Abstract:Peptide-based supramolecular hydrogels have been extensively explored in biomaterials owing to their unique bioactive, stimulus-responsive, and biocompatible features. However, peptide-based hydrogels often have low mechanical stability with storage moduli of 10-1000 Pa. They are susceptible to mechanical destruction and solvent erosion, greatly hindering their practical application. Here, we present a photo-cross-linking strategy to enhance the mechanical stability of a peptide-based hydrogel by 10(4)-fold with a storage modulus of ~100 kPa, which is one of the highest reported so far for hydrogels made of small peptide molecules. This method is based on the ruthenium-complex-catalyzed conversion of tyrosine to dityrosine upon light irradiation. The reinforcement of the hydrogel through photo-cross-linking can be achieved within 2 min thanks to the fast reaction kinetics. The enhancement of the mechanical stability was due to the formation of a densely entangled fibrous network of peptide dimers through a dityrosine linkage. We showed that in order to implement this method successfully, the peptide sequence should be rationally designed to avoid the cross talk between self-assembly and cross-linking. This method is convenient and versatile for the enhancement of the mechanical stability of tyrosine-containing peptide-based hydrogels. We anticipate that the photo-cross-linked supramolecular hydrogels with much improved mechanical stability will find broad applications in tissue engineering and drug controlled release.

Abstract:The review describes the use of enzyme catalysis and self-assembly, two essential and ubiquitous processes in biology, to create molecular nanofibers of peptide derivatives at the supramolecular level as potential nanomedicines and biomaterials. In particular, we discuss the use of enzymes to instruct the self-assembly of small peptide derivatives for hydrogelation, which takes place in vitro or in vivo, extra- or intracellularly as a new strategy to detect the activity of enzymes, screen enzyme inhibitors, type bacteria, develop drug delivery systems, enhance the activity and stability of enzymes, and control the fete of cells. Abstract:Dipeptide self-assembled hydrogels have potential biomedical applications because of their great biocompatibility, bioactivity, and tunable physicochemical properties, which can be modulated in the molecular level by design of amino acid sequences. Herein, a series of dipeptides (Fmoc-FL, -YL, -LL, and -YA) are designed to form shear-thinning hydrogels with self-healing and tunable mechanical properties by adjusting the synergetic effect of hydrophobic interactions (π-π stacking and hydrophobic effect) and hydrogen bonds of peptides through substitution of amino acid residues. The enhancement of hydrophobic interactions is a primary factor to promote mechanical rigidity of hydrogels, and strong hydrogen-bonding interactions between molecules contribute to the instantaneous self-healing property, which is supported by experimental studies (FTIR, CD, SEM, AFM, and rheology) and molecular dynamics simulations. The injectable dipeptide hydrogels were certified as an ideal endoscopic submucosal dissection filler to make operation convenient and secure in mice and living mini-pig's experiments with a longer duration time, higher stiffness, and lower inflammatory response than commercial clinical fillers.

Abstract:While supramolecular hydrogels have received grow-ing interest due to their unique dynamic features, their relatively weak mechanical properties have largely limited their biomedical applications. In this study, we propose and demonstrate a strategy to reinforce the mechanical properties of supramolecu-lar hydrogel by introducing polymeric multiple-unit linker (PMUL), which incorporates multiple supramo-lecular units into a polymeric backbone to crosslink supramolecular hydrogel. We demonstrated that PMUL can effectively improve the kinetic stability of supramolecular crosslinkers through multiple-unit in-teraction in a DNA supramolecular hydrogel model system, thus leading to higher mechanical strength. Meanwhile, the dynamic features of the supramolec-ular hydrogels have been well preserved, including shear-thinning, self-healing properties, and revers-ible thermal responsiveness. This strategy offers a simple but effective way for mechanical reinforce-ment of supramolecular hydrogels to construct nov-el biomaterials.

