Abstract:We report a label-free fluorescence turn-on approach for the sensitive and selective sensing of silver ion Ag(+) based on Ag(+) induced conformational change of cytosine-rich single-stranded DNA. A silver specific oligonucleotide (SSO) undergoes a structural transition from single-stranded DNA (ssDNA) to a C-Ag(+)-C mediated hairpin structure, which can be recognized by a double-stranded DNA (dsDNA) intercalator Sybr Green I (SG). The linear response range for Ag(+) detection was from 1 nM to 100 nM. The method presented here is highly sensitive and a limit of detection of 1 nM was obtained. More importantly, this optical method was successfully used to identify complex sample mixtures.

Abstract:Water-soluble cadmium telluride quantum dots capped with mercaptosuccinic acid (MSA-CdTe QDs) were prepared for the selective and sensitive fluorescence probing of silver(I) ion. The fluorescence intensity of the MSA-CdTe QDs could be quenched only in the presence of Ag+, and the relative fluorescence intensity was decreased as a linear function of Ag+ concentration. Moreover, the proposed sensor was free from common interfering metal ions. The MSA-CdTe QDs were 3.0 nm in particle size and the maximum photoluminescence wavelength was 558 nm. Better sensitivity was obtained with 4.5 x 10(-4) mol L-1 MSA-CdTe QDs in 200 mmol L-1 phosphate buffer (pH 9.0) for a reaction time of 30 min. A linear response between the relative fluorescence intensity, and the concentration of Ag+ was obtained over the range 0.4 to 8 mmol L-1, with a relative standard deviation of 3.6% (n = 5) for 1.5 mmol L-1 and a detection limit of 54 nmol L-1. The proposed method was applied for the determination of Ag+ in two fixer samples at the mmol L-1 level. The spike recovery at a spiked level of 15 mmol L-1 varying from 89.4% to 92.3%, and good agreement between the results by the proposed and AAS methods proved the accuracy of the proposed method.

Abstract:A simple method for the synthesis of water-soluble and well-dispersed fluorescent DNA-dots under mild conditions is reported. The solution of DNA-dots shows blue fluorescence and the luminescence lifetime equals 2.74 ns, with a quantum yield up to 7.5%. It is attractive that DNA-dots can be used as an effective fluorescent probe for the detection of iron ions with relatively good selectivity and sensitivity in an aqueous solution as well as in biological imaging applications.

Abstract:A sensitive photoelectrochemical (PEC) biosensor for silver ions (Ag+) was developed based on Zn-Co doped C and CdS quantum dots (CdS QDs) nanomaterials. Hydrophobic modified sodium alginate (HMA), which could stabilize and improve the PEC performance of CdS QDs, was also used for the construction of PEC sensors. Specially, Zn-Co doped C, CdS QDs and HMA were sequentially modified onto the electrode surface via the drop-coated method, and the C base rich DNA strand was then immobilized onto the modified electrode. As the C base in DNA specifically recognized Ag+, it formed the C-Ag+-C complex in the presence of Ag+, which created a spatial steric hindrance, resulting in a reduced PEC response. The sensing platform is sensitive to Ag+ in the range of 10.0 fM to 0.10 μM, with a limit of detection of 3.99 fM. This work offers an ideal platform to determine trace heavy metal ions in environmental monitoring and bioanalysis.

Abstract:A novel fluorescent Ag+ sensor was developed based on the label-free silver (I) specific oligonucleotide (SSO) and Thioflavine T (ThT) monomer-excimer switch. C-rich SSO which contain C-C mismatched base pairs can selectively bind to Ag+ ions and the formed duplexes which constructed by C-Ag+-C structure are thermally stabilized without largely altering the double helical structure. ThT give very weak fluorescent in bulk solution and/or in the presence of SSO. However ThT shows high fluorescence in the presence of SSO and Ag+ at the same time mainly because ThT excimer, which has the high quantum yield, formed and stabilized in the minor or major groove. Based on the discovery, we developed the novel Ag+ sensor. Under the optimum condition, the selectivity of this system for Ag+ over other metal ions in aqueous solution is remarkably high, and Ag+ can be quantified over the dynamic range of 30–450 nM, with a limit of detection of ~16 nM and a linear correlation coefficient of 0.995.

