Abstract:A new highly stereoselective construction of the side chain of the C-24 and C-25 oxysterols has been achieved by using desmosterol acetate as the starting material and an improved Sharpless catalytic asymmetric dihydroxylation with 100% d.e. (diastereomeric excess) as the key step. The result is much better than the usual asymmetric dihydroxylation procedure. t-Butyl nitrite/2,2,6,6-tetramethylpiperidine N-oxyl radical/FeCl3 catalyst system was developed to activate molecular oxygen for the aerobic oxidation of 24-hydroxycholesterol and the 24-ketocholesterol was obtained in 86.2% yield. The oxidation system has never been reported before. The mechanism for the catalytic aerobic oxidation was also proposed.

Abstract:Polycyclic sultams are efficiently synthesized through a palladium-catalyzed tandem reaction of 1-bromo-2-(cyclopropylidenemethyl)benzenes with 2-alkynylbenzenesulfonamides. The transformation proceeds through double carbometallation with excellent chemoselectivity and regioselectivity, leading to a range of 7,7a-dihydro-6H-benzo[f]indeno[1,2-d][1,2] thiazepine 5,5-dioxides in moderate to good yields. During the reaction process, three new bonds and two rings including the seven-membered sultam ring are formed.

Abstract:Methyl 3-oxo-2,6,6-trimethylcyclohex-1-ene-1-carboxylate was synthesised over eight steps from the starting material of citral instead of α-ionone. KMnO4, HC(OEt)3 and O2/TEMPO/CuCl were substituted for NaIO4, CH3I and pyridinium chlorochromate, respectively. Every step was simplified and gave a 48% overall yield. The procedure is suitable for large scale production.

Abstract:A series of structurally diverse 4-trifluoromethyl β-sultams was conveniently synthesized via the sulfa-Staudinger cycloadditions of 2,2,2-trifluoroethanesulfonyl chloride with different imines under very mild conditions. The products were obtained in yields up to 75%, and in cis/trans ratios ranging from 59:41 to 12:78.

Abstract:The generation of thioesters through thioesterification of carboxylic acid has significant synthetic value given the easy availability of various carboxylic acids and the extensive application of thioesters. An economical and rapid thioester synthesis approach is still needed. Herein, we achieved thioesterification of carboxylic acids with valuable glycosyl thiol, aromatic and aliphatic thiols mediated by sulfonyl fluoride (SO2F2). The compatibility of numerous carboxylic acid substrates was tested, including (hetero)aromatic and aliphatic carboxylic acids and the reaction proceeded smoothly under mild conditions at good yields. Late‐stage modification of drug molecules rac‐Naproxen and Loxoprofen proved feasible. Besides, the gram‐scale reaction demonstrated the utility of this protocol.

Abstract:sulfomethyltetrazole 5 thiol disodium salt, a side chain of cefonicid,was synthesized from sodium bisulfite,ammonium hydroxide and formaldehyde through five steps of reactions. The yield of product was 10.8% in the optimum condition. The process was more economic in which the iodomethane is substituted to the bromoethane in the second reaction.

Abstract:Cyclic sulfonamides (sultams) play a unique role in drug discovery and synthetic chemistry. A direct synthesis of sultams by an intramolecular C(sp(3))-H amidation reaction using an iron complex in situ derived from Fe(ClO4)(2) and aminopyridine ligand is reported. This strategy features a readily available catalyst and tolerates a broad variety of substrates as demonstrated by 22 examples (up to 89% yield). A one-pot iron-catalyzed amidation/oxidation procedure for the synthesis of cyclic N-sulfonyl ketimines is also realized with up to 92% yield (eight examples). The synthetic utility of the method is validated by a gram-scale reaction and derivatization of the products to ring-fused sultams.

Abstract:β-Chiral sulfones are structural motifs widely found in natural products and bioactive molecules, while also serving as important intermediates for the synthesis of valuable chiral scaffolds. In contrast to the rapid growth of sulfur dioxide insertion chemistry over the last decade, enantioselective catalytic variants for accessing β-chiral sulfones, especially those bearing a quaternary carbon stereocenter, remain rare. Herein, we report an enantioselective copper/bisoxazoline catalyzed oxysulfonylation of alkenes to yield isoxazolinyl-containing β-chiral sulfones bearing quaternary carbon stereocenters with moderate to excellent yields and up to 96:4 er. The advantage and irreplaceability of this sulfur dioxide insertion catalytic system have been demonstrated by achieving transformations clearly unavailable with the previously reported sulfonyl chloride system. Additionally, the recoverability of the new-developed bisoxazoline ligand L9 and its reusability without any erosion of the catalytic activity and enantioselectivity further demonstrate the usefulness of the protocol.

Abstract:The solid-phase synthesis of sultams from resin-bound amino acids is described. The sulfonylation of the resin-bound primary amines afforded the requisite secondary amines, after which the intramolecular Buchwald–Hartwig-type coupling forms the C–S bond. A final alkylation on the sulfonamides followed by cleavage provided the corresponding seven-membered sultams.

Abstract:An efficient total synthesis of (S)-14-azacamptothecin has been accomplished in 10 steps and 56% overall yield from 5H-pyrano[4,3-d]pyrimidine 8. A mild Hendrickson reagent-triggered intramolecular cascade cyclization, a highly enantioselective dihydroxylation, and an efficient palladium-catalyzed transformation of an O-allyl into N-allyl group are the key steps in the synthesis. This work provides a much higher overall yield than the previous achievement and shows sound flexibility for the further applications that will lead to new bioactive analogues.

Abstract:The core of benzo[f][1,2]thiazonin-5(4H)-one 3,3-dioxide is efficiently assembled through a palladium-catalyzed tandem reaction of 2-alkynylbenzenesulfonamides with 2-(2-bromobenzylidene)cyclobutanones. A range of polycycles with a nine-membered sultam ring are generated in moderate to good yields. During the transformation, three new bonds are formed via a tandem process with the construction of a nine-membered sultam ring. Double carbometallation is believed to be involved with the ring-opening of cyclobutanone.

Abstract:Sulfur oxides are widespread and gaining increasing attention in pharmaceutical and biotechnology discoveries owing to the heightened activities despite sulfur dioxide (SO2) is one of the most important gaseous air pollutants. Considerable research efforts have been devoted to deal with sulfur oxides in a greener and sustainable way. Herein, we report a visible-light-promoted four-component radical-polar crossover tandem cyclization involved twofold insertion of SO2, and the formation of S(IV) and S(IV) centers in one procedure. Various types of functionalized β-sulfonyl sultines were provided from the easy-to-access C(sp2)-H and C(sp3)-H building blocks with high group compatibility under mild and operationally simple conditions. Gram-scale preparation and late-stage transformations demonstrate opportunities to rapidly construct molecular complexity. Mechanistic studies supported a SO2 bi-inserted radical-polar crossover cyclization relay sequence, which allows for a more efficient utilization of SO2 and is also more environmentally friendly.