Abstract:In this work, the mechanical behavior of sparsely cross-linked, neutral polyacrylamide (PAAm) hydrogels containing densely cross-linked polyelectrolyte microgels of poly(2-acrylamido-2-methylpropanesulfonic sodium) (PNaAMPS) were studied systematically by varying the formulations. The microgel-reinforced (MR) hydrogels have a two-phase composite structure, where the disperse phase is the rigid double-network (DN) microgels, and the continuous phase is the soft PAAm matrix. At the optimal formulation, the MR gels showed high mechanical strength and toughness, comparable to conventional DN hydrogels. The two critical parameters for the substantial enhancement of mechanical strength and toughness of MR gels are the concentration of PNaAMPS microgel and the molar ratio of the PAAm to the PNaAMPS in the microgel phase. Selective dyeing of the embedded microgels in MR gels allowed for visualization of the deformation of microgels, and we found that the local strain of microgels was much smaller than the global strain applied on MR gels; this indicates that isostress model (Reuss's model) is more suitable than isostrain model (Voigt's model) for this composite system.

Abstract:Learning from the stabilization of protein structures, we combine aromatic-aromatic and electrostatic interactions to tailor the elasticity of supramolecular hydrogels. Specifically, calcium ions allow interfiber cross-links among the supramolecular nanofibers of small peptides that consist of multiple carboxylic acid and aromatic groups. A small change in the concentration of calcium increases the elasticity of hydrogels over several orders of magnitude. This simple approach not only demonstrates a biomimetic design of materials but also confirms that the combination of multiple aromatic-aromatic interactions and multiple calcium salt bridges is a feasible way to control the mechanical properties of soft nanomaterials.

Abstract:Supramolecular hydrogels exhibit promising potential in biological and clinical fields due to their special dy-namic properties. However, most existing supramolecular hydrogels suffer from poor mechanical strength, which severely limits their applications. Here in this study, the Kinetically Interlocking Multiple-Units (KIMU) strategy was applied to the hyaluronan networks by introducing different supramolecular interaction motifs in an organized and alternative manner. Our strategy successfully elevated the energy barrier of crosslinker dissoci-ation to 103.0 kJ mol-1 and increased the storage modulus of hydrogels by 78 % with the intrinsic dynamic properties preserved. It can be expected that this method would bring a convenient and effective route to fabricate novel supramolecular materials with excellent mechanical properties.

Abstract:Polymer-supramolecular double-network hydrogels (PS-DN hydrogels) often shows much improved recovery rates than conventional double-network hydrogels because of the fast self-assembling properties, making them attractive candidates for tissue engineering and flexible electronics. However, as the supramolecular network is dynamic and susceptible to break under low strains, the overall mechanical properties of PS-DN hydrogels are still limited. Here, we report the mechanical properties for PS-DN hydrogels can be significantly improved by tuning the supramolecular network structures. A single amino acid change of the self-assembling peptide can tune the assembled structures from nanofiber to nanoribbon. Such a microscopic structural change can greatly increase the Young's modulus (107.4 kPa), fracture stress (0.48 MPa), and toughness (0.38 MJ m-3) of the PS-DN hydrogels. Moreover, the structural change also leads to slightly faster recovery rates (< 1 s). We propose that such dramatically different mechanical properties can be understood by the impact of individual peptide rupture events on the overall network connectivity in the two scenarios. Our study may provide new inspirations for combining high mechanical strength and fast recovery in double network hydrogels by tuning the supramolecular network structures.This article is protected by copyright. Abstract:Peptides as unique supramolecular nanofibrous materials are favored choices to fabricate hydrogel for artificial extracellular matrices. Herein, we devised an amphiphilic polypeptide that could be motivated by multiple substances with positively charged group at physiological temperature and pH to assemble supramolecular nanofiber hydrogels since the electrostatic repulsion between monomers was attenuated. The hydrogelation phenomenon and spectroscopy were compatible with molecular dynamics simulations (MDs) results for revealing the assembly mechanism of this peptide, confirming that the behavior of assembling and the typical secondary structure of the assemblies was β -sheet. The impact of peptide concentration on the mechanical properties of hydrogels were described using rheology. We found that the peptide formed elastic gels from 0.3 wt% onwards in practical use and that these recovered by themselves after shear thinning that indicated they were injectable hydrogels. The successful establishment of cell spheres with high cell viability during 15 days culture in 3D space demonstrated the nanofiber hydrogels had potential in cell model construction and microtissue culture. Overall, the superior biologically friendly gel-forming conditions and excellent biological properties of this supramolecular assembly peptide material have invigorated the progress of biomedical fields such as tissue regeneration based on in vitro 3D culture.

