughput. This study presents a facile digital microfluidic chip driven by centrifugal force for digital PCR analysis. Interestingly, regardless of the hydrophilicity or hydrophobicity of the inner chip surface, an efficient digitization process can be achieved. PCR reagents introduced into the inlet can be allocated to 9600 microchambers and subsequently isolated by the immiscible phase (silicone oil). The centrifugal priming approach offers a facile means to achieve high-throughput analysis. The design was further employed for the quantification of nucleic acids using digital PCR. The calculated result exhibited a strong correlation with the measured value at the concentrations from 1 copy/mu L to 1000 copies/mu L (R-2 = 0.99). Additionally, the chip also allowed digital multiplexed analysis, thereby indicating its potential for multi-target detection applications.

Abstract:Due to the difficulty in sample introduction, the presence of air bubbles and the complicated fabrication with high cost, there are no commercial products of microfluidic polymerase chain reaction (microPCR) on the market yet. In this study, an air bubble resistant and disposable microPCR chip was developed. A simple fabrication method using Norland Optical Adhesive 81 (NOA81) as the substrates was employed to prepare the microPCR chip. The temperature of the microPCR chip was controlled using a self-developed portable and programmable PCR apparatus. The simulation results indicate that the developed microPCR chip has good performance in temperature stability even in the presence of air bubbles. The DNA was successfully amplified without adding bovine serum albumin, implicating that the developed microPCR chip has low adsorption of DNA polymerase. The good performance of microPCR chip attributed to the property of N0A81, the fabrication method and the irreversible sealing. The portable and programmable PCR device combined with the disposable microPCR chip is suitable for the amplification of both short and long DNA fragments and Short Tandem Repeats (STRs). It offers a practical approach for portable forensic test, combining the cost benefits of small volume reagent with the disposable polymer microPCR chip. (C) 2015 Elsevier B.V. All rights reserved.

Abstract:Polymerase chain reaction (PCR) has been considered as the gold standard for detecting nucleic acids. The simple PCR system is of great significance for medical applications in remote areas, especially for the developing countries. Herein, we proposed a low-cost self-assembled platform for microchamber PCR. The working principle is rotating the chamber PCR microfluidic chip between two heaters with fixed temperature to solve the problem of low temperature variation rate. The system consists of two temperature controllers, a screw slide rail, a chamber array microfluidic chip and a self-built software. Such a system can be constructed at a cost of about US$60. The micro chamber PCR can be finished by rotating the microfluidic chip between two heaters with fixed temperature. Results demonstrated that the sensitivity of the temperature controller is 0.1°C. The relative error of the duration for the microfluidic chip was 0.02 s. Finally, we successfully finished amplification of the target gene of Porphyromonas gingivalis in the chamber PCR microfluidic chip within 35 min and on-site detection of its PCR products by fluorescence. The chip consisted of 3200 cylindrical chambers. The volume of reagent in each volume is as low as 0.628 nL. This work provides an effective method to reduce the amplification time required for micro chamber PCR.

Abstract:Rapid heat transfer is crucial for an efficient polymerase chain reaction (PCR), and this makes temperature control one of the most essential features in a micro-PCR system, which always includes a heater and a sensor composing a closed-loop. Yet, the fabrication of the heater and the sensor often prevented micro-PCR systems from achieving both cost-effectiveness and fabrication-easiness. For most of the early researches micromachining techniques were used to allow sensors and heaters be integrated on a silicon or glass chip. However, the cost prevented them from wide applications. The work described in this paper is part of our effort to solve the cost/fabrication dilemma. An innovative digital temperature control system was developed by introducing a heater/sensor switching procedure. Only one temperature controlling element fabricated by flexible printed circuit technology was utilized in the constructed PCR device with minimum fabrication steps. The glass chip-based device was made from low cost materials and assembled with adhesive bonding. Through seemingly simple steps, we obtained both disposability and portability at the same time. Temperature stability within ±0.3 °C and a transitional rate of 8 °C/s during heating/cooling was achieved. A 244 bp DNA fragment of hepatitis C virus was successfully amplified in our device by a three-stage thermal cycling process. Further improvement was assisted by finite element analysis, and demonstrated by experiment.

