Establishment of Real-Time FQ-PCR for Quantitative Detection of Human Cytomegalovirus and Its Preliminary Clinical Application

Abstract:Objective: To establish a rapid, sensitive, and specific real-time fluorescent quantitative polymerase chain reaction (FQ-PCR) method for quantitative detection of human cytomegalovirus (HCMV). Methods: HCMV IE (immediately gene) fragment was chosen to build FQ-PCR reaction system by creating upstream and downstream primers and TaqMan probe. Plasmids containing target fragments of HCMV were conducted as the external standards to develop the real-time FQ-PCR. Additional reference strains from the bacteria and virus were used to test the specificity of primers and probes, and fifty clinical specimens were quantitative detected with real-time FQ-PCR. Results: The quantitative range of the assays was 1010-106 copy/L, R=0.997, and the correlation was well. The concentration of standard coefficient of variation of Ct values was less than 5%, and the reproducibility was well. And specificity was determined by non-amplificated different human pathogens, including bacteria and virus. Fifty clinical specimens collected from patients were assayed by this method, and the detection rate of HCMV was 86% (43/50). Conclusion: The real time FQ-PCR was demonstrated as a rapid, specific and convenient method, and it is helpful in the study of the pathogenesis and epidemiology of HCMV.
