Abstract:A new dolabellane diterpene 1, named as hydroclathrol, has been isolated from the alga Hydroclathrus tenuis. Its structure has been determined on the basis of spectral analysis as 1, 4, 8-trimethyl-14-isopropyl-bicyclo [9, 3, 0]-4(Z), 8(E), 11(Z)-tetradeca triene-6-ol.

Abstract:Twelve new members (1-12) of the dolabellane family, co-occurring with three related known diterpenoids (13-15), were isolated from the Xisha soft coral Clavularia viridis. Their structures were determined by extensive spectroscopic analysis, modified Mosher's method, and X-ray diffraction analysis. Clavuperoxylides A (3) and B (4) represent the first examples of dolabellanes containing peroxyl groups, especially the novel peroxide bridge in 4, whereas clavufuranolides A-C (9-11) are the first example of dolabellane diterpenoids comprising a tetrahydrofuran ring. The possible biogenetic relationship of all the isolates was proposed. In bioassay, several compounds exhibited considerable cytotoxicity against A549 and P388 cell lines. Compound 7 exhibited inhibitory activity against protein tyrosine phosphatases 1B (PTP1B), an anti-diabetic target, representing the first report of PTP1B inhibitory activity for dolabellane diterpenoids.

Abstract:Ternifolide A (1), a new diterpenoid featuring a unique 10-membered lactone ring formed between C-6 and C-15, along with ternifolide B (2), a nor-diterpenoid, and ternifolide C (3) were isolated from the leaves of Isodon ternifolius. Both H-8 and H-9 being alpha-orientations in compound 1 were found for the first time. The absolute configurations of 1 and 3 were confirmed by X-ray diffraction study. Compounds 1 and 3 were evaluated for their cytotoxicity.

Abstract:The acetone extract of Dicranopteris dichotoma afforded two new tetranorclerodanes, 18-hydroxyaylthonic acid (1) and 18-oxo-aylthonic acid (2), and four new clerodane-type diterpene glycosides, (6S,13S)-6-O-[6-O-acetyl-beta-d-glucopyranosyl-(1-->4)-alpha-l-rhamnopyranosyl]cleroda-3,14-dien-13-ol (3), (6S,13S)-6-O-[4-O-acetyl-beta-d-glucopyranosyl-(1-->4)-alpha-l-rhamnopyranosyl]cleroda-3,14-dien-13-ol (4), 6-O-[6-O-acetyl-beta-d-glucopyranos-yl-(1-->4)-alpha-l-rhamnopyranosyl]-(13E)-cleroda-3,13-dien-15-ol (5), and 6-O-[beta-d-glucopyranosyl]-(1-->4)-alpha-l-rhamnopyranosyl-(13E)-cleroda-3,13-dien-15-ol (6), together with two known compounds, aylthonic acid (7) and (6S,13S)-cleroda-3,14-diene-6,13-diol (8). The structures of these new compounds were established by a combination of 1D and 2D NMR techniques, MS, and acid hydrolysis. Compound 8 showed modest anti-HIV-1 activity.

Abstract:Five undescribed dolabellane-type diterpenoids (1-5), together with three related known ones (6-8), were isolated from the soft coral Clavularia viridis. Their structures and stereochemistry were elucidated by extensive spectroscopic analysis and NMR calculation with DP4+ probability analysis. The absolute configurations of 1 and 5 were unambiguously determined by X-ray crystallographic analysis. A plausible biosynthetic connection between undescribed compounds 1-5 was proposed.

Abstract:A phytochemical investigation of the leaves of Callicarpa macrophylla led to the isolation of five new diterpenoids (1-5), macrophypenes A-E and nine known analogues (6-14). The structures of 1-5 were established on the basis of extensive analysis of NMR spectroscopic data, X-ay diffraction data, and experimental and calculated electronic circular dichroism spectra. Compound 1 is a spiroditerpenoid with a novel skeleton, and compound 5 is a rare ent-abietane diterpenoid possessing a peroxide bridge. Compounds 1, 5-7, and 11-14 stimulate nerve growth factor mediated neurite outgrowth from PC12 cells.

The structure of the new compound, as well as its absolute configuration, was established by means of spectroscopic data analyses, including 2D NMR spectroscopy and X-ray structural analysis.

Abstract:A new caryolane sesquiterpene, 1-methoxycaryol-9-one (1), along with eight known compounds (29), was isolated from the whole plant of Heteropappus altaicus. The structures were elucidated on the basis of comprehensive 1D- and 2D-NMR analysis and high-resolution mass spectrometry. The C-13-NMR data of compound 2 are reported for the first time.

