Abstract:The arcuate nucleus is a crucial hypothalamic brain region involved in regulating body weight homeostasis. Neurons within the arcuate nucleus respond to peripheral metabolic signals, such as leptin, and relay these signals via neuronal projections to brain regions both within and outside the hypothalamus, ultimately causing changes in an animal's behaviour and physiology. There is a substantial amount of evidence to indicate that leptin is intimately involved with the postnatal development of arcuate nucleus melanocortin circuitry. Further, it is clear that leptin signalling directly in the arcuate nucleus is required for circuitry development. However, as leptin receptor long isoform (Leprb) mRNA is expressed in multiple nuclei within the developing hypothalamus, including the postsynaptic target regions of arcuate melanocortin projections, this raises the possibility that leptin also signals in these nuclei to promote circuitry development. Here, we used RT‐qPCR and RNAscope® to reveal the spatio‐temporal pattern of Leprb mRNA in the early postnatal mouse hypothalamus. We found that Leprb mRNA expression increased significantly in the arcuate nucleus, ventromedial nucleus and paraventricular nucleus of the hypothalamus from P8, in concert with the leptin surge. In the dorsomedial nucleus of the hypothalamus, increases in Leprb mRNA were slightly later, increasing significantly from P12. Using duplex RNAscope®, we found Leprb co‐expressed with Sim1, Pou3f2, Mc4r and Bdnf in the paraventricular nucleus at P8. Together, these data suggest that leptin may signal in a subset of neurons postsynaptic to arcuate melanocortin neurons, as well as within the arcuate nucleus itself, to promote the formation of arcuate melanocortin circuitry during the early postnatal period.

Abstract:An extreme-ultraviolet (EUV) radiation source near the 13-nm wavelength generated in a small (1.1 J) pinch plasma is presented. The ignition of the plasma occurs in a pseudosparklike electrode geometry, which allows for omitting a switch between the storage capacity and the electrode system and for low inductive coupling of the electrically stored energy to the plasma. Thus energies of only a few joules are sufficient to create current pulses in the range of several kiloamperes, which lead to a compression and a heating of the plasmas to electron densities of more than 10(17) cm(-3) and temperatures of several tens of electron volts, which is necessary for emission in the EUV range. As an example, the emission spectrum of an oxygen plasma in the 11-18-nm range is presented. Transitions of beryllium- and lithium-like oxygen ions can be identified. Current waveform and time-resolved measurements of the EUV emission are discussed. In initial experiments a repetitive operation at nearly 0.2 kHz could be demonstrated. Additionally, the broadband emission of a xenon plasma generated in a 2.2-J discharge is presented.
O2 sensing at the mammalian carotid body: why multiple O2 sensors and multiple transmitters?

Abstract:Carotid bodies are the sensory organs for detecting systemic hypoxia and the ensuing reflexes prevent the development of tissue/cellular hypoxia. Although every mammalian cell responds to hypoxia, O2 sensing by the carotid body is unique in that it responds instantaneously (within seconds) to even a modest drop in arterial PO2. Sensing hypoxia in the carotid body requires an initial transduction step involving O2 sensor(s) and transmitter(s) for subsequent activation of the afferent nerve ending. This brief review focuses on: (a) whether the transduction involves ‘single’ or ‘multiple’ O2 sensors; (b) the identity of the excitatory transmitter(s) responsible for afferent nerve activation by hypoxia; and (c) whether inhibitory transmitters have any functional role. The currently proposed O2 sensors include various haem‐containing proteins, and a variety of O2‐sensitive K+ channels. It is proposed that the transduction involves an ensemble of, and interactions between, haem‐containing proteins and O2‐sensitive K+‐channel proteins functioning as a ‘chemosome’; the former for conferring sensitivity to wide range of PO2 values and the latter for the rapidity of the response. Hypoxia releases both excitatory and inhibitory transmitters from the carotid body. ATP is emerging as an important excitatory transmitter for afferent nerve activation by hypoxia. Whereas the inhibitory messengers act in concert with excitatory transmitters like a ‘push–pull’ mechanism to prevent over excitation, conferring the ‘slowly adapting’ nature of the afferent nerve activation during prolonged hypoxia. Further studies are needed to test the interactions between putative O2 sensors and excitatory and inhibitory transmitters in the carotid body.

