Abstract:Stem rust, caused by Puccinia graminis f.sp. tritici, is one of the most dangerous rust diseases on wheat. Through physiological, biochemical, and molecular analysis, the relationship between the change in resistance of 15 wheat cultivars to stem rust disease and the response of 41 stem rust resistance genes (Sr,s) as well as TTKSK, TTKST, and TTTSK races was explained. Some cultivars and Sr genes, such as Gemmeiza-9, Gemmeiza-11, Sids-13, Sakha-94, Misr-1, Misr-2, Sr31, and Sr38, became susceptible to infection. Other new cultivars include Mir-3 and Sakha-95, and Sr genes 13, 37, 40, GT, and FR*2/SRTT3-SRTT3-SR10 remain resistant. Some resistance genes have been identified in these resistant cultivars: Sr2, Sr13, Sr24, Sr36, and Sr40. Sr31 was not detected in any cultivars. Reactive oxygen species such as hydrogen peroxide and superoxide, enzymes activities (catalase, peroxidase, and polyphenoloxidase), and electrolyte leakage were increased in the highly susceptible cultivars, while they decreased in the resistant ones. Anatomical characteristics such as the thickness of the epidermis, ground tissue, phloem tissue and vascular bundle diameter in the midrib were decreased in susceptible cultivars compared with resistant cultivars. Our results indicated that some races (TTKSK, TTKST, and TTTSK) appeared for the first time in Egypt and many other countries, which broke the resistant cultivars. The wheat rust breeding program must rely on land races and pyramiding genes in order to develop new resistance genes that will survive for a very long time.

Induction of multiple defense responses in wheat plants against stripe rust using mycorrhizal fungi and Streptomyces viridosporus HH1

Abstract:Stripe rust, caused by Puccinia striiformis f. sp. tritici , is one of the most devastating diseases of wheat worldwide. This study dealt with investigating the biocontrol activity of mycorrhizal colonization and/or application of Streptomyces viridosporus HH1 against stripe rust of wheat. In vitro, data showed that S. viridosporus HH1 had a strong anti-spore germination effect on uredospores of P. striiformis (90% reduction) A 84.2% reduction in the disease severity was achieved in the infected wheat plants, which were colonized with mycorrhizal fungi and were sprayed with S. viridosporus HH1. Molecular investigation revealed that mycorrhizal colonization and application of S. viridosporus HH1 upregulated the defensive genes ethylene response factor protein 3 ( JERF3 ) by 20.7-fold, chitinase II (13.6-fold), and β -1,3-glucanase (9.8-fold). Furthermore, mycorrhizal colonization and application of S. viridosporus HH1 led to a significant increase in the activity of the antioxidant enzymes peroxidase (52.3%), polyphenol oxidase (106.9%), and catalase (87.9%). The phenolic content also increased due to mycorrhizal colonization and application of S. viridosporus HH1, while the lipid peroxidation decreased in the infected wheat leaves. A mitigation in the adverse effects of infection on the photosynthetic pigments in wheat leaves was noticed. Observations from transmission electron microscopy showed that different hypersensitivity reactions were induced on the ultrastructural level in the infected wheat cells due to this treatment. In addition, a significant growth-promoting effect was also reported. It can be concluded that mycorrhizal colonization and spraying with S. viridosporus HH1 have a promising biocontrol activity against stripe rust in wheat. Field evaluation is necessary in the future studies before the use recommendation.

