Abstract:An unexpected process from tetrahydrodiazirino[3,1-a]isoquinolines and diversified sulfonyl chlorides RSO2Cl (R=aryl, alkyl) to deliver highly yields 4,5-dihydro-3H-benzo[d][1,2]diazepine skeleton compounds was disclosed via N-insertion ring-opening reaction. The structural framework of 2,3-benzodiazepines is unambiguously confirmed by single-crystal X-ray crystallographic analysis. The present protocol features mild reaction conditions, high yields and feasibility of large-scale synthesis and catalyst-free access to benzo[2,3]diazepines skeleton. In contrast, azomethine imines were produced by employing tetrahydrodiazirino[3,1-a]isoquinoline reacts with aroyl chlorides under the same conditions.

Abstract:A new method for synthesis of synthesis 6-hydroxy-3, 4-dihydro-2(lH) quinolinone was investigated with higher yield and simpler operation. The title compound was prepared starting from 3-chloropropionyl chloride, via cyclizing, nitration, reduction, diazotization and hydrolysis. The total yield is 67.7%, and the yield of each step is between 86.5% -99.6%. 6-Hydroxy-3, 4-dihydro-2(1H)quinolinone was characterized by MS and 1H NMR.

Abstract:3,4-Dihydro-5-hydroxy-1(2H)-isoquinolinone(1) was synthesized from isoquinoline(2) by a three-step reaction that isoquinoline5-sulfonic acid(3) was prepared by the reaction of fuming sulfuric acid with 2,1,5-dihydroxy-isoquinoline(4) was obtained from 3 in a melted mixture of sodium hydroxide and potassium hydroxide and hydrogenation of 4 to 1 was fulfilled in the presence of 5% PdC.The overall yield of 1 was 47.6%.The structure of 1 was identified by ~1H NMR and MS.

Abstract:Rh(III)-catalyzed C-H activation/annulation of benzimidates with 4-diazoisochroman-3-imines furnished 8-alkoxy-5H-isochromeno[3,4-c]isoquinolines in moderate to excellent yields with a broad range of substrate scope. The reaction was carried out under mild reaction conditions and could be scaled up with practical usage. Similar reaction between benzimidates and 4-diazoisoquinolin-3-ones provided 1-alkoxy-4-arylisoquinolin-3-ols in excellent yields. Moreover, the synthesized products could be conveniently transformed to the corresponding heterocycles with a 1,8-naphthyridinone or isochromenopyridinone core, which are privileged structures in medicinal chemistry.

Abstract:A one-pot procedure has been developed to prepare the derivatives of 2-arylimidazo(2, 1-a)-isoquinolines (2a similar to 2b), 2-arylimidazo(1, 2-a) quinolines (4a similar to 4b) and 2-arylimidazo(1, 2-a) phthalazines (6a similar to 6c) in moderate yields (51% similar to 62%) by the reaction of isoquinolinium salts, quinolinium salts or phdialazinium salts with ammonium acetate in the presence of tetrakispyridine cobalt(II) dichromate (TPCD).

Abstract:In this study, a synthetic method to access benzo[4,5]imidazo[2,1‐a]isoquinolines is developed by the reaction of 2‐(2‐bromoaryl)benzimidazoles with 1,3‐diketones under basic conditions. A reaction pathway involving the initial formation of a C–C coupled intermediate by nucleophilic aromatic substitution and subsequent deacylative cyclocondensation is proposed for this process.

Abstract:A mild direct synthesis of chiral imidazo[2,1‐a]isoquinolin‐3‐one derivatives with dihydroisoquinolines and N‐substituted amino acids have been developed. By the use of easily accessible DCC as condensation reagent at ambient temperature, a series of imidazo[2,1‐a]isoquinolin‐3‐ones can be assembled readily in low to good yields with low to excellent dr values. Interestingly, the use of N‐substituted L‐Threonine and L‐Serine afforded oxazino[2,3‐a]isoquinolin‐4‐one derivatives with good diastereoselectivities. A mild direct synthesis of imidazo[2,1‐a]isoquinolin‐3‐one derivatives with dihydroisoquinolines and N‐substituted amino acids have been developed by the use of DCC as condensation reagent at ambient temperature.

Abstract:A simple synthesis of benzo[d,e]imidazo[2,1-a]isoquinolines is described. Heating a mixture of acenaphthoquinone, a benzil, and ammonium acetate under solvent-free conditions afforded 9,10-diaryl-7H-benzo[d,e]imidazo[2,1-a]isoquinolin-7-ones in good to excellent yields.

Abstract:Using benzaldehydes, NaN(SiMe(3))(2), and N-acylpyrroles, an operationally simple tandem method to produce a wide array of 3,4-dihydroisoquinolones is presented (37 examples, yields up to 98%). A unique feature of this method stems from the sequential aminobenzylation of aldehydes and transamidation of the corresponding N-(trimethylsilyl)imines in one pot. In this process, three new bonds are generated (one C-C and two C-N bonds).

