Abstract:A convenient method for the synthesis of polyamides containing N-methylpyrrole (Py) and N-methylimidazole (Im) in solution has been developed. Most of the building blocks have been prepared by a haloform reaction in a simple way that column chromatography can be avoided. By use of the DCC/HOBT coupling reaction, the building blocks prepared have been effectively connected to construct a variety of subchains and polyamides without employing amino protection and deprotection. By use of the present method, an eight-ring polyamide, PyPyPyPy gamma PyImImPy beta Dp (gamma is gamma-aminobutyric acid, beta is beta-alanine, Dp is N,N-dimethylpropyldiamine), has been synthesized by the coupling of two four-ring subchains in one step.

Abstract:The pyrrole-imidazole (Py-Im) polyamides represent the only available class of synthetic small molecules which can be designed to recognize virtually any predetermined B-DNA sequence in minor groove and permeate into nucleus to regulate gene expression in vitro & in vivo due to affinities and specificities, and equal or exceed natural eukaryotic transcriptional regulatory proteins. They are mainly composed of N-methylpyrrole (Py), N-methylimidazole (Im), N-methyl-3-hydroxypyrrole (Hp) amino acids and aliphatic chain compounds including aliphatic amino acids. In these moieties, an aliphatic chain as a part of constructing polyamides and their conjugates plays a very important role in extending and specifically recognizing predetermined DNA sequences, linking functioned bioactive molecules to polyamides, and gene regulation. Understanding the role of aliphatic chains in polyamides and their conjugates may help us to better design suitable polyamides to be applied in DNA recognition, which can speed up research of polyamides as a gene-targeted clinical drug. In this paper, we review the application of aliphatic chains of polyamides and their conjugates in polyamides binding to B-DNA minor groove and analyze existing problems.
Electrospray Ionization Mass Spectrometric Analysis of Noncovalent Complexes Between Dna and Polyamides Containing N-Methylpyrrole and N-Methylimidazole

Abstract:Electrospray ionization mass spectrometry was utilized to investigate the noncovalent complexes between novel polyamides and DNA containing the TCCT sequence. We analyzed the noncovalent binding of the polyamides with the DNA and assessed their relative affinities and stoichiometry. The results confirm that hairpin polyamides have higher binding affinities than three-ring polyamides. The hairpin polyamide (PyPyPyPygammaPyImImPybetaDp) has the highest affinity, and the beta-linked polyamide (PyPyPybetaImImImbetaDp) shows a dominant 1:2 binding stoichiometry. Two groups of competition experiments were undertaken to compare the binding affinities of the duplex DNA with different polyamides directly. The affinity scale thus obtained for the group-1 polyamides is PyPyPyPygammaPyImImPybetaDp > PyPyPybetaImImImbetaDp approximate to PyPyPygammaImImImbetaDp much greater than PyPyPybetaDp > PyImImbetaDp approximate to ImImPybetaDp, and the order for the group-2 polyamides is PyPyPygammaImImImbetaDp > PyPyPygammaImImImbetaOEt > PyPyPygammaImImImbetaCOOH. Abstract:The new aromatic m-catenated diamine, N-(3, 5-diaminophenyl)-4-(naphthalen-8-yloxy)benzamide was synthesized from 1- naphthol and 4- fluoro benzonitrile. A series of novel aromatic polyamides; containing pendant bulky naphthalene-8-oxybenzamide unit; was synthesized from the diamine and several structurally different aromatic diacids such as 4,4'-Oxydibenzoic acid, Naphthalene-2, 6-dicarboxylic acid, 4,4'-Sulfonyldibenzoic acid, Biphenyl-4,4'-dicarboxylic acid, and 4,4'-(perfluoropropane-2, 2-diyl) dibenzoic acid via direct Yamazaki's phosphorylative polycondensation method using triphenyl phosphite (TPP) as a condensing agent in pyridine (Py)/LiCl/1-methyl-2-pyrrolidone (NMP) solvent system. These new kink structured polyamides were characterized by inherent viscosities, solubility in organic solvents, FT-IR, DSC, TGA and XRD techniques. The polyamides were obtained in good yields and had inherent viscosities in range 0.35 and 0.55 dL/g. These polyamides showed good solubility in amide type polar aprotic solvents namely N, N' dimethyl formamide (DMF), 1-methyl-2-pyrrolidone (NMP), N, N' dimethyl acetamide (DMAc), dimethyl sulfoxide (DMSO) and partly soluble in m-cresol, tetrahydrofuron (THF). Polyamides showed glass-transition temperatures in the range 207 °C-245 °C indicating their processability at these temperatures. The 10% weight loss temperatures as determined by the thermogravimetric analysis were more than 413 °C, indicating a good thermal stability. X-ray diffraction patterns of these polyamides indicated the amorphous nature, which is reflected in solubility behavior.