Abstract:AgRP neurons in the arcuate nucleus of the hypothalamus (ARC) coordinate homeostatic changes in appetite associated with fluctuations in food availability and leptin signaling. Identifying the relevant transcriptional regulatory pathways in these neurons has been a priority, yet such attempts have been stymied due to their low abundance and the rich cellular diversity of the ARC. Here we generated AgRP neuron-specific transcriptomic and chromatin accessibility profiles from male mice during three distinct hunger states of satiety, fasting-induced hunger, and leptin-induced hunger suppression. Cis-regulatory analysis of these integrated datasets enabled the identification of 18 putative hunger-promoting and 29 putative hunger-suppressing transcriptional regulators in AgRP neurons, 16 of which were predicted to be transcriptional effectors of leptin. Within our dataset, Interferon regulatory factor 3 (IRF3) emerged as a leading candidate mediator of leptin-induced hunger-suppression. Measures of IRF3 activation in vitro and in vivo reveal an increase in IRF3 nuclear occupancy following leptin administration. Finally, gain- and loss-of-function experiments in vivo confirm the role of IRF3 in mediating the acute satiety-evoking effects of leptin in AgRP neurons. Thus, our findings identify IRF3 as a key mediator of the acute hunger-suppressing effects of leptin in AgRP neurons.

Abstract:Objective: In order to assess the role of neuropeptide Y (NPY) in the rat of diet-induced obesity (DIO), the change of the expression of NPY in the arcuate nucleus (ARC) of hypothalamus was observed, in association with the diet composition and individual sensitivity to the weight promoting action of the high fat (HIF) diet. Methods: (a) After fed HIF diet for 14 weeks, rats showing higher body weight and lower body weight were assigned as DIO and diet resistant (DR) group, respectively. Food intake was measured daily, for 4 weeks. (b) NPY expression was detected in ARC of the hypothalamus via immunohistochemistry. Results: (a) About 50% rats developed DIO on an HIF diet. (b) DIO rats were accompanied by a marked higher energy intake and up-regulation of NPY expression in ARC. Conclusion: Up-regulation of NPY expression in the hypothalamic ARC of the DIO rats in response to an HIF diet is associated with increased weight gain and obesity.
GABAergic leptin receptor-expressing neurons in the dorsomedial hypothalamus project to brown adipose tissue-related neurons in the paraventricular nucleus of mice

Abstract:Brown adipose tissue (BAT) contributes to energy homeostasis via nonshivering thermogenesis. The BAT is densely innervated by the sympathetic nervous system (SNS) and activity of pre-autonomic neurons modulates the sympathetic outflow. Leptin, an adipocyte hormone, alters energy homeostasis and thermogenesis of BAT via several neuronal circuits; however, the cellular effects of leptin on interscapular BAT (iBAT)-related neurons in the hypothalamus remain to be determined. In this study, we used pseudorabies virus (PRV) to identify iBAT-related neurons in the paraventricular nucleus (PVN) of the hypothalamus and test the hypothesis that iBAT-related PVN neurons are modulated by leptin. Inoculation of iBAT with PRV in leptin receptor reporter mice (Lepr:EGFP) demonstrated that a population of iBAT-related PVN neurons expresses Lepr receptors. Our electrophysiological findings revealed that leptin application caused hyperpolarization in some of iBAT-related PVN neurons. Bath application of leptin also modulated excitatory and inhibitory neurotransmission to most of iBAT-related PVN neurons. Using channel rhodopsin assisted circuit mapping we found that GABAergic and glutamatergic Lepr-expressing neurons in the dorsomedial hypothalamus/dorsal hypothalamic area (dDMH/DHA) project to PVN neurons; however, connected iBAT-related PVN neurons receive exclusively inhibitory signals from Lepr-expressing dDMH/DHA neurons.
Leptin Activated Hypothalamic BNC2 Neurons Acutely Suppress Food Intake

