Abstract:Two new clerodane diterpenoids ( 1 and 2 ), a new pyran-2-one derivative ( 3 ), along with five known compounds ( 4 ‒ 8 ), were isolated from Croton crassifolius . Notably, crassifolin X ( 1 ) is a novel clerodane diterpenoid, characterized with a peculiar δ -lactone core being formed between C-1 and C-4. Their structures, including absolute configurations, were established on the basis of spectroscopic methods (UV, IR, HRESIMS and NMR), and circular dichroism experiments. In addition, all compounds were evaluated for their anti-neuroinflammatory activities based on the expression of TNF- α and IL-6 levels on LPS-induced BV2 cells, and compounds 1 ‒ 3 and 5 showed potential anti-neuroinflammatory activity.

Abstract:Eight previously undescribed diterpenoids, crolaevinoids A-H, including two halimanes, four clerodanes, and two laevinanes, along with six known analogues were isolated from the twigs of Croton laevigatus. The structures of the previously undescribed were elucidated by spectroscopic analysis, and their absolute configurations were determined by combination of a single crystal X-ray diffraction and CD analysis (exciton chirality and Rh-2(OCOCF3)(4)-induced methods). Crolaevinoids A and B represent the first halimane diterpenoids with a unique lactone bridge between C-12 and C-17. All compounds were evaluated for their inhibitory effects on the nitric oxide (NO) production induced by lipopolysaccharide (LPS) in RAW264.7 macrophage cells. Furocrotinsulolide A and 3,4,15,16-diepoxy-cleroda-13(16),14-diene-12,17-olide exhibited pronounced inhibition of NO production with IC50 values of 10.4 +/- 0.8 and 6.0 +/- 1.0 mu M, respectively, being more potent than the positive control, quercetin (IC50 = 13.1 +/- 1.9 mu M). Abstract:Phytochemical investigation of the leaves and twigs of Croton yanhuii led to the isolation of seven highly modified nor-clerodane diterpenoids (1-7), including three new ones, croyanoids A-C (1-3), along with four known analogues (4-7). Compound 1 incorporates a 5,12-epoxy ring, forming a unique cage-like, 6/6/6/5-fused tetracyclic ring system. Their structures were established by extensive spectroscopic analysis, and the absolute configurations of 1-4 were determined by a combination of circular dichroism (CD) analysis and single-crystal X-ray diffraction. All compounds were tested in an array of bioassays, but were inactive. Crotoeurin A (7), a nor-clerodane dimer with a high yield of 0.2‰ isolated in current study, was considered as a chemotaxonomic marker for this species.

Abstract:Fifteen new labdane-type diterpenoids, sublyratins A-O (1-15), along with four known analogues (16-19) were isolated from the aerial parts of Croton sublyratus. Their structural assignments were challenging due to the stereoisomeric features evident and were achieved by analyzing comprehensively the spectroscopic data and electronic circular dichroism spectra and using X-ray crystallographic analysis. Compounds 9 and 16-18 displayed cytotoxic activity against the HL-60 cell line with IC50 values of 1.5-2.8 μM.

Abstract:The phytochemical investigation of the leaves and twigs of Croton yunnanensis resulted in the isolation of eight new clerodane furanoditerpenoids, named croyunfuranoids A-H（1-8）, along with three known analogs（9-11）. The structures of these compounds were elucidated using spectroscopic analyses, and their absolute configurations were determined through a combination of electronic circular dichroism（ECD） calculations and single-crystal X-ray diffraction. Notably, Croyunfuranoid D（4） is identified as a rare 18,19-dinor-clerodane diterpenoid. Additionally, the structure of a previously reported diterpenoid, crotonyunnan B, was revised.All isolated compounds were evaluated for their inhibitory activities on nitric oxide（NO） production in LPS-induced RAW 264.7 macrophages. Among them, compounds 5 and 6 demonstrated significant inhibitory effects, with IC 50 values of 20.33 ± 2.31 and 22.80 ±1.31 μmol·L -1 , respectively.