Abstract:A precursor of a β-peptide derivative transforms into a hydrogelator and forms supramolecular hydrogels upon phosphatase-catalyzed dephosphorylation. This new strategy works in vitro and in vivo (see image). The enzymatic hydrogelation in vivo indicates that the β-peptide-based hydrogel exhibits a longer lifetime than that of α-peptide hydrogel in a subcutaneous environment. Such an excellent in vivo stability renders the β-peptide-based hydrogel as a potential candidate for biomedical applications.

Abstract:Supramolecular hydrogels display unique dynamic features such as self-healing and injectability which are endowed by their non-covalent crosslinking. These extraordinary dynamic properties enable them to be broadly applied in various fields, but also decrease their mechanical strength. This inherent contradiction between mechanical strength and dynamic features has posed great challenge to their practical applications. This article provides an overview of emerging techniques for mechanical strengthening of supramolecular hydrogels with preserved dynamic features. Inspired by the "strong means slow" principle of transient supramolecular networks, particular focus is placed on how the exchange kinetics of the supramolecular crosslinkers can be engineered to tune their mechanical strength. Some other effective strengthening strategies are also reviewed, which are usually combined to realize the coexistence of mechanical strength and dynamics. Meanwhile, we point out the future development of supramolecular hydrogels with unique mechanical properties and their potential challenges.

Abstract:Peptide-based hydrogels have gained considerable attention as a compelling platform for various biomedical applications in recent years. Their attractiveness stems from their ability to seamlessly integrate diverse properties, such as biocompatibility, biodegradability, easily adjustable hydrophilicity/hydrophobicity, and other functionalities. However, a significant drawback is that most of the functional self-assembling peptides cannot form robust hydrogels suitable for biological applications. In this study, we present the synthesis of novel peptide-PEG conjugates and explore their comprehensive hydrogel properties. The hydrogel comprises double networks, with the first network formed through the self-assembly of peptides to create a β-sheet secondary structure. The second network is established through covalent bond formation via N-hydroxysuccinimide chemistry between peptides and a 4-arm PEG to form a covalently linked network. Importantly, our findings reveal that this hydrogel formation method can be applied to other peptides containing lysine-rich sequences. Upon encapsulation of the hydrogel with antimicrobial peptides, the hydrogel retained high bacterial killing efficiency while showing minimum cytotoxicity toward mammalian cells. We hope that this method opens new avenues for the development of a novel class of peptide-polymer hydrogel materials with enhanced performance in biomedical contexts, particularly in reducing the potential for infection in applications of tissue regeneration and drug delivery.

Abstract:Proteins are classified as biopolymers which share similar structural features with semi-crystalline polymers. Although their unique biocompatibility facilitates the universal applications of protein-based hydrogels in the biomedical field, the mechanical performances of protein-based hydrogels fall short of practical requirements. Conventional strategies for enhancing mechanical properties focus on forming regularly folded secondary structures as analogs of crystalline regions. This concept is based on proteins as the analogy of semi-crystalline polymers, in which crystalline regions profoundly contribute to the mechanical performances. Even though the contribution of the amorphous region is equally weighted for semi-crystalline polymers, their capacity to improve the mechanical performances of protein-based structures is still undervalued. Herein, the potential of promoting the mechanical performances is explored by controlling the state of amorphous regions in protein-based hydrogels. A fibril protein is chosen, regenerated silk fibroin (RSF), as a model molecule for its similar viscoelasticity with a semi-crystalline polymer. The amorphous regions in the RSF hydrogels are transformed from extended to entangled states through a double-crosslinking method. The formation of entanglement integrates new physically crosslinked points for remarkable improvement in mechanical performances. A robust hydrogel is not only developed but also intended to provide new insights into the structural-property relationship of protein-based hydrogels.
A General Protein Unfolding‐Chemical Coupling Strategy for Pure Protein Hydrogels with Mechanically Strong and Multifunctional Properties

