Abstract:A simple and efficient procedure is presented for Aza-Michael additions of various amines with conjugate alkenes bearing electron withdrawing group catalyzed by polystyrene-supported aluminum chloride (Ps-AlCl3) without the use of any solvents. The catalyst shows high catalytic activity for both aromatic amines and aliphatic amines. Chemoselective additions of the two types of amines with conjugate alkenes are achieved. Regioselective additions of two different amino groups in one molecule proceed smoothly. Ps-AlCl3 has better recyclability and can be reused several times without apparent loss of activity.

Abstract:A series of chiral tridentate ligands derived from readily available enantiopure l-proline were designed and synthesized. The ligands together with Cu(OAc)2 were successfully used in asymmetric Henry reactions. Various structurally divergent aldehydes and nitromethane were converted into versatile β-nitro alcohols in MeOH at room temperature with very good yields (up to 85%) and enantioselectivities (up to 86%).

Abstract:Near-critical water (NCW) has drawn increasing attention as an environmentally benign medium for organic chemical reactions, especially acid/base catalysis without added acid or base. However, problems such as low reaction rates and poor selectivity exist for organic syntheses in NCW that greatly hinder the application of this attractive technique in industrially interesting processes. We have developed a novel solvent system to accelerate base-catalyzed reactions by the addition of NH3 to NCW. The reactions demonstrated include hydrolysis of cinnamaldehyde and aldol condensation of benzaldehyde with acetaldehyde. The reaction kinetics of the two reactions were determined. The effects of the NH3 concentration and temperature on the reactions were also investigated. The pOH of NH3-enriched NCW was estimated. The results demonstrate the feasibility of using NH3-enriched NCW as a sustainable reaction medium for the acceleration of base-catalyzed reactions.

Abstract:Henry reactions and Knoevenagel reactions, used to prepare substituted olefins, have been limited by complex catalyst separation. In this article, methoxyl propylamine acetate ionic liquid was used as an environmentally benign catalyst for these reactions under solvent-free condition for the first time. This ionic liquid was shown to effectively catalyze Henry reactions and Knoevenagel reaction of active nitromethane compounds with various aldehydes. Yields from the catalyzed reactions were over 99% under solvent free condition. The process is highly effective, environmentally benign, and very selective. Furthermore, methoxyl propylamine acetate ionic liquid was conveniently separated with the products and easily recycled to catalyze Knoevenagel reaction again with excellent yields.

Abstract:A robust biocatalyst for green Henry reaction was achieved. Based on the fact that Henry reaction requires a base for proton transfer, we firstly proposed that the catalytic triad of lipase could play this role. The distance between the substrate and the catalytic center and the surrounding amino acid interaction network were used as the criterion. Benzaldehyde and nitromethane were used as the model reaction, RNL (Lipase from Rhizopus niveus) was considered to be the best Henry reaction catalyst via a molecular dynamics simulation. Then experiments demonstrated that RNL has a yield of 48 % using model substrate in water. Further, in order to increase product yield, the chemical modifier 1, 2-cyclohexanedione (CHD) was used to modify Arg on RNL. As a result, RNL (CHD) increased the activity of catalyzing Henry reaction and had a broad spectrum of substrates, the yield of the product was as high as 67–99 %.

Abstract:The novel efficient procedure has been developed for the conjugate addition of amines to electron deficient alkenes. A series of hydrotalcite-like materials were synthesized as catalyst for the conjugate addition of amines and alkenes. After optimizing the reaction conditions, ZnAl-LDHs (3 : 1) was chosen as the best catalyst for the reaction. The results showed that the catalyst worked very well for the conjugate addition of amines to electron deficient alkenes with the excellent yields in several minutes. Operational simplicity, no solvent, low cost of the catalyst, high yields, reusability, excellent chemoselectivity, wide applicability are the key features of this method.

Abstract:Nitroalkanes underwent fast additions to a variety of structurally diverse aldehydes under the catalysis of d-aminoacylase in DMSO. The influences of reaction conditions including solvents, temperature, enzyme concentration and molar ratio of substrates were systematically investigated. Seventeen products were obtained in short time with moderate to high yields. It is the first report on hydrolase-catalyzed fast Henry reaction in organic solvent.

Abstract:A highly enantioselective Henry reaction of β,γ-unsaturated α-ketoesters with nitromethane in water by virtue of chiral copper complexes has been developed. A series of unsaturated β-nitro-α-hydroxy esters bearing tetrasubstituted carbon stereocenters were obtained exclusively with high yields and excellent enantioselectivities. This method could avoid tedious anaerobic anhydrous manipulation and reduce the environmental pollution caused by organic solvents.

K2CO3 supported on different carriers have been synthesized for the conjugate addition of amines and alkenes. After optimizing the reaction conditions, K2CO3/MgO was chosen as the most efficient catalyst for the reactions. The results showed that the catalyst was very efficient for the conjugate addition of amines to electron deficient alkenes with the excellent yields in several minutes. Operational simplicity, without need of any solvent, low cost of the catalyst used, high yields, reusability, excellent chemoselectivity, and applicability to large-scale reactions are the key features of this methodology.

Abstract:It was found that chitosan supported L-proline can be used as a heterogeneous organocatalyst to catalyze the asymmetric Henry reaction with different substrates in various media. The catalyst showed better performance in water and aqueous micelles. Unexpectedly, it was found that the chitosan itself can also catalyze Henry reaction. The catalytic performance of chitosan was well studied under various conditions. Similar to the chitosan supported L-proline, chitosan showed better performance in water and aqueous micelles than in the organic solvents, especially, a yield of 99% with a dr ratio of 1 : 2.2 was obtained in Span-40 aqueous micelle.

Abstract:A highly anti-selective asymmetric Henry reaction has been developed, affording synthetically versatile beta-nitroalcohols in a predominately anti-selective manner (mostly above 15:1) and excellent ee values (mostly above 95?%). Moreover, the anti-selective Henry reaction was carried out in the presence of water for the first time with up to 99?% ee. The catalytic mechanism was proposed based on the detection of the intermediates by extractive electrospray ionization mass spectrometry (EESI-MS). Furthermore, the anti adducts have been successfully transformed into the biochemically important (+)-spisulosine and a pyrroloisoquinoline derivative.

Abstract:Several novel chiral tridentate ligands containing thiazoline were efficiently synthesized from commercially available l=cysteine in high yield. These ligands were subsequently applied to the asymmetric Henry reaction of nitromethane and various aldehydes. It was found that the structures of the thiazoline ligands had a significant influence on the enantioselectivity. It was shown that the optimal catalyst for this reaction was a ligand complexed with CuCl, which was formed from chiral thiazoline with chiral aminoalcohol. At −20°C, with 10 mol% of this ligand, a product with (S)-configuration was isolated in 93% yield and 98% enantiomeric excess. Copyright © 2015 John Wiley & Sons, Ltd.