Abstract:Croton sublyratus (Euphorbiaceae) is a traditional medicinal plant used by the Thai populace to treat helminthic infections and dermatologic conditions. In present study, eight new labdane‐type diterpenoids, crotonoids A–H (1–8) and one known analogue (9) were isolated from the aerial parts of C. sublyratus. Compounds 6 and 7 belong to the rare class of 14,15‐dinor‐labdane diterpenoids. Compound 8 exhibited a rare 14,15,17‐trinor‐labdane skeleton. The structures of all these diterpenoids were elucidated by spectroscopic data analysis, electronic circular dichroism calculations, and single‐crystal X‐ray diffraction analysis. Compound 9 exhibited moderate anti‐inflammatory activity via the inhibition of NO production in lipopolysaccharide‐induced RAW 264.7 cells.

Abstract:Laeviganoids A-T (1-20), 20 new ent-clerodane-type diterpenoids featuring a 2-furanone (1-3) or a furan (4-20) ring, as well as six analogues (21-26), were isolated from the roots of Croton laevigatus. Their structures were determined by spectroscopic data analysis, experimental electronic circular dichroism measurements, and X-ray crystallographic studies. Compounds 4-6, 16, 21-24, and 26 could influence the anti-inflammatory protumoral phenotype of macrophages. Among these compounds, 21 and 26 are the most potent, as evidenced by consistently downregulating the classic anti-inflammatory cytokine IL-10 and upregulating the classic pro-inflammatory cytokine TNF-α on the secretion level in RAW 264.7 cells.

Abstract:Three new clerodane type diterpenoids (2-4), together with one known analogues (1), were isolated from the tuberous roots of Tinospora sagittata (Oliv.) Gagnep. Compound 3 is an unusual clerodane diterpenoid with the carbonyl functionality at C-3, which represents the first example from this diterpenoid compounds class. The structures were established on the basis of spectroscopic data interpretation. The absolute configuration of 1 was determined for the first time by single-crystal X-ray diffraction analysis with CuKα irradiation. These isolates were evaluated for their cytotoxic activities against five human cancer cell lines and inhibitory activities on LPS-induced NO production in RAW264.7 cells.

The structures of these new compounds were established by a combination of 1D and 2D NMR techniques, MS, and acid hydrolysis. Compound 8 showed modest anti-HIV-1 activity.

Abstract:A phytochemical investigation of the leaves of Callicarpa macrophylla led to the isolation of five new diterpenoids (1-5), macrophypenes A-E and nine known analogues (6-14). The structures of 1-5 were established on the basis of extensive analysis of NMR spectroscopic data, X-ay diffraction data, and experimental and calculated electronic circular dichroism spectra. Compound 1 is a spiroditerpenoid with a novel skeleton, and compound 5 is a rare ent-abietane diterpenoid possessing a peroxide bridge. Compounds 1, 5-7, and 11-14 stimulate nerve growth factor mediated neurite outgrowth from PC12 cells.

Abstract:A new guaiane-type sesquiterpene glycoside, torilolone 8-O-beta-D-glucopyranoside (1), together with a known analogue compound, torilolone 11-O-beta-D-glucopyranoside (2), was isolated from the fruits of Daucus carota L. Their chemical structures were elucidated on the basis of MS, NMR spectroscopic analyses coupled with chemical degradation. The cytotoxic activities of two isolated compounds were evaluated against two human gastric cancer cell lines BGC-823 and AGS using MTT assay.

Abstract:As part of our continuing efforts toward the discovery of the biologically active diterpenoids from medicinal plants, six new C‐ring aromatized abietane diterpenoids, clerodenoids A–F (1–6), were isolated from Clerodendrum chinense. These C‐ring aromatized compounds mainly belonged to the rearranged abietane skeletons of 17(15→16)‐abeo‐abietane (2–4) and 17(15→16),18(4→3)‐diabeo‐abietane (5 and 6). Their structures were elucidated by extensive spectroscopic data, X‐ray crystallography, and ECD calculation. Abstract:A series of cyclohexylethanoids, including two new compounds, 1-hydroxy-1-(8-palmitoyloxyethyl) cyclohexanone (1) and 5-O-butyl cleroindin D (2), together with five known ones, rengyolone (3), cleroindin C (4), cleroindin B (5), rengyol (6) and isorengyol (7), were isolated from the leaves of Clerodendrum trichotomum. Their structures were elucidated on the basis of spectroscopic analyses and by comparison of their NMR data with those in the literatures.