Abstract:Stripe rust, induced by Puccinia striiformis f. sp. tritici , is the most harmful and prevalent disease in temperate regions worldwide, affecting wheat production areas globally. An effective strategy for controlling the disease involves enhancing genetic resistance against stripe rust, achieved through Egyptian breeding efforts not previously conducted on wheat genotypes. The resistance level to stripe rust in thirty-eight wheat genotypes was assessed using marker-assisted selection methods. The investigation suggests that wheat breeding programs can utilize slow-rusting Yr genes, which are effective resistance genes, to develop novel genotypes with stripe rust resistance through marker-assisted breeding. Based on the four disease responses of the wheat genotypes under investigation, the results categorized the genotypes into three groups. The first group included resistant genotypes, the second group exhibited a slow-rusting character with the lowest disease symptom rates, and the last group displayed the highest disease characteristics rates throughout the three seasons, comprising fast-rusting genotypes. The rust-resistant genes identified were Yr5 , Yr9 , Yr10 , Yr15 , Yr17 , Yr18 , Yr26 , Yr29 , Yr30 , and Yr36 . Genes Yr26 , Yr30 , and Yr36 were present in all genotypes. Genotypes Misr3, Misr4, Giza168, Giza167, Giza170, Giza171, Gemmeiza9, and Gemmeiza10 carried the Yr9 gene. Only one genotype, Sids13, was found to have the Yr17 gene. Genes Yr18 and Yr29 were identified in Sids14, Giza168, Giza170, Gemmeiza9, and Gemmeiza10. However, none of the wheat genotypes showed the presence of Yr5 , Yr10 , or Yr15 . Several backcrossing generations were conducted to introduce the Yr5 and Yr10 genes into susceptible genotypes (Misr1, Misr2, and Gemmeiza11). These genotypes are cultivated globally and are known for producing high-quality flour, making them of great importance to farmers. The study demonstrates significant potential for enhancing wheat genotypes for stripe rust resistance and increased production.

Abstract:Wheat stripe rust (Puccinia striiformis f. sp. tritici, Pst), is one of the most serious diseases of wheat (Triticum aestivum L.) worldwide. To gain a better understanding of the protective mechanism against stripe rust at the adult plant stage, the differences in photosystem II and antioxidant enzymatic systems between susceptible and resistant wheat in response to stripe rust disease (P. striiformis) were investigated. We found that chlorophyll fluorescence and the activities of the antioxidant enzymes were higher in resistant wheat than in susceptible wheat after stripe rust infection. Compared with the susceptible wheat, the resistant wheat accumulated a higher level of D1 protein and a lower level of reactive oxygen species after infection. Furthermore, our results demonstrate that D1 and light-harvesting complex II (LHCII) phosphorylation are involved in the resistance to stripe rust in wheat. The CP29 protein was phosphorylated under stripe rust infection, like its phosphorylation in other monocots under environmental stresses. More extensive damages occur on the thylakoid membranes in the susceptible wheat compared with the resistant wheat. The findings provide evidence that thylakoid protein phosphorylation and antioxidant enzyme systems play important roles in plant responses and defense to biotic stress.

Abstract:Leaf rust caused by Puccinia triticina (Pt) is one of the most impactful diseases causing substantial losses in common wheat (Triticum aestivum L.) crops. In adult plants resistant to Pt, a horizontal adult plant resistance (APR) is observed: APR protects the plant against multiple pathogen races and is distinguished by durable persistence under production conditions. The Lr46/Yr29 locus was mapped to chromosome 1B of common wheat genome, but the identity of the underlying gene has not been demonstrated although several candidate genes have been proposed. This study aimed to analyze the expression of nine candidate genes located at the Lr46/Yr29 locus and their four complementary miRNAs (tae-miR5384-3p, tae-miR9780, tae-miR9775, and tae-miR164), in response to Pt infection. The plant materials tested included five reference cultivars in which the molecular marker csLV46G22 associated with the Lr46/Yr29-based Pt resistance was identified, as well as one susceptible control cultivar. Biotic stress was induced in adult plants by inoculation with fungal spores under controlled conditions. Plant material was sampled before and at 6, 12, 24, 48 hours post inoculation (hpi). Differences in expression of candidate genes at the Lr46/Yr29 locus were analyzed by qRT-PCR and showed that the expression of the genes varied at the analyzed time points. The highest expression of Lr46/Yr29 candidate genes (Lr46-Glu1, Lr46-Glu2, Lr46-Glu3, Lr46-RLK1, Lr46-RLK2, Lr46-RLK3, Lr46-RLK4, Lr46-Snex, and Lr46-WRKY) occurred at 12 and 24 hpi and such expression profiles were obtained only for one candidate gene among the nine genes analyzed (Lr46-Glu2), indicating that it may be a contributing factor in the resistance response to Pt infection.

