Abstract:In this paper, polyethylene glycol (PEG) of different molecular weight was grafted on the polyethylene terephthalate (PET, Dacron) films by plasma surface grafting modification. The competitive adsorption relation of plasma (fibrinogen and albumin) adsorbing on materials surface was analyzed in light of surface energy and interface free energy. The results indicated that the PET films grafted PEG long chain molecular possesses the characteristic of preferentially adsorbing albumin and this adsorption tendency of grafted PEG6000 sample is most distinct. The platelet adhesion tests of the PET films whose surfaces were pre-set in contact with fibrinogen and albumin indicated that the surface adsorbing albumin can distinctly inhibit platelet adhesion and aggregation and possess favorable blood compatibility, but the surface adsorbing fibrinogen can enhance platelet adhesion and aggregation.

Abstract:In this paper, polyethylene glycol (PEG) of various different molecular weights was grafted onto PET films using plasma surface grafting modification. The surface structure of PEG-grafted PET films was analyzed by attenuated total reflection Fourier transform infrared spectroscopy (ATR-FTIR) and X-ray photoelectron spectroscopy (XPS), suggesting that the surface structure and composition of PET films changes due to the presence of PEG. Blood compatibility was characterized by in vitro platelet adhesion experiments and coagulation factors. The tests of platelet adhesion and coagulation factors in vitro suggest that PEG grafted onto polymer surfaces can improve the blood compatibility of PET films remarkably. The modified PET films were pre-coated with albumin and fibrinogen respectively; platelet adhesion tests in vitro then indicated that samples pre-coated with albumin have better blood compatibility than with fibrinogen, resulting in the conclusion that the albumin can improve blood compatibility. The contact angle of PEG-grafted films was measured by the sessile drop method and the surface free energy and interface free energy were induced. It is indicated that the PEG-grafted PET films have the characteristic of preferentially adsorbing albumin.

Abstract:Two methacrylate monomers, oligo(ethylene glycol) methyl ether methacrylate (OEGMA; MW=300gmol−1, poly(ethylene glycol) (PEG) side chains of average length n=4.5) and 2-methacryloyloxyethyl phosphorylcholine (MPC; MW=295gmol−1), were grafted from silicon wafer surfaces via surface-initiated atom transfer radical polymerization. The grafted surfaces were used as model PEG and phosphorylcholine surface systems to allow comparison of the effectiveness of these two motifs in the prevention of plasma protein adsorption and platelet adhesion. It was found that at high graft density fibrinogen adsorption from plasma on the poly(MPC) and poly(OEGMA) surfaces for a given graft chain length was comparable and extremely low. At low graft density, poly(OEGMA) was slightly more effective than poly(MPC) in resisting fibrinogen adsorption from plasma. Flowing whole blood experiments showed that at low graft density the poly(OEGMA) surfaces were more resistant to fibrinogen adsorption and platelet adhesion than the poly(MPC) surfaces. At high graft density, both the poly(MPC) and poly(OEGMA) surfaces were highly resistant to fibrinogen and platelets. Immunoblots of proteins eluted from the surfaces after contact with human plasma were probed with antibodies against a range of proteins, including the contact phase clotting factors, fibrinogen, albumin, complement C3, IgG, vitronectin and apolipoprotein A-I. The blot responses were weak on the poly(MPC) and poly(OEGMA) surfaces at low graft density and zero at high graft density, again indicating strongly protein resistant properties for these surfaces. Since the side chains of the poly(OEGMA) are about 50% greater in size than those of poly(MPC), the difference in protein resistance between the poly(MPC) and poly(OEGMA) surfaces at low graft density may be due to the difference in surface coverage of the two graft types.

Abstract:Argon plasma-induced graft polymerization of poly(ethylene glycol) methyl ether methacrylate (PEGMA) on H2 plasma-pretreated poly(tetrafluoroethylene) (PTFE) films was carried out to render the PTFE surfaces hydrophilic and protein-repellent. The surface microstructure and composition of the PEGMA graft-polymerized PTFE surfaces from plasma induction were characterized by water contact angle, X-ray photoelectron spectroscopy (XPS), Fourier-transform infrared spectroscopy (FTIR) and atomic force microscopy (AFM) measurements. The extent of cross-linking in the plasma-polymerized PEGMA (pp-PEGMA) was estimated by gel fraction determination. In general, an appropriate RF power of approximately 15 W and glow discharge time of 60 s for polymerization produced a high graft yield of pp-PEGMA on the H2 plasma-pretreated PTFE surface (the pp-PEGMA-g-PTFE surface). The hydrophilicity of the PTFE surfaces was greatly enhanced by the presence of pp-PEGMA. The PTFE surface with a high density of the grafted pp-PEGMA was very effective in preventing bovine serum albumin (BSA) protein adsorption.

