Abstract:Aim: To observe the biocompatibility of β-tricalcium phosphate (TCP)/calcium polyphosphate fiber (CPPF)/poly-levolactic acid (PLLA) composite as a novel bone tissue engineered scaffold by observing the morphology and proliferation of in vitro cultured cells. Methods: The experiment was carried out in the experimental center of Medical College and Institute of Endemic Diseases, Xi'an Jiaotong University from January 2002 to January 2004. 1 β-TCP/CPPF/PLLA scaffold was prepared by the Institute of Composite Materials, Lanzhou Jiaotong University; CPPF was (15.0±1.2) μm in diameter with (3.0± 0.46)% mean biodegradation rate, (1.0±0.2) GPa tensile strength, and (48.0±7.6) GPa; the mean molecular mass of PLLA was 27.6×104. The weight ratio of scaffold composite was β-TCP:CPPF:PLLA=2:3:5. 2 With the proportion of 1 g composite:10 mL extraction mediator, β-TCP/CPPF/PLLA scaffold was extracted by using DMEM containing 0.1 volume fracture fetal bovine serum at 37°C for 24 to 72 hours to prepare the extraction fluid, which was filtered before using. The bone marrow stromal cells (BMSCs) were divided into 3 groups and cultured with β-TCP/CPPF/PLLA maceration extract (experiment group), DMEM culture medium (negative control group) and 6.4 g/L Phenol solutions (positive control group), and the dilution of the maceration extract was divided into 100%, 75%, 50%, and 25%, respectively. 3 The growth and change condition of BMSCs were observed by phase contrast microscope. The growth and proliferation of BMSCs were investigated with MTT method to determine A value and calculate the proliferation rate of cells. Results: 1 On days 1, 2 and 3, the cell number of the positive control group displayed noticeable decrease, and the cells were of pyknosis even disaggregation; the cell number of the experiment group and negative control group was remarkably increased with normal call appearance. 2 At each time point of culture, the cytotoxicity of all groups were grade 0 with more than 80% cell proliferation rate except the positive control group was grade 4 with 0-20% cell proliferation rate; except the positive control group, the A values of experiment groups and negative control group were increased with culture days, and the A value of the positive control group on days 1, 2 and 3 was much lower than other groups (P < 0.01);. there were no significant differences between negative control group and experiment group (P > 0.05). Conclusion: β-TCP/CPPF/PLLA scaffold composite has a negligible influence and differentiation of BMSCs. It has good cell compatibility and no cytotoxicity.