Abstract:Protein-based hydrogels have attracted great attention due to their excellent biocompatible properties, but often suffer from weak mechanical strength. Conventional strengthening strategies for protein-based hydrogels are to introduce nanoparticles or synthetic polymers for improving their mechanical strength, but often compromise their biocompatibility. Here, a new, general, protein unfolding-chemical coupling (PNC) strategy is developed to fabricate pure protein hydrogels without any additives to achieve both high mechanical strength and excellent cell biocompatibility. This PNC strategy combines thermal-induced protein unfolding/gelation to form a physically-crosslinked network and a -NH2/-COOH coupling reaction to generate a chemicallycrosslinked network. Using bovine serum albumin (BSA) as a globular protein, PNC-BSA hydrogels show macroscopic transparency, high stability, high mechanical properties (compressive/tensile strength of 115/0.43 MPa), fast stiffness/toughness recovery of 85%/91% at room temperature, good fatigue resistance, and low cell cytotoxicity and red blood cell hemolysis. More importantly, the PNC strategy can be not only generally applied to silk fibroin, ovalbumin, and milk albumin protein to form different, high strength protein hydrogels, but also modified with PEDOT/PSS nanoparticles as strain sensors and fluorescent fillers as color sensors. This work demonstrates a new, universal, PNC method to prepare high strength, multi-functional, pure protein hydrogels beyond a few available today.

Abstract:Hydrogels are a class of biomaterials used in the field of tissue engineering and drug delivery. Many tissue engineering applications depend on the material properties of hydrogel scaffolds, such as mechanical stiffness, pore size, and interconnectivity. In this work, we describe the synthesis of peptide/polymer hybrid double-network (DN) hydrogels composed of supramolecular and covalent polymers. The DN hydrogels were prepared by combining the self-assembled pentafluorobenzyl diphenylalanyl aspartic acid (PFB-FFD) tripeptide for the first network and the polymeric PNIPAM-PEGDA copolymer for the second network. During this process, self-assembled peptide nanostructures are cross-linked to the polyacrylamide group in the polymer network through non-covalent interactions. The PNIPAM-PEGDA:PFB-FFD hydrogel exhibited higher mechanical stiffness ( G ′ ∼2 kPa) than the PNIPAM-PEGDA copolymer. Moreover, PNIPAM-PEGDA:PFB-FFD hydrogel shows a decrease in pore size (∼1.2 μm) compared to the original copolymer (∼5.2 μm), with the structural framework of highly interconnected fibrous peptide network. The mechanical stiffness of hydrogels was systematically investigated by rheological analysis in response to various variables, including UV exposure time, concentration of peptides, and amino acid functionalization. Modulating the time of UV irradiation resulted in PNIPAM-PEGDA:PFB-FFD hydrogels with a four-fold increase in stiffness. The influence of amino acid side chains and terminal charge of peptides on the strength of DN hydrogels was also investigated using pentafluorobenzyl diphenylalanyl lysine (PFB-FFK). Interestingly, PFB-FFK, which has an amine group on the side chain, does not exhibit the DN structures. The mechanical properties and pore sizes of PNIPAM-PEGDA:PFB-FFK hydrogel were very similar to those of the PNIPAM-PEGDA copolymer due to poor cross-linking. The biocompatibility of the hydrogel materials was tested with the hMSC cell line using the MTT method, and the results indicate that the materials are non-toxic and potentially useful for biological applications.