Abstract:Leptin is an adipose tissue hormone that maintains homeostatic control of adipose tissue mass by regulating the activity of specific neural populations controlling appetite and metabolism . Leptin regulates food intake by inhibiting orexigenic agouti-related protein (AGRP) neurons and activating anorexigenic pro-opiomelanocortin (POMC) neurons . However, while AGRP neurons regulate food intake on a rapid time scale, acute activation of POMC neurons has only a minimal effect . This has raised the possibility that there is a heretofore unidentified leptin-regulated neural population that suppresses appetite on a rapid time scale. Here, we report the discovery of a novel population of leptin-target neurons expressing basonuclin 2 ( ) that acutely suppress appetite by directly inhibiting AGRP neurons. Opposite to the effect of AGRP activation, BNC2 neuronal activation elicited a place preference indicative of positive valence in hungry but not fed mice. The activity of BNC2 neurons is finely tuned by leptin, sensory food cues, and nutritional status. Finally, deleting leptin receptors in BNC2 neurons caused marked hyperphagia and obesity, similar to that observed in a leptin receptor knockout in AGRP neurons. These data indicate that BNC2-expressing neurons are a key component of the neural circuit that maintains energy balance, thus filling an important gap in our understanding of the regulation of food intake and leptin action.

Adiponectin Potentiates the Acute Effects of Leptin in Arcuate Pomc Neurons

Abstract:Objective: Adiponectin receptors (AdipoRs) are located on neurons of the hypothalamus involved in metabolic regulation - including arcuate proopiomelanocortin (Pomc) and Neuropeptide Y/Agouti-related peptide (NPY/AgRP) neurons. AdipoRs play a critical role in regulating glucose and fatty acid metabolism by initiating several signaling cascades overlapping with Leptin receptors (LepRs). However, the mechanism by which adiponectin regulates cellular activity in the brain remains undefined.Methods: In order to resolve this issue, we utilized neuron-specific transgenic mouse models to identify Pomc and NPY/AgRP neurons which express LepRs for patch-clamp electrophysiology experiments.Results: We found that leptin and adiponectin synergistically activated melanocortin neurons in the arcuate nucleus. Conversely, NPY/AgRP neurons were inhibited in response to adiponectin. The adiponectin-induced depolarization of arcuate Pomc neurons occurred via activation of Phosphoinositide-3-kinase (PI3K) signaling, independent of 5' AMP-activated protein kinase (AMPK) activity. Adiponectin also activated melanocortin neurons at various physiological glucose levels.Conclusions: Our results demonstrate a requirement for PI3K signaling in the acute adiponectin-induced effects on the cellular activity of arcuate melanocortin neurons. Moreover, these data provide evidence for PI3K as a substrate for both leptin and adiponectin to regulate energy balance and glucose metabolism via melanocortin activity. (C) 2016 The Author(s). Abstract:This study demonstrates the role between the VMN PACAP neurones and PAC1R expressed by POMC and NPY/AgRP neurones in homeostatic control of energy balance. This study furthers the investigation of how pituitary adenylate cyclase activating polypeptide (PACAP) and the PAC1 receptor (PAC1R) regulate the homeostatic energy balance circuitry. We hypothesized that apoptotic ablation of PACAP neurones in the hypothalamic ventromedial nucleus (VMN) would affect both energy intake and energy expenditure. We also hypothesized that selective PAC1R knockdown would impair the PACAP‐induced excitation in anorexigenic proopiomelanocortin (POMC) neurones and inhibition of orexigenic neuropeptide Y (NPY)/agouti‐related peptide (AgRP) neurones in the hypothalamic arcuate nucleus (ARC). The results show CASPASE‐3‐induced ablation of VMN PACAP neurones leads to increased energy intake and meal frequency as well as decreased energy expenditure in lean animals. The effects were more robust in obese males, whereas we saw the opposite effects in obese females. We then utilized visualized whole‐cell patch clamp recordings in hypothalamic slices. PAC1R knockdown in POMC neurones diminishes the PACAP‐induced depolarization, increase in firing, decreases in energy intake and meal size, as well as increases in CO2 production and O2 consumption. Similarly, the lack of expression of the PAC1R in NPY/AgRP neurones greatly attenuates the PACAP‐induced hyperpolarization, suppression of firing, decreases in energy intake and meal frequency, as well as increases in energy expenditure. The PACAP response in NPY/AgRP neurones switched from predominantly inhibitory to excitatory in fasted animals. Finally, the anorexigenic effect of PACAP was potentiated when oestradiol was injected into the ARC in ovariectomized females. This study demonstrates the critical role of anorexigenic VMN PACAP neurones and the PAC1R in exciting POMC and inhibiting NPY/AgRP neurons to control homeostatic feeding.