Abstract:This paper reports on a simple and practicable biochip for polymerase chain reaction (PCR). The micro biochip (12mm×12mm) is a hybrid type which is composed of a polydimethylsiloxane (PDMS) cover with a hexagonal chamber and a glass substrate integrated with platinum (Pt) microheater and microsensor. Bubble formation has been reported as a big problem with PCR chips. In this paper we present a simple and practicable operation schedule to get a bubble free chip, in which a successful PCR process was guaranteed.

Abstract:This study proposes a rapid and inexpensive thermocycler that enables rapid heating of samples using a thin glass chip and a cheap chip resistor to overcome the on-site diagnostic limitations of polymerase chain reaction (PCR). Microchip PCR devices have emerged to miniaturize conventional PCR systems and reduce operation time and cost. In general, PCR microchips require a thin-film heater fabricated through a semiconductor process, which is a complicated process, resulting in high costs. Therefore, this investigation substituted a general chip resistor for a thin-film heater. The proposed thermocycler consists of a compact glass microchip of 12.5 mm × 12.5 mm × 2 mm that could hold a 2 μL PCR sample and a surface-mounted chip resistor of 6432 size (6.4 mm × 3.2 mm). Improving heat transfer from the chip resistor heater to the PCR reaction chamber in the microchip was accomplished via the design and fabrication of a three-dimensional chip structure using selective laser-induced etching, a rapid prototyping technique that allowed to be embedded. The fabricated PCR microchip was combined with a thermistor temperature sensor, a blower fan, and a microcontroller. The assembled thermocycler could heat the sample at a maximum rate of 28.8 °C/s per second. When compared with a commercially available PCR apparatus running the same PCR protocol, the total PCR operating time with a DNA sample was reduced by about 20%.

Abstract:This study presents a novel microfabricated polymerase chain reaction (PCR) chip based on silicon. A microheater utilizing doped semiconductors as heating resistors and a temperature sensor made of Pt are integrated on the chip to make up a thermal module. The micro-stationary PCR chip is fabricated on a silicon wafer using photolithography, wet etching and ion implantation technology. The package is created without complex processes. Three types of configurations for the microheater are designed and simulated to analyze the temperature distribution by the finite element analysis so as to enhance the temperature uniformity in the reaction chamber. With this approach, the microheater is optimized. Finally, the simulation results are validated by infrared images from experiments.

Abstract:Real-time polymerase chain reaction (real-time PCR) tests were successfully conducted in an aluminum-based microfluidic chip developed in this work. The reaction chamber was coated with silicone-modified epoxy resin to isolate the reaction system from metal surfaces, preventing the metal ions from interfering with the reaction process. The patterned aluminum substrate was bonded with a hydroxylated glass mask using silicone sealant at room temperature. The effect of thermal expansion was counteracted by the elasticity of cured silicone. With the heating process closely monitored, real-time PCR testing in reaction chambers proceeded smoothly, and the results show similar quantification cycle values to those of traditional test sets. Scanning electron microscope (SEM) and atomic force microscopy (AFM) images showed that the surface of the reaction chamber was smoothly coated, illustrating the promising coating and isolating properties. Energy-dispersive X-ray spectroscopy (EDS), X-ray photoelectron spectroscopy (XPS), and inductively coupled plasma-optical emission spectrometer (ICP-OES) showed that no metal ions escaped from the metal to the chip surface. Fourier-transform infrared spectroscopy (FTIR) was used to check the surface chemical state before and after tests, and the unchanged infrared absorption peaks indicated the unreacted, antifouling surface. The limit of detection (LOD) of at least two copies can be obtained in this chip.