Abstract:This highlight summarizes recent advances in the enzymatic formation of supramolecular hydrogels, a new method for the creation of biofunctional soft matter. Using phosphatase, thermolysin, β-lactamase, and phosphatase/kinase as examples, we illustrate the design and application of enzyme catalyzed or regulated formation of supramolecular hydrogels. This approach provides a new strategy to detect the presence of enzymes, screen enzyme inhibitors, assist biomineralization, assay the types of bacteria, and aid the development of smart drug delivery systems. The strategy of using enzymes to control the self-assembly of small molecules described in this highlight will lead to the further development of new materials for biomedical applications and improve understanding of molecular self-assembly in water.

Abstract:Biological load-bearing tissues are strong, tough, and recoverable under periodic mechanical loads. However, such features have rarely been achieved simultaneously in the same synthetic hydrogels. Here, we use a force-coupled enzymatic reaction to tune a strong covalent peptide linkage to a reversible bond. Based on this concept we engineered double network hydrogels that combine high mechanical strength and reversible mechanical recovery in the same hydrogels. Specifically, we found that a peptide ligase, sortase A, can promote the proteolysis of peptides under force. The peptide bond can be re-ligated by the same enzyme in the absence of force. This allows the sacrificial network in the double-network hydrogels to be ruptured and rebuilt reversibly. Our results demonstrate a general approach for precisely controlling the mechanical and dynamic properties of hydrogels at the molecular level.

Abstract:A design scheme for preparing PAM/APEI hydrogel with the dissipate energy mechanism under the loads. Appropriate mechanical properties is one of the prerequisites for the applications of hydrogels, where cross‐linker is a crucial component in manipulating such mechanical property. Herein, acryloyl‐functionalized polyethyleneimine (APEI) was innovatively used as novel functional cross‐links to optimize the mechanical properties of polyacrylamide (PAM) hydrogels. APEI cross‐links were synthesized by simply adding acryloyl chloride into polyethyleneimine in aqueous solution. The chemical structures of APEI were confirmed by Fourier transform infrared and nuclear magnetic resonance spectra. APEI possessed massive highly acryloyl functional groups that can serve as multifunctional cross‐linking centers for the polymerization of monomers via covalent bonding interactions. These covalent bonds are favorable for constructing a firm 3D cross‐linked networks for hydrogels to withstand deformations, endowing hydrogels with the improved mechanical properties. The tensile strength with an elongation at break for the hydrogels can reach 234 kPa and 18.4‐fold, respectively. In addition, even the hydrogels can bear cyclic mechanical loads and deformations for 50 cycles at 5‐fold stain, indicating a good fatigue resistance. The deformability of APEI cross‐links clusters under stretching was responsible for the stretchability of PAM/APEI hydrogels. This work provides a simple strategy to enhance the mechanical properties of hydrogels via the development of novel APEI chemical cross‐linker.

Abstract:Self-assembled peptide-based nanomaterials have exhibited wide application potential in the fields of materials science, nanodevices, biomedicine, tissue engineering, biosensors, energy storage, environmental science, and others. Due to their porous structure, strong mechanical stability, high biocompatibility, and easy functionalization, three-dimensional self-assembled peptide hydrogels revealed promising potential in bio-related applications. To present the advances in this interesting topic, we present a review on the synthesis and functionalization of peptide hydrogels, as well as their applications in drug delivery, antibacterial materials, cell culture, biomineralization, bone tissue engineering, and biosensors. Specifically, we focus on the fabrication methods of peptide hydrogels through physical, chemical, and biological stimulations. In addition, the functional design of peptide hydrogels by incorporation with polymers, DNA, protein, nanoparticles, and carbon materials is introduced and discussed in detail. It is expected that this work will be helpful not only for the design and synthesis of various peptide-based nanostructures and nanomaterials, but also for the structural and functional tailoring of peptide-based nanomaterials to meet specific demands.