Abstract:A study was made of the relative sensitivity and specificity of presumptive and confirmatory media for the most probable number enumeration of enterococci in frozen vegetables. Azide dextrose broth yielded the highest numbers of confirmable enterococci and its sensitivity was shown to be comparable to nonselective media. The use of ethyl violet azide broth as a confirmatory medium resulted in a significant number of false positive tests. Growth in broth containing 6.5% sodium chloride incubated at 45 C for 2 days was found to be a more specific confirmatory test for enterococci.

Abstract:Angiotensin receptors are present in a number of organs and systems including heart, kidney, gonad, and placenta; pituitary and adrenal glands; the peripheral vessels, and the central nervous system. This octapeptide exerts diverse effects that include induction of cell hypertrophy and/or hyperplasia and a stimulation of hormone synthesis and ion transport in the heart, kidney, and adrenal, primarily through type 1 (AT1) receptors. In the kidney, several heterogeneous cell populations--endothelial, epithelial, and vascular--carry AT1 receptors. Some studies suggest that AT2 receptors are also functional, but the cell type carrying this receptor and the nature of its specific function have not been fully elucidated. Although studies indicate that AT1 receptors are affected in response to physiological and pathophysiological manipulations, the functional significance of these modulations remains largely uncertain. Nevertheless, recent human genetic studies indicate that polymorphisms in AT1 receptors, as well as in other angiotensin-related genes, have significant impact on organ remodeling processes of the heart and the kidney.

Abstract:Background: Recent studies reveal perineuronal nets (PNNs), extracellular matrix specializations that restrict the plasticity of neurons, are influenced by pubertal status. Here we show that PNN are changed by gonadectomy (GDX) both in male and female mice in medial amygdala (MeA) and arcuate nucleus (Arc). Method: 49 wild type mice are divided into 16 groups by sex, gonadectomy, diet, nutritional status (fed or fasted groups). Fat and lean body mass were assessed by QMR at baseline and before perfusion. Body weight, food intake, and blood glucose were measured weekly for five weeks. Images were captured in MeA, Arc, paraventricular hypothalamus (PVH), anterior hypothalamus (AH), and an intense WFA positive radial area near PVH, which we named ‘periPVH’ after labelling of Wisteria floribunda agglutinin (WFA). Results: In MeA region, female mice have significantly lower total fluorescence intensity (TFI) (Estimate, 95% Confidence Interval: –448887.02, –578807.10 – –318966.94; p <0.0001) and total number of PNNs (–319.67, –379.34258 – –259.99075: p<0.0001) than male mice. Notably, in female, OVX mice have lower mean fluorescence intensity (MFI) (Mean ± SD: 14.5 ± 3.3 vs. 19.6 ± 4.1; p = 0.003) and TFI (64907.8 (58406.6–96971.3) vs. 123613.9 (83545.9–149131.5); p = 0.015) than sham mice, while the total number of PNNs (Mean ± SD: 63 ± 23 vs. 55 ± 3; p = 0.391) shows no significant difference. Male castration mice have lower total number of PNNs (382 ± 141 vs. 55 ± 22; p <0.0001) and TFI (439343.5 (366431.4–787461.9) vs. 168416.3 (141273.0–258020.8); p <0.0001) than sham mice, while the MFI shows no significant difference. In Arc, mice experiencing GDX have significantly lower MFI (–3.63, –5.61303 – –1.63967; p <0.001), TFI (–28525.8, –45357.46 – –11694.16; P<0.01) or total number of PNNs (–15.3, –29.24 – –1.41; p =0.036) than mice not. PVH, AH, and periPVH don’t show associations with sex, GDX, diet or nutritional status in our observation. Abstract:Selective leptin resistance (SLR) is a pathological state caused by prolonged high fat diet (HFD) feeding in which cardiovascular responses to leptin are preserved, but metabolic effects are attenuated. Thus, SLR is hypothesized to contribute to obesity-associated hypertension, but the mechanisms causing SLR are unclear. Desensitization of leptin signaling in cells of the arcuate nucleus (ARC) of the hypothalamus has been proposed as one of potential mechanisms contributing to SLR. Tanycytes, located along the walls of the third ventricle, play critical roles in the regulation of energy balance through the transport of blood-borne metabolic hormones such as leptin and ghrelin into the cerebrospinal fluid. Previous studies have reported that 10 weeks of 45% HFD (D12451) feeding is sufficient to cause SLR in wildtype C57BL/6J mice. To examine the transcriptomic changes induced by prolonged HFD exposure in tanycytes, we performed single-nucleus RNA-sequencing on nuclei isolated from ARC of ad libitum fed male C57BL/6J mice fed chow diet (7013; CD, n=7) or HFD (n=8) from 8 to 18 weeks of age. At 18 weeks, fat mass (+10.1±1.4 vs +3.2±0.4g, p<0.01) and plasma leptin (486±148 vs 119±43 pg/mL, p<0.05) were increased in mice fed HFD vs CD, energy expenditure was decreased (p<0.05), and caloric intake was normal after correction for body mass. After pooling replicates and filtering out nuclei with low quality reads, a total of 6,621 (CD) & 14,396 (HFD) nuclei remained. Unbiased clustering (Seurat) identified 23 major clusters of cell types within the arcuate. Ingenuity Pathway Analysis indicated enrichment (z-score +0.45, p<0.05) of the canonical "Leptin Signaling in Obesity" pathway in β2 tanycytes (which transport circulating leptin into the cerebrospinal fluid), but such pathways were de-enriched/inhibited in downstream neuronal clusters such as Agouti-related peptide ( Agrp ) neurons (z-score -1.6, p=0.009) and unchanged in neurons expressing proopiomelanocortin ( Pomc ) (p=0.2). Collectively, these results lead us to hypothesize that SLR is associated with normal transport of circulating leptin into the arcuate nucleus, while the primary dysfunction that causes SLR instead occurs within selected neuronal populations such as Agrp neurons.