Abstract:A real-time polymerase chain reaction ( PCR ) micro total analysis system (μ-TAS ) integrated nucleic acid extraction, PCR amplification and real-time-fluorescent PCR detection on a same microfluidic chip was prepared for the fully automated and on-chip analysis of nucleic acid. The proposed method had the advantage such as low sample consumption, fast analysis and simple operation and so on. Micromachining technology was used to fabricate the anodic molds of integrated nucleic acid extraction microfluidic chip. A polydimethylsiloxane (PDMS) substrate with 3D channels was manufactured by a combination of molds and an injection molding method. The glass substrate and the chip were bonded together using a plasma treatment. The μ-TAS included a microfluidic control device whose micro fluidic velocity ( 0-10 mL/min ) could be adjusted, a TEC platform which the precision of temperature control was 0. 1℃ and a CCD detection module. The DNA of human blood was extracted by using a silica gel membrane method on the microfluidic chip. The processes of DNA extraction and detection were preset in the μ-TAS. Human blood lysate ( 20 μL ) were driven to the extraction chamber and was then washed. The fluidic drive speed was 2 mL/min. DNA and PCR reagents were mixed and then were driven into the PCR chamber. The fluidic drive speed was 1 mL/min. The GAPDH gene in extracted genome DNA was amplified by PCR and detected. The amplified product was verified by melting analysis. The results of nucleic acid extraction method on the chip were compared to those obtained using a standard manual centrifuge extraction method. The amplification curves were obvious. Ct values of the chip method were 25 . 3 and 26 . 9 . The denaturation temperature of all the melting was 89 . 9 ℃. The results validated that the chip-based method and device realized the extraction of nucleic acid, amplification and detection automatically.

Abstract:Microfluidic chips (μFC) are emerging as powerful tools in chemistry, biochemistry, nanotechnology and biotechnology. The microscale size, possibility of integration and high-throughput present huge technical potential to facilitate the research of cell behavior by creating in vivo-like microenvironments. Here, we have developed a new method for rapid fabrication of μFC with Norland Optical Adhesive 81 (NOA81) for multiple cell culture with high efficiency. The proposed method is more suitable for the early structure exploration stage of μFC than existing procedures since no templates are needed and fast fabrication methods are presented. Simple PDMS-NOA81-linked microvalves were embedded in the μFC to control or block the fluid flow effectively, which significantly broadened the applications of μFC. Various types of cells were integrated into the chip and normal viabilities were maintained up to 1 week. Besides, concentration gradient was generated to investigate the cells in the μFC responded to drug stimulation. The cells appeared different in terms of shape and proliferation that strongly demonstrated the potential application of our μFC in online drug delivery. The high biocompatibility of NOA81 and its facile fabrication (μFC) promise its use in various cell analyses, such as cell-cell interactions or tissue engineering.

Abstract:PCR technology has been widely applied to biomedicine and other related fields. PCR biochip shortens reaction time and reduces expensive reagents quantities. If the PCR biochip can be integrated with capillary electrophoresis chip, then the microminiaturization of biochemical analysis system will be possible. A micro PCR biochip based on silicon is designed and fabricated by MEMS technology in this paper. In order to reduce the processing complicacy, the temperature sensor and the micro heater were fabricated with chromium metal film. Since the characteristics of metal membrane is closely related to the performance of the material itself and the fabrication technology, the impact of fabrication technology on the membrane performance is studied in this paper. The results indicate the rational domain design, suitable processing condition and appropriate controlling system are preferable to the requirements of PCR reaction. (c) 2004 Elsevier B.V. Abstract:Point-of-care detection for pathogen is of critical need for wide epidemic warning and medical diagnosis. In this work, we have designed and developed a fully portable and integrated microchip based real-time polymerase chain reaction machine for rapid pathogen detection. The instrument consists of three functional components including heating, optical, and electrical modules, which are integrated into a portable compact box. The microchip is consumable material replaceable to meet various detection needs. Consequently, we demonstrated the outstanding performance of this portable machine for rapid detection of Salmonella and Escherichia coli O157:H7 with the advantage of time-saving (∼25 min), less samples consumption, portability, and user-friendly operation.

Abstract:Gene is the genetic code of human beings, and only 0.1‰ difference among individuals but creates a wealthy diversity. Thus it is significant in practicality to explore the difference. Polymerase chain reaction (PCR) is one of the technologies mostly used in genetic research. But traditional thermal cyclers are not only time- and energy-consuming but also difficult to be integrated and portable. The combination of microfluidics and PCR can lessen the reaction volume significantly and increase the reaction efficiency with easy integration and miniaturization. In this review, we briefly introduced microchamber PCR chip and continuous-flow PCR chip according to their structures, as well the detection methods including capillary electrophoresis, fluorescence, electrochemistry and DNA hybridization array. Finally, we summarized the recent advance and pointed out the future development of microfluidic PCR chip.