Abstract:With the help of a silane coupling agent, poly(ethylene glycol) (PEG), a well-biocompatable agent, was grafted onto the surface of ultrahigh molecular weight polyethylene (UHMWPE) by ultraviolet initiation. Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy analysis proved the success of PEG grafting. Water contact angle measurement showed that the modified UHMWPE was obviously improved in surface hydrophilicity and thermogravimetric analysis result showed that its thermostability did not decline even it was pretreated by strong acids. Then, the protein adsorption of the modified UHMWPE was investigated using three model proteins including bovine serum albumin, lysozyme, and fibrinogen. Rabbit blood was used to study the platelet adhesion on the surface of modified UHMWPE. The results indicated that the quantity of protein adsorption on the modified UHMWPE grafted PEG reduced apparently for all the model proteins while there was some specific differences or exceptions among them. It was ascribed to the changed surface chemical composition, surface hydrophilicity and surface topography after modification. The adhesive ability of blood platelets on the modified surface of UHMWPE decreased after PEG grafting. Owing to the improved resistance to fibrinogen adsorption and platelet adhesion, the surface modification might endow the UHWMPE surface better anticoagulation ability according to clotting mechanism.

Abstract:Poly(tetrafluoroethylene) (PTFE) films with surface grafted poly(ethylene glycol) (PEG) chains were prepared by two methods: (1) UV-induced graft copolymerization of methoxy poly(ethylene glycol) monomethacrylate (PEGMA) onto the plasma-pretreated PTFE films; and (2) coupling of the hydroxyl groups of PEG via ester linkages with the carbonyl chloride groups which were introduced onto the acrylic acid (AAc) graft-copolymerized PTFE surface through reaction with thionyl chloride (SOCl2). The UV-induced graft copolymerization of PEGMA onto the plasma-pretreated PTFE film was explored with different macromonomer concentrations and different UV graft copolymerization time, The coupling reaction, on the other hand, was explored with PEG of different molecular weights. The surface microstructures and compositions of the PEG-modified PTFE films from both processes were characterized by contact angle, X-ray photoelectron spectroscopy (XPS), and atomic force microscopy (AFM) measurements. In general, higher macromonomer concentration and longer UV graft copolymerization time led to a higher graft yield for the UV-induced graft copolymerization with PEGMA. Contact angle measurements revealed that the hydrophilicity of the PTFE film surface was greatly enhanced by the grafting of the PEG chains. The PTFE surface with a high density of grafted PEG was very effective in preventing bovine serum albumin adsorption.

Abstract:Ar plasma pretreated polyterafluoroethylene(PTFE) films were subjected to further surface modification by grafting acrylic acid(AAc) on the surface.The surface composition and microstructure of the modified films were characterized by XPS and ATR-FTIR.It was shown that PAAc formed on the surface and the PTFE-g-PAAc films were obtained ultimately.The study confirmed that PTFE-g-PAAc films have a great improvement in surface wettability and surface stability.This approach can extend the application fields of the polymer to some relative fields and have significance for other high polymer materials.
Preparation and Characterization of Nonfouling Polymer Brushes on Poly(Ethylene Terephthalate) Film Surfaces

Abstract:In this study, a surface grafting of nonfouling poly(ethylene glycol) methyl ether acrylate (PEGMA) on poly(ethylene terephthalate) (PET) was carried out via surface-initiated atom-transfer radical polymerization (SI-ATRP) to improve hemocompatibility of polymer based biomaterials. To do this, the coupling agent with hydroxyl groups for the ATRP initiator was first anchored on the surface of PET films using photochemical method, and then these hydroxyl groups were esterified by bromoisobutyryl bromide, from which PET with various main chain lengths of PEGMA was prepared. The structures and properties of modified PET surfaces were investigated using water contact angle (WAC), ATR-FTIR, X-ray photoelectron spectroscopy (XPS) and Atomic force microscopy (AFM). The molecular weights of the free polymer from solution were determined by gel permeation chromatography (GPC). These results indicated that grafting of PEGMA on PET film is a simple way to change its surface properties. The protein adsorption resistance on the surfaces of PET was primarily evaluated by an enzyme-linked immunosorbent assay (ELISA). The result demonstrated that the protein adsorption could be well suppressed by poly(PEGMA) brush structure on the surface of PET. This work provides a new approach for polymers to enhance their biocompatibility.