Abstract:Wheat stripe rust ( Puccinia striiformis f. sp. tritici, Pst ) is the most damaging wheat disease, causing substantial losses in global wheat production and productivity. Our study aimed to unravel the complex reciprocity between reactive oxygen species and the antioxidant defense system as a source of resistance against stripe rust in diploid, tetraploid and hexaploid wheat genotypes. The significant genetic variability for stripe rust in the materials under study was evident as the genotypes showed contrasting responses during both the adult and seedling stages. Our thorough perspective on the biochemical responses of wheat genotypes to stripe rust infection revealed distinct patterns in oxidative damage, antioxidant enzymes and photosynthetic pigments. Principal component analysis revealed inverse correlations between antioxidants and ROS, underscoring their key function in maintaining the cellular redox balance and protecting plants against oxidative damage. Diploid ( Ae. tauschii ) wild wheat exhibited a better biochemical defense system and greater resistance to stripe rust than the tetraploid ( T. durum ) and hexaploid ( Triticum aestivum ) wheat genotypes. The antioxidant enzyme activity of durum wheat was moderate compared to diploid and hexaploid wheat genotypes. The hexaploid wheat genotypes exhibited increased ROS production, reduced antioxidant enzyme activity and decreased photosynthetic pigment levels. This study enhances understanding of the antioxidant defense system across different wheat ploidies facing stripe rust, serving as a valuable strategy for improving crop disease resistance. This study validated the biochemical response of stripe rust-resistant and susceptible candidate genotypes, which will be used to develop genetic resources for discovering stripe rust resistance genes in wheat.

Abstract:Crop diseases reduce wheat yields by ~25% globally and thus pose a major threat to global food security1. Genetic resistance can reduce crop losses in the field and can be selected through the use of molecular markers. However, genetic resistance often breaks down following changes in pathogen virulence, as experienced with the wheat yellow (stripe) rust fungus Puccinia striiformis f. sp. tritici (Pst)2. This highlights the need to (1) identify genes that, alone or in combination, provide broad-spectrum resistance, and (2) increase our understanding of the underlying molecular modes of action. Here we report the isolation and characterization of three major yellow rust resistance genes (Yr7, Yr5 and YrSP) from hexaploid wheat (Triticum aestivum), each having a distinct recognition specificity. We show that Yr5, which remains effective to a broad range of Pst isolates worldwide, is closely related yet distinct from Yr7, whereas YrSP is a truncated version of Yr5 with 99.8% sequence identity. All three Yr genes belong to a complex resistance gene cluster on chromosome 2B encoding nucleotide-binding and leucine-rich repeat proteins (NLRs) with a non-canonical N-terminal zinc-finger BED domain3 that is distinct from those found in non-NLR wheat proteins. We developed diagnostic markers to accelerate haplotype analysis and for marker-assisted selection to expedite the stacking of the non-allelic Yr genes. Our results provide evidence that the BED-NLR gene architecture can provide effective field-based resistance to important fungal diseases such as wheat yellow rust.

Abstract:Stripe (yellow, YR) rust bringing about by fungal pathogen of Puccinia striforms Westend Westend. is a serious wheat disease that poses substantial threats to the global wheat yield. Currently, the appearance of virulent fungus races is overcomed restricted by the studed resistance genes of wheat. Seach and identification of new genetic sources with durable resistance genes can allow efficiently to incorporate these target genes into germplasm pools. To increase spring wheat genetic diversity, on the back phone of parental variety Kazakhstanskay-19 (WT) characterizing by rust resistance through physical mutagenesis bythe various gamma irradiation doses, namely 300-, 350-, and 400 Gy, new M 3 mutant lines were created. From these new mutant resources, the 75 genotypes having higher yield-associated parameters such as grains number and weight per main spike comparing the parent variety were selected by phenotyping as the adult plant resistant (APR) under rust infected field trial. To measure resistance of these breeding lines at seedling stage by microscopy through the number of haustorial mother cells formation induced by YR treatment with the isolate of Puccinia striforms "Warrior" as temporary responses was carried out. Almost all of spring wheat mutant lines (93.0%, 70 samples from 75 mutant lines) identified as APR to YR had a strong association with SR which is developed on longer time of infection. The most effective dose of irradiation to generate the genetic variation was 350 Gy with mean of 60.08 ( p<0.005) according to Anova analysis.
Effectiveness of plant extracts for repressing stem rust disease severity of wheat caused by Puccinia triticina Eriks under field conditions

