Genetic diversity of Sichuan elite wheat cultivars based on microsatellites and STS-PCR markers

Abstract:The genetic diversity among 40 Sichuan elite wheat cultivars, which had once popularized above 66700 ha per year during recent 50 years, was evaluated by microsatellites and STS-PCR markers. In microsatellites analysis, 33 out of 46 makers (71.7%) showed polymorphism. A total of 110 microsatellite alleles were detected. The number of alleles ranged from 1 to 8, with an average of 2.4 per locus. The microsatellite-derived genetic similarities (microsatellite-GS) ranged from 0.451 to 0.767, with the mean of 0.601. In STS-PCR analysis, no polymorphism was observed by directly detected the amplified products of the 13 STS-PCR markers. Upon digestion with HinfI and HhaI, 12 out of 13 markers (92.3%) and 23 out of 26 marker/enzyme combinations (88.5%) revealed polymorphisms. A total of 92 bands were observed in 26 marker/enzyme combinations, with 3.5 bands per marker/enzyme combination. Eighty out of 92 bands (87.0%) were polymorphic, among which 1 to 8 polymorphic bands were generated by each informative marker/enzyme combination. The STS-PCR-based genetic similarity (STS-PCR-GS) ranged from 0.478 to 0.989, with the mean of 0.753. The cluster analysis indicated that all 40 genotypes could be distinguished by both microsatellite and STS-PCR markers. Comparative analysis based on microsatellite and STS-PCR marker revealed that the genetic similarity among the cultivars released after 1970 were increased and get more closely related, especially among the cultivars released after 1990. These results suggested that it is very urgent to increase the genetic diversity of Sichuan wheat cultivars.