Abstract:Synthesis of poly(α-amino acid)s bearing carboxyl groups is a critical pathway to prepare biomaterials to simulate functional proteins. The traditional approaches call for carboxyl-protected monomers to prevent degradation of monomers or wrong linkage. In this contribution, we synthesize N -carboxypentyl glycine N -thiocarboxyanhydride (CPG-NTA) and iminodiacetic acid N -thiocarboxyanhydride (IDA-NTA) without protection. Initiated by amines, CPG-NTA directly polymerizes into polyCPG bearing unprotected carboxyl groups with controlled molecular weight (2.8–9.3 kg mol −1 ) and low dispersities (1.08–1.12). Block and random copolymerizations of CPG-NTA with N -ethyl glycine N -thiocarboxyanhydride (NEG-NTA) demonstrate its versatile construction of complicated polypeptoids. On the contrary, IDA-NTA transforms amines into cyclic IDA dimer-capped species with carboxyl end group in decent yields (>89%) regio-selectively. Density functional theory calculation elucidates that IDA repeating unit is prone to cyclize to be the six-membered ring product with low ΔG . The polymer is a good adhesive reagent to various materials with adhesive strength of 33–229 kPa.

Abstract:A new aromatic diiodide compound, 1,2 - bis(4 - iodophenyl) - 4,5 - diphenyl - 1 H - imidazole was synthesized by a one - pot sequential diazotization-iodination reaction using 1,2 - bis(4 - aminophenyl) - 4,5 - diphenyl - 1 H - imidazole as the starting material in a KI/NaNO 2 / p - TsOH/CH 3 CN system at room temperature. A series of aromatic polyamides (PAs) containing tetraphenyl-imidazolyl units were prepared in good yields via a Pd(PPh 3 ) 2 Cl 2 catalyzed carbonylation polymerization of this diiodide, diamines and carbon monoxide in DMAc in the presence of PPh 3 and 1,8-diazabicyclo[5,4,0]-7-undecene. These new PAs with inherent viscosities of 0.63–0.72 dL/g were amorphous and easily soluble in amide type aprotic solvents such as N - methyl - 2 - pyrrolidone (NMP), N,N '-dimethylacetamide (DMAc), N,N ′ - dimethylformamide (DMF), dimethyl sulfoxide (DMSO) and pyridine at room temperature. Thermal analyses data of the resulting PAs up to 800 °C in nitrogen showed their high thermal behavior with 5% weight loss temperature of 425–517 °C and char yield of 43–58%. The present PAs could be classified as "self-extinguishing polymers" based on the limiting oxygen index in the 34.7–40.7 range. Abstract:Halophenyl derivatives were linked to different termini of polyamide containing three imidazole rings to give novel conjugates for examining the influence of the location of linking on DNA-cleavage efficiency. The DNA cleavage activities of the conjugates were compared on supercoiled DNA (pBluescript SK) under UV irradiation at 365 nm. It was found that bromophenyl-ImImImbetaDp (2), which is the conjugate of bromophenyl motif directly linked to the N-terminus of polyamide, had the highest photocleavage efficiency among six novel conjugates. In addition, it was found that the polyamide containing three N-methylimidazole rings could greatly enhance the photocleavage efficiency of these conjugates.