Abstract:Background Stem rust caused by Puccinia triticina Erikss is one of the most devastating fungal foliar diseases, and causes substantial yield losses to wheat crops cultivated under field conditions. This study aimed to evaluate the efficacy of plant extract-based foliar sprays for wheat with carnation, ginger and cinnamon on the severity of stem rust infection of three wheat cultivars cultivated under field conditions for two consecutive winter seasons of (2020/21 and 2021/22). Results In both seasons our data showed that all treatments were associated with some degree of disease suppression recorded as a range of 2.3–8.6% compared to 15.3–24.5% in untreated control treatment. The plant extract treatment was more effective than Amistar, a commercial fungicide. They recorded disease severity as: scores ranged between 2.3 and 3.2%; 3.0–4.5%, and 3.0–6% for wheat cvs . Giza 1, Misr 2, and Sids 14, in respective order. Meanwhile, scores of 4.1, 7.5, and 8.6% were recorded at fungicidal treatment. Spaying wheat plants with carnation extract showed the lowest disease prevalence followed by ginger and cinnamon extract, respectively. Increased in grain yield was associated with reduced disease severity. Conclusions The present investigation demonstrated the efficacy using plant extracts against foliar diseases caused by Puccinia and it was suggested that they may be an alternative to traditional chemical treatment.

Abstract:Stem rust caused by the pathogen Puccinia graminis f. sp. tritici is a destructive fungal disease-causing major grain yield losses in wheat. Therefore, understanding the plant defence regulation and function in response to the pathogen attack is required. As such, an untargeted LC-MS-based metabolomics approach was employed as a tool to dissect and understand the biochemical responses of Koonap (resistant) and Morocco (susceptible) wheat varieties infected with two different races of P. graminis (2SA88 [TTKSF] and 2SA107 [PTKST]). Data was generated from the infected and non-infected control plants harvested at 14- and 21- days post-inoculation (dpi), with 3 biological replicates per sample under a controlled environment. Chemo-metric tools such as principal component analysis (PCA), orthogonal projection to latent structures-discriminant analysis (OPLS-DA) were used to highlight the metabolic changes using LC-MS data of the methanolic extracts generated from the two wheat varieties. Molecular networking in Global Natural Product Social (GNPS) was further used to analyse biological networks between the perturbed metabolites. PCA and OPLS-DA analysis showed cluster separations between the varieties, infection races and the time-points. Distinct biochemical changes were also observed between the races and time-points. Metabolites were identified and classified using base peak intensities (BPI) and single ion extracted chromatograms from samples, and the most affected metabolites included flavonoids, carboxylic acids and alkaloids. Network analysis also showed high expression of metabolites from thiamine and glyoxylate, such as flavonoid glycosides, suggesting multi-faceted defence response strategy by understudied wheat varieties towards P. graminis pathogen infection. Overall, the study provided the insights of the biochemical changes in the expression of wheat metabolites in response to stem rust infection.

Abstract:Wheat stripe rust (Puccinia striiformis f. sp. tritici, Pst) is the most destructive wheat disease and a major problem for the productivity of wheat in the world. To obtain a better understanding about different effects of redox homeostasis and photosystem (PS) to Pst infection in wheat, we investigated the differences in photosynthesis and the antioxidant defense system in wheat cultivar Chuanmai42 (CM42) in response to two Chinese Pst races known as CYR32 and V26. The results showed that V26-infected wheat accumulated a higher reactive oxygen species (ROS), cell death, and energy dissipation than CYR32-infected wheat when compared with the control. Furthermore, we found that the activities of three antioxidant enzymes (APX, GR, and GPX) and four resistance-related enzymes in CYR32-infected wheat were significantly higher than that in V26-infected wheat. In addition, quantitative RT-PCR indicated that the expression levels of two genes associated with resistant stripe rust in CYR32-infected wheat were clearly higher than that in V26-infected wheat. Compared with CYR32-infected wheat, lower photochemical efficiencies were observed in V26-infected wheat at the adult stage. Meanwhile, only a marked decline in D1 protein was observed in V26-infected wheat. We therefore deduced that wheat with stripe rust resistance could maintain high resistance and photosynthetic capacity by regulating the antioxidant system, disease-resistant related enzymes and genes, and the levels of PSII reaction center proteins.

