Abstract:Recently, we have developed highly regio- and stereoselective carbometalation of 2-alkynols and 2,3-allenols. The organometallacyclic intermediates may be trapped with I(2) to afford 3-iodoalk-2-en-1-ols. These 3-iodoalk-2-en-1-ols may readily undergo the Sonogashira coupling with terminal propargyl alcohols to form 4-alkyn-2-ene-1,6-diols. Subsequent cycloisomerization in DMA or CH(2)Cl(2) with Au(PPh(3))Cl and AgOTf as the catalyst would afford polysubstituted 2-(1-alkenyl)furans; with PdCl(2) as the catalyst and the reaction in DMA in the presence of allylic bromides, the same substrates afforded polysubstituted 2-(1,4-alkadienyl)furans. In both types of catalyzed cyclization reactions, the elimination of H(2)O promoted the aromatization to form the furan ring. Different alkyl or aryl groups could be introduced into different positions of furans due to the substituent-loading capability of 3-iodoalkenols and diversity of the terminal propargyl alcohols and allylic bromides.

Abstract:A general route to either cis- or trans-2, 5-disubstituted tetrahydrofurans was described by using the nucleophilic addition of organolithium derivatives to tricyclic lactones, followed by a highly stereocontrolled acid-assisted reduction with sodium cyanoborohydride of the hemiketals formed. The stereoselectivity observed can be rationalized by the preferential approach of the hydride on the less hindered face of an oxonium ion intermediate. The cis- or trans-2, 5-disubstituted tetrahydrofurans were provided by the hot decomposition and then hydrogenation of tricycle compounds produced.

Abstract:A copper‐catalyzed olefinic C(sp2)–H activation/carbene insertion/ester hydrolysis/cyclization sequence has been developed for the synthesis of substituted 2(5H)‐furanones employing readily available enaminones and diazo esters as substrates. This method features operationally simple and good functional group tolerance. The transformation could be scaled up to gram quantities and the obtained 2(5H)‐furanone derivatives could be easily converted to a wide range of useful synthetic building blocks, indicative of the applicability of the synthetic strategy. Isotope labeling experiments demonstrated the ester group of diazo esters was hydrolyzed during the reaction.

Abstract:A novel method for the synthesis of benzofuran analogues via condensation of 2-hydroxybenzaldehyde or 2-hydroxyphenyl ketone with methyl 4-bromocrotonate was reported. Moderate to good yield was obtained with cesium carbonate as base in DMF at 70 degrees C. The new-generated double bond was (E)-configuration determined by X-ray diffraction. All benzofuran derivatives were determined by H-1 NMR, C-13 NMR and HRMS techniques.

Abstract:A bioinspired photo-Fries/imine capture cascade reaction was developed in continuous-flow mode, which facilitated the rapid construction of a series of diversely functionalized 2,7-heterocycle-fused tetrahydrocarbazoles, the ubiquitous core structures embedded in strychnos and akuammiline-type monoterpene indole alkaloids. The synthetic utility of this novel method has been preliminarily explored by the first total synthesis of (+)-alsmaphorazine C and formal synthesis of (+)-strictamine in a concise and efficient manner.

Abstract:1,2- cis C -Aryl furanosides are prevalent in nature and exhibit significant biological activities. The 1,2- cis configuration is less favorable in terms of stereoelectronic and steric effects, making the synthesis of this type of skeleton highly challenging. Traditional methods for the synthesis of 1,2- cis C -aryl furanosides usually require complicated protection manipulations, resulting in lengthy synthetic routes and low overall efficiency. Here, we report a simple and highly applicable procedure for the synthesis of 1,2- cis C -aryl furanosides from unprotected aldoses via Petasis reaction and subsequent deaminative cyclization. Unprotected aldose mediated Petasis reactions yield linear 1,2- trans 1-aryl polyhydroxy amines. Halogenation of the amine motif activates the conventionally inert C–N bond and triggers the key stereoinvertive intramolecular substitution process, affording 1,2- cis C -aryl furanosides with excellent chemo- and diastereoselectivity. This procedure does not require the use of any sensitive reagents, and can be conducted in one-pot without precautions against oxygen or moisture, offering a streamlined approach to 1,2- cis C -aryl furanoside natural products and bioactive agents.

Abstract:In AgSbF6/Rh2(OAc)4/DCE system, two-component diastereoselective reactions of 2-diazo-3,5-dioxo-6-ynoates (phosphonates and sulphones) and alkenes provided easy access to 2-cyclopropyl-γ-pyrones through 6-endo-dig cyclization-cyclopropanation. In AgOAc/Rh2(OAc)4/Et3N/DCE system, two-component reaction of 2-diazo-3,5-dioxo-7-aryl-6-ynoates and alkenes afforded 2-cyclopropyl-2-alkylene-3(2H)-furanones through 5-exo-dig cyclization-cyclopropanation. The possible mechanism of reaction is discussed. A simple procedure and mild conditions are significant features of this strategy.

