Abstract:Using layered γ-zirconium phosphate (γ-ZrP) for preparation of immobilized hemoglobin (Hb) was investigated. We discussed the effects of pH, ionic strength and concentration of amines on the intercalation and immobilization behaviors. The adsorption, desorption, catalytic activity, thermal stability of the inorganic solid-protein nanocomposites were also studied. This fundamental research was carried out aiming at comparing the influence of different surface properties on the Hb immobilization after γ-ZrP has been pre-intercalated with different kinds of amine or without amine.

Abstract:The fact that different gamma-zirconium phosphate (gamma-ZrP) preintercalation method induced varied degree and type of conformational change of the adsorption protein was confirmed by characterization techniques including circular dichroism (CD), fourier transform infrared spectroscopy (FTIR) and X-ray powder diffraction (XRD) analysis. The results indicated that the association of hemoglobin with gamma-ZrP preintercalated using butylamine was correlated with conformational change in the secondary structure of the protein. gamma-ZrP which was preintercalated with tetra (n-butylammonium) hydroxide caused the conformational change of Hemoglobin in both the secondary structure and the tertiary structure. X-ray powder diffraction analysis was used to analyze the crystalline structure of the nanocomposites prepared by relamination. The adsorption isotherms of Hemoglobin on different matrices were set up and fitted with Langmuir and Freundlich equations.

Abstract:This paper reports layer-by-layer films fabricated with hemoglobin (Hb) and gold nanoparticles (AuNPs) as well as their applications in carrying oxygen. The characterization of {Hb/AuNPs}(n) multilayer films at different layers revealed that the formation of films was step-by-step. Meanwhile, the study showed that the {Hb/AuNPs}(n) modified glass carbon electrode can maintain the biological activity of Hb. The capacity of carrying oxygen was associated with the number of assembled layers. The {Hb/AuNPs}(3) multilayer films displayed a good storing and releasing ability of oxygen. The study can provide an alternative method for construction of hemoglobin function interface with high-efficiency oxygen carrying capacity. (C) 2015 Elsevier B.V. All rights reserved.

Abstract:The potential toxicity towards human kidneys of hemoglobin (Hb), when used directly, has severely limited its application as a red blood cell substitute and in cancer treatments. In this work, a novel hemoglobin-polymer conjugate was prepared by a reaction between the lysine amino groups of Hb and the carboxyl groups of a copolymer, poly(N-isopropylacrylamide) grafted carboxylated dextran (HOOC-Dex-g-PNIPAAm), which was synthesized by single electron transfer living radical polymerization (SET-LRP) and post-carboxylation. The conjugate was characterized by FTIR, H-1 NMR, SDS-PAGE, DLS, fluorescence spectroscopy and TEM. Results showed it had a relatively low critical micelle concentration (CMC) and could form stable spherical nanoparticles upon heating above the LCST. The redox activity and gas-binding capacity of the Hb conjugate were subsequently confirmed by UV-vis spectroscopy, indicating the retention of Hb bioactivity after conjugation. Furthermore, dextran with different numbers of PNIPAAm chains was synthesized for comparison. It revealed that at 37 degrees C, the temperature above the LCST, the conjugation of Hb to the copolymer Dex-g-PNIPAAm could improve the stability of Hb which increased with the number of PNIPAAm chains.

Abstract:A new method was reported to modify layered zirconium phosphate (ZrP) with thermoresponsive polymer PNIPAM (poly N-isopropylacrylamide). PNIPAM was proved to be covalently grafted onto ZrP. (60)Co γ-rays irradiation produced peroxide groups on the surface which, upon heating, initiated free radical polymerization and subsequent attachment of PNIPAM.