Abstract:Wheat stem rust, which is caused by Puccinia graminis f. sp. tritici (Pgt), is a highly destructive disease that affects wheat crops on a global scale. In this study, the reactions of 150 bread wheat varieties were evaluated for natural Pgt infection at the adult-plant stage in the 2019-2020 and 2020-2021 growing seasons, and they were analyzed using specific molecular markers to detect stem rust resistance genes (Sr22, Sr24, Sr25, Sr26, Sr31, Sr38, Sr50, and Sr57). Based on phenotypic data, the majority of the varieties (62%) were resistant or moderately resistant to natural Pgt infection. According to molecular results, it was identified that Sr57 was present in 103 varieties, Sr50 in nine varieties, Sr25 in six varieties, and Sr22, Sr31, and Sr38 in one variety each. Additionally, their combinations Sr25 + Sr50, Sr31 + Sr57, Sr38 + Sr50, and Sr38 + Sr57 were detected in these varieties. On the other hand, Sr24 and Sr26 were not identified. In addition, many varieties had low stem rust scores, including a large minority that lacked Sr57. These varieties must have useful resistance to stem rust and could be the basis for selecting greater, possibly durable resistance.
Characterization of non-host resistance in broad bean to the wheat stripe rust pathogen

Abstract:Background: Non-host resistance (NHR) confers plant species immunity against the majority of microbial pathogens and represents the most robust and durable form of plant resistance in nature. As one of the main genera of rust fungi with economic and biological importance, Puccinia infects almost all cereals but is unable to cause diseases on legumes. Little is known about the mechanism of this kind of effective defense in legumes to these non-host pathogens. Results: In this study, the basis of NHR in broad bean (Vicia faba L.) against the wheat stripe rust pathogen, Puccinia striiformis f. sp. tritici (Pst), was characterized. No visible symptoms were observed on broad bean leaves inoculated with Pst. Microscopic observations showed that successful location of stomata and haustoria formation were significantly reduced in Pst infection of broad bean. Attempted infection induced the formation of papillae, cell wall thickening, production of reactive oxygen species, callose deposition and accumulation of phenolic compounds in plant cell walls. The few Pst haustoria that did form in broad bean cells were encased in reactive oxygen and callose materials and those cells elicited cell death. Furthermore, a total of seven defense-related genes were identified and found to be up-regulated during the Pst infection. Conclusions: The results indicate that NHR in broad bean against Pst results from a continuum of layered defenses, including basic incompatibility, structural and chemical strengthening of cell wall, posthaustorial hypersensitive response and induction of several defense-related genes, demonstrating the multi-layered feature of NHR. This work also provides useful information for further determination of resistance mechanisms in broad bean to rust fungi, especially the adapted important broad bean rust pathogen, Uromyces viciae-fabae, because of strong similarity and association between NHR of plants to unadapted pathogens and basal resistance of plants to adapted pathogens.

Abstract:Zymoseptoria tritici is the causal agent of Septoria tritici blotch (STB), one of the most economically destructive wheat foliar diseases. In this study, we explore the physiological and molecular changes elicited in two wheat cultivars with divergent responses (Taichung 29 = susceptible, and Shafir = resistant) upon infection by Z. tritici. Our aim is to uncover novel insights into the intricate mechanisms that govern wheat defense against Z. tritici infection. Our quantitative histopathological study showed that H2O2 accumulated in the resistant cultivar to a higher degree compared to the susceptible cultivar at the biotrophic and switching phase. Additionally, we combined qPCR with a targeted quantitative HPLC technique to evaluate the expression profiles of 13 defense-related genes and profile the polyphenolic compounds induced differentially in the STB susceptible and resistant cultivar. Our finding indicated that five out of 13 genes were strongly up-regulated in the resistant cultivar compared with that of the susceptible one at eight days post-inoculation (dpi), corresponding to the transition phase present in the infection process of Z. tritici. Finally, our targeted HPLC analysis demonstrated that the traced phenolic compounds were highly elevated in the susceptible cultivar infected by Z. tritici compared with that of the resistant cultivar. In conclusion, our comprehensive analysis unveils a robust defense response in the resistant wheat cultivar Shafir, characterized by heightened H2O2 accumulation, significant up-regulation of key defense-related genes during the transition phase, and a distinct profile of polyphenolic compounds, shedding light on the intricate mechanisms contributing to its resistance against Z. tritici, thereby providing valuable insights for the development of more resilient wheat varieties.
Assessing performance and stability of yellow rust resistance, heat tolerance, and agronomic performance in diverse bread wheat genotypes for enhancing resilience to climate change under Egyptian conditions

