Abstract:Bupleurum chinense DC. is a well-known and very important traditional Chinese drug. It is often used to treat common cold with fever, alternating chill and fever, feeling of fullness and oppression in the chest. A new saikosaponin was isolated from Bupleurum chinense DC. and its structure was identified as 3-O-[beta-D-glucopyranosyl (1-->3)-beta-D-fucopyranosyl]-3 beta, 16 alpha, 23, 28-tetrahydroxy-olean-11, 13 (18)-dien-30-oic acid 30-O-[xylityl(1-->1)]-beta-D-glucopyranosyl-6-ester on the basis of chemical and spectral evidence. In addition, three known saikosaponins were isolated and characterized as saikosaponin 1, 2-O-acetyl-saikosaponin b2 and 2-O-acetyl-saikosaponin a. All the compounds were isolated from Bupleurum chinense DC. for the first time.

Abstract:Seven new saikosaponins (1-7), together with four known ones (8 - 11), were isolated from the roots of Bupleurum chinense. Their structures were elucidated by comprehensive spectroscopic analysis and chemical methods. All obtained saikosaponins were oleanane-type, except for compound 11, which was the first lupane-type saikosaponin isolated from the genus Bupleurum. Compound 1 represented the first example of saikosaponins bearing a hydroxyl group at C-15, while compound 4 was a saikosaponin containing acetoxy group at C-23 rarely found in genus Bupleurum. Compounds 6 and 7, bearing an n-butoxy moiety at C-11, were confirmed to be artifacts by LC-MS experiment. All the isolates were evaluated for their inhibitory effects towards nitric oxide production induced by lipopolysaccharide in BV-2 microglial cells, and saikosaponins A (9) and D (10) exhibited the most potent activity, which were comparable to the positive control dexamethasone.

Abstract:A new steroidal glycoside (1), (25R)-14α, 17α-hydroxyspirost-5-en-3β-yl 3-O-α-L-rhamnpyranosyl-(1 → 2)-β-D-glucopyranosyl-(1 → 3)-β-D-glucopyranoside, together with three known steroidal glycosides, (25R)-3β-hydroxyspirost-5-en-1β-yl-3-O-α-L-rhamnopyranosyl-(1 → 2)-O-β-D-xylopyranosyl-(1 → 3)-α-L-arabinopyranoside (2), Cixi-ophiopogon B (3) and Cixi-ophiopogon A (4), were obtained from the tuberous roots of Ophiopogon japonicus (Liliaceae). Compound 2 was isolated from the Ophiopogon genus for the first time. Their structures were identified on the basis of extensive mass and nuclear magnetic resonance spectroscopic analysis.

Abstract:One new chromone, (2′S,4′S)-5′-hydroxy-5-O-methylvisamminol ( 1 ), was isolated from the roots of Saposhnikovia divaricata. The structure of compound 1 was identified by spectroscopic methods including 1D, 2D NMR, and HR-ESI-MS data. The absolute configuration of compound 1 was elucidated by electronic circular dichroism (ECD) calculation. Compound 1 showed moderate cytotoxic activity against human prostatic cancer PC-3 and breast cancer MCF-7 cell lines, with IC 50 values of 52.66 ± 1.73 and 83.12 ± 8.50 μM, respectively.

Abstract:A new chromone glycoside, 3,5,7-trihydroxylchromone-3-O-α-L-arabinopyranoside ( 1 ), together with quercetin ( 2 ), (+)-catechin ( 3 ), (−)-epi-catechin ( 4 ) were isolated from the aerial parts of Rhododendron spinuliferum . The structure of 1 was elucidated on the basis of spectroscopic and 2D-NMR spectral analysis. In addition, 1 exhibited mild inhibitory effect on NO production in LPS-stimulated RAW264.7 cells.

Abstract:In this study, a new chromone, six coumarins, and a phenolic compound were isolated from a MeOH extract of an Angelica dahurica stems. Through the use of various spectral techniques, the structures of the isolated compounds were determined to be scopoletin ( 1 ), xanthyletin ( 2 ), xanthotoxin ( 3 ), hopeyhopin ( 4 ), thamnosmonin ( 5 ), 6-[(1S, 2R)-2,3-dihydroxy-1-methoxy-3-methylbutyl]-7-methoxycoumarin ( 6 ), decursidate ( 7 ), and new compound 8 , named dahuricalol. Among the isolatled and identified compounds, (S)-5-hydroxy-8-(hydroxymethyl)-2,2-dimethyl-6-oxo-3,4-dihydro-2H,6H-pyrano[3,2-g]chromen-3-yl (Z)-2-(hydroxymethyl)but-2-enoate ( 8 ) was isolated from plant for the first time.

Abstract:Two new steroidal saponins, chinenoside VI (1) and neomacrostemonoside D (8b), together with ten known saponins were isolated from the bulbs of Allium chinense. On the basis of chemical evidence and spectral analysis (IR, H-1-NMR C-13-NMR H-1-H-1 COSY, HMQC, HMBC and SI-MS), chinenoside VI was established as (25R)-24-O-beta-D-glucopyranosyl-3 beta,24 beta-dihydroxy-5 alpha-spirost 3-O-alpha-arabinopyranosyl-(1-->6)-beta-D-glucopyranoside, and neomacrostemonoside D was elucidated as neotigogenin 3-O-beta-D-glucopyranosyl-(1-->2)[beta-D-glucopyranosyl-(1-->3)]-(6-acetyl-beta-D-glucopyranosyl)-(1-->4)-beta-D-galactopyranoside, respectively.
Transcriptome Analysis of Bupleurum Chinense Focusing on Genes Involved in the Biosynthesis of Saikosaponins.