Abstract:A heme protein hemoglobin (Hb) was reacted with preexfoliated layered α-zirconium phosphate (α-ZrP) platelets. An X-ray diffraction (XRD) pattern of small range showed that the exfoliated α-ZrP platelets reassembled after the addition of Hb molecules, with the protein intercalated between the layers. UV–Vis and Fourier transform infrared (FTIR) spectra analysis displayed that no significant denaturation occurred to the intercalated protein. The bioactivity of Hb was also investigated by testing the electrochemical properties of the Hb/α-ZrP composite. Results showed that the intercalation of Hb into the layered material not only improved the thermal stability of Hb but also enhanced the direct electron transfer ability between protein molecules and electrode. The protein still showed bioactivity after treatment at a temperature as high as 85°C. A pair of well-defined redox peaks at approximately –0.37 and –0.32V was observed on the cyclic voltammograms (CVs) of the Hb/α-ZrP composite modified electrode, and the electrode reactions showed a surface-controlled process with a single proton transfer. The resultant biosensor constructed by the Hb/α-ZrP composite displayed an excellent response to the reduction of hydrogen peroxide (H2O2) with good reproducibility.

Abstract:For the purpose of preparing pillared phosphates, the intercalation of n-alkylamine and n-amino-alcohol in layered alpha-Zr(HPO4)(2) . H2O(alpha-ZrP) and its effect on the structure and properties of the layered phosphate have been investigated in detail, It was found that the delamination degree is related with the nature and the amount of amine added, and the extent of HPO42- hydrolysis is dependent both on the basicity of the amine and the delamination degree, The delaminated sheets are metastable, and they may recover their ordered arrangement and experience phase transformations in air, The XRD results and the approximate calculation of the model structures of the intercalates show that a bilayer orientation of the n-alkylamine and n-amino-alcohol molecules is formed when the intercalates are saturated, After long time exposure in air, the intercalates of methylamine and ethylamine will lose about 50% of the amine and transfer to a monolayer orientation, but the intercalates of n-propylamine, n-butylamine and n-animo-alcohol still keep a bilayer orientation.

Abstract:Layered compound zirconium bis(monohydrogenphosphate)(alpha-ZrP) intercalated with rare earth complex Eu(DBM)(3)phen was prepared. The pre-intercalation of p-methoxyaniline into alpha-ZrP makes the interlayer separation large enough to exchange PMA with europium complex, thus, the luminescent assembly was prepared. This was confirmed by X-ray diffraction, UV-visible spectra and elemental analysis. The fluorescence spectra and lifetime of the assembly were also presented.

Abstract:Affinity membranes were prepared with poly(vinylidene fluoride) (PVDF) hollow fibers or flat membranes as the matrix, 1,6-hexanediamine (HDA) as the space arm, and amino acid—l-phenylalanine (l-Phe), l-tryptophane (l-Trp) and l-histidine (l-His) as the hydrophobic ligands, respectively. X-ray photoelectron spectroscopy (XPS) analysis results indicated that the contents of oxygen and nitrogen increased from 5.6% to 16.5% and 0.6% to 3.0% on the membrane surface after l-Phe modification, respectively, while fluoride content decreased from 38.3% to 23.0%. The effects of operation parameters such as pH value and ionic strength, on the human γ-globulin (HGG) adsorption capacity of the affinity membranes were investigated in a batch system. The adsorption phenomena appeared to follow a typical Langmuir adsorption isotherm under the optimal condition, where the maximum adsorption capacity (qm) of l-Phe affinity membrane for γ-globulin was 0.318mg/cm2 membrane and the equilibrium constant (Kd) value was found to be 0.453mg/mL solution from the Scatchard plot. Forty hollow fibers with a membrane area of 0.0188m2 in the membrane module adsorb 21mg HGG from 10mL human plasma with a purity of 83.9% in a single-pass mode. And the affinity membrane represented good stability throughout repeated adsorption–elution cycles.

