Abstract:Twelve new nortriterpenoids, namely wuweizidilactones J-P (1-7), schindilactones I-K (8-10), preschisanartanin N (11), and schisdilactone J (12), as well as 23 known analogues, were isolated from the leaves and stems of Schisandra chinensis. The absolute configurations of 1 and 11 were established by calculated electronic circular dichroism (ECD) spectra and that of 7 was confirmed by single-crystal X-ray diffraction. The absolute configurations of 2-6, 8, and 9 were determined by an empirical comparison of their experimental ECD spectra with that of 1. It is the first time that the ECD spectra of 18(13 -> 14)-abeoschiartane-, 18-norschiartane-, and schisanartane-type nortriterpenoids possessing an alpha,beta-unsaturated-gamma-lactone moiety are presented, this method providing an alternative means of obtaining absolute configurational assignments for these types of nortriterpenoids. All isolates were evaluated for their anti-acetylcholinesterase and anti-butyrylcholinesterase activities, and compounds 8, 13, 23, and 31 show anti-acetylcholinesterase activity at concentrations of 50 mu M, with 12.7, 10.7, 16.6, and 32.1% inhibition, respectively. Abstract:Norditerpenes are considered to be a common and widely studied class of bioactive compounds in plants, exhibiting a wide array of complex and diverse structural types and originating from various sources. Based on the number of carbons, norditerpenes can be categorized into C19, C18, C17, and C16 compounds. Up to now, 557 norditerpenes and their derivatives have been found in studies published between 2010 and 2023, distributed in 51 families and 132 species, with the largest number in Lamiaceae, Euphorbiaceae, and Cephalotaxaceae. These norditerpenes display versatile biological activities, including anti-tumor, anti-inflammatory, antimicrobial, and antioxidant properties, as well as inhibitory effects against HIV and α-glucosidase, and can be considered as an important source of treatment for a variety of diseases that had a high commercial value. This review provides a comprehensive summary of the plant sources, chemical structures, and biological activities of norditerpenes derived from natural sources, serving as a valuable reference for further research development and application in this field.

Abstract:Schicagenins A-C (1-3), three unprecedented nortriterpenoids characterized with a tetracyclic oxa-cage motif and C(9) side chain, were discovered from the leaves and stems of Schisandra chinensis. Their structures were determined on the basis of extensive spectroscopic analysis, and the absolute stereochemistries were established by single-crystal X-ray diffraction and CD experiments. A plausible biosynthetic pathway of 1-3 was also discussed.

Abstract:Several novel bioactive components isolated from Chinese medicinal plants will be presented. These include novel maytansinoid tumor inhibitors, some new ent-kaurane and rosane diterpenoids from Mallotus anomalus Meer et Chun (Euphorbiaceae), as well as novel insecticide, stemona alkaloids from Stemona parviflora C. H. Wright (Stemonaceae). Both are native plants of Hainan island, China. 2D NMR techniques such as mono and hetero-COSY, NOESY, COLOC as well as 1H-NMR line broadening effect were utilized for structure elucidation. The separation techniques, structure elucidations and bioassay results will be reported.

Abstract:Two new 18-norschiartane-type schinortriterpenoids, namely wuwezidilactones Q (1) and R (2) were isolated from the stems of Schisandra lancifolia. Their structures were determined on the basis of extensive spectroscopic analysis. The absolute configurations of the new compounds were further determined by ROESY and an empirical comparison of their experimental ECD spectra with literature.

Abstract:Eight known norditerpenoid alkaloids and one diterpenoid alkaloid were isolated from the mother liquid after seperating lappaconitine from the roots of Aconitum sinomontamum Nakai. They are lappaconitine (1), N-deacetyllappaconitine (2), lappaconine (3), lappaconidine (4), ranaconitine (5), N-deacetylranaconitine (6), 8-O-acetylexcelsine (7), excelsine (8) and septatisine (9). Five known norditerpenoid alkaloids and one diterpenoid alkaloid were isolated from the whole plant of A. racemulosum Franch var. pengzhouense w. J. Zhang et G. H. Chen. They are condelphine (10), isotalatizidine (11), 14-O-acetylvirescenine (12), virescenine (13), anthranoyllycoctonine (14) and atistinum hydrochloride (15). The structures of all these alkaloids were identified on the basis of spectral data and chemical properties.