Abstract:The Ritter reaction of dialkyl benzyl carbinols with 3-methoxy- and 3-phenoxypropanenitriles afforded the corresponding 1-(2-methoxyethyl)- and 1-(2-phenoxyethyl)-3,3-dialkyl-3,4-dihydroisoquinolines. Likewise, benzo[ f ]isoquinoline derivatives were synthesized from 2-methyl-3-(naphthalen-1-yl)propan-2-ol. The obtained compounds were shown to have the imine structure, but they exhibited enamine properties. In particular, their reactions with oxalyl chloride resulted in pyrrole annulation.

Abstract:The reaction of 1-(3,4-dimethoxy)phenylcycloalkanemethane- and 4-(3,4-dimethoxyphenyl)tetrahydropyran-4-methanamines with benzo[ d ][1,3]dioxol-5-carbonyl chloride was used to synthesize N -substituted benzo[ d ][1,3]dioxol-5-carboxamides. The cyclization of the latter with phosphorus oxychloride gave dihydroisoquinolines, whose subsequent reduction with sodium borohydride to spiro-substituted tetrahydroisoquinolines followed by Eschweiler–Clark reaction gave N -methyl derivatives. The condensation of the starting amines with benzo[ d ][1,3]dioxol-5-carbaldehyde formed Schiff bases, which were reduced with sodium borohydride to obtain the corresponding secondary amines―acyclic analogs of tetrahydroisoquinolines. The cyclization of the latter products under Eschweiler–Clark reaction resulted in the synthesis of N -benzodioxolmethyl-substituted tetrahydroisoquinolines.

Abstract:A Rh(III)-catalyzed reaction of N-methoxybenzamides and 4-diazoisochroman-3-imines is described. This method offers a rapid entry to 3-amino-4-arylisoquinolinone architectures from readily available starting materials under mild reaction conditions. A one-pot cascade C-H bond activation/formal [4 + 2] cycloaddition mechanism is proposed. The synthesized 3-amino-4-arylisoquinolinones can be conveniently converted into dibenzo[c,f][1,8]naphthyridines.

Abstract:A multicomponent synthesis of tetrahydroisoquinolines from carboxylic acids, alkynyl ethers, and dihydroisoquinolines is described. This process features readily available starting materials, simple experimental procedures for achievement of molecule complexity, and structural diversity. The preliminary control experiment and crossover reaction provide important insight into the reaction mechanism. The formed tetrahydroisoquinolines could be transformed to an array of compounds.

Abstract:It was found that 4-hydroxy-2-butenoic ester (11) could not react with 3,4-dihydro-isoquinoline (4a). Individual addition reactions of γ-mercapto-α,β-unsaturated esters (18) and -unsaturated amide (19) with 3,4-dihydroisoquinolines (4) were carried out under appropriate conditions to provide the corresponding thiazolo[2,3-α]isoquinoline derivatives with good yields (up to 87%) and significant diastereomeric selectivity. The mechanism of the crucial reaction was discussed.

Abstract:Nine 1,4-dihydropyridine Derivatives,2,6-dimethyl-3,5-diethoxycarbonyl-4-(substituted benzyl)-1,4-dihydropyridines(4a~4i),were synthesized by Hantzsch reaction of single replace phenylacetaldehyde which was prepared from single replace phenylacetic acid by esterification in SOCl2/MeOH,reduction in NaBH4/MeOH,then oxidation with Dess-Martin periodinane.The structures were characterized by 1H NMR,13C NMR and MS.4b~4i were new compounds.

Abstract:Synthesis of isoquinolines based on efficient C-C and C-N bond formation through Rh(III)-catalyzed C-H activation and subsequent intramolecular cyclization is reported. Diazodiesters serving as a C-2 source in the newly formed heterocycles are first demonstrated. Additionally, the Rh(III)-catalyzed direct C-H activation/cyclization of benzimidates with diazoketoesters is also described.

Abstract:In this article, a hitherto unreported catalyst-free ring expansion reaction of tetrahydroisoquinolines with o-alkynylarylaldehydes for the construction of dibenzo[b,d]azepine skeleton is described. Using air as a "green" oxidant, some important biologically active dibenzo[b,d]azepines were produced with favorable tolerance of functional groups and a wide scope of substrates. The synthetic potential was further confirmed by facile scale-up reactions and the synthesis of isoindoline derivatives.

Abstract:An efficient approach to access functionalized dihydro-[1,3]oxazino[4,3-a] isoindole and tetrahydroisoquinoline skeletons has been developed through the addition-cyclization process of ynamides 8 with N-acyliminium ions generated from N,O-acetals 6,7. The reactions were conducted under the catalysis of Cu(OTf)(2), and a number of functionalized dihydro-[1,3]oxazino[4,3-a] isoindoles 9a-9y and tetrahydroisoquinolines 10a-10g, 11a-11p were generated in 48-98% yields. When chiral ynamides 8n-8u were used, optically pure products 11a-11p could be obtained with good to excellent yields and diastereoselectivities. (C) 2021 Elsevier Ltd. All rights reserved.
