Abstract:Thirteen 1-deoxynojirimycin (DNJ) derivatives of five different skeletal structures were designed and synthesized. The newly synthesized compounds were evaluated using an in vitro alpha-glucosidase assay, and kinetic parameters (Ki, IC50) were measured. Some DNJ derivatives showed weak alpha-glucosidase inhibitory activities, and the compounds 1-(3-benzyloxy-2-hydroxypropy1)2-hydroxymethyl-piperidine-3,4,5-triol (2a) and 1-{3[1-(4-fluoropheny1)-1H-[1,2,3]triazol-4-ylmethoxy]-2-hydroxypropyl}-2-hydroxymethyl-piperidine-3,4,5-triol (13d) showed activities comparable to that of DNJ. While 2a was found to be a reversible, non-competitive inhibitor of alpha-glucosidase with a Ki value of 1.56 x 10(-4) M and an IC50 value of 3.07 x 10(-4) M, 13d was a reversible, competitive inhibitor of alpha-glucosidase with a Ki value of 2.08 x 10(-4) M and an IC50 value of 3.31 x 10(-4) M.

Abstract:To obtain α-glucosidase inhibitors with high activity, 19 NB-DNJDs (N-benzyl-deoxynojirimycin derivatives) were designed and synthesized. The results indicated that the 19 NB-DNJDs displayed different inhibitory activities towards α-glucosidase in vitro. Compound 18a (1-(4-hydroxy-3-methoxybenzyl)-2-(hydroxymethyl) piperidine-3,4,5-triol) showed the highest activity, with an IC50 value of 0.207 ± 0.11 mM, followed by 18b (1-(3-bromo-4-hydroxy-5-methoxybenzyl)-2-(hydroxymethyl) piperidine-3,4,5-triol, IC50: 0.276 ± 0.13 mM). Both IC50 values of 18a and 18b were significantly lower than that of acarbose (IC50: 0.353 ± 0.09 mM). According to the structure-activity analysis, substitution of the benzyl and bromine groups on the benzene ring decreased the inhibition activity, while methoxy and hydroxyl group substitution increased the activity, especially with the hydroxyl group substitution. Molecular docking results showed that three hydrogen bonds were formed between compound 18a and amino acids in the active site of α-glucosidase. Additionally, an arene‒arene interaction was also modelled between the phenyl ring of compound 18a and Arg 315. The three hydrogen bonds and the arene‒arene interaction resulted in a low binding energy (–5.8 kcal/mol) and gave 18a a higher inhibition activity. Consequently, compound 18a is a promising candidate as a new α-glucosidase inhibitor for the treatment of type II diabetes.

Abstract:A facile and efficient synthesis of 6-O-methyl-1-deoxynojirimycin 4 from commercially available methyl alpha-D-glucopyranoside in 10 steps and 25% overall yield was reported. The synthetic strategy was based on the regioselective protection/deprotection at 6-O-position and cyclization as the main reaction steps. Compound 4 was evaluated for its inhibitory effect (IC50) on alpha-glucosidase using an in vitro assay. It showed improved inhibitory activity compared to 1-deoxynojirimycin and acarbose.

Abstract:A series of N-alkylated deoxynojirimycin (DNJ) derivatives connected to a terminal tertiary amine at the alkyl chains of various lengths were prepared. These novel synthetic compounds were assessed for preliminary glucosidase inhibition and anticancer activities in vitro. Potent and selective inhibition was observed among them. Compound 7d (IC50 = 0.052 mM) showed improved and selective inhibitory activity against β-glucosidase compared to DNJ (IC50 = 0.65 mM). In addition, analysis of the kinetics of enzyme inhibition by using Lineweaver–Burk plots indicated that 7d inhibited β-glucosidase in a competitive manner, suggesting that 7d was expected to bind to the active site of β-glucosidase. Compounds 8b and 8c were found to be moderate and selective inhibitors of α-glucosidase. Nevertheless, none of compounds inhibited the growth of B16F10 melanoma cells.