Abstract:Efficient route: A novel Rh(I)-catalyzed regio- and stereospecific carbonylation reaction of (1-alkynyl)cyclopropyl ketones by selective activation of a carbon-carbon sigma bond of the cyclopropane ring was demonstrated (see scheme). This method provides a general, efficient, stereoselective route to synthesise 1,3,5-trisubstituted and 1,3,5,6-tetrasubstituted 5,6-dihydrocyclopenta[c]furan-4-one with convertible functional groups.

Abstract:HHerein, we disclosed the asymmetric construction of an oxa‐quaternary stereocenter via an intramolecular oxa‐Michael (IOM) reaction in β‐substituted ortho‐hydroxymethyl chalcone by the formation of 1,1‐disubstituted‐1,3‐dihydroisobenzofuran using cinchona alkaloid‐based chiral amino‐squaramide catalyst. Both the (E‐ and Z)‐β‐substituted ortho‐hydroxymethyl chalcone provide (S)‐ and (R)‐enantiomers of the 1,1‐disubstituted‐1,3‐dihydroisobenzofuran with excellent stereospecificity. In general, excellent yields (up to 95%) and enantioselectivity (up to 98% ee) were obtained. Furthermore, the resulting 1,1‐disubstituted isobenzofuran or phthalan was converted to corresponding chiral 3,3‐disubstituted phthalides without losing the enantioselectivity. This methodology provides the core moiety of the (S)‐citalopram drug.

Abstract:The construction of fused [5.5.0] bicyclic heterocycles with precise regio-, stereo-, and enantioselective control remains a significant challenge in asymmetric catalysis. In this work, we introduce a novel gold-catalyzed asymmetric [8+4] cycloaddition of 1-(1-alkynyl)cyclopropyl ketones with simple tropones, yielding highly functionalized cyclohepta[b]furo[3,4-d]oxepine derivatives with excellent diastereo- and enantioselectivity (38 examples, all >20:1 dr, up to 95% ee). Additionally, an efficient kinetic resolution (KR) process is achieved (s factor up to 104). The gram-scale synthesis and subsequent synthetic transformations of the cycloadducts further demonstrate the synthetic potential of this method. Furthermore, the cycloadduct can undergo a [1,5]-H shift under acid catalysis, adding another dimension to the structural diversity.

Abstract:A novel catalyst‐ and solvent‐controlled dimerization of aliphatic alkynones through tandem Michael addition/rearrangement/cyclization sequence has been developed. The elaborated protocol provides a convenient chemo‐, regio‐ and stereoselective access to rare 3(2H)‐furanones decorated with 2‐exo‐methylene and 4‐ethenyl functionalities. A comprehensive density functional theory (DFT) study of possible reaction mechanism including ab initio molecular dynamics (AIMD) simulations and quantum theory of atoms in molecules (QTAIM) analysis of various substrate/catalyst/solvent complexes has revealed the Janus‐like behavior of the employed sodium tert‐butoxide/toluene system that is responsible both for the chemoselectivity of the initial addition step and the inhibition of the final cyclization step.

Abstract:A palladium-catalyzed domino cyclization/cycloisomerization reaction of alkyne-tethered carbamoyl chlorides with (E)-β-chloroenones is reported. This reaction proceeds via a syn-carbopalladation of the alkyne, followed by a vinyl-PdII-catalyzed cycloisomerization of the (E)-β-chloroenone cascade, which provides an efficient method to synthesize furan-linked methylene oxindoles. The reaction features stereodefined vinyl-PdII species, high to excellent 5-exo/6-endo selectivity, excellent Z/E selectivity, and the sequential formation of three bonds and bis-heterocycles. The strategy for the synthesis of furan-containing benzofurans has also been demonstrated.

Abstract:The first sequential Corey-Chaykovsky cyclopropanation/Cloke-Wilson rearrangement between propargyl sulfonium salts and acrylonitrile derivatives has been developed, affording the tetra-substituted 2,3-dihydrofurans in generally excellent yields (57-98%) with good diastereoselectivities (7:1-18:1). In addition, chiral propargyl sulfonium salt is also suitable for this strategy, giving the optically active 2,3-dihydrofurans with good enantioselectivities. This reaction sequence was designed upon in situ generated 10π-conjugated structures from the dearomatization of indole fragments and subsequent intramolecular 1,6-addition.