Abstract:Amphiphilic block copolymers poly (ethylene glycol)-co-poly (glycidyl methacrylate) (PEG-co-P(GMA)) were synthesized via atom transfer radical polymerization (ATRP). Postpolymerization decoration of reactive epoxy groups in the P(GMA) block with N-(3-amino propyl) imidazole (API) resulted in a new kind of water soluble copolymer PEG-P (GMA-API). The weight-average molecular weight and polydispersity of PEG-P(GMA-API) were determined to be 17175 and 1. 08 with H-1-NMR and GPC,respectively. The resulted copolymers could self-assemble into spherical nano-micelles directly in an aqueous medium. The critical micellization concentration (CMC) of the micelles was 4 x 10(-2) g/L, which was significantly influenced by amino and imidazole groups. Dynamic light scattering (DLS) study showed the diameter of the micelles was about 40 nm and the micelle surface was always positively charged when the pH of the solution increased from 4 to 9. The resulted nano-micelles had the capacity to encapsulate hemoglobin (Hb) in aqueous solution with the encapsulation efficiency of 32. 4 wt% at pH 7. 4. The features of Hb-based micelles were spherical with a diameter of 40 nm, which was larger than that of blank micelles (30 nm). The Hb-based micelles displayed a reversible change in the UV-Visible absorption spectrum from the deoxy form to the oxygen-binding one, in response to the partial oxygen pressure of the atmosphere. The characteristic absorption peaks of the encapsulated Hb in different gas-binding states showed no significant change comparing with those of the free Hb. The Hb encapsulated PEG-P (GMA-API) nano-micelles showed the potential application as artificial oxygen carriers for transfusion.

Abstract:Layered inorganic materials are widely used as fillers in nanocomposites due to their unique properties such as mechanical performance, thermal properties, flame resistance, barrier properties, corrosion resistance, catalytic activity, and multifunctionality. Among them, layered α-zirconium phosphate (α-ZrP: Zr(HPO 4 ) 2 H 2 O) is one of the representative inorganic materials and unique properties with P–O–H groups that can exchange inorganic and organic cations and intercalate basic molecules. This paper deals with the use of this compound as an adsorbent for heavy metal cations in wastewater. Samples were prepared by hydrothermal treatment at various Zr/P ratio, water amounts, and temperatures. Furthermore, an attempt was made to widen the interlayer distance of the sample using n-butylamine. The α- zirconium phosphate prepared at 160°C for 20 h with Zr/ P = 1/2 adsorbed the most amount of metal ions. Furthermore, interlayer amine modification was effective. Then, the behaviour of the adsorption process was observed by varying the time of adsorption from 1 h to 50 h, in order to clarify the change in adsorption of the samples over time.

Abstract:A new inorganic–organic nanocomposite of polyacrylamide (PAM) and gamma-zirconium phosphate (γ-ZrP) was prepared by intercalative polymerization. Intercalation of acrylamide (AM) monomer in γ-ZrP was investigated by sonochemical and refluxing methods. High-intensity ultrasound does not induce the polymerization of AM but enhances greatly the intercalation rate. On the other hand, ultrasound also enhanced hydrolyzation of –CONH2 and shrinkage of PAM gel. The interlayer distance of AM-intercalated γ-ZrP is 16.4 Å. The polymerization and intercalation of AM occurred nearly at the same time by refluxing method. The same γ-ZrP/PAM nanocomposites were obtained with (NH4)2S2O8 treatment.
Preparation and Utilization of a Highly Discriminative Absorbent Imprinted with Fetal Hemoglobin

Abstract:Development in hemoglobin-based oxygen carriers (HBOCs) that may be used as alternatives to donated blood requires an extensive supply of highly pure hemoglobin (Hb) preparations. Therefore, it is essential to fabricate inexpensive, stable and highly selective absorbents for Hb purification. Molecular imprinting is an attractive technology for preparing such materials for targeted molecular recognition and rapid separations. In this case study, we developed human fetal hemoglobin (HbF)-imprinted polymer beads through the fusion of surface imprinting and Pickering emulsion polymerization. HbF was firstly covalently coupled to silica nanoparticles through its surface-exposed amino groups. The particle-supported HbF molecules were subsequently employed as templates for the synthesis of molecularly imprinted polymers (MIPs) with high selectivity for Hb. After removing the silica support and HbF, the resulting MIPs underwent equilibrium and kinetic binding experiments with both adult Hb (HbA) and HbF. These surface-imprinted MIPs exhibited excellent selectivity for both HbA and HbF, facilitating the one-step isolation of recombinant Hb from crude biological samples. The saturation capacities of HbA and HbF were found to be 15.4 and 17.1 mg/g polymer, respectively. The present study opens new possibilities for designed resins for tailored protein purification, separation and analysis.

