Abstract:Mixed-mode chromatography is a new kind of chromatography technology for bioseparation.The special ligand combines two or more interaction modes,commonly hydrophobic and electrostatic interactions.Mixed-mode chromatography can directly capture target protein from moderate-ionic-strength feedstock without the need of dilution or other additives,which certainly improves the process efficiency and reduces the cost of primary recovery in the downstream process.This paper reviews the current advances in mixed-mode chromatography,as well as technical history and some applications.The future development and application areas are also discussed.

Abstract:Expanded bed adsorption (EBA) is an innovative chromatography technology that allows the introduction of particle containing feedstock without the risk of blocking the bed. Compared with normal chromatography, two factors should be considered in EBA process design: (1) the fluid side transport can not be neglected due to the increased void fraction in the bed; (2) the influence of suspended biomass in the feedstock should be seriously considered. With the appropriate methods of predicting the sorption efficiency and characterizing biomass influences in EBA, a parallel process design strategy is proposed that considers the efficient adsorption of target protein, together with the negative effects of biomass in the feedstock. The strategy was applied to the EBA separation of recombinant formate dehydrogenases from E. coli homogenate. Good results were obtained with a purification factor of 6 and a yield of 85%. In addition, the process should be scaled-up.
High-throughput screening methods for protein adsorption evaluation with microtiter filter plate

Abstract:Aiming at the process optimization of chromatographic separation, the high-throughput screening methods of protein adsorption with microtiter filter plate were established for initial resin screening, adsorption properties evaluation, adsorption isotherms and adsorption kinetics measurement, and adsorption and elution conditions optimization. Firstly, the operation parameters of 96-well filter plate were optimized, and with two ion exchangers and two mixed-mode resins as the model, the bovine serum albumin adsorption was investigated with different resins and liquid conditions by means of microtiter filer plate. The binding capacity maps were obtained and the appropriate adsorption-desorption conditions were determined. Further, the microtiter filter plate was applied to measure the adsorption isotherms and adsorption kinetics with four resins at specific adsorption conditions, and the adsorption parameters were obtained. Finally, the elution conditions were optimized using microtiter filter plate. The microscale results were comparable to that with packed-bed separation, indicating that the methods developed in the present work were practicable. The results demonstrated that the high-throughput process development of protein adsorption with microtiter filer plate was feasible and can be used to fast screen the best reins and liquid conditions. New methods showed the advantages of low resource consumption, large experimental fluxes, short developing period, wide application and high stability, which would be one new method for development of the protein separation process.
Process design and application of twin-column continuous chromatography for antibody affinity separation

Abstract:In order to improve process throughput and reduce operation cost of Protein A affinity chromatography, a twin-column continuous chromatography method was studied for monoclonal antibody (mAb) separation. Effects of residence time in the column and loading protein concentration were investigated, and the separation conditions were optimized for mAb capture from cell culture broth. Two typical Protein A affinity resin (MabSelect SuRe and UniMab) were used and the breakthrough curves were measured under different residence times and IgG concentrations, and their effects on the productivity, resin capacity utilization and buffer consumption were evaluated. The results indicate that the twin-column continuous chromatography method has higher productivity and capacity utilization, and lower buffer consumption when comparing to traditional batch chromatography at low residence times. The separation performance of continuous chromatography for mAb capture from cell culture broth was similar to batch chromatography with purity > 97%, yield > 87%, aggregates < 2% and host cell proteins < 300 ppm. However, the capacity utilization increased from 20% ~30% to 70% ~80%, and the buffer consumption reduced to 1/5~1/4 of traditional batch processes. The results demonstrate that the twin-column continuous chromatography method shows great potential for the mAb production.

Abstract:Ionic liquids have exhibited unique selective dissolution and absorption properties and provide new ways for gas separation and purification.Immobilizing ionic liquid into polymers or porous inorganic supports generates the so-called immobilized or supported ionic liquids which possess the advantages of both the ionic liquids and the support materials.For applications in gas separation,they are characterized by continuous process,competitive or lower cost,higher sorption and disorption rate.In this paper,the research progresses in the applications of the immobilized ionic liquids including particles and membranes in the gas separation and purification are reviewed.

Abstract:Hydrocarbon separation is one of the most critically important and complex industrial separation processes, offering versatile bulk chemicals and vital support to the national economy. Traditional separation technologies, such as cryogenic distillation and solvent extraction, are energy-intensive and cause serious environmental stress. Moreover, the growth of industries and technologies and the greater requirements for products (e.g., purity) lead to challenges that cannot be met using traditional separation methods. Adsorptive and membrane-based separations are recognized as energy-efficient alternatives by which to revolutionize the current energy-intensive conditions and satisfy the new demands. This critical review presents the recent progress in metal-organic frameworks (MOFs) and related membranes (e.g., continuous MOF membranes and mixed-matrix membranes) for hydrocarbon separation. The contributions of the underlying separation mechanisms (e.g., enthalpy-driven thermodynamic equilibrium, molecular sieving, kinetic separation based on molecular size, and combined mechanisms) and the adopted strategies (e.g., defect and microstructure control, membrane thickness and interfacial compatibility) to the breaking of trade-off (e.g., permeability/selectivity and capacity/selectivity) and the design of novel materials and processing technologies are discussed. Moreover, this review also summarizes the potential barriers that exist from the academic to the ultimate industrial implementations and the prospects of future development.