Abstract:BACKGROUND:Bupleurum chinense DC. is a widely used traditional Chinese medicinal plant. Saikosaponins are the major bioactive constituents of B. chinense, but relatively little is known about saikosaponin biosynthesis. The 454 pyrosequencing technology provides a promising opportunity for finding novel genes that participate in plant metabolism. Consequently, this technology may help to identify the candidate genes involved in the saikosaponin biosynthetic pathway.RESULTS:One-quarter of the 454 pyrosequencing runs produced a total of 195, 088 high-quality reads, with an average read length of 356 bases (NCBI SRA accession SRA039388). A de novo assembly generated 24, 037 unique sequences (22, 748 contigs and 1, 289 singletons), 12, 649 (52.6%) of which were annotated against three public protein databases using a basic local alignment search tool (E-value ≤1e-10). All unique sequences were compared with NCBI expressed sequence tags (ESTs) (237) and encoding sequences (44) from the Bupleurum genus, and with a Sanger-sequenced EST dataset (3, 111). The 23, 173 (96.4%) unique sequences obtained in the present study represent novel Bupleurum genes. The ESTs of genes related to saikosaponin biosynthesis were found to encode known enzymes that catalyze the formation of the saikosaponin backbone; 246 cytochrome P450 (P450s) and 102 glycosyltransferases (GTs) unique sequences were also found in the 454 dataset. Full length cDNAs of 7 P450s and 7 uridine diphosphate GTs (UGTs) were verified by reverse transcriptase polymerase chain reaction or by cloning using 5' and/or 3' rapid amplification of cDNA ends. Two P450s and three UGTs were identified as the most likely candidates involved in saikosaponin biosynthesis. This finding was based on the coordinate up-regulation of their expression with β-AS in methyl jasmonate-treated adventitious roots and on their similar expression patterns with β-AS in various B. chinense tissues.CONCLUSIONS:A collection of high-quality ESTs for B. chinense obtained by 454 pyrosequencing is provided here for the first time. These data should aid further research on the functional genomics of B. chinense and other Bupleurum species. The candidate genes for enzymes involved in saikosaponin biosynthesis, especially the P450s and UGTs, that were revealed provide a substantial foundation for follow-up research on the metabolism and regulation of the saikosaponins.

Abstract:Five chromone glycosides were isolated from the water-soluble portions of 70% EtOH extract of the roots of Saposhnikovia divaricata, including two new chromone glycosides 1 and 2. The structures of the chromone glycosides were identified as (3'S)-3'-O-β-d-apiofuranosyl-(1 → 6)-β-d-glucopyranosylhamaudol (1), (2'S)-4'-O-β-d-apiofuranosyl-(1 → 6)-β-d-glucopyranosylvisamminol (2), 3'-O-glucopyranosylhamaudol (3), 4'-O-β-d-glucopyranosylvisamminol (4), and 4'-O-β-d-glucopyranosyl-5-O-methylvisamminol (5) on the basis of extensive spectroscopic methods, and the absolute configurations of the new compounds were elucidated by the electronic circular dichroism (ECD) calculation and acid hydrolysis. The cytotoxic activities of the glycosides 1 - 5 against three human cancer cell lines (PC-3, SK-OV-3, and H460) were evaluated. The result showed that compounds 1 - 5 had weak cytotoxic activities against the human cancer cell lines with IC50 values in the range of 48.54 ± 0.80 - 94.25 ± 1.45 μm.

Abstract:Four new flavonoids (1–4) and fourteen known compounds (5–18), were isolated from the aerial part of Bupleurum chinense DC. The structural determination of the new flavonoids was accomplished using comprehensive spectroscopic methods, including 1D and 2D NMR spectra with references to the literatures, as well as high-resolution mass spectrometric analysis. The anti-proliferative activities of the flavonoids (1–18) against HeLa cells were evaluated using the MTT assay with cisplatin as the positive control.

Abstract:Two new cholestanol glycosides (1 and 2) were obtained from the roots and rhizomes of Smilacina henryi. Their structures were determined as 5 alpha a-cholest-9(11)-ene-3 beta, 26-dihydroxy-16, 22-dione 3-O-beta-D-glucopyranosyl-(1 -> 2)-beta-D-galactopyrano side (1), 5 alpha-cholest-9(11)-ene-3 beta, 26-dihydroxy-16, 22-dione 3-O-beta-D-glucopyranosyl-(1 -> 2)-[beta-D-glucopyranosyl-(1 -> 3)]-beta-D -galactopyrano side (2), by physicochemical properties and spectroscopic methods. In addition, the isolated glycosides were tested for their cytotoxic activity against human HepG2 tumor cells and compound 2 showed moderate activity with IC50 value of 59.32 mu M.