Abstract:In this study, a sensitive ratiometric fluorescent nanosensor was constructed using a facile one-pot method by encapsulating carbon dots (CDs) and cadmium telluride quantum dots (CdTe QDs) into the pore cavities of a metal-organic framework (ZIF-8). In this nanosensor (CD/CdTe QD@ZIF-8), the fluorescence attributed to CdTe QDs was quenched by silver ions (Ag+), and the fluorescence intensity of CDs did not change. The introduction of ZIF-8 into the system can not only adsorb Ag+ but also easily separate CDs and CdTe QDs from the matrix. The developed CD/CdTe QD@ZIF-8 composite used as a ratiometric fluorescent probe exhibited high sensitivity and selectivity towards Ag+. The working linear range was 0.1-20 μM with a limit of detection (LOD) of 1.49 nM. Finally, the proposed nanosensor was applied to determine Ag+ in lake water with satisfactory results.

Abstract:Improving the sensitivity of the fluorescence method is crucial in biosensor fields. In this study, an ultrasensitive and excellent selectivity method was developed for the detection of trace Cu2+ ions using 3-mercaptopropionic acid capped CdSSe/CdS/ZnS quantum dots (MPA-CdSSe/CdS/ZnS QDs) as a fluorescent probe. Through energy band engineering to optimize the interior inorganic structure and passivate the surface defects, the photoluminescence quantum yield of the MPA-CdSSe/CdS/ZnS QDs in aqueous solution had as high as 70 %. The X-ray diffraction (XRD) and Transmission Electron Microscope (TEM) analysis revealed the prepared CdSSe/CdS/ZnS QDs were cubic zinc blende structure with average diameter 7.2 nm. Further, the fluorescence of the MPA-CdSSe/CdS/ZnS QDs was significantly quenched by the Cu2+ with fast response time through an efficient photo-induced electron transfer (PET) progress. By optimizing the thickness of CdS shell, the MPA-CdSSe/CdS/ZnS QDs probe with appropriate CdS shell thickness exhibited excellent linear linearity and sensitivity for the detection of Cu2+ ions with limit of detection as low as 0.05 nM. Additionally, ions interference experiments demonstrated the good selectivity of the probe towards Cu2+. Furthermore, this sensing system could also be applied to the measurement of Cu2+ ions in real water samples with satisfactory recoveries. This work provided new insights to design and synthesize high-performance QDs-based fluorescence probes with controllable exciton emission behavior for their applications in biosensor and biological fields.

Abstract:A simple, low-cost and universal fluorescence “signal on” sensing strategy has been developed based on functional nucleic acids via energy transfer between DNA-templated silver nanoclusters (DNA-AgNCs) and gold nanorods (GNRs). Functional nucleic acids were used as highly selective recognition probes, in which an aptamer was used to detect small molecules represented by tetracycline and DNAzyme was used to detect heavy metal ions represented by Pb2+. The fluorescent DNA-AgNCs were synthesized by the designed oligonucleotide sequences, which consisted of three parts, including the AgNCs synthesis template C6G5C6, spacer T5 and complementary sequences of the aptamer or enzyme strand. The difference in electrostatic interactions between ss/dsDNA and positively charged GNRs leads to energy transfer with different efficiencies between ss/dsDNA-AgNCs and GNRs. The analytes represented by tetracycline and Pb2+ can destroy the dsDNA structure and reduce the energy transfer efficiency between DNA-AgNCs and GNRs, thus achieving fluorescence recovery and a "signal-on" analytical strategy. This strategy has excellent specificity and sensitivity with LODs of 106.3 pM for tetracycline and 58.9 pM for Pb2+ and has been successfully applied to detect tetracycline in milk and Pb2+ in river water. Using DNA-AgNCs formed in situ as signal probes, this strategy does not require labels or modifications and can be completed in 40 min without complex analytical instruments. Moreover, this strategy can be extended to detect other targets by replacing functional nucleic acid sequences. Therefore, it has promising prospects in the sensitive, simple and rapid detection of contaminants in food and environment samples.