Abstract:AIM: To study the alkaloids from the roots of Aconitum sinomontanum of Ranunculaceae. METHODS: The alkaloids were isolated using silica gel chromatography (column and chromatodron), and the structures were elucidated on the basis of IR, MS, 1HNMR, 13CNMR, 1H-1H COSY, HMQC and HMBC analysis. RESULTS: Twenty seven compounds have been isolated from the total alkaloids, two of which are sinomontanines D (1) and E (2). CONCLUSION: Sinomontanines D (1) and E (2) are new bisnorditerpenoid alkaloids.

Abstract:A chemical investigation of the 95% EtOH extract of the seeds of Cephalotaxus fortunei var. alpina led to the isolation of nine new cephalotane‐type norditerpenoids, ceforalides A—I (1—9), and two known analogues (10 and 11). Compounds 1—8 belong to a rare class of A‐ring‐contracted cephalotane‐type norditerpenoids, of which compound 8 incorporates a unique tetrasubstituted 2,5‐cyclohexadienone A‐ring. Compound 9 is a rare 13,14‐seco‐17‐nor‐cephalotane‐type diterpenoid. Their structures were determined based on NMR, HRESIMS, ECD, and X‐ray diffraction analysis. Biological tests revealed compounds 10 and 11 displayed moderate antimalarial activity. Nine new cephalotane‐type norditerpenoids, ceforalides A—I (1—9), and two known analogues (10 and 11) were isolated from Cephalotaxus fortunei var. alpina. Compound 8 incorporates a unique tetrasubstituted 2,5‐cyclohexadienone A‐ring. Compounds 10 and 11 displayed antimalarial activity.

Abstract:Tetracyclic norditerpene dilactones are an important class of terpenoids that have been isolated from both terrestrial and marine sources, typically from Podocarpus plants and from filamentous fungi. This class of molecules shares a common 6/6/6/5 tetracyclic ring skeleton, which possesses a densely oxygenated carbon framework and contiguous stereocenters. What's more challenging for synthetic chemists are the consecutive sp2-hybridized carbon centers, which exacerbates the strain/rigidity of the whole molecule. In addition, many of these molecules display promising biological activities, such as antitumor, insecticidal, anti-feedant, allelopathic, and antibiotic activities. The unique structures and interesting biological profiles of norditerpene dilactones have attracted considerable attention from synthetic chemists. Herein we summarize the synthetic efforts with respect to tetranorditerpene dilactones.

Abstract:A systematic phytochemical investigation on Abies forrestii afforded two new and 20 known compounds. Abieseconordines A and B (1 and 2) are the first two examples of norditerpenes with a novel 18-nor-5,10?:?9,10-disecoabietane skeleton. Their structures were established mainly by analysis of 1D- and 2D-NMR spectroscopic data. In addition, electronic circular-dichroism calculations and molecular-orbital analysis were utilized to confirm the absolute configuration of 1. Both compounds exhibited a potent effect in a bioassay inhibiting LPS-stimulated NO production in RAW264.7 macrophages.

Abstract:Fourteen new norditerpenoids (abiesanordines A–N, 1–14), including a novel podocarpene bearing a rare enolic structure (abiesanordine A, 1), were isolated from Abies georgei together with eight known ones. Their structures were determined mainly by detailed analysis of 1D and 2D NMR spectroscopic data including HSQC, DQF COSY, HMBC, and NOESY. All the isolates were tested for inhibitory activities against LPS-induced NO production in RAW264.7 macrophages, abiesanordine I (9) showed the strongest activity with the IC50 value of 17.0μg/mL. Furthermore, it exhibited no cytotoxicity against RAW264.7 macrophages under the concentration of 50μg/mL.