Abstract:High-intensity ultrasound was applied to the intercalation process of a-zirconium phosphate (alpha-ZrP) with alkylamine, which can increase the reaction rate obviously. In this paper, methylamine was chosen as a colloidal reagent for the exfoliation of a-ZrP, the intercalated compound of hexadecyltrimethyl ammonium bromide(C16) with alpha-ZrP, which could be abbreviated to OZrP, was prepared by "exfoliation-intercalation method", and its structure, morphology and hydrophobic property were characterized and investigated by XRD, TEM, SEM, IR. The results indicated that the OZrP retained layered structure with the d-spacing of 2.9nm. Interestingly, the OZrP presented well hydrophobic property, which may be used as a promising precursor for preparing polymer/alpha-ZrP nanocomposites.

Abstract:Assorted trypsin immobilization trials were thoroughly conducted and compared to identify an efficient enzyme immobilization at the galleries of amine preintercalated γ-zirconium phosphate. That was, (a) direct trypsin binding by physical adsorption, (b) coupling of enzyme to glutaraldehyde (GA) activated carrier, (c) trypsin immobilization by GA crosslinking, (d) incorporating bovine serum albumin (BSA) as a spacer during the immobilization by covalently coupling to GA activated carrier or by covalent trypsin crosslinking with GA. We reported here that covalent immobilization of trypsin by using GA as a crosslinking agent and BSA as a spacer was superior to other immobilization strategies investigated. In this study, the experimental parameters in each step of the immobilization procedure, such as the concentration of the preintercalation agent, the crosslinking agent, the spacer and trypsin, etc., were investigated to improve the efficiency of trypsin immobilization. A reasonable enzymatic activity recovery at 75.6% and improved thermal stability were achieved. The optimum pH value and optimum temperature of immobilized enzyme were determined.
Layered assembly of PEGylated graphene oxide and Mg-Al layered double hydroxide nanosheets with superior adsorption capacity and selective isolation of hemoglobin

Abstract:This study developed a novel organic-inorganic hybrid composite, shortly as GO-PEG-LDHs, by self-assembly of exfoliated Mg-Al layer double hydroxide (LDHs) on the polyethylene glycol (PEG) grafted graphene oxide (GO) to achieve the selective adsorption of hemoglobin (Hb). The prepared GO-PEG-LDHs has a hierarchical structure with a homogeneous loading of exfoliated LDHs nano-sheets on its surface. The adsorption test reveals that GO-PEG-LDHs exhibits an adsorption efficiency of 95.03% for Hb and 3.45% for bovine serum albumin (BSA). The adsorption of Hb follows the Langmuir model, with an ultrahigh adsorption capacity of 55248.6 mg/g, which is higher than any previously reported materials. Meanwhile, the adsorbed Hb can be efficiently recovered through elution with a 50 mM Tris-HCl buffer, with an elution efficiency of 80.77%. Circular dichroism (CD) spectra indicate no conformational change for Hb during the process of adsorption/desorption. Furthermore, the composite demonstrates the ability to selectively isolate Hb in the presence of interfering protein BSA, indicating its potential for practical applications.
Hemoglobin on phosphonic acid terminated self-assembled monolayers at a gold electrode: immobilization, direct electrochemistry, and electrocatalysis.

Abstract:Phosphonic acid (-PO3H2) terminated self-assembled monolayers (SAMs) on a gold surface were used as a functional interface to immobilize hemoglobin (Hb). In situ surface-enhanced infrared absorption spectroscopy (SEIRAS) measurements show that Hb immobilization is a sluggish process due to formation of multilayer Hb structures on the PO3H2-terminated SAMs. as revealed by ellipsometry, atomic force microscopy (AFM). and cyclic voltammetry (CV). In the multilayered Hb film the innermost Hb molecules can directly exchange electrons With the electrode, whereas Hb beyond this layer communicates electronically With the electrode via protein-protein electron exchange. In ad clition, electrochemical measurements indicate that immobilization of Hb on the PO3H2-terminated SAMs is not driven by the electrostatic interaction, but likely by hydrogen-bonding interaction. The immobilized Hb molecules show excellent bioelectrocatalytic activity towards hydrogen peroxide, that is, the PO3H2-terminated SAMs are promising for construction of third-generation biosensors.