Abstract:A series of novel copolycondensation type polyimides containing pyridine ring were successfully prepared via two-step reaction from mixed diamines and three dianhydrides,respectively. The chemical structures of the resulting polymers were characterized by Fourier transform infrared spectrum(FT-IR) spectroscopy and 1H nuclear magnetic resonance spectroscopy (1H-NMR). Thermal stability,solubility and some optical properties of the obtained polymers were studied by thermogravimetry analyzer (TGA),solubility test,UV-vis spectroscopy and fluorescence spectroscopy. It is found that the polyimides,which are introduced aromatic heterocycle (pyridine ring),bulky pendant substituent (phenyl) and flexible linkage (ether bond) by copolycondensation,have excellent thermal stability (10% thermal decomposition temperature under N2 is above 520 ℃),and present better solubility and certain optical activity (UV-vis absorption bands appeared in the range of 200 nm～400 nm,and emission spectra exhibited in the range of 485 nm～489 nm as excited at 425 nm) at the same time.

Abstract:In order to develop an optimal subunit as a T-recognition element in hairpin polyamides, 15 novel chirality-modified polyamides containing (R)-a,beta-diaminopropionic acid (R beta?a-NH?2), (S)-a,beta-diaminopropionic acid (S beta?a-NH?2), (1R,3S)-3-aminocyclopentanecarboxylic acid (RSCp), (1S,3R)-3-amino-cyclopentanecarboxylic acid (RSCp), (1R,3R)-3-aminocyclopentanecarboxylic acid (RRCp) and (1S,3S)-3-amino-cyclopentanecarboxylic acid (SSCp) residues were synthesized. Their binding characteristics to DNA sequences 5'-TGCNCAT-3'/3'-ACGN'GTA-5' (N.N'=A.T, T.A, G.C and C.G) were systemically studied by surface plasmon resonance (SPR) and molecular simulation (MSim) techniques. SPR showed that polyamide 4, AcIm-S beta?a-NH?2-ImPy-?-ImPy-beta-Py-beta Dp (beta/S beta?a-NH?2 pair), bound to a DNA sequence containing a core binding site of 5'-TGCACAT-3' with a dissociation equilibrium constant (KD) of 4.5 x 10(-8)?m. This was a tenfold improvement in specificity over 5'-TGCTCAT-3' (KD=4.5 x 10(-7)?M). MSim studies supported the SPR results. More importantly, for the first time, we found that chiral 3-aminocyclopentanecarboxylic acids in polyamides can be employed as base readers with only a small decrease in binding affinity to DNA. In particular, SPR showed that polyamide 9 (RRCp/beta pair) had a 15-fold binding preference for 5'-TGCTCAT-3' over 5'-TGCACAT-3'. A large difference in standard free energy change for A.T over T.A was determined (??Go=5.9 kJ?mol-1), as was a twofold decrease in interaction energy by MSim. Moreover, a 1:1 stoichiometry (9 to 5'-TGCTCAT-3'/3'-ACGAGTA-5') was shown by MSim to be optimal for the chiral five-membered cycle to fit the minor groove. Collectively, the study suggests that the (S)-a-amino-beta-aminopropionic acid and (1R,3R)-3-aminocyclopentanecarboxylic acid can serve as a T-recognition element, and the stereochemistry and the nature of these subunits significantly influence binding properties in these recognition events. Subunit (1R,3R)-3-aminocyclopentanecarboxylic acid broadens our scope to design novel polyamides.

Abstract:Poly(aryl imino) containing biphenyl group was synthesized by the condensation polymerization of 4,4'-Dibromobiphenyl and aromatic diamines via Palladium-catalyzed aryl amination reaction The structure of the polymers synthesized was characterized by means of FT-IR,1H-NMR spectroscopy and elemental analysis,and the testing results show a good agreement with the proposed structure.DSC and TG measurements showed that the polymers possess good thermal stability(Tg>250℃;TD>510℃).