Abstract:Yellow rust and heat stress adversatively impact the growth and production of bread wheat in particular under rising adverse environmental conditions. Stability of grain yield is a pivotal purpose of plant breeders to improve wheat production and ensure global food security especially under abrupt climate change. The objective of this study was to assess the performance and stability of diverse bread wheat genotypes for yellow rust resistance, heat stress, and yield traits. The studied genotypes were evaluated in two different locations under two sowing dates (timely and late sowing) during two growing seasons. The obtained results displayed significant differences among the tested locations, sowing dates, and genotypes for most measured traits. The yellow rust measurements evaluated under the field conditions including final rust severity (FRS), the average coefficient of infection (ACI), and area under disease progress curve (AUDPC) revealed that Giza-171, Misr-1, Gemmeiza-12, Shandweel-1, Sids-13, Line-1, Line-2, and Line-55 had better resistance. Based on heat sensitivity measurements, Line-1 and Line-2 followed by Line-35, Shandweel-1 and Line-55 were classified as more tolerant to heat stress compared with the remaining genotypes. The genotypes Line-55, Gemmeiza-12, Giza-171, Line-1, Line-2, and Misr-1 were able to maintain acceptable agronomic performance under timely and late sowing dates in all evaluated environments. Different statistical procedures were employed to explore the adaptability and stability of tested genotypes i.e. , joint regression, stratified ranking, Wricke's Ecovalence values, cultivar superiority, additive main effects, and multiplicative interaction (AMMI), AMMI stability value, and genotype plus genotype-by-environment interaction (GGE). The applied stability parameters were quite similar for describing the stability of the evaluated wheat genotypes. The results indicated that Gemmeiza-12, Giza-171, Sids-12, Sids-13, Misr-1 Shandweel-1, Line-1, Line-2, and Line-55 were desirable and stable. The heatmap and hierarchical clustering were exploited for dividing the evaluated bread wheat genotypes into different clusters based on yellow rust resistance measurements, heat tolerance indices, and agronomic performance. Line-1 and Line-2 had the best performance for all rust resistance, heat tolerance, and agronomic performance followed by Giza-171, Line-55, Line-35, Gemmeiza-12, Shandweel-1, Misr-1, and Sids-13. In conclusion, our findings provide evidence of utilizing promising genotypes in rust resistance, heat tolerance, and agronomic performance in breeding programs for improving wheat grain yield stability mainly under climate change.

Abstract:Abstract Stripe or yellow rust (YR) caused by Puccinia striiformis Westend. f. sp. tritici Erikss. is a persistent biotic-stress threatening global wheat production. To broaden our understanding of the shared genetic basis of YR resistance across multi-site and multi-year evaluations, we performed a large genome-wide association study using 43,706 YR observations on 23,346 wheat lines from the International Maize and Wheat Improvement Center evaluated between 2013 and 2019 at sites in India, Kenya and Mexico, against predominant races prevalent in the countries. We identified 114 repeatable markers tagging 20 quantitative trait loci (QTL) associated with YR on ten chromosomes including 1D, 2A, 2B, 2D, 3A, 4A, 4D, 5A, 5B and 6B, among which four QTL, QYr.cim-2DL.2 , QYr.cim-2AS.1 , QYr.cim-2BS.2 and QYr.cim-2BS.3 were significant in more than ten datasets. Furthermore, we report YR-associated allelic fingerprints for the largest panel of wheat breeding lines (52,067 lines) till date, creating substantial opportunities for YR favorable allele enrichment using molecular markers. Overall, the markers and fingerprints reported in this study provide excellent insights into the genetic architecture of YR resistance in different geographical regions, time-periods and wheat germplasm and are a huge resource to the global wheat breeding community for accelerating YR resistance breeding efforts.