Abstract:Direct electrochemistry and thermal stability of hemoglobin (Hb) immobilized on a nanometer-sized zirconium dioxide (ZrO2) modified pyrolytic graphite (PG) electrode were studied. The immobilized Hb displayed a couple of stable and well-defined redox peaks with an electron transfer rate constant of (7.90 +/- 0.93)s(-1) and a formal potential of -0.361 V (-0.12 V versus NHE) in 0.1M pH 7.0 PBS. Both nanometer-sized ZrO2 and dimethyl sulfoxide (DMSO) could accelerate the electron transfer between Hb and the electrode. Spectroscopy analysis of the Hb/ZrO2/DMSO film showed that the immobilized Hb could retain its natural structure. This modified electrode showed a high thermal stability up to 74 degrees C and an electrocatalytic activity to the reduction of hydrogen peroxide (H2O2) without the aid of an electron mediator. The electrocatalytic response showed a linear dependence on the H2O2 concentration ranging from 1.5 to 30.2 microM with a detection limit of 0.14 microM at 3sigma. The apparent Michaelis-Menten constant KMapp for H2O2 sensor was estimated to be (0.31 +/- 0.02) mM, showing a high affinity.
A novel PEG-mediated approach to entrap hemoglobin (Hb) within ZIF-8 nanoparticles: Balancing crystalline structure, Hb content and functionality

Abstract:Hemoglobin (Hb)-based oxygen carriers are investigated as a potential alternative or supplement to regular blood transfusions, particularly in critical and life-threatening scenarios. These include situations like severe trauma in remote areas, battlefield conditions, instances where blood transfusion is not feasible due to compatibility concerns, or when patients decline transfusions based on religious beliefs. This study introduces a novel method utilizing poly(ethylene glycol) (PEG) to entrap Hb within ZIF-8 nanoparticles (i.e., Hb@ZIF-8 NPs). Through meticulous screening, we achieved Hb@ZIF-8 NPs with a record-high Hb concentration of 34 mg mL-1. These NPs, sized at 168 nm, displayed exceptional properties: a remarkable 95 % oxyhemoglobin content, excellent encapsulation efficiency of 85 %, and resistance to Hb oxidation into methemoglobin (metHb). The addition of PEG emerged as a crucial factor amplifying Hb entrapment within ZIF-8, especially at higher Hb concentrations, reaching an unprecedented 34 mg mL-1. Importantly, PEG exhibited a protective effect, preventing metHb conversion in Hb@ZIF-8 NPs at elevated Hb concentrations.

Abstract:Stable films of clay and hemoglobin (Hb) were assembled layer-by-layer on various solid substrates by alternate adsorption of negatively charged clay platelets from their aqueous dispersions and positively charged Hb from pH 4.5 buffers. Cyclic voltammetry (CV), quartz crystal microbalance and UV-vis spectroscopy were used to monitor film growth. CV of {clay/Hb}(n) films on pyrolytic graphite electrodes showed a pair of well-defined, nearly reversible peaks at about -0.27 V versus saturated calomel electrode (-0.02 V vs normal hydrogen electrode) at pH 5.5, characteristic of the HbFe(III)/Fe-II redox couples. Although the amount of Hb adsorbed in each bilayer was essentially the same, the fraction of electroactive Hb decreased with the number of clay/Hb bilayers (n). Electroactivity of Hb extended to six clay/Hb bilayers. The Soret absorption band of Hb in {clay/Hb}(6) films showed that Hb retained its secondary structure similar to its native state in the medium pH range. A Bragg peak in X-ray diffraction for {clay/Hb}(20) films suggested a partly ordered, layered structure of the films, but without full Hb intercalation. {Clay/Hb}(6) film electrodes catalyzed the electrochemical reduction of trichloroacetic acid, oxygen, and hydrogen peroxide.
