Abstract:Eleven new triterpenes, cavalerols A - K (1-11, resp), ten of which were derivatives of hopanc and one was derivative of dammarane, were isolated from the twigs of Eurycorymbus cavaleriei Their structures were elucidated by spectroscopic methods including 2D-NMR analysis. Cavalerol D (4) and cavalerol F (6). cavalerol B (2), and cavalerol G (7) were two pairs of isomers, and silver ion was introduced for their differentiation by multi-stage tandem mass spectrometry And nine compounds, except Sand 10, were tested for Timone reductase (OR) induction activities in vitro, and results showed that compounds 2, 4, and 7 exhibited moderate induction activities with CD values of 862, 9 13, and 256 mu g/ml. respectively, and compounds 6 and 8 showed cytotoxicity against hepa 1c1c7 cell line with IC50 values of no more than 1 mu g/ml

Abstract:Four compounds, including one new lignan cavaol H (1), one new coumarinolignoid 5′-hydroxycleomiscosin B (4) and two known lignans (2–3) were isolated from the 95% ethanol extract of the twigs of Eurycorymbus cavaleriei. Their structures were established on the basis of various spectroscopic analyses including 1D-(1H, 13C, and DEPT) and 2D-NMR (COSY, HMQC, and HMBC). The absolute stereochemistry of the two known lignans was firstly reported in this article by 1H NMR studies on Mosher's ester derivatives. In the present study, quinone reductase induction activities of compounds 1–4 were assayed, compound 4 showed moderate quinone reductase induction with concentration to double the enzyme activity (CD) of 10.5 ± 0.8 μg/mL. Then we established LC-MS-MS analysis of glutathione (GSH) incubation with compound 4 to explain whether compound 4 induced quinone reductase through alkylating of the sulfhydryl groups of Keap1. Reconstructed selected ion chromatogram (SIC) of m/z 706 after compound 4 incubation with GSH was different from that with Tris–HCl buffer solution, which meant the quinone reductase induction activity of compound 4 attributed to alkylating the sulfhydryl groups of Keap1.

Abstract:From the fruits of Celastrus orbiculatus Thunb. a new beta-dihydroagarofuran sesquiterpene ester named 1beta,2beta,13-triacetoxy-9alpha-cinnamoyloxy-beta-dihydroagarofuran (1) has been isolated along with two known compounds, 6alpha-acetoxy-1beta, 9beta-dibenzoyloxy-8beta-hydroxy-beta-dihydroagarofuran (2), and 1beta, 6alpha-diacetoxy-9beta-benzoyloxy-8beta-hydroxy-beta-dihydroagarofuran ( 3). Their structures were elucidated on the basis of spectroscopic data.

Abstract:Phytochemical investigation of the fruits of Daucus carota L. resulted in the isolation of a new sesquiterpene named as daucucarotol (1). Its structure was elucidated on the basis of 1D and 2D NMR experiments, coupled with MS studies. To our knowledge, compound 1 is the first example for a natural eudesmane sesquiterpene with a hydroxymethyl group located at a methine carbon rather than a usual quaternary carbon in the two fused six-membered ringsystems.

Abstract:As a part of our interest in biologically active germacranolides from the genus Carpesium (Compositae), we have investigated the constituents of Carpesium cernuum. This paper reports the isolation and structural elucidation of a new germacranolide, cemolide A (Compound 1), from the herb. The structure of Compound 1 was determined as 2α,3β-dihydroxy-9-angeloxygermacra-4-en-6,12-olide on the basis of spectral evidence. The skeleton of Compound 1 was elucidation by IR, MS, 1H and 13C NMR, COSY, HMQC and HMBC experiments. The stereochemistry of Compound 1 was deduced by ROESY spectral data. Finally, the procedures of extraction and isolation were described in detail.

