Origin of afferent fibers to arcuate nucleus of the hypothalamus as studied by the horseradish peroxidase method

Abstract:The afferent connections of arcuate nucleus have been examined in the rat by mapping the distribution of labelled neurons following iontophoretic injection of horseradish peroxidase (HRP) into the unilateral arcuate nucleus of the hypothalamus by ventral approach. Neurons labelled by the transported enzyme shown by tetramethyl benzidine (TMB) and 3,3-diaminobenzidine (DAB) staining could be observed in ipsilateral nucleus hypothalamicus anterior, nucleus hypothalamicus lateralus, nucleus hypothalamicus periventricularis, nucleus hypothalamicus ventro-medialis, nucleus preopticus medialis, nucleus amygdaloideus, nucleus striae terminalis and subiculum. A few labelled neurons were also detected bilaterally in nucleus supraopticus, nucleus suprachiasmaticus, nucleus paraventricularis and nucleus preopticus, pars suprachiasmatica and contralateral arcuate nucleus. No labelled neurons were identified in other structures. In general, the afferent fiber connections defined in the present study provide anatomical details for understanding the physiological function of arcuate nucleus.