Abstract:Adverse energy states exert a potent suppressive influence on the reproductive axis by inhibiting the pulsatile release of gonadotrophin‐releasing hormone and luteinizing hormone. One potential mechanism underlying this involves the metabolic‐sensing proopiomelanocortin and agouti‐related peptide/neuropeptide Y (AgRP/NPY) neuronal populations directly controlling the activity of the arcuate nucleus kisspeptin neurons comprising the GnRH pulse generator. Using acute brain slice electrophysiology and calcium imaging approaches in Kiss1‐GFP and Kiss1‐GCaMP6 mice, we have examined here whether NPY and α‐melanocyte stimulating hormone (αMSH) provide a direct modulatory influence on the activity of arcuate kisspeptin neurons in the adult mouse. NPY was found to exert a potent suppressive influence upon the neurokinin B‐evoked firing of approximately half of arcuate kisspeptin neurons in both sexes. This effect was blocked partially by the NPY1R antagonist BIBO 3304 while the NPY5R antagonist L152,804 was ineffective. NPY also suppressed the NKB‐evoked increase in intracellular calcium levels in the presence of tetrodotoxin and amino acid receptor antagonists indicating that the inhibitory effects of NPY are direct on kisspeptin neurons. In contrast, no effects of αMSH were found on the excitability of arcuate kisspeptin neurons. These studies provide further evidence supporting the hypothesis that AgRP/NPY neurons link energy status and LH pulsatility by demonstrating that NPY has a direct suppressive influence upon the activity of a sub‐population of arcuate kisspeptin neurons.

Identification of neuronal subpopulations that project from hypothalamus to both liver and adipose tissue polysynaptically

Abstract:The autonomic nervous system regulates fuel availability and energy storage in the liver, adipose tissue, and other organs; however, the molecular components of this neural circuit are poorly understood. We sought to identify neural populations that project from the CNS indirectly through multisynaptic pathways to liver and epididymal white fat in mice using pseudorabies virus strains expressing different reporters together with BAC transgenesis and immunohistochemistry. Neurons common to both circuits were identified in subpopulations of the paraventricular nucleus of the hypothalamus (PVH) by double labeling with markers expressed in viruses injected in both sites. The lateral hypothalamus and arcuate nucleus of the hypothalamus and brainstem regions (nucleus of the solitary tract and A5 region) also project to both tissues but are labeled at later times. Connections from these same sites to the PVH were evident after direct injection of virus into the PVH, suggesting that these regions lie upstream of the PVH in a common pathway to liver and adipose tissue (two metabolically active organs). These common populations of brainstem and hypothalamic neurons express neuropeptide Y and proopiomelanocortin in the arcuate nucleus, melanin-concentrating hormone, and orexin in the lateral hypothalamus and in the corticotrophin-releasing hormone and oxytocin in the PVH. The delineation of this circuitry will facilitate a functional analysis of the possible role of these potential command-like neurons to modulate autonomic outflow and coordinate metabolic responses in liver and adipose tissue.
Leptin differentially increases sympathetic nerve activity and its baroreflex regulation in female rats: role of oestrogen

