Abstract:A concise and efficient synthetic approach was developed for the synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones via a one-pot, three-component reaction of 4-aminocoumarin, arylglyoxal monohydrates, and aromatic amines promoted by KHSO4 in toluene under reflux condition. The attractive features of this protocol are operational simplicity, short reaction time, easy purification of products and good to excellent yields.

Abstract:A mild, efficient, and environmentally benign one-pot synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones by a three-component domino reaction of 4-aminocoumarins, arylglyoxal monohydrates, and 1,3-dicarbonyl compounds in refluxing ethanol without the use of catalyst is reported. The present protocol features operational simplicity, short reaction time, good yields, and the absence of aqueous workup procedure and chromatographic separation. [GRAPHICS] .

Abstract:A novel tandem one-pot method for the synthesis of polysubstituted pyridine derivatives has been developed via Knoevenagel-type reaction and subsequent 675 electrocyclization of the Blaise reaction intermediates and 3-formylchromones. Abstract:A one-pot two-step reaction of 4-aminocoumarin with arylglyoxal monohydrates and p-toluenesulfonates has been developed, which offers 3-alkoxy-substituted chromeno[4,3-b]pyrrol-4(1H)-ones in moderate to good yields. At first, arylglyoxal monohydrates and 4-aminocoumarin were converted into 3-hydroxychromeno[4,3-b]pyrrol-4(1H)-one intermediates in AcOH under reflux. Then the title compounds in an in situ reaction of the intermediates with p-toluenesulfonates catalyzed by DBU in refluxing toluene were obtained.

Abstract:A series of polysubstituted pyrroles were rapidly synthesized in moderate to good yields via catalyst-free one-pot three-component reaction of 1,3-dicarbonyl compounds with primary amines and nitrostyrenes by using liquid-assisted grinding. This protocol provides several advantages over the conventional chemical synthesis, such as simple work-up procedure, wide substrate scope, short reaction time, and environmental friendliness. Structures of polysubstituted pyrroles were confirmed by H-1 NMR, C-13 NMR, and ESI-MS studies.

Abstract:A three-component reaction of one molecule of imine and two molecules of alkynoates is realized with the catalysis of Cu(I)–Fe(III) in a sequential manner to allow the direct synthesis of functionalized pyrroles, during which, one C–N bond and two C–C bonds are formed with high atom economy. This method benefits from easily available starting materials, low-cost catalysts, and convenient operations.

Abstract:A facile and convenient synthesis of substituted pyridines has been developed via a one-pot multicomponent reaction of easily available 1,3-dicarbonyl compounds, aromatic aldehydes, malononitrile and alcohol in the presence of NaOH under mild conditions. A series of functionalized pyridines were thus obtained by this multicomponent reaction, in which four new bonds were formed in a highly chemo- and regioselective manner, and alcohol played dual roles as both reactant and reaction medium. Particularly valuable features of this protocol including mild conditions, simple execution, broad substrate scope, and good yields of products make it an efficient and promising synthetic strategy to build pyridine skeleton.

Abstract:A convenient zinc(II) chloride-catalyzed regioselective propargylation/amination/cycloisomerization process has been developed for the synthesis of substituted pyrrole derivatives from propargylic acetates, enoxysilanes and primary amines. Various aromatic and aliphatic propargylic acetates participate well in the reaction, providing the propargylation/amination/cycloisomerization products in good yields with complete regioselectivity. The one-pot multicomponent coupling reaction furnishes substituted pyrroles in high yields by circumventing the intermediates’ isolation. Zinc(II) chloride acts as a multifunctional catalyst and catalyzes three mechanistically distinct processes in a single-pot. The protocol developed has been extended to the synthesis of N-bridgehead pyrroles containing polycyclic fragments.

Abstract:We describe an unprecedented Pd(OCOCF3)(2)-catalyzed cascade process for the synthesis of highly functionalized 1,2,3,5-tetra-substituted pyrroles with high efficiency. Unlike documented methods relying on preformed enamines and active halogenated terminal alkenes, the process employs simple 2-alkenal-dicarbonyls and primary amines in 'one-pot'.

Abstract:Highly substituted novel 4H-pyrimido[1,6-a] pyrimidines were prepared by a trifluoromethanesulfonic acid catalyzed one-pot three-component condensation of 4-aminopyrimidines, aldehydes, and β-ketoesters. A preliminary feasibility study was undertaken on these compounds, to assess the potential production of a library of further diversified compounds by nucleophilic replacement of Cl(R(1)) or by reaction of electrophiles with the NH(2)(R(2)) group.

Abstract:N-(Hetero)aryl-4,5-unsubstituted pyrroles were synthesized from (hetero)arylamines, 1,3-dicarbonyl compounds, and α-bromoacetaldehyde acetal by using aluminum(III) chloride as a Lewis acid catalyst through [1 + 2 + 2] annulation. This new versatile methodology provides a wide scope for the synthesis of different functional N-(hetero)aryl-4,5-unsubstituted pyrrole scaffolds, which can be further derived to access multisubstituted pyrrole-3-carboxamides. In the presence of 1.2 equiv of KI, a polysubstituted pyrazolo[3,4-b]pyridine derivative was also successfully synthesized.

Abstract:A green and highly efficient method for the regioselective synthesis of imidazo[1,2-a] pyridine derivatives has been developed by annulation of heterocyclic ketene aminals (HKAs) and beta-oxodithioesters (ODEs) as building blocks with aldehydes under solvent-free conditions using Et3N as the catalyst. The present green synthesis shows fascinating properties such as high regioselectivity, concise one-pot methodology, short reaction times, easy purification and avoids the use of transition metals. This methodology provides an alternative approach for easy access to highly substituted imidazo[1,2-a]pyridines in good yields.

Abstract:An efficient one-pot synthesis of isoquinolines and heterocycle-fused pyridines by three-component reaction of aryl ketones, hydroxylamine, and alkynes is developed. The reaction involves condensation of aryl ketones and hydroxylamine, rhodium(III)-catalyzed C-H bond activation of the in situ generated aryl ketone oximes, and cyclization with internal alkynes. This protocol enables rapid assembly of multisubstituted isoquinolines as well as γ-carbolines, furo[2,3-c]pyridines, thieno[2,3-c]pyridines, and benzofuro[2,3-c]pyridines from readily available substrates.