Abstract:Comb-like polyamides bearing different contents of tetraaniline pendant groups were synthesized via nucleophilic polymerization coupled with a post-functionalization method, and exhibit excellent solubility, good thermal stability and interesting spectroscopic properties. Their electronic properties, including conductivity, dielectricity, electroactivity, electrochromism and anticorrosion, were effectively adjusted and controlled by importing different numbers of tetraaniline pendant groups.

Single Component Polymerization of Diisocyanoacetates Toward Polyimidazoles

Abstract:The polymerization of triple-bond building blocks will generate functional polymers owing to their unsaturated backbones, which facilitate the electron delocation within the main chains. Currently, the research focus in this area is the alkyne-based polymerization, and the carbon-nitrogen triple-bond based polymerization to produce stable nitrogen-containing polymers is rarely reported. In this paper, a new polymerization of diisocyanoacetate was successfully established. The silver acetate-catalyzed polymerization of diisocyanoacetate in the acetonitrile or DMF readily produced soluble polyimidazoles with high molecular weights (M-w up to 32500) in excellent yields (up to 94%) at room temperature after 2 h. Moreover, this polymerization performed in a single component fashion, which shows remarkable advantages over the traditional two- or multicomponent ones. In addition, the resultant polyimidazoles could be postfunctionalized to yield ionized polyelectrolytes and could feature the aggregation-induced emission (AIE) characteristics by incorporation of an AIE-active tetraphenylethene moiety in its polymer chains. This single component polymerization will become a powerful tool for the preparation of polyimidazoles and be potentially applicable in materials and biological fields.

Abstract:This study investigates the synthesis and characterization of new diamines with carbazole and imidazole pendant groups, leading to the development of polyamides and polyimides. The polyamides were synthesized by reacting diamines with carboxylic acids using polyamidation and phosphorylation techniques, while polyimides were prepared by combining diamines and dianhydrides. FT–IR and 1H-NMR analyses were used for comprehensive structural elucidation of diamine, polyamides, and polyimides. The polyamides and polyimides with inherent viscosities ranging from 0.48 to 0.64 dL/g showed notable thermal stability. The T 10 % weight loss values were between 287 and 340 °C for polyamides and 321–381 °C for polyimides. The percentage of char residue varied between 33 and 56% at 700 °C. Additionally, the maximum emission wavelengths of polymers ranged from 410 to 436 nm. The presence of bulky carbazole and imidazole pendant groups facilitated the good solubility of the polymers in common organic solvents. The antibacterial properties of synthesized polyamides and polyimides against both Gram-negative and Gram-positive bacteria and fungi were investigated.

Abstract:New soluble polyimides were synthesized from different 4,4′-diaminodiphenylmethane monomers with different alkyl substituents [4′4′-diamino-3,3′-dimethyl-diphenyl-methane, DDMDPM; 4,4′-methylene -bis(2-ethyl-6-methylaniline), MBEMA; and 4,4′-methylene-bis(2,6-diethylaniline), MBDEA] in one-step with the poly(amic acid)s prepared from the polyaddition of 4,4′-oxydiphthalic anhydride (ODPA). The structures of the synthesized polyimides were confirmed by Fourier transform infrared (FTIR) and nuclear magnetic resonance (NMR) spectroscopy measurements. The sizes and numbers of alkyl substituents affected the thermal properties, dielectric constants, and solubilities of the polyimides. The obtained polyimides were soluble in various solvents, such as N-methyl-2-pyrrolidone (NMP), N,N-dimethylacetamide (DMAc), N,N-dimethylformamide (DMF), tetrahydrofuran (THF), dichloromethane (DCM), and chloroform (CClH3). These polyimides exhibited excellent thermal stability with a decomposition temperature (Td) greater than 500 °C and relatively low coefficients of thermal expansion and dielectric constants.