Abstract:1-Deoxynojirimycin(DNJ) is an effective α-glucosidase inhibitor,being able to lower blood glucose efficiently.Nojirimycin was firstly found in streptomyces,while DNJ was firstly isolated from the root bark of mulberry tree.DNJ has been isolated and identified from mulberry(Morus L.),dayflower(Commelina communis),hyacinth(Hyacinthus orientalis) and adenophora(Adenophora triphylla var.japonica),among which mulberry has the highest content of DNJ.DNJ content of mulberry varies greatly with mulberry varieties,medicament portions,seasons,climates,geographical and soil conditions,leaf positions,and development stages etc.In microorganisms,DNJ was isolated from various species of streptomycete and bacillus.It was also discovered that two endophytes from mulberry,Stenotrophomonas maltophilia and Micrococcus luteus,could produce DNJ stably.So far,fermentation conditions for many microorganisms with DNJ production have been investigated.In insect,besides silkworm(Bombyx mori) which has rich content of DNJ,other mulberry monophagous insect species or oligophagous ones including Bombyx mandarina,Rondotia menciana,Phthonandria atrineata,and Diaphani apyloalis also have high content of DNJ.DNJ in insect body comes from their food,and DNJ content in silkworm body varies with silkworm species,developmental periods,diet,tissue and organ etc.It was also noticed that absorption and concentration of DNJ in silkworm body changed periodically with the advance of larval instar.Researches aiming to elucidate biosynthetic pathway of DNJ in streptomycete,bacillus,and dayflower have been conducted.It was found that DNJ synthetic pathway is different in different species.Important progress has been made in chemosynthesis of DNJ.Three major synthetic methods have been developed and some synthesized DNJ derivatives have been used in clinic.

Abstract:1-deoxynojirimycin (DNJ) is a well-known α-glucosidase inhibitor. A series of phenyltriazole-deoxynojirimycin hybrids containing C4 and C6 (4 and 6 methylenes, respectively) linkers were synthesized. These novel compounds were assessed for preliminary glucosidase inhibition and cytotoxicity tests in vitro. Among them, compounds 12–14 and 16–20 (IC50: 105 ± 9–11 ± 1 μM) were more active than deoxynojirimycin (DNJ, IC50 = 155 ± 15 μM). The kinetics of enzyme inhibition measured by using Lineweaver–Burk plots indicated that compounds 18 and 19 were competitive inhibitors. In addition, a molecular docking study of α-glucosidase revealed that the interaction modes and the orientations of compound 18 and DNJ were clearly different. Furthermore, in tissue culture, HL60 cell compounds showed no cytotoxicity at low concentrations. When the concentration reached 50 μM, only compound 20 exhibited cytotoxicity. The structure–activity relationships exhibit that the length of the linker and the nature of 4-position substituents on the phenyl have a significant effect on the inhibitory potency of glucosidases and cytotoxicity.

Abstract:In order to discover highly potent alpha- glucosidase inhibitors, 20 xanthone-benzenesulfonamide derivatives were designed and synthesized. As expected, most of these derivatives exhibited good to excellent in-hibitory activities, the IC50 values of these xanthone sulfonamides varied from 0.4 +/- 0.05 to 52.2 +/- 2.1 mu M. Especially, compound 4ag showed the most potent activity (IC50 = 0.4 mu M, with 111-fold more potent than the positive control 1-deoxynojirimycin (DNJ), which is a leap compared to our previous xanthone compound. It was noteworthy that these derivatives can be competitive, noncompetitive or un-competitive inhibitors, while these inhibitory types differed by the alteration of the substituents on the benzene ring of benzenesulfonamide. To give insight into this interesting effect, further docking study was performed, indicating the flexible conformations introduced by the sulfonamide structure affect the binding site and binding mode of these compounds with the enzyme. Furthermore the uncompetitive inhibitors showed significantly stronger binding interactions with the enzyme-substrate complex than the enzyme. Additionally, the competitive inhibitor 4aj (1 mu M) exhibited a similar glucose-level decreasing and glucose up-take increasing effect com pared to DNJ (5 mu M), the positive control in HepG2 cells at five-fold lower concentration, revealing that this compound has promising hypoglycemic effect at the cellular level. Abstract:A series of deoxynojirimycin-triazole hybrid iminosugars had been prepared and investigated against a panel of glucosidases. Compound 10f (IC50 = 0.063 +/- 0.006 mM) exhibited improved inhibitory potency against alpha-glucosidase compared to 1-deoxynojirimycin (IC50 = 0.155 +/- 0.015 mM). Moreover, analysis of the kinetics of enzyme inhibition by using Lineweaver-Burk plots indicated that 10f inhibited alpha-glucosidase in a competitive manner. Compound 10m displayed moderate inhibition of alpha-glucosidase and beta-glucosidase. The structure-activity relationships indicated that the introduction of the phenyl group and the nature of 4-position substituents on the phenyl ring had significant effects on the glucosidase inhibitory potency of the compounds.