Abstract:Abstract Wheat stem rust causes by Puccinia graminis f. sp. tritici is a serious threat to wheat production all over the world. This study evaluated the efficacy of Bacillus subtilis , B. polymyxa and B. megaterium , Eugenol and leaf extract of Artemisia cina against the disease, and the involved mechanisms in disease resistance. B. subtilis and Eugenol treatments inhibited spore germination and increased incubation and latent periods than other treatments. Additionally, the lowest infection type, pustule length and pustule width, and receptivity (no. of pustules) were recorded in case of B. subtilis and Eugenol treatments compared with the check control. The best results in disease assessment were achieved by foliar spray application 24 h before and after inoculation. Abnormalities, lysis, collapse, and shrinking of urediniospores were recorded in treated leaves. The activities of antioxidant defense enzymes, i.e., catalase (CAT), peroxidase (POX), and polyphenoloxidase (PPO), were significantly increased in B. subtilis and Eugenol treatments. On the other hand, electrolyte leakage was decreased. The active compounds in B. subtilis were revealed by utilizing GC-MS analysis.

Abstract:Stripe rust (yellow rust), caused by Puccinia striiformis f. sp. tritici (Pst), is considered as a major threat to the wheat production worldwide, including Pakistan. In the current study, 80 wheat landraces collected from different agroecological zones of Pakistan were screened for their stripe rust resistance. Molecular markers were used to identify/postulate the stripe rust genes in these landraces. The results indicated that 46 (57.5%) landraces had all-stage resistance, and only 4 (5%) landraces showed adult-plant resistance. The possible stripe rust resistance genes in these entries were postulated by the using already reported linked molecular markers of all-stage resistance and adult plant stage resistance genes (Yr4 Yr5, Yr10, Yr15 and Yr24/26 and adult-plant resistance genes Yr18, Yr29 and Yr32). Molecular analysis indicated that resistant genes Yr4, Yr5, Yr10, Yr15, Yr17, Yr18, Yr26/24, Yr29 and Yr32 were found in 16(20%), 12(15%), 4(5%), 15(18.75%), 16(20%), 13(16.25%), 39(48.75%), 20(25%) and 7(7.5%) of landraces, respectively. Majority of the landraces showed the presence of more than one gene. These lines could be used effectively in the wheat breeding programs.

Abstract:A rigid triphosphine PPh2C6H4-PPh-C6H4PPh2 () reacted with Cu(+) and a stoichiometric amount of terminal alkyne under basic conditions to give a family of copper(i) alkynyl compounds [Cu()C[triple bond, length as m-dash]CR]. The number of terminal -C[triple bond, length as m-dash]CH groups in the starting ligand determines the nuclearity of the resulting complexes giving mono- (, R = Ph; , R = C6H4OMe; , R = C6H4NO2; , R = C6H4CF3; , R = 2-pyridyl), di- (R = -(C6H4)n-, n = 1 (), n = 2, (), n = 3 ()) and trinuclear complexes (, R = 1,3,5-(C6H4)3-C6H3; , R = 1,3,5-(C6H4-4-C2C6H4)3-C6H3). In all the complexes the Cu(i) centers are found in a distorted tetrahedral environment that is achieved by tridentate coordination of the ligand and σ-bonding to the alkynyl function. The crystal structures of , and were estimated by single crystal X-ray diffraction analysis. The (31)P, (1)H and (1)H-(1)H COSY NMR spectroscopy confirms that all the molecules remain intact in solution. The photophysical studies carried out in the solid state at 298 and 77 K revealed moderate to weak orange luminescence (Φem up to 19%), tentatively assigned to thermally activated delayed fluorescence for the mononuclear complexes. The quantum yields of emission of demonstrated strong dependence on the nature of the alkynyl ligand, the role of which in the electronic transitions was elucidated by TD-DFT computational studies.