Abstract:Obesity and hypertension are commonly associated, and activation of the sympathetic nervous system is considered to be a major contributor, at least in part due to the central actions of leptin. However, while leptin increases sympathetic nerve activity (SNA) in males, whether leptin is equally effective in females is unknown. Here, we show that intracerebroventricular (i.c.v.) leptin increases lumbar (LSNA) and renal (RSNA) SNA and baroreflex control of LSNA and RSNA in α-chloralose anaesthetized female rats, but only during pro-oestrus. In contrast, i.c.v. leptin increased basal and baroreflex control of splanchnic SNA (SSNA) and heart rate (HR) in rats in both the pro-oestrus and dioestrus states. The effects of leptin on basal LSNA, RSNA, SSNA and HR were similar in males and pro-oestrus females; however, i.c.v. leptin increased mean arterial pressure (MAP) only in males. Leptin did not alter LSNA or HR in ovariectomized rats, but its effects were normalized with 4 days of oestrogen treatment. Bilateral nanoinjection of SHU9119 into the paraventricular nucleus of the hypothalamus (PVN), to block α-melanocyte-stimulating hormone (α-MSH) type 3 and 4 receptors, decreased LSNA in leptin-treated pro-oestrus but not dioestrus rats. Unlike leptin, i.c.v. insulin infusion increased basal and baroreflex control of LSNA and HR similarly in pro-oestrus and dioestrus rats; these responses did not differ from those in male rats. We conclude that, in female rats, leptin's stimulatory effects on SNA are differentially enhanced by oestrogen, at least in part via an increase in α-MSH activity in the PVN. These data further suggest that the actions of leptin and insulin to increase the activity of various sympathetic nerves occur via different neuronal pathways or cellular mechanisms. These results may explain the poor correlation in females of SNA with adiposity, or of MAP with leptin.

Abstract:Perineuronal nets (PNNs) are widely present in the hypothalamus, and are thought to provide physical protection and ion buffering for neurons and regulate their synaptic plasticity and intracellular signaling. Recent evidence indicates that PNNs in the mediobasal hypothalamus play an important role in the regulation of glucose homeostasis. However, whether and how hypothalamic PNNs are regulated are not fully understood. In the present study, we examined whether PNNs in various hypothalamic regions in mice can be regulated by sex, gonadal hormones, dietary interventions, or their interactions. We demonstrated that gonadal hormones are required to maintain normal PNNs in the arcuate nucleus of hypothalamus in both male and female mice. In addition, PNNs in the terete hypothalamic nucleus display a sexual dimorphism with females higher than males, and high-fat diet feeding increases terete PNNs only in female mice but not in male mice. On the other hand, PNNs in other hypothalamic regions are not influenced by sex, gonadal hormones or dietary interventions. In summary, we demonstrated that hypothalamic PNNs are regulated in a region-specific manner and these results provide a framework to further investigate the potential functions of PNNs in regulating energy/glucose homeostasis at the interplay of sex, gonadal hormones and diets.

Abstract:OBJECTIVES The aim of this study was to assess the prevalence of infection by HCV, HBV, HDV and HIV and their biological and histopathological patterns in 104 intravenous drug users. METHODS AND RESULTS Seventy-five patients (72%) had anti-HCV antibodies. Transmission was rapid because 33% of those who had been drug users for 6 months or less had anti-HCV antibodies. The contamination rate was very high because 90% of those who had been drug users for 2 years or less had anti-HCV antibodies. Thirty-four (33%) had an HBV marker, and 6 were HBs Ag carriers. None of the patients had anti-HDV antibodies. Only one patient had anti-HIV antibodies. Twenty-five anti-HCV antibody positive drug users underwent liver biopsy. Seven (28%) had normal ALAT levels and 18 (72%) had permanently or intermittently elevated ALAT levels. The mean histological activity on the Knodell index was 4.1 (range: 1-8). CONCLUSIONS This study indicates that contamination by HCV is almost inevitable after 2 years of intravenous drug use. The low prevalence of HBV, HDV, and HIV infection might be explained by a low endemic state of these viruses in our area.