Abstract:Background: Bioactive compounds from various natural sources have been attracting more and more attention, owing to their broad diversity of functionalities and availabilities. However, many of the bioactive compounds often exist at an extremely low concentration in a mixture so that massive harvesting is needed to obtain sufficient amounts for their practical usage. Thus, effective fractionation or separation technologies are essential for the screening and production of the bioactive compound products. The applicatons of conventional processes such as extraction, distillation and lyophilisation, etc. may be tedious, have high energy consumption or cause denature or degradation of the bioactive compounds. Membrane separation processes operate at ambient temperature, without the need for heating and therefore with less energy consumption. The "cold" separation technology also prevents the possible degradation of the bioactive compounds. The separation process is mainly physical and both fractions (permeate and retentate) of the membrane processes may be recovered. Thus, using membrane separation technology is a promising approach to concentrate and separate bioactive compounds. Methods: A comprehensive survey of membrane operations used for the separation of bioactive compounds is conducted. The available and established membrane separation processes are introduced and reviewed. Results: The most frequently used membrane processes are the pressure driven ones, including microfiltration (MF), ultrafiltration (UF) and nanofiltration (NF). They are applied either individually as a single sieve or in combination as an integrated membrane array to meet the different requirements in the separation of bioactive compounds. Other new membrane processes with multiple functions have also been developed and employed for the separation or fractionation of bioactive compounds. The hybrid electrodialysis (ED)-UF membrane process, for example has been used to provide a solution for the separation of biomolecules with similar molecular weights but different surface electrical properties. In contrast, the affinity membrane technology is shown to have the advantages of increasing the separation efficiency at low operational pressures through selectively adsorbing bioactive compounds during the filtration process. Conclusion: Individual membranes or membrane arrays are effectively used to separate bioactive compounds or achieve multiple fractionation of them with different molecule weights or sizes. Pressure driven membrane processes are highly efficient and widely used. Membrane fouling, especially irreversible organic and biological fouling, is the inevitable problem. Multifunctional membranes and affinity membranes provide the possibility of effectively separating bioactive compounds that are similar in sizes but different in other physical and chemical properties. Surface modification methods are of great potential to increase membrane separation efficiency as well as reduce the problem of membrane fouling. Developing membranes and optimizing the operational parameters specifically for the applications of separation of various bioactive compounds should be taken as an important part of ongoing or future membrane research in this field.

Abstract:Bioactive natural products are a main source of new drugs, functional foods and food additives. The separation of bioactive natural products plays an important role in transformation and use of biomass. The isolation and purification of bioactive principle from a complex matrix is often inherent bottleneck for the utilization of natural products, so a series of extraction and separation techniques have been developed. This review covers recent advances in the separation of bioactive natural products with an emphasis on their solubility and diffusion coefficients, recent extraction techniques and isolation techniques. This overview of recent technological advances, discussion of pertinent problems and prospect of current methodologies in the separation of bioactive natural products may provide a driving force for development of novel separation techniques.

Abstract:The combination of membranes and microfluidic systems has been applied in areas such as bioscience, environmental and analytical chemistry, and in the past time research into the integration of membrane mass transfer control functions into microfluidic devices has shown a significant growth trend. The multiple uses of membrane-based microfluidic systems have been demonstrated in published reports. The aim of this paper is to provide an overview of the development process in this field, starting with the basic terminology of microfluidic and membrane processes and outlining membrane-based microfluidic platforms that combine the two, followed by a classification of membrane integration on a chip based on the preparation methods used to combine the two. Three membrane functions that membranes can perform in microfluidic systems are then discussed; they are separation and enrichment of substances, immobilization and culturing of cells, and fluid control of pumps and valves. Finally, several possible research directions and related issues for membrane-based microfluidic platforms are presented, providing a deeper reference for future research combining membranes with microfluidic platforms.

