(Pst) in the world, except an isolate found in India and another in Australia (Nagarajan 1986; Wellings and McIntosh 1990). In June 2017, a stripe rust sample was obtained from a farmer’s winter wheat (cultivar unknown) field at the adult-plant stage of growth in Guancun, Xinjie town, Baoji, Shaanxi, China. When the sample was tested on seedlings of the Chinese set of 19 differentials including Triticum spelta ‘Album’ (TSA), the Yr5 donor (Chen et al. 2009), plus Yr single-gene lines in the Avocet S (AvS) background including the Yr5 near-isogenic line (AvSYr5NIL) and genotypes possessing one or more Yr genes using the standard procedures (Wang et al. 2016), it was virulent with infection type (IT) 9 on AvSYr5NIL and also virulent with IT 7 on TSA. Such Yr5 virulence was never seen with previous Pst isolates collected in China. Several single-uredinium isolates were obtained from TSA or AvSYr5NIL and tested on the above wheat genotypes; they all produced similar results and confirmed the Yr5 virulence. Single-uredinium isolate TSA-6 was selected to represent the Yr5-virulence race. On the set of Chinese differentials (Chen et al. 2009), the isolate was avirulent (IT 0 to 6) on Zhong 4 and Guinong 22 and virulent (IT 7 to 9) on Trigo Eureka, Fulhard, Lutescens 128, Mentana, Virgilio, Abbondanza, Early Premium, Funo, Danish 1, Jubilejina 2, Fengchan 3, Lovrin 13, Kangyin 655, Suwon 11, Lovrin 10, Hybrid 46, and TSA. Based on this avirulence/virulence pattern, the isolate was identified as a new race, temporarily named TSA-6. On the Yr single-gene lines and supplemental genotypes, the isolate was virulent (IT 7 to 9) on Yr1, Yr5, Yr6, Yr9, Yr17, Yr27, Yr28, Yr44, YrSP, Yr76, Siete Cerros T66 (Yr2), TP981 (Yr25), Vilmorin23 (Yr3, YrV23), Hybrid 46 (Yr4, YrH46), and Avocet A (YrA or Yr73 and Yr74) but was avirulent (IT 0 to 6) on Yr7, Yr8, Yr10, Yr15, Yr24, Yr26, Yr32, Yr43, YrTr1, and YrExp2. To make sure that the TSA and AvSYr5NIL plants are correct, DNA samples from infected leaves were tested with the sequence-tagged site (STS) marker closely linked to the Yr5 locus using primers Yr5STS-7 (5′-GTACAATTCACCTAGAGT-3′) and Yr5STS-8 (5′-GCAAGTTTTCTCCCTATT-3′) (Chen et al. 2003); both TSA and AvSYr5NIL had the band linked to the resistance allele, whereas AvS had the band linked to the susceptible allele. Thus, virulence to Yr5 was confirmed, and the new race virulent to Yr5 was identified. To the best of our knowledge, this is the first detection of a Pst race virulent to Yr5 in China. In our tests of 165 wheat cultivars, 111 wheat cultivars (67.3%) were susceptible to this race at seedling stage. In our 170 field samples collected from western Shaanxi and 39 from Huzhu, Qinghai, in 2018, we detected five samples (2.39%) virulent to Yr5 with the same avirulence/virulence pattern as TSA-6. Although most currently grown wheat cultivars do not have Yr5, the gene has been recently used in breeding programs in China. Therefore, continually monitoring Yr5 virulence is needed to assess the risk of the virulent race or races to wheat breeding programs and production in China and other countries.The author(s) declare no conflict of interest.Reference:Chen, W.
