Abstract:An unexpected process from tetrahydrodiazirino[3,1-a]isoquinolines and diversified sulfonyl chlorides RSO2Cl (R=aryl, alkyl) to deliver highly yields 4,5-dihydro-3H-benzo[d][1,2]diazepine skeleton compounds was disclosed via N-insertion ring-opening reaction. The structural framework of 2,3-benzodiazepines is unambiguously confirmed by single-crystal X-ray crystallographic analysis. The present protocol features mild reaction conditions, high yields and feasibility of large-scale synthesis and catalyst-free access to benzo[2,3]diazepines skeleton. In contrast, azomethine imines were produced by employing tetrahydrodiazirino[3,1-a]isoquinoline reacts with aroyl chlorides under the same conditions.

Abstract:A cost‐effective protocol has been used for the synthesis of novel benzo[g]benzo[4,5]imidazo[2,1-b]quinazoline derivatives. This domino, one‐pot, three‐component reaction was carried out between 2-hydroxy-1,4-naphthoquinone, aromatic aldehydes, and 2-aminobenzimidazole in the presence of piperidine in DMF solvent. This protocol has several advantages such as a convenient operational process, easy access to raw material, good yields and simple work-up. The structure of these compounds were characterized by spectroscopic techniques such as IR, 1 H-NMR, 13 C-NMR and CHN analysis.

Abstract:The La(OTf)(3)-catalyzed [3+2] cycloaddition reactions for the synthesis of benzo[d]oxazoles/benzofurans via quinones and 1,2-di-tert-butyl-3-(cyanimino)diaziridine (1,3-di-tert-butyl-2-cyanoguanidine)/vinyl azides have been explored. A series of 5-hydroxybenzofuran-4-carboxylic acid derivatives and 5-hydroxybenzo[d]oxazole-4-carboxylic acid derivatives were conveniently obtained with high yields and good stereoselectivities, which could be used for further transformations to valuable compounds.

Abstract:A Rh(III)-catalyzed reaction of N-methoxybenzamides and 4-diazoisochroman-3-imines is described. This method offers a rapid entry to 3-amino-4-arylisoquinolinone architectures from readily available starting materials under mild reaction conditions. A one-pot cascade C-H bond activation/formal [4 + 2] cycloaddition mechanism is proposed. The synthesized 3-amino-4-arylisoquinolinones can be conveniently converted into dibenzo[c,f][1,8]naphthyridines.

Abstract:A Rh(III)-catalyzed C-H activation/cyclization cascade was developed for the first divergent synthesis of pyrazolo[1,5-a]quinazolines through a [5 + 1] annulation reaction exclusively. The one-pot procedure is recognized for its broad substrate scope, functional group tolerance, and high atom economy. Mechanistic studies reveal the reaction pathway, addressing current limitations. Notably, this catalytic transition metal-assisted tandem annulation smoothly proceeds through the reaction of substituted phenyl-1H-pyrazol-5-amine with an alkyne ester or amide, where a one ring carbon is provided by the alkynoate building block. This transformation highlights the potential of transition metal-catalyzed methods for synthesizing diverse pyrazolo[1,5-a]quinazoline frameworks.

Abstract:A new efficient synthesis of polysubstituted benzo[d]pyrido[1′,2':1,2]imidazo[4,5-f][1,3]diazepines and pyrido[2′,1':2,3]imidazo[1,5-c]quinazolin-7-ium salts via sequential Groebke-Blackburn-Bienaymé/Staudinger/aza-Wittig/carbodiimide-mediated annulation has been developed. The three-component Groebke-Blackburn-Bienaymé reactions of 2-azidobenzaldehydes 1 , isocyanides 2 , and 2-aminopyridines 3 produced the azide intermediates 4 , which were treated sequentially with triphenylphosphine and isocyanates to give polysubstituted benzo[d]pyrido[1′,2':1,2]imidazo[4,5-f][1,3]diazepines and pyrido[2′,1':2,3]imidazo[1,5-c]quinazolin-7-ium chlorides in good yields.

Abstract:We herein described a nickel-catalyzed cyclization of N-(o-ethynylaryl)acrylamides for the selective synthesis of dihydrocyclobuta[c]quinolin-3-ones and benzo[b]azocin-2-ones. The two varied products could be easily obtained by tuning the reaction temperature. This reaction features easy temperature-control, high efficiency, and gram-scale synthesis.

Abstract:Diverse quinazolinone-[2,3]-fused polycyclic skeletons occupy a prominent position in drug discovery. Even with currently available methods there still remain unmet needs for flexible access to such structures. Herein, we have explored a mild "one pot" procedure for the construction of various quinazolinone-[2,3]-fused polycycles. The procedure involves Pd-catalyzed carbonylation of N -(2-iodophenyl)acetamides, release of the masked terminal amine, and two sequential and spontaneous cyclizations. This generally applicable approach features easy assembly of precursors from readily available starting materials, mild reaction conditions, non-cumbersome operation, and polycyclic diversity.

Abstract:An oxidative [3+1+1] convergent domino cyclization is disclosed. This protocol enables to get quinoline, quinoxaline, quinazolin‐4(3H)‐one and benzo[d]thiazole attached 2,4,5‐trisubstituted oxazoles from methyl azaarenes, benzoins and NH 4 OAc in the presence of iodine and molecular sieves without any metal catalyst.