Abstract:This study is to investigate the chemical constituents from the whole plant Corydalis edulis. The chemical constituents were separated and purified by macroporous resin D101, silica gel, Sephadex LH-20, ODS, and semi-preparative HPLC. Their structures were determined by physicochemical properties and spectroscopic data. Four compounds were isolated from the dichloromethane and water extracts of the whole plant C. edulis, and identified as 6'-β-D-xylosylicariside B2(1),(3S,5R,6S,7E)-5,6-epoxy-3-hydroxy-7-megastigmen-9-one(2), loliolide(3), and 5,5'-dimethoxybiphenyl-2,2'-diol(4), respectively. Compound 1 is a new compound, of which the absolute configuration was established by electronic circular dichroism(ECD) calculations. Compound 4 is obtained from the plants of Papaveraceae family for the first time. Compounds 2 and 3 are firstly isolated from the Corydalis genus.

Abstract:A new tremulane sesquiterpene, ceriponol P, has been isolated from the cultures of Ceriporia lacerate, together with three known metabolites 3-indolecarbaldehyde, 3-methyl-2-indolinone, and daucosterol. Its structure was established by detailed analysis of its spectroscopic data, especially 1D, 2D NMR and HR-ESI-MS data. Ceriponol P represents the first example of monocyclic tremulane sesquiterpene. In biological assays, ceriponol P did not exhibit any significant cytotoxicity against HeLa, HepG2, and SGC 7901 cell lines at a concentration of 20 μM. All the known compounds were reported for the first time from the genus Ceriporia.

Abstract:Three unusual sesterterpenoids featuring unprecedented rearranged colquhounane (C25) and tetranorcolquhounane (C21) frameworks, colquhounoids E (1) and F (3) and norcolquhounoid F (2), were isolated from a Lamiaceae medicinal plant Colquhounia coccinea var. mollis. Their structures were elucidated by spectroscopic analysis and quantum chemical calculations. A biomimetic inspired regioselective cyclopropane cleavage was achieved under acidic conditions. The immunosuppressive activities of these new sesterterpenoids were also evaluated.

Abstract:The current study aimed to investigate the chemical components of the Chinese mangrove plant Cerbera manghas L., collected in Hainan Island.Repeated column chromatography was performed for the isolation and purification.Compounds were identified by the analysis of their 1D and 2D NMR data as well as IR and MS spectral data.A new linear monoterpene,named (2E,6S)-8-hydroxy-2-hydroxymethyl-6-rnethyl-2-octenoic acid(1),together with cerbinal(2),cerberidol(3),2α,3β,24-trihydroxyolea- 12en-28oic acid(4),4,4,9,7-tetrahydroxy-3,3-dimethoxy-7,9-epoxylignan(5),(-)-olovil(6),(+)-cycloolivil(7),naringenin-7- glucoside(8),aromadendrin(9),naringenine(10),and(+)-dihydroquercetin(11) were isolated from the EtOH and CH_2Cl_2 extracts. Abstract:Hedychium coronarium, a perennial herb belonging to the family Zingiberaceae, is cultivated as a garden plant or cut flower as well as for medicine and aromatic oil. Its flowers emit a fresh and inviting scent, which is mainly because of monoterpenes present in the profile of the floral volatiles. However, fragrance produced as a result of monoterpenes has not been well studied. In the present study, two novel terpene synthase (TPS) genes (HcTPS7 and HcTPS8) were isolated to study the biosynthesis of monoterpenes in H. coronarium. In vitro characterization showed that the recombinant HcTPS7 was capable of generating sabinene as its main product, in addition to nine sub-products from geranyl diphosphate (GPP). Recombinant HcTPS8 almost specifically catalyzed the formation of linalool from GPP, while it converted farnesyl diphosphate (FPP) to α-bergamotene, cis-α-bisabolene, β-farnesene and other ten sesquiterpenes. Subcellular localization experiments revealed that HcTPS7 and HcTPS8 were located in plastids. Real-time PCR analyses showed that HcTPS7 and HcTPS8 genes were highly expressed in petals and sepals, but were almost undetectable in vegetative organs. The changes of their expression levels in petals were positively correlated with the emission patterns of sabinene and linalool, respectively, during flower development. The results indicated that HcTPS7 and HcTPS8 were involved in the biosynthesis of sabinene and linalool in H. coronarium flowers. Results on these two TPSs first characterized from H. coronarium provide new insights into molecular mechanisms of terpene biosynthesis in this species and also lay the basis for biotechnological modification of floral scent profile in Hedychium.
