Abstract:Herein we report an electrochemical strategy for the construction of polysubstituted oxazoles from easily available ketones and acetonitrile under room temperature. The method shows high efficiency, a broad substrate scope, and requires no external chemical oxidant. Mechanistic investigations suggest that the reaction proceeds through a Ritter-type reaction/oxidative cyclization using acetonitrile as the reactant and solvent. X-ray crystallographic analysis reveals that the reaction involves the attack of acetonitrile on the carbonyl carbon instead of the α-carbon.

Abstract:Electrochemical synthesis has been rapidly developing over the past few years. Here, we report a practical and eco-friendly electrocatalytic isomerization of allylic alcohols to their corresponding carbonyl compounds. This reaction can be carried out in undivided cells without the addition of external chemical oxidants and metal catalysts. Moreover, this reaction features a broad substrate scope including challenging allylic alcohols bearing tri- and tetra-substituted olefins and affords straightforward access to diverse β-aryl-ketones. Mechanistic investigations suggest that the reactions proceed through a radical process. This study represents a unique example in which electrochemistry enables hydrogen atom transfer in organic allylic alcohol substrates using a simple organocatalyst.

Abstract:C–H/N–H cross-coupling has become a key technology for the selective conjugation of azole drug molecules. However, the development of new synthetic models and green chemical methods is imperative to enhance the construction of multi-functional compounds and compounds with unique functional groups. We herein reported an electrochemical synthesis of α-tetrazolyl ketones with excellent yields and broad substrate scope, encompassing electron-donating and electron-withdrawing group of aryl ketones, heterocycles, alkyl and various ketone drugs. It was further proved that α-iodoketone was involved in this transformation of the reaction as a critical intermediate.

Abstract:An efficient strategy for carbazoles and 1,2,4-triarylpyrroles synthesis from anilines and cyclohexanones or acetophenones under transition-metal-free conditions is developed. A variety of disubstituted 9-arylcarbazoles were synthesized in moderate to good yields promoted by KI/I-2 using anilines as the nitrogen and aryl source. Meanwhile, a variety of 1,2,4-triarylpyrroles were also selectively synthesized from anilines and acetophenones in the presence of KI alone.

Abstract:Transition‐metal‐free access to carbazoles was achieved through base‐mediated intramolecular C–C bond formation. Reactions of N‐substituted o‐halodiarylamines with potassium tert‐butoxide in the presence of ethylene glycol or 1,10‐phenanthroline provided carbazoles in moderate to excellent yields. This transformation may proceed through a radical pathway according to a control experiment with a radical scavenger.

Abstract:The Cu-catalyzed novel aerobic oxidative esterification reaction of 1,3-diones for the synthesis of α-ketoesters has been developed. This method combines C-C σ-bond cleavage, dioxygen activation and oxidative C-H bond functionalization, as well as provides a practical, neutral, and mild synthetic approach to α-ketoesters which are important units in many biologically active compounds and useful precursors in a variety of functional group transformations. Abstract:The construction of an aryl ketone structural unit by means of catalytic carbon–carbon coupling reactions represents the state-of-the-art in organic chemistry. Herein we achieved the direct deoxygenative ketone synthesis in aqueous solution from readily available aromatic carboxylic acids and alkenes, affording structurally diverse ketones in moderate to good yields. Visible-light photoredox catalysis enables the direct deoxygenation of acids as acyl sources with triphenylphosphine and represents a distinct perspective on activation. The synthetic robustness is supported by the late-stage modification of several pharmaceutical compounds and complex molecules. This ketone synthetic strategy is further applied to the synthesis of the drug zolpidem in three steps with 50% total yield and a concise construction of cyclophane-braced 18–20 membered macrocycloketones. It represents not only the advancement for the streamlined synthesis of aromatic ketones from feedstock chemicals, but also a photoredox radical activation mode beyond the redox potential of carboxylic acids.

Abstract:We report a set of electrochemically regulated protocols for the divergent synthesis of ketones and β-keto esters from the same β-hydroxycarboxylic acid starting materials. Enabled by electrochemical control, the anodic oxidation of carboxylic acids proceeded in either a one-electron or a two-electron pathway, leading to a 1,4-aryl transfer or a semipinacol-type 1,2-group transfer product with excellent chemoselectivity. The 1,4-aryl transfer represents an unprecedented example of carbon-to-oxygen group transfer proceeding via a radical mechanism. In contrast to previously reported radical group transfer reactions, this 1,4-group transfer process features the migration of electron-rich aryl substituents. Furthermore, with these chemoselective electrochemical oxidation protocols, a range of ketones and β-keto esters including those possessing a challenging-to-access medium-sized ring could be synthesized in excellent yields.

Abstract:A new type of alkyne esterification was developed under electrochemical conditions using alkyne aldehydes and alcohol compounds as starting materials and carbene and iodine as mediators. Through this method, external oxidants and transition metals are excluded and the alkyne bond is fully preserved. This approach is compatible with a wide range of substrates and various functional groups and provides a green and mild new route for directly converting alkyne aldehydes into alkyne esters.

Abstract:C(sp3)–H/O–H cross-coupling represents an ideal strategy for constructing C(sp3)-O bonds but remains challenging due to the relatively inert of C(sp3)−H bonds. Herein we report a facile and novel electrochemical oxidative C(sp3)–H/O–H cross-coupling method, leading to various useful α-acyloxyketones with moderate to excellent yields under transition metal- and oxidant free conditions. A series of carboxylic acids including aromatic and aliphatic acids and ketones are compatible with this reaction.

Abstract:This study describes a catalyst-free α-acyloxylation of ketones and a KBr-mediated α-acyloxylation of cyclic ethers. These conversions are effectively mediated by hypervalent iodine(III) reagents serving dual roles as the oxidant and nucleophilic source. Consequently, esters are produced directly in moderate to excellent yields. The proposed method features good functional group compatibility, a broad substrate scope, and high synthetic efficiency and is remarkably environmentally friendly.

Abstract:The synthesis of 4-sulfonyl-1,7-diesters was well-developed, under the open-vessel condition, by K2CO3-mediated double alkylation of α-sulfonyl o-hydroxyacetophenones with acrylates, and tandem debenzoylation of the resulting α,α-disubstituted o-hydroxyacetophenones. A plausible mechanism is proposed and discussed here. This high-yield protocol provides a highly effective intermolecular alkylation and intramolecular debenzoylation via the formation of two carbon-carbon single (C-C) bonds and the cleavage of carbon-carbon single (C-C) bond.
