Abstract:Rh(III)-catalyzed C(sp(3)) - H bond aminocarbonylation of 8-methylquinolines and isocyanates has been realized under mild conditions. This approach is applicable to different aryl and alkyl isocyanates, leading to the synthesis of various alpha-quinolinyl amide compounds in moderate to excellent yields. A plausible mechanism for this transformation is proposed according to the experimental results obtained.

Abstract:The preparation of a new class of aliphatic amino iodane(III) reagents has been realized with the N‐TMS‐amine and acetoxybenziodazole participated in the formation of N‐I bond. The amino‐containing iodane(III) reagent 2d was characterized by single‐crystal X‐ray diffraction, which revealed the expected hypervalent iodane distorted T‐shaped geometry. A practical copper‐catalyzed, directed electrophilic amination of aryl amines employing amino‐iodane(III) as amination agents was disclosed that proceeded smoothly without external additives.

Abstract:A series of 4-spiro-cyclohexadienonyl-β-lactam-3-carbonitriles, 2,7-dioxo-1-azaspiro[3.5]nona-5,8-diene-3-carbonitriles, was synthesized in satisfactory to excellent yields via the intramolecular nucleophilic cyclization of N-(p-hydroxyphenyl)cyanoacetamides with iodobenzene diacetate (IBD) as oxidant and potassium hydroxide as base. Acetic 4-spiro-cyclohexadienonyl-β-lactam-3-carbimidic anhydrides were obtained when organic base triethylamine was applied instead of potassium hydroxide. The mechanisms of the intramolecular nucleophilic cyclization and formation of acetic β-lactam-3-carbimidic anhydrides were proposed. The cyclization is a sequence of nucleophilic ipso addition and oxidative dearomatization. The formation of acetic carbimidic anhydrides is an acid-catalyzed acetate addition to the nitriles.

Abstract:An additive-assisted iridium-catalyzed directed C(sp3)-H amination with 2,2,2-trichloroethoxycarbonyl azide as an amino source is reported. Both carboxylate anions and the corresponding cations in the additives are crucial to achieve satisfactory efficiency. Sodium acetate or n-pentanoic acid can promote the amination of various primary C(sp3)-H bonds adjacent to secondary, tertiary, and quaternary carbons in ketoximes or N-aromatic heterocycles, respectively, providing a practical route to versatile β-amino ketoxime and N-heteroaryl ethanamine derivatives. The amination products can be treated as isocyanate analogues and can be converted to other useful amino functionalities. An iridacyclic compound was isolated and identified as a plausible intermediate.

Abstract:Here, we report the selective C(sp3)-C(sp3) cleavage/alkynylation of cycloalkylamides for the aminoalkyne synthesis under mild photoredox catalysis conditions. γ- and δ-Aminoalkynes with versatile alkyne and amine substituents are efficiently constructed from cyclopropylamides and cyclobutylamides via amidyl radicals enabled by hypervalent iodine(III) reagents. The catalytic amount of cyclic iodine(III) BI'-OAc facilitated the single-electron oxidation and ring-opening alkynylation of cycloalkylamides, which were investigated by a series of mechanistic probing experiments. Abstract:The condensation of N-acylated amino acids with polymer-supported cyclic malonic acid ester was carried out in the presence of DCC/DMAP. The cyclisation of the intermediate resin, by heating in an organic solvent, gave the N-protected tetramic acid derivatives in good yields and high purity.

Abstract:A novel zinc-catalyzed hypervalent iodine mediated cascade transformation of anilines and aldehydes to benzimidazoles was developed. A series of 1,2-disubstituted benzimidazoles were achieved with aromatic, heteroaromatic, and aliphatic aldehydes. In this procedure, initial condensation of anilines with aldehydes afforded N-arylimine intermediates which underwent C-H/C-H amination/annulation to generate the desired products in moderate to excellent yields. The reaction features an extremely simple reaction system, and good functional group toleration with water as the only byproduct.

Abstract:A method for the construction of cyclophane-braced peptide macrocycles via Pd-catalyzed aminoquinoline-directed intramolecular C(sp<sup>3</sup>)–H arylation with aryl iodides is developed. Unlike our previous AQ-directed exo-type intramolecular C–H arylation of long alkyl tails, this endo-type C–H cyclization reaction takes places on the β-methyl group of <i>N</i>-methyl alanine at the C-termini of peptides. Abstract:The development of C(sp3 )-H functionalizations of free carboxylic acids has provided a wide range of versatile C-C and C-Y (Y=heteroatom) bond-forming reactions. Additionally, C-H functionalizations have lent themselves to the one-step preparation of a number of valuable synthetic motifs that are often difficult to prepare through conventional methods. Herein, we report a β- or γ-C(sp3 )-H carbonylation of free carboxylic acids using Mo(CO)6 as a convenient solid CO source and enabled by a bidentate ligand, leading to convenient syntheses of cyclic anhydrides. Among these, the succinic anhydride products are versatile stepping stones for the mono-selective introduction of various functional groups at the β position of the parent acids by decarboxylative functionalizations, thus providing a divergent strategy to synthesize a myriad of carboxylic acids inaccessible by previous β-C-H activation reactions. The enantioselective carbonylation of free cyclopropanecarboxylic acids has also been achieved using a chiral bidentate thioether ligand.

Abstract:A hypervalent iodine(III) reagent plays a novel role as an efficient coupling reagent to promote the direct condensation between carboxylic acids and alcohols or amines to provide esters, macrocyclic lactones, amides, as well as peptides without racemization. The regeneration of iodosodilactone (1) can also be readily achieved. The intermediate acyloxyphosphonium ion C from the activation of a carboxylic acid is thought to be involved in the present esterification reaction.

Abstract:The cobalt(II) complex of 3,5-Di(t)Bu-IbuPhyrin, [Co(P1)], is an effective catalyst for intramolecular amination of electron-deficient C H bonds, including those adjacent to electron-withdrawing CO2R, C(O)NR2, C(O)R, and CN groups, in excellent yields with high regio- and stereoselectivity. Abstract:The Iodine-mediated oxidative intramolecular amination of anilines via cleavage of unactivated (sp3)C-H and N-H bonds for the production of indolines is described. This transition-metal-free approach provides a straightforward strategy for producing (sp3)C-N bonds for use in the preferential functionalization of unactivated (sp3)C-H bonds over (sp2)C-H bonds. The reaction could be performed on a gram scale for the synthesis of functionalized indolines.

Abstract:Amide compounds are important organic compounds, which play an important role in biomedical chemistry, materials science, life science, and other fields. The synthesis of α-CF3 amides, especially compounds containing 3-(trifluoromethyl)-1,3,4,5-tetrahydro-2H-benzo[b][1,4]diazepine-2-one, has long been a challenge due to the tensile properties and instability of the rings. Here, we report an example of palladium-catalyzed carbonylation of CF3-containing olefin to form α-CF3 acrylamide. By controlling the ligands, we can get different amide compounds as products. This method has good substrate adaptability and functional group tolerance.