Abstract:Simple and convenient assays with quantum dots (QDs) as the labels for DNA detection are developed. The probe DNA modified with thiol was first immobilized on a pretreated Au electrode, and then the complementary DNA (cDNA) oligonucleotides were hybridized with the immobilized probes by immersing the probe-modified Au electrode into the cDNA oligonucleotide solution. Finally, the avidin-modified QDs were bound to the biosensor in the presence of biotin-modified cDNA. The fabrication process for the biosensor was monitored by electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). Different from the traditional sandwich-structure strategy, the QDs bind to the target DNA directly via the biotin-avidin-system. By observing the ECL signal and determination of the cadmium component in QDs, the DNA hybridization event was detected by ECL and square wave anodic stripping voltammetric technique (SWASV) respectively. For SWASV detection, the signal linearly increased with the increase of the logarithm of the cDNA concentration over the range of 50 nM similar to 5 mu M. The minimum detectable concentration is 50 pM. For ECL, it showed wider linearity range over 5 nM similar to 5 mu M and lower detectable concentration of 10 pM. This indicated that the ECL assay could be comparable to the conventional electrochemical assay. Furthermore, this biosensor possesses high selectivity over different sequences of target DNA oligonucleotides.

Abstract:A new strategy to combine isothermal exponential amplification (IEA) with CdTe quantum dots (QDs) functionalized silica nanospheres as a label was designed for highly sensitive fluorescent detection of target DNA. A well plate was used as a support to immobilize a molecular beacon (MB) as a recognition probe and perform the IEA procedure. After the MB recognized target DNA and opened its cycle, the stem part could hybridize with a primer to initiate the polymerization of the DNA strand, which led to the release of target to open another MB molecule and start the next cycle of strand-replacement polymerization. Meanwhile, the formed double-stranded DNA was recognized by nicking endonuclease, leading to an endonuclease-based strand-replacement polymerization, which produced a DNA trigger to open more MB. The opened MB molecules were finally bound to the label by biotin-streptavidin coupling. Upon a dissolving process, the released cadmium cation could sensitize the fluorescent emission of Rhod-5N to achieve cascade signal amplification. The proposed method could detect target DNA ranging from 10(-17) to 10(-11) mol L-1 with a detection limit down to similar to 50 copies. It also showed high selectivity. This highly sensitive and specific assay has potential to become a promising DNA quantification method in biomedical research and clinical diagnosis.

Abstract:Anovel luminescence `Turn-On' nanoplatform for the sensitive sensing of Ag+ was fabricated based on luminescence resonance energy transfer technique between sodium citrate functionalized upconversion nanoparticles (Cit-UCNPs, energy donor) and graphene quantum dots (GQDs, energy acceptor). Amino-labeled single-strandedDNA(NH2-ssDNA) containing a number of cytosine (C) was conjugated on the surface of the Cit-UCNPs to capture Ag+ ions. Due to the p-p stacking interaction betweenNH(2)-ssDNA and GQDs, the upconversion luminescence can be quenched. However, upon the addition of Ag+, the p-p stacking interaction weakens due to the formation of the hairpin structure of C-Ag+-Con the UCNPs. As a result, GQDs will leave the surface of the UCNPs and the upconversion luminescence can be enhanced (Turn-On). Based on this fact, the sensor was developed for the detection of Ag+ with a linear concentration range from 2 x 10(-4) to 1 mu Mand a detection limit as low as 60 pM. The assay method is fairly simple with high selectivity and sensitivity, which can be used for the determination of Ag+ in environmental water samples.

