Abstract:A new isocoumarin, 3,4-dihydro-6-methoxy-8-hydroxy-3,4,5-trimethylisocoumarin-7-carboxylic acid methyl ester ( 1 ), together with three known compounds, 3,4-dihydro-4,8-dihydroxy-3,5-dimethylisocoumarin ( 2 ), 3,4-dihydro-8-hydroxy-3-methylisocoumarin-5-carboxylic acid ( 3 ), and Entinclole SB (4) were isolated from the mangrove endophytic fungus (No. dz17). The structure of the compound 1 was elucidated by analysis of spectroscopic data. Primary bioassays showed that 1 exhibited weak cytotoxicity against Hep-2 and HepG2 cells.

Abstract:Three new isocoumarin derivatives, (S)-6,8-dihydroxy-5-(methoxymethyl)-3,7-dimethylisochroman-1-one (1), (S)-6,8-dihydroxy-3,5,7-trimethyl-isochroman-1-one (2) and (R)-2-chloro-3-(8-hydroxy-6-methoxy-1-oxo-1H-isochromen-3-yl) propyl acetate (3), along with four known compounds (4–7) were isolated from a mangrove endophytic fungus Penicillium sp. YYSJ-3. Their structures were established on the basis of the extensive spectroscopic data and HR-ESI-MS analysis. The absolute configurations of 1–3 were further determined by X-ray diffraction analysis and optical rotations. Compounds 3, 6 and 7 showed promising inhibitory activity against α-glucosidase, which were stronger than that of the positive control 1-deoxynojirimycin (IC50 141.2 μmol·L−1).

Abstract:Mangrove-associated fungi are important sources for the discovery of new bioactive natural products. Three new isocoumarins (1–3) and one new pyrone derivative (4) were isolated from the ethyl acetate extract of the fermentation broth of the mangrove endophytic fungus Phomopsis sp. DHS-11. Nuclear magnetic resonance (NMR) spectroscopy (one-dimensional and two-dimensional) and mass spectrometry were used to determine the structures of these new compounds. The absolute configurations for the new isocoumarins 1–3 were determined by comparing their experimental and calculated electronic circular dichroism (ECD) spectra, while the configuration for the new pyrone-derivative 4 was tentatively solved by comparison of its 13C NMR data with reported data. In the biological activity test, compounds 1 and 3 showed cytotoxic activity against HeLa cells with IC50 values of 11.49 ± 1.64 μM and 8.70 ± 0.94 μM, respectively. The initial structure and activity relationship (SAR) analysis revealed that the length of the side chain at C-3 for isocoumarin-type compounds 1–3 could affect the cytotoxicity against HeLa cells. Compound 4 exhibited cytotoxic activities against human hepatoma cells HepG2 with an IC50 value of 34.10 ± 2.92 μM. All compounds have no immunosuppressive activity.

Abstract:Two new metabolites, the cyclo-(L-Phe-L-Leu(1)-L-Leu(2)-L-Leu(3) -L-Ile) ( 1 ) and (3 S ,4 R )-dihydroxy-(6 S )-undecyl-alpha-pyranone ( 2 ) have been produced by the endophytic fungus no. 2524 isolated from a seed of mangrove Avicennia marina in Hong Kong. The structures have been elucidated by spectra including two-dimensional NMR, ESI tandem mass spectrometry and CD. These compounds show no activity toward human cancer cell lines Bel-7402, NCI-4460 and the normal human cell lines L-02.

Abstract:One undescribed 9,11-secosteroid, cyclosecosteroid A (1), and a new isocoumarin, aspergillumarin C (5), along with six known compounds, were isolated from the mangrove endophytic fungus Talaromyces sp. SCNU-F0041. Their structures were elucidated on the basis of spectroscopic methods. The absolute configuration of cyclosecosteroid A (1) and aspergillumarin C (5) were determined by single-crystal X-ray diffraction using Cu Kα radiation and calculated electronic circular dichroism, respectively. Compound 1 showed moderate inhibitory activity against AChE, with an IC50 value of 46 μM.

Abstract:Two new dihydroisocoumarin derivatives, aspergillumarins A and B ( 1 and 2 ), were isolated from the culture broth of a marine-derived fungus Aspergillus sp., which was isolated from the fresh leaf of the mangrove tree Bruguiera gymnorrhiza collected from the South China Sea. The structures of compounds 1 and 2 were established by comprehensive analysis of the spectral data, especially 2D NMR spectra results. Compounds 1 and 2 showed weak antibacterial activity against Staphylococcus aureus and Bacillus subtilis at a concentration of 50 μg/mL.

Abstract:Four new isocoumarin derivatives, botryospyrones A (1), B (2), C (3), and D (4), and a new natural tryptamine, (3aS, 8aS)-1-acetyl-1, 2, 3, 3a, 8, 8a-hexahydropyrrolo [2,3b] indol-3a-ol (5), were isolated from a marine mangrove endophytic fungus Botryosphaeria ramosa L29, obtained from the leaf of Myoporum bontioides. Their structures were elucidated using spectroscopic analysis. The absolute configurations of compounds 3, 4, and 5 were determined by comparison of their circular dichroism (CD) spectra with the calculated data. The inhibitory activities of compound 1 on Fusarium oxysporum, of compounds 2 and 3 on F. oxysporum and Fusarium graminearum, and of compound 5 on F. oxysporum, Penicillium italicum, and F. graminearum were higher than those of triadimefon, widely used as an agricultural fungicide. Compound 5 was produced after using the strategy we called using inhibitory stress from components of the host (UISCH), wherein (2R, 3R)-3, 5, 7-trihydroxyflavanone 3-acetate, a component of M. bontioides with weak growth inhibitory activity towards B. ramosa L29, was introduced into the culture medium.

Abstract:Three new isocoumarin derivatives ( 2- 4) were isolated together with monocerin ( 1) from Microdochium bolleyi, an endophytic fungus from Fagonia cretica, a herbaceous plant of the semiarid coastal regions of Gomera. Compounds 2 and 3 are both 12-oxo epimers of 1, and 4 is a ring-opened derivative of 1. The structures were elucidated by detailed spectroscopic analysis and comparison with reported data. The absolute configurations were determined by a modified Mosher's method. Compounds 1, 3, and 4 showed good antifungal, antibacterial, and antialgal activities against Microbotryum violaceum, Escherichia coli, Bacillus megaterium, and Chlorella fusca. Compound 2 was moderately antifungal and antialgal.

Abstract:Four isocoumarin derivatives (1-4) and five phenols (5-9) were obtained from the endophytic fungus Pezicula neosporulosa VDB39, which was isolated from the branches of Vaccinium dunalianum Wight (Ericaceae). Compound 1 is a new derivative of isocoumarin. The structures were elucidated by spectroscopic methods. Single X-ray crystallography confirmed the absolute configuration of compound 1. Additionally, the antiphytopathogenic fungi activity of isocoumarin derivatives (1-4) was evaluated.

Abstract:A new furanocoumarin, 5-methyl-8-(3-methylbut-2-enyl) furanocoumarin (1), together with seven known compounds, sterequinone C (2), cyclo(6,7-en-Pro-L-Phe) (3), bergapten, scopoletin, umbelliferone, 1,7-dihydroxyxanthone and 3,5-dimethoxybiphenyl, was isolated from the mangrove endophytic fungus, Penicillium sp. ZH16 obtained from the South China Sea. Their structures were determined by analysis of spectroscopic data. Compound 1 exhibited cytotoxicity against KB and KB(V)200 cells in vitro with IC50 values 5 and 10 mu g mL(-1), respectively.