Abstract:The influence of different surface modifications with poly(ethyleneglycol) (PEG) layers on the adsorption of fibrinogen and the adhesion and activation of macrophage-like human leukocytes was investigated. Poly(ethylene terephthalate) (PET) was modified using pulsed AC plasma polymerization with two types of starting monomers to generate: 1) a reactive acid surface using maleic anhydride (MAH) as monomer, and 2) a PEG-like surface using diethyleneglycol methyl vinyl ether (DEGVE) as monomer. The MAH surface was used as a reactive platform to graft linear chains of non-fouling mPEG via an intermediate layer of poly(ethyleneimine) (PEI) under lower critical solution temperature (LCST) conditions of the mPEG. The DEGVE monomer is used to create PEG-like layers by use of low power plasma conditions. The ability of the surfaces to resist protein adsorption was investigated quantitatively using 125I-radiolabeled human fibrinogen, and the conformation of the adsorbed protein was tested using an anti-fibrinogen monoclonal antibody in an enzyme-linked immunosorbent assay. The results showed that PEGylated surfaces adsorbed significantly less (up to 90% less) fibrinogen, and that unfolding of adsorbed fibrinogen was more pronounced on the linear mPEG layers than on the PEG-like plasma polymer surfaces. Adhesion of in-vitro differentiated macrophage-like U937 cells was reduced on both the PEG-like plasma polymer surfaces and the linear mPEG layers compared to the unmodified PET surface, but cells adhering to the PEG-like plasma polymer surfaces secreted less tumor necrosis factor-α (TNF- α) than cells adhering to the linear mPEG layers. In conclusion, the method for preparing non-fouling surfaces for long-term implanted devices influence surface-induced cellular responses of the host.
Surface Characterization and Platelet Adhesion Studies of Plasma Polymerized Allylamine Films Deposited on Polyethylene Terephthalate

Abstract:The plasma polymerized allylamine films were fabricated to modify the surfaces of the polyethylene terephthalate(PET) by pulsed and continued plasma polymerization technique,then the molecules of heparin were immobilized on the polyallylamine films by chemical reaction.The surface properties of the film were determined by attenuated total reflection Fourier transform infrared spectroscopy(ATR-FTIR),X-ray photoelectron spectroscopy(XPS),contact angle measurement,and chemical analysis.The content of the primary amine group of pulsed plasma polymerized allylamine film(PPAa-P) and the concentration of heparin immobilized on the PPAa-P films are 1.40% and 4.07μg/cm2,which were higher than those on the continued plasma polymerized allylamine film(PPAa-C) and the heparin-immobilized PPAa-C film.Compared to PPAa-C film,PPAa-P film was better hydrophilicit}.The anticoagulative property of the PET,heparin-immobilized PPAa-P(H-PPAa-P) and heparin-immobilized PPAa-C(H-PPAa-C) films were evaluated via in vitro platelet adhesion.Compared with the untreated PET,platelet adhesion and activation on the surfaces H-PPAa-P and H-PPAa-C films were remarkably decreased.Especially,the surface of H-PPAa-P was found to be much better anticoagulative effectivity.
Plasma graft polymerization of Acrylic Acid and immobilization of Heparin to improve blood compatibility of Polyethylene terephthalate (PET).

Abstract:Polymeric materials have been attracted the attention of researchers in various R&D applications. In this research, attempts were made to evaluate the changes in blood compatibility of polyethylene terephthalate (PET) by grafting acrylic acid (AAc) and immobilizing heparin by employing two-step plasma treatment. The PET surface was modified by using a novel method named “two-step plasma treatments” (TSPT). While first plasma is creating active sites for grafting, the second plasma polymerizing pre-adsorbed reactive monomer onto the surface of films. Finally, heparin immobilization was performed in the presence of 1-ethyl-3-(dimethylaminopropyl) carbodiimide. All films were characterized by attenuated total reflection Fourier transformed infrared (ATR-FTIR) spectroscopy and scanning electron microscopy (SEM). The surface hydrophilicity of films was studied by water contact angle test and blood compatibility evaluated by Lactate dehydrogenase method (LDH Test). In vitro studies based on LDH results demonstrared that platelet adhesion onto the modified surfaces with heparin was drastically reduced in comparison with unmodified PET.