Abstract:BACKGROUND: As an emerging and attractive low-dimensional functional materials, Ti3C2 MXene quantum dots (QDs) enlarge the toolbox of fluorescence sensing. However, monochromatic fluorescence, which only provide one single signal, is often beset by challenges such as false-positive readouts and limitations in selectivity. Consequently, to improve the sensing accuracy by means of cross-verified dual-signal authentication, the endeavor to engineer dual-mode nanoprobes based on Ti3C2 QDs, incorporating both the capability of fluorescence and an alternative sensing mechanism, emerges as a compelling avenue.RESULTS: Here, based on the alterations in colorimetric and fluorescent signals of Ti3C2 QDs with the addition of Ag+, we propose a dual-mode sensor obviating the necessity for nanoprobe labeling. Owing to the decent reducibility of Ti3C2 QDs, Ag+ is adsorbed and reduced, resulting in the generation of plasmonic Ag nanoparticles (NPs), which simultaneously trigger colorimetric responses of the solution and enhance the fluorescent emission of Ti3C2 QDs. The confluence of colorimetry and fluorometry within this strategy optimally harnesses the modulating role of the acquired Ag NPs on the reducing capability and fluorescence characteristics of Ti3C2 QDs. The equilibrium imparts versatility and promising prospects to this analyte-triggered label-free method, which enables a remarkable specificity and an excellent detecting limit (0.45 μM) for Ag+.SIGNIFICANCE: The balance between reducibility and fluorescence of Ti3C2 QDs for dual-mode detection is inventively demonstrated. With the exemplification of a direct influence of both features of the nanoprobe via the introduction of analytes, this study opens the feasibility of the analyte-perturbed felicitous equilibrium, which endows label-free methods with versatility and promising prospects. This design may evoke more biosensing strategies with the function of double-signal mutual verification.

Abstract:Silver ion (Ag+) is one of the most common heavy metal ions that cause environmental pollution and affect human health, and therefore, its detection is of great importance in the field of analytical chemistry. Here, we report an 8-nucleotide (nt) minidumbbell DNA-based sensor (M-DNA) for Ag+ detection. The minidumbbell contained a unique reverse wobble C·C mispair in the minor groove, which served as the binding site for Ag+. The M-DNA sensor could achieve a detection limit of 2.1 nM and sense Ag+ in real environmental samples with high accuracy. More importantly, the M-DNA sensor exhibited advantages of fast kinetics and easy operation owing to the usage of an ultrashort oligonucleotide. The minidumbbell represents a new and minimal non-B DNA structural motif for Ag+ sensing, allowing for the further development of on-site environmental Ag+ detection devices.

Abstract:Due to the high importance of detecting DNA with both fast speed and high sensitivity, we proposed a new dsDNA detection method relying on a novel single-color fluorescence “off–on” switch system. Water-soluble glutathione capped CdTe QDs (emission at 605nm) was prepared for taking advantage of the readily tunable emission property of QDs. Initially, QDs was completely quenched by the Ru(phen)2(dppz)2+, as the spontaneous formation of QDs-Ru assembling dyads. Then, in the case of the addition of dsDNA, the Ru(phen)2(dppz)2+ was removed away from the CdTe QDs, producing free CdTe QDs and the Ru-dsDNA complex. Both of them could be excited at the same wavelength and emit overlaid fluorescence. This single-color fluorescence “off–on” signal was sensitive to the concentration of dsDNA. Native dsDNA with the concentration of 10pg/mL could be detected when 0.5nM CdTe QDs was used, and ssDNA, RNA or BSA had no interference on it. With this system, the dsDNA samples of hepatitis B virus (HBV) patients were tested. The results were in good agreement with those detected by fluorescence quantitative PCR (P>0.05), and for those samples with very low DNA concentrations, this system could provide more accurate results, demonstrating the possible clinical applicability of this “off–on” switch system. For this system, chemical conjugation or labeling of probes is not required, and unmodified native DNA targets could be detected in less than half an hour. Therefore, a simple, fast, sensitive, low cost, highly selective and practically applicable detection system for dsDNA has been described.

Abstract:In the present paper, we report that CdSe quantum dots (QDs) can act as the coreactant of Ru(bpy)32+ electrogenerated chemiluminescence (ECL) in neutral condition. Strong anodic ECL signal was observed at ~1.10 V at CdSe QDs modified glassy carbon electrode (CdSe/GCE), which might be mainly attributed to the apparent electrocatalytic effect of QDs on the oxidation of Ru(bpy)32+. Ru(bpy)32+ can be intercalated into the loop of hairpin DNA through the electrostatic interaction to fabricate a probe. When the probe was bound to the CdSe QDs modified on the GCE, the intense ECL signal was obtained. The more Ru(bpy)32+ can be intercalated when DNA loop has larger diameter and the stronger ECL signal can be observed. The loop of hairpin DNA can be opened in the presence of target DNA to release the immobilized Ru(bpy)32+, which can result in the decrease of ECL signal. The decreased ECL signal varied linearly with the concentration of target DNA, which showed the ECL biosensor can be used in the sensitive detection of DNA.