Abstract:A series of novel xanthone derivatives 6–16 having non-coplanar and flexible structures were synthesized as potent α-glucosidase inhibitors. Biological evaluation indicated that compounds 6–12 bearing one or two naphthol moieties exhibited up to 30-fold enhanced activities compared with their corresponding parent compounds 2–5, whereas compounds 13–16 bearing one dihydroxylnaphthalenyl group showed decreased activities compared with their corresponding analogs 6–9 having one naphthol group. Among them, compounds 7–8, 10–12 and 15 were more active than 1-deoxynojirimycin, a well-known inhibitor for α-glucosidase. The structure–activity correlations suggested that inhibiting of α-glucosidase was a result of multiple interactions with the enzyme, including π-stacking, hydrophobic effect and conformational flexibility due to the structural non-coplanarity. In addition, compounds 4, 8 and 15 showed non-competitive inhibition.

Abstract:Two series of compounds (chalcones and bis-chalcones) were designed, synthesized, and evaluated as α-glucosidase inhibitors (AGIs) with 1-deoxynojirimycin as positive control in vitro. Most of the compounds with two or four hydroxyl groups showed better inhibitory activities than 1-deoxynojirimycin towards α-glucosidase with noncompetitive mechanism. Moreover, most of the hydroxy bis-chalcones exhibit good α-glucosidase inhibitory activities in enzyme test. Inspiringly, bis-chalcones 2g (at 1 μM concentration) has stronger effect than 1-deoxynojirimycin on reducing the glucose level in HepG-2 cells (human liver cancer cell line).

Abstract:A series of novel quinazoline-1-deoxynojirimycin hybrids were designed, synthesized and evaluated for their inhibitory activities against two drug target enzymes, epidermal growth factor receptor (EGFR) tyrosine kinase and α-glucosidase. Some synthesized compounds exhibited significantly inhibitory activities against the tested enzymes. Comparing with reference compounds gefitinib and lapatinib, compounds 7d, 8d, 9b and 9d showed higher inhibitory activities against EGFR (IC50: 1.79–10.71nM). Meanwhile the inhibitory activities of 7d, 8d and 9c against α-glucosidase (IC50=0.14, 0.09 and 0.25µM, respectively) were obvious higher than that of miglitol (IC50=2.43µM), a clinical using α-glucosidase inhibitor. Interestingly, compound 9d as a dual inhibitor showed high inhibitory activity to EGFRwt tyrosine kinase (IC50=1.79nM), also to α-glucosidase (IC50=0.39µM). The work could be very useful starting point for developing a new series of enzyme inhibitors targeting EGFR and/or α-glucosidase.

Abstract:1-Deoxynojirimycin (DNJ, C₆H13NO₄, 163.17 g/mol), an alkaloid azasugar or iminosugar, is a biologically active natural compound that exists in mulberry leaves and Commelina communis (dayflower) as well as from several bacterial strains such as Bacillus and Streptomyces species. Deoxynojirimycin possesses antihyperglycemic, anti-obesity, and antiviral features. Therefore, the aim of this detailed review article is to summarize the existing knowledge on occurrence, extraction, purification, determination, chemistry, and bioactivities of DNJ, so that researchers may use it to explore future perspectives of research on DNJ. Moreover, possible molecular targets of DNJ will also be investigated using suitable in silico approach.

Abstract:We recently described the discovery of oxygenated N-alkyl deoxynojirimycin (DNJ) derivative 7 (CM-10-18) with antiviral activity against dengue virus (DENY) infection both in vitro and in vivo. This imino sugar was promising but had an EC50 against DENY in BHK cells of 6.5 mu M, which limited its use in in vivo. Compound 7 presented structural opportunities for activity relationship analysis, which we exploited and report here. These structure activity relationship studies led to analogues 2h, 2l, 3j, 3l, 3v, and 4b-4c with nanomolar antiviral activity (EC50 = 0.3-0.5 mu M) against DENY infection, while maintaining low cytotoxicity (CC50 > 500 mu M, SI > 1000). In male Sprague-Dawley rats, compound 3l was well tolerated at a dose up to 200 mg/kg and displayed desirable PK profiles, with significantly improved bioavailability (F = 92 +/- 4%).