Abstract:The surfaces of polyethylene terephthalate (PET) were modified by oxygen plasma-induced and ultraviolet (UV)-assisted acrylic acid (AAc) grafting polymerization, and the carboxyl (COOH) groups on the PET surface was 5.29×10−9mol/cm2. Then using the COOH as reacting sites, the molecules of gelatin and bovine serum albumin (BSA) were further co-immobilized on the PET surface. The modified PET surfaces were characterized by X-ray photoelectron spectroscopy (XPS) and surface chemical quantitative analysis. The results showed that the molecules of gelatin and albumin were immobilized on the PET surface. The concentration of gelatin on the gelatin-immobilized PET surface was 2.02μg/cm2. For the gelatin-immobilized PET surface, the human umbilical vein endothelial cells (HUVECs) culture attachment and proliferation ratios were improved, but the anticoagulation became worse proved by platelet adhesion test in vitro and the lactate dehydrogense (LDH) test. After further co-immobilization of albumin with gelatin biomolecules on the PET surface (PET-Gel–BSA), the percent of platelet adhesion in vitro decreased 28% than that on the gelatin-immobilized PET surface, and the cell density on the PET-Gel–BSA film (1.08×105cells/cm2) was significantly higher than that on the control PET surface. This investigation tries to find a method which can construct the anticoagulant surface before the endothelium formation and also accelerate the endothelialization of polymer surface.
Surface Modification on Polyethylene Terephthalate Films with 2-Methacryloyloxyethyl Phosphorylcholine.

Abstract:In this study, the surface of polyethylene terephthalate (PET) was modified to improve the protein and cell adhesion behavior with low temperature ammonia plasma treatment followed by 2-methacryloyloxyethyl phosphorylcholine (MPC) grafting. The x-ray photoelectron spectroscopy (XPS) results showed that the -COO(-), -N-C=O and -P-O-H groups were successfully incorporated onto the sample surface after MPC grafting. Furthermore, formation of new bonds, -N= and N-H on the sample surface grafted with MPC was recorded by Fourier transform infrared spectroscopy (FTIR). A large number of spherical particles at submicron to nanometer scale were also observed on the surface by atomic force microscopy (AFM). The cell adhesion experiments on PET film surfaces were evaluated and the highly hydrophilic surfaces could not promote cell adhesion and spreading. All results achieved in this study have clearly indicated that the method combining low temperature ammonia plasma treatment and MPC grafting is an effective way of producing a suitably hydrophilic PET surface with the capability of weakening the protein adsorption greatly.
Modification of Polycarbonateurethane Surface with Poly (ethylene Glycol) Monoacrylate and Phosphorylcholine Glyceraldehyde for Anti-Platelet Adhesion

Abstract:Poly(ethylene glycol) monoacrylate (PEGMA) is grafted onto polycarbonateurethane (PCU) surface via ultraviolet initiated photopolymerization. The hydroxyl groups of poly(PEGMA) on the surface react with one NCO group of isophorone diisocyanate (IPDI) and another NCO group of IPDI is then hydrolyzed to form amino terminal group, which is further grafted with phosphorylcholine glyceraldehyde to establish a biocompatible hydrophilic structure on the surface. Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy confirm the successful grafting of both PEG and phosphorylcholine functional groups on the surface. The decrease of the water contact angle for the modified film is caused by synergic effect of PEG and phosphorylcholine, which both have the high hydrophilicity. Furthermore, the number of platelets adhered is relative low on the synergetically modified PCU film compared with the PCU film modified only by poly(PEGMA). Our synergic modification method using both PEG and phosphorylcholine may be applied in surface modification of blood-contacting biomaterials and some relevant devices.
Effect of chain density and conformation on protein adsorption at PEG-grafted polyurethane surfaces

Abstract:Polyurethanes were modified using monobenzyloxy polyethylene glycol (BPEG) which possesses a bulky hydrophobic benzyloxy group at one end and a hydroxyl group at the other end as a preconstructed BPEG layer, and poly(ethylene glycol) (PEG) and monomethoxyl poly(ethylene glycol) (MPEG) with various chain lengths as fillers. Our objective was to investigate the effect of PEG graft density and conformation on protein adsorption at PEGlated surface. The graft density was estimated by a chemical titration method. The combination of ATR-FTIR, AFM and titration results provide evidences that the graft density can be increased by backfilling PEG or MPEG to a BPEG layer. However, fibrinogen and albumin adsorption significantly increased on all surfaces after PEG or MPEG backfilling. We conclude that the conformation of hydrophobic benzyloxy end groups of the BPEG layer plays a key role. The benzyloxy end groups of preconstructed PEG chains stretch to the surface after PEG backfilling, which possibly accounts for the observed increase in protein adsorption. The BPEG conformation change after backfilling with PEG or MPEG was also suggested by contact angles. Additionally, protein adsorption was slightly influenced by the length of filler, suggesting a change in surface morphology.

