Abstract:AIM To investigate the chemical composition of the root of Salvia przewalskii Maxim. METHODS Compounds were isolated by silica gel column chromatography. Structures of these compounds were elucidated by spectral analysis (EI-MS, FAB-MS, 1HNMR, 13CNMR, 1H-1H COSY, 1H-13C COSY, HMBC, NOESY) and phytochemical properties. RESULTS Eight compounds were isolated and identified as: tanshinone II-A (I), crypotanshinone (II), przewaquinone A (III), sugiol (IV), ursolic acid (V), 2 alpha, 3 alpha-dihydroxy urs-12-ene-28-acid (VI), oleanolic acid (VII), and neo-przewaquinone A (VIII). CONCLUSION Compound VIII is a new compound, and compound II, IV, V, VI and VII are isolated from this plant for the first time.

Abstract:Three new compounds including one ortho-quinone-type icetexane diterpenoid, i.e., przewalskin E (1), one abietane dinorditerpenoid. i.e., przewalskin F (2), and one 7,20-epoxyabietane diterpenoid. i.e., przewalskin G (3), were isolated from Salvia przewalskii MAXIM. The identification and structure elucidation of these compounds were based on 1D- and 2D-NMR spectral-data analysis. It is noteworthy that przewalskin E (1), featured with an 11,12-dioxoicetexane skeleton, represents the second example of a naturally occurring ortho-quinone-type icetexane diterpenoid from Salvia plants.

Abstract:In order to investigate the biologically active constituents of the Salvia przewalskii Maxim., a new diterpenoid, named isoganxinonic acid A (1), together with y-linolenic acid (2), tanshinol B (3), cryptotanshinone (4), paeoniflorin (5), protocatechualdehyde (6), protocatechuic acid (7), caffeic acid (8), rosmarinic acid (9) and salvianolic acid B (10), were isolated from 50% ethanol extract of the roots and rhizomes of S. przewalskii and their structures were identified by MS and NMR spectra. Compound 5 was reported from Labiatae for the first time.

Abstract:Salvia przewalskii Maxim. (Labiatae) is widely distributed in the western areas of China, such as Gansu, Sichuan, Yunnan and Tibet provinces. And it has been used as a substitute in Chinese folk medicine for S. miltiorrhiza Bge. (Dan Shen). The mainly chemical constituents of S. przewalskii are diterpenoids and phenolic acids. In this work, isolation and purification were carried out repeatedly by silica gel, Sephadex LH-20, ODS C(18) and MCI gel column chromatography. Two new diterpenoids, named ganxintriol A(1) and ganxintriol B(2), and a new monoterpenoid glycoside, named ganxinoside A(3), were obtained from 50% ethanol extract of the roots and rhizomes of S. przewalskii. Their structures were established on the basis of physical methods, chemical methods and spectral data.

Abstract:Two C₂₁- and C₂₂-terpenoids, salviprzols A (1) and B (2), together with 24 known compounds including 17 diterpenoids (3-19), a triterpenoid (20), and 6 phenolic derivatives (21-26), were isolated from the roots of Salvia przewalskii Maxim. Salviprzols A and B represented a new subtype of C₂₃-terpenoids featured by an additional 2-oxopropyl moiety at C-12 and a rare γ-hydroxyl-α-methyl-α,β-unsaturated-γ-lactone ring system. Their structures were elucidated by extensive spectroscopic analyses, and the structure of 2 was confirmed by a single-crystal X-ray diffraction crystallography. The cytotoxic activities of the new isolates were tested. A plausible biosynthetic pathway for 1 and 2 was also proposed.

Abstract:Salvia przewalskii Maxim is a perennial plant from the genus Salvia (family Lamiaceae). The roots of S. przewalskii were long used as a traditional herb to treat blood circulation related illnesses in China. As part of our continuing interest in polycyclic natural products from medicinal plants, two unprecedented adducts comprised of a dinor-diterpenoid and a 9′-nor-rosmarinic acid derivative, linked by a 1,4-benzodioxane motif (1 and 2), were isolated from the roots of S. przewalskii. Their structures were established by extensive spectroscopic approaches including 1D, 2D NMR, and HRFABMS. Their cytotoxic activities against five human tumor cell lines were evaluated.

Abstract:A new secoiridoid named chinensisol (1) along with twenty known compounds were isolated from the 95% ethanol extract of Cortex fraxini. Their structures were elucidated on the basis of NMR, MS, IR and UV spectral evidences. The quinone reductase (QR) inducing activities of the compounds were evaluated and the results showed that compounds 1, 9 and 14 had moderate QR inducing activities with CD values (concentration required to double the specific activity of QR) of 72.4 ± 7.7, 34.3 ± 3.3 and 42.0 ± 0.4 μM respectively.