Abstract:LC-MS/MS-based molecular networking annotation coupled 1H NMR detection allowed the depiction of the soft coral Clavularia viridis to produce a profile of dolabellane-type diterpenoids. Chromatographic separation of the EtOAc fraction resulted in the isolation of 12 undescribed dolabellane-type diterpenoids, namely, clavirolides J-U (1-12). Their structures were characterized by the extensive analysis of the spectroscopic data, including the calculated ECD and X-ray diffraction for the configurational assignments. Clavirolides J-K are characterized by a 1,11- and 5,9-fused tricyclic tetradecane scaffold fused with a α,β-unsaturated-δ-lactone, and clavirolide L possesses a 1,11- and 3,5-fused tricyclic tetradecane scaffold, which extend the dolabellane-type scaffolds. Clavirolides L and G showed significant inhibition against HIV-1 without RT enzyme inhibition, providing additional non-nucleosides with different mechanisms from efavirenz.

Abstract:Two highly oxygenated nor-clerodane diterpenoids, crocleropenes A and B (1 and 2), together with four known compounds (3-6) were isolated from the leaves and twigs of Croton caudatus. Their structures were elucidated by combination of extensive spectroscopic analysis and electronic circular dichroism (ECD) calculations. 1 and 2 represent the first examples of nor-clerodane-3,5(10)-diene diterpenoids. Compounds 1 and 2 exhibited weak cytotoxicity in vitro against MCF-7 cancer cells with IC50 values of 35.8 and 40.2 μM, respectively. [Formula: see text].

Abstract:A series of cyclohexylethanoids, including two new compounds, 1-hydroxy-1-(8-palmitoyloxyethyl) cyclohexanone (1) and 5-O-butyl cleroindin D (2), together with five known ones, rengyolone (3), cleroindin C (4), cleroindin B (5), rengyol (6) and isorengyol (7), were isolated from the leaves of Clerodendrum trichotomum. Their structures were elucidated on the basis of spectroscopic analyses and by comparison of their NMR data with those in the literatures. Abstract:A novel diterpenoid with an unprecedented carbon skeleton, rhodomollanol A (1), and a new grayanane diterpenoid, rhodomollein XXXI (2), were isolated from the leaves of Rhododendron molle. Their structures were elucidated using comprehensive spectroscopic methods and single-crystal X-ray diffraction. Compound 1 possesses a unique cis/trans/trans/cis/cis-fused 3/5/7/5/5/5 hexacyclic ring system featuring a rare 7-oxabicyclo[4.2.1]nonane core decorated with three cyclopentane units. The plausible biosynthetic pathway for 1 was proposed. Compound 1 exhibited moderate PTP1B inhibitory activity.

Abstract:Phytochemical investigation of the leaves and twigs of Croton yanhuii led to the isolation of seven highly modified nor-clerodane diterpenoids (1-7), including three new ones, croyanoids A-C (1-3), along with four known analogues (4-7). Compound 1 incorporates a 5,12-epoxy ring, forming a unique cage-like, 6/6/6/5-fused tetracyclic ring system. Their structures were established by extensive spectroscopic analysis, and the absolute configurations of 1-4 were determined by a combination of circular dichroism (CD) analysis and single-crystal X-ray diffraction. All compounds were tested in an array of bioassays, but were inactive. Crotoeurin A (7), a nor-clerodane dimer with a high yield of 0.2‰ isolated in current study, was considered as a chemotaxonomic marker for this species.

Compounds 1 and 2 exhibited an agonistic effect on pregnane X receptor at 10 mu M.

Abstract:Eight previously undescribed diterpenoids, crolaevinoids A-H, including two halimanes, four clerodanes, and two laevinanes, along with six known analogues were isolated from the twigs of Croton laevigatus. The structures of the previously undescribed were elucidated by spectroscopic analysis, and their absolute configurations were determined by combination of a single crystal X-ray diffraction and CD analysis (exciton chirality and Rh-2(OCOCF3)(4)-induced methods). Crolaevinoids A and B represent the first halimane diterpenoids with a unique lactone bridge between C-12 and C-17. All compounds were evaluated for their inhibitory effects on the nitric oxide (NO) production induced by lipopolysaccharide (LPS) in RAW264.7 macrophage cells. Furocrotinsulolide A and 3,4,15,16-diepoxy-cleroda-13(16),14-diene-12,17-olide exhibited pronounced inhibition of NO production with IC50 values of 10.4 +/- 0.8 and 6.0 +/- 1.0 mu M, respectively, being more potent than the positive control, quercetin (IC50 = 13.1 +/- 1.9 mu M).