Abstract:All-aqueous phase separation systems play essential roles in bioanalytical and biochemical applications. Compared to conventional oil and organic solvent-based systems, these systems are characterized by their rich bulk and interfacial properties, offering superior biocompatibility. In particular, phase separation in all-aqueous systems facilitates the creation of compartments with specific physicochemical properties, and therefore largely enhances the accessibility of the systems. In addition, the all-aqueous compartments have diverse affinities, with an important property known as partitioning, which can concentrate (bio)molecules toward distinct immiscible phases. This partitioning affinity imparts all-aqueous interfaces with selective permeability, enabling the controlled enrichment of target (bio)molecules. This review introduces the basic principles and applications of partitioning-induced interfacial phenomena in a typical all-aqueous system, namely aqueous two-phase systems (ATPSs); these applications include interfacial chemical reactions, bioprinting, and assembly, as well as bio-sensing and detection. The primary challenges associated with designing all-aqueous phase separation systems and several future directions are also discussed, such as the stabilization of aqueous interfaces, the handling of low-volume samples, and exploration of suitable ATPSs compositions with the efficient protocol.

Abstract:With the rapid development of biotechnology, the importance of microfluidic bioseparation and bioassay in biomedicine, clinical diagnosis, and other fields has become increasingly prominent. Microfluidic technology, with its significant advantages of high throughput, automated operation, and low sample consumption, has brought new breakthroughs in the field of biological separation and bioassay. In this paper, the latest research progress in microfluidic technology in the field of bioseparation and bioassay is reviewed. Then, we focus on the methods of bioseparation including active separation, passive separation, and hybrid separation. At the same time, the latest research results of our group in particle separation are introduced. Finally, some application examples or methods for bioassay after particle separation are listed, and the current challenges and future prospects of bioseparation and bioassay are discussed.

Abstract:Traditional Chineae medicine (TCM) is often composed of a variety of natural medicines. Its composition is complex, and many of its components can not be analyzed and identified. The first step in the rational application of TCM is to successfully separate the effective components which is also a great inspiration for the development of new drugs. Among the many separation technologies of TCM, the traditional heating concentration separation technology has high energy consumption and low efficiency. As a new separation technology, membrane separation technology has the characteristics of simple operation, high efficiency, environment-friendly and so on. The separation effect of high molecular weight difference solution is better. The applications of several main membrane separation technologies such as microfiltration, nanofiltration, ultrafiltration and reverse osmosis are reviewed, the methods of restoring membrane flux after membrane fouling are discussed, and their large-scale industrial applications in the future are prospected and summarized.

Abstract:<p>Polymerized high internal phase emulsions (polyHIPEs) as highly porous adsorption materials have received increasing attention and wide applications in separation science in recent years due to their remarkable merits such as highly interconnected porosity, high permeability, good thermal and chemical stability, and tailorable chemistry. In this review, we attempt to introduce some strategies to utilize polyHIPEs for separation science, and highlight the recent advances made in the applications of polyHIPEs for diverse separation of small organic molecules, carbon dioxide, metal ions, proteins and other interesting targets. Abstract:Modern biomass and organic waste are becoming major, carbon-neutral sources of fine chemicals, biomolecules and fuels to replace fossil fuel products. As a consequence, there is a strong demand for advanced separation techniques to purify biomolecules occurring in complex mixtures, because actual downstream processing accounts for a major part of the overall cost. Here we review the mechanism, methods and applications of three-phase partitioning. Methods include macroaffinity ligand-, metal ion-, enzyme-, ultrasound-, ionic liquid- and microwave-assisted partitioning. Applications to the purification of recombinant and conjugated proteins, enzymes, carbohydrate polymers, lipids and pigment are presented. Three-phase partitioning precipitates biomolecules from aqueous solutions using t -butanol and ammonium sulphate. This technique is applicable for both upstream and downstream with unrefined samples, and downstream, when a simple and scalable step is required. The yield of the biomolecules reaches 90% and higher, and the maximum purification degree is 12–40-fold. Three-phase partitioning allows about 70% of potential savings over standard chromatographic procedures.

Abstract:Membrane processes are increasingly reported for various applications in both upstream and downstream technology, such as microfiltration, ultrafiltration, emerging processes as membrane chromatography, high performance tangential flow filtration and electrophoretic membrane contactor. Membrane-based processes are playing critical role in the field of separation/purification of biotechnological products. Membranes became an integral part of biotechnology and improvements in membrane technology are now focused on high resolution of bioproduct. In bioseparation, applications of membrane technologies include protein production/purification, protein-virus separation. This manuscript provides an overview of recent developments and published literature in membrane technology, focusing on special characteristics of the membranes and membrane-based processes that are now used for the production and purification of proteins.

Abstract:Using membrane separation has broader application prospects; however, its low mass transfer efficiency has limited its development. Progress with studies on improving the selectivity and permeability of new hollow fiber membranes has been slow and difficult thus far. Additionally, improving the structure of the hollow fiber membrane module to improve the fluid flow performance of the flow field in the membrane module and enhance mass transfer is a common research strategy. In this study, a series of innovative designs of membrane modules developed by scholars in the field of hollow fiber membrane applications were reviewed. Various successful optimization designs can provide new insights for researchers on membrane separation.