Abstract:This study aimed to report the structure elucidation of the compounds isolated from Salvia miltiorrhiza, and their biological evaluations. Ten undescribed diterpenoid quinones and 10 known analogues were isolated from the dried roots of S. miltiorrhiza. Their structures were elucidated by extensive analysis, including nuclear magnetic resonance, high-resolution mass spectra, and ultraviolet and infrared spectra. Their absolute configurations were determined by comparing the experimental and calculated electronic circular dichroism spectra. In the evaluation of bioactivities, Salvianolactone acid I, epi-danshenspiroketallactone F, danshinspiroketallactone, grandifolia G, and 2H-Naphtho [1,8-bc]furan (10 μM) significantly increased cell viability and decreased the nuclear transport of p-P65 in lipopolysaccharide-induced bronchial epithelial cells. It was concluded that the diterpenoid quinones might belong to potent targeted lung-protective agents.

Abstract:The antibacierial (staphylococcus aureus) activities of 23 diterpene quinones from the Salvia przewalskii Maxim, the artificial synthesis ones and that of the metabolism in pigs were examined in an effort to study the quantitative structure activity relationship with the Free-Wilson method. With the application of the multipiy regression, the Free-Wilson pattern was established by the dummy (indicator) variable of the structural fragments or substituents. The results showed: (1) The diketone moiety played the fundamental role in antibacterial activities, ortho-quinones was higher active than para-quinones. (2) The single bond existed between the C15 and the C16 at D-ring led to higher activity. (3) The saturation at the A-ring led to higher activity. (4) The components adjacent to nitrogen substituents at C15 led to higher activity. (5)The hydroxylation or dehydrogenation at A-ring led to less activity. The research results can provide the theoretical basis for developing a new antibacterial drug from the Salvia diterpene quinones.

Abstract:Fifteen undescribed diterpenoid quinones salviamilthone A-O ( 1 – 15 ), together with three known diterpenoid quinones ( 16 – 18 ), were isolated from the roots of Salvia miltiorrhiza Bunge. Their structures were elucidated using 1D and 2D NMR data, while the relative and absolute configurations were confirmed by NOESY correlations and comparison between experimental and calculated ECD spectra. In the evaluation of bioactivities, salviamilthone J ( 10 ), salviamone ( 18 ) (10 μM) significantly increased cell viability and decreased the expression of IL-1β in lipopolysaccharide-induced BEAS-2B cells. These data provide the molecular justification for the usage of Salvia miltiorrhiza in treating acute lung injury.

Abstract:One new chromone, (2′S,4′S)-5′-hydroxy-5-O-methylvisamminol ( 1 ), was isolated from the roots of Saposhnikovia divaricata. The structure of compound 1 was identified by spectroscopic methods including 1D, 2D NMR, and HR-ESI-MS data. The absolute configuration of compound 1 was elucidated by electronic circular dichroism (ECD) calculation. Compound 1 showed moderate cytotoxic activity against human prostatic cancer PC-3 and breast cancer MCF-7 cell lines, with IC 50 values of 52.66 ± 1.73 and 83.12 ± 8.50 μM, respectively.

Abstract:(±)-Salvicatone A (1), a C27-meroterpenoid featuring a unique 6/6/6/6/6-pentacyclic carbon skeleton with a 7,8,8a,9,10,10a-hexahydropyren-1 (6H)-one motif, was isolated from the roots and rhizomes of Salvia castanea Diels f. tomentosa Stib. Its structure was characterized by comprehensive spectroscopic analyses along with computer-assisted structure elucidation, including ACD/structure elucidator and quantum chemical calculations with 1H/13C NMR and electronic circular dichroism. Biogenetically, compound 1 was constructed from decarboxylation following [4 + 2] Diels-Alder cycloaddition reaction between caffeic acid and miltirone analogue. Bioassays showed that (-)-1 and (+)-1 inhibited nitric oxide production in lipopolysaccharide-induced RAW264.7 macrophage cells with an IC50 value of 6.48 ± 1.25 and 15.76 ± 5.55 μM, respectively. The structure-based virtual screening based on the pharmacophores in ePharmaLib, as well as the molecular docking and molecular dynamics simulations study, implied that (-)-1 and (+)-1 may potentially bind to retinoic acid receptor-related orphan receptor C to exert anti-inflammatory activities.
