A New Method for Synthesis of Boronic Acid Adducts of Technetium(Iii) Dioxime

Abstract:BATOs are a family of technetium clathrochelates. The first BATOs were prepared by Treher et al. in 1989 [1], Since then a large number of BATO complexes have been synthesized, and many of them were obtained in Single crystal forms suitable for structural determination. The molecule of these complexes contains three bidentate dioxime ligands capped by a boronic acid moiety and an anionic ligand at the axial Position. The coordination polyhedron may be approximately described as a capped trigonal prism [1]. Some BATOs reveal a tendency to be concentrated in myocardium or brain. Two of them have recently been commercially available as the cerebral and myocardial perfusion radiopharmaceuticals [2, 3]. BATOs are primarily synthesized by direct reduction of pertechnetate with stannous chloride in the presence of a dioxime and a boronic acid. The yield is usually 20 -30% (of Tc). Ligand exchange reactions are also used for preparation of BATOs from Tc(dioxime)3(n-OH)SnCl3 and RB(OH)2 [4], and of the rhenium analogues from ReCl3(CH3CN)(PPh3)2 and RB(0H)2 [5]. The yield is also not very high. The low yield prevents a füll utihzation of the valuable pertechnetates. We report here a new method for the synthesis of BATOs from [Tc(SC(NH2)2)6]Cl3 • 4 H2O, a dioxime, and a boronic acid. By this method we synthesized four new BATOs: TcCl(BDO)3BC6H4CH3, TcCKBDO),BQH4NH2, TcCl(MeCDO)3BC6H4CH3, and TcCl(MeCD0)3BC6H4NH2, where BDO is benzil dioxime, and MeCDO 4-methyl cyclohexanedione dioxime.