Abstract:We utilise pulsed AC plasma polymerisation to create thin films that either contain reactive acid functional groups (by deposition of maleic anhydride (MAH) followed by hydrolysis) or are poly(ethylene glycol) (PEG)-like in nature (by using diethylene glycol vinyl ether (DEGVE) as monomer). The MAH films were further modified with PEG chains using a two-step wet chemical method. For the DEGVE films the plasma power was varied in order to change the degree of monomer fragmentation and thus retention of PEG-like character. The chemistry of the surfaces was determined using Fourier-transform infrared spectroscopy (FTIR), X-ray photoelectron spectroscopy (XPS), and time-of-flight secondary ion mass spectroscopy (ToF SIMS). Significant reduction (up to 90%) in protein adsorption was achieved.

Abstract:Inspired by the composition of adhesive proteins in mussels, we used self-polymerized dopamine to form a thin and surface-adherent polydopamine layer onto poly(ethylene terephthalate) (PET) sheet, followed by covalently grafting lysine (Lys) to improve hemocompatibility and anti-biofouling property. The obtained surfaces were characterized by water contact angle measurements, attenuated total reflectance Fourier transform infrared spectroscopy, and X-ray photoelectron spectroscopy analysis. The results of platelet adhesion and protein adsorption tests showed that Lys-immobilized PET was endowed with improved resistance to nonspecific protein adsorption and platelet adhesion. Cell assay results showed that PET-g-Lys surface could greatly inhibit NIH 3T3 cell adhesion. These works provide a facile hemocompatible and anti-fouling surface for biomedical applications.

Abstract:A surface modification technique was developed for the covalent immobilization of poly(vinyl alcohol) (PVA) hydrogel onto poly(ethylene terephthalate) (PET) to improve the biocompatibility of the film. The PET film was first graft copolymerized with poly(ethylene glycol) monomethacrylate (PEGMA) in the presence of ethylene glycol dimethacrylate (EGDMA) as crosslinker, and then oxidized with a mixture of acetic anhydride (Ac2O) and dimethyl sulfoxide (DMSO) to produce aldehyde groups on the PET surface. Finally, the prepared PVA solution was cast onto the film and covalently immobilized on the film through the reaction between the aldehyde groups on the PET film and the hydroxyl groups of PVA. The good attachment of the PVA layer to the PET film was confirmed by observing the cross-section of the PET-PVA film using scanning electron microscopy (SEM). Heparin was immobilized on the PVA layered PET using two different methods, physical entrapment and covalent bonding, to further improve the biocompatibility of the film. Attenuated total reflectance (ATR) FT-IR spectroscopy and X-ray photoelectron spectroscopy (XPS) were used to characterize the chemical composition of the surface modified films. The biocompatibility of the various surface modified PET films was evaluated using plasma recalcification time (PRT) and platelet adhesion.

Abstract:We report a study involving chitosan chains immobilized on poly (ethylene terephthalate) (PET) films by plasma and ultraviolet (UV) grafting modification. The surface structure of the modified PET is determined by means of attenuated total reflection Fourier transform infraed spectroscopy (ATR-FTIR) and X-ray photoelectron spectroscopy (XPS). The results show that the chains of chitosan are successfully grafted on the surface of PET. Platelet adhesion evaluation in vitro is conducted to examine the blood compatibility in vitro. Scanning electron microscopy (SEM) and optical microscopy reveal that the amounts of adhered, aggregated and morphologically changed platelets are reduced on the chitosan-immobilized PET films. The number of platelet adhered on the modified film is reduced by almost 48% compared to the amount of platelets on the untreated film. Our result thus shows that chitosan immobilized on the PET surface improves blood compatibility.

Abstract:It remains a challenge to endow antithrombotic ability to polymeric material by surface grafting without disturbing the bulk properties of the substrate. A heparin induced functional layer within 80 nm was fabricated and covalently grafted on polyethylene terephthalate surface via carbene chemistry (Hep-g-PET). Heparin was oxidized with the minimum antithrombrin sequence remained creating aldehyde group on the chain terminal. Oxidized heparin was then covalently decorated on the poly (amidoamine) (PAMAM) grafting PET substrate. The interface between blood and PET was improved by the surface functionality, and the account of the attached platelets reduced to 29±12.1% of its initial value. The bulk properties of the functionalized film were hardly influenced, and the visible light transmittance kept more than 96%. The tethered layer also showed the ability to kill attached S. aureus and E. coli efficiently. Functionalized membrane showed negligible ex-vivo cell cytotoxicity and low hemolysis ratio. Hep-g-PET was implant in between mice skin and muscle, and showed outstanding histology response. The influences from the layer thickness and the heparin chain length were explored. Strategies reported in this work may help to improve the design of polymeric implant bio-devices.
