Abstract:An orthogonal dispersive spectral-domain optical coherence tomography (SDOCT) system based on a spectrometer consisting of a high spectral resolution virtually-imaged phased array (VIPA) and a low resolution diffraction grating is developed. Two-dimensional (2D) dispersion generated by the combination of the VIPA and the grating in conjunction with a 2D CCD leads to an improved performance of the spectrometer. Ultrahigh spectral resolution of 0.002 nm within a free spectrum range of 50 nm is realized, providing the spectrometer with a spectral sampling rate up to similar to 10(5). The developed SDOCT realizes an imaging depth over 80 mm, which is the longest depth range ever achieved by SDOCT. The increased spectral sampling rate also results in a high signal-to-noise ratio of the SDOCT system. The application of the developed system is further illustrated by quantitative phase imaging of a glass plate and an optical lens. (C) 2012 Optical Society of America

Abstract:We present a high-speed spectral domain optical coherence tomography (SD-OCT) system at 830 nm wavelength which is consisted of a fiber based Michelson interferometer and a custom-built spectrometer. The designed resolution of the spectrometer is about 67.4pm which limits the maximum detection depth 2.56mm in air. And the 35us exposure time of the high speed line scan CCD makes real-time imaging possible. Furthermore, a novel method of spectrometer calibration is put forward. The method can remove the influence of dispersion mismatch, thus accurately determine the distribution of wavelength on the line scan CCD, which leads to a precise interpolation and a subsequent better contrast image.

Abstract:The high frequency ultrasonic wave transmitting along UV-induced fiber Bragg grating, produces a long periodic ultrasonic grating to modulate the fiber Bragg grating, which is called as ultrasonic superstructure fiber grating. The acoustic-optic characteristics of the ultrasonic superstructure fiber grating are analyzed by using mode coupling theory. It is found that there are many individual peaks with same interval in the reflective spectra of ultrasonic superstructure fiber grating, and the interval of reflective peaks depends on the ultrasonic frequency. It is therefore possible to tune the reflective wavelength of first-order peak in the reflective spectra through changing ultrasonic frequency, so as to realize fast-tunable acoustic-optic filter with wide tunable range. Compared with conventional tunable acoustic-optic filters the present filter offers more narrow bandwidth and faster tuning speed.
Development of a high speed spectral domain optical coherence tomography system at 835 nm

Abstract:We develop a high speed multi-functional spectral domain (SD-OCT) system, using a broadband light source centered at 835nm and a custom-built spectrometer with a spectral resolution of 0.0674nm. After precise spectral calibration of the spectrometer, non-uniform discrete Fourier transform (NDFT) of the acquired spectral data is adopted for image reconstruction. In vivo imaging of human finger from volunteer at A-scan rate of 29 kHz with different reconstruction methods reveal that with the NDFT method, improved sensitivity fall-off especially at large depth is achieved in contrast to the conventional discrete Fourier transform (DFT) with interpolation method. And the phase information of the spectral data is used for functional extension to velocity mapping, Doppler images of rats' cerebral arteries at A-scan rate of 20 kHz are demonstrated. Furthermore, based on the spatial sinusoidal modulation in the reference arm, the complex spectral interferogram can be retrieved by harmonics analysis and digital synchronous demodulation which is free of mirror image and the coherence noise. Double imaging depth range on fresh shrimp at A-scan rate of 10 kHz is achieved and the resulting complex conjugate rejection ratio is up to 41dB.
High-speed Wide Tuning Range Wavelength Swept Laser Around 1310 Nm for Frequency Domain OCT

Abstract:Optical coherence tomography (OCT) is an emerging cross-sectional imaging technology. It uses broadband light sources to achieve axial image resolutions on a few microns scale. In this paper, the principle of Fourier domain mode-locked (FDML) wavelength swept laser for swept source optical coherence tomography (SS-OCT) is presented and crucial parameters of the laser system are discussed. On analysis of different methods and system configurations, a new configuration of swept laser system is put forward. We theoretically demonstrate a high-speed, broad bandwidth, Fourier domain mode-locked (FDML) wavelength swept laser around 1310 nm for high-resolution frequency domain optical coherence tomography (FD-OCT). The laser has a fiber ring cavity with a semiconductor optical amplifier gain medium. Intra-cavity mode selection is achieved with an in-fiber tunable fiber Fabry-Perot filter. The laser is designed to generate 32 kHz sweeping rate with a tuning range of 165 nm full width, 3 dB sweep range of 91 nm. In SS-OCT applications the swept laser will achieve about 8.3 μm axial resolution in air and 5.9 μm axial resolution in biological tissue. The instantaneous linewidth is 0.103 nm corresponding to a coherence length of several mm, enabling OCT imaging over a large axial range. The swept laser implemented in the FD-OCT system can provide information of biological tissue imaging in real time.

Abstract:We develop a high speed spectral domain optical coherence tomography (SD-OCT) system based on a custom-built spectrometer and non-uniform discrete Fourier transform (NDFT) to realize minimized depth dependent sensitivity fall-off. After precise spectral calibration of the spectrometer, NDFT of the acquired spectral data is adopted for image reconstruction. The spectrometer is able to measure a wavelength range of about 138 nm with a spectral resolution of 0.0674 nm at central wavelength of 835 nm, corresponding to an axial imaging range of 2.56 mm in air. Zemax simulations and sensitivity fall-off measurements under two alignment states of the spectrometer are given. Both theoretical simulations and experiments are done to study the depth dependent sensitivity of the developed system based on NDFT in contrast to those based on conventional discrete Fourier transform (DFT) with and without interpolation. In vivo imaging on human finger from volunteer is conducted at A-scan rate of 29 kHz and reconstruction is done based on different methods. The comparing results confirm that reconstruction method based on NDFT indeed improves sensitivity especially at large depth while maintaining the coherence-function-limited depth resolution.

Abstract:We propose a novel spectral-domain optical coherence tomography (SD-OCT) equipped with a Fresnel spectrometer, which utilizes a Fresnel zone plate (FZP) as both dispersion and focusing optics and thus spreads the spectral interferogram evenly in wavenumber domain because of the proportional relation between the focal length of the FZP and the wavenumber. With no need of the conversion calculation from wavelength to wavenumber in conventional SD-OCT, this new design is favorable for fast imaging with high resolution. As only a FZP and CCD are used, the Fresnel spectrometer is simple and compact. It is experimentally shown that its performance is as good as that of numerical interpolation in conventional SD-OCT. Imaging of bio-tissue by Fresnel SD-OCT is also demonstrated.

Abstract:The invention relates to the spectral optical coherence imaging system based on optic computation. The spectral optical coherence imaging system based on the optic computation is characterized by comprising an optic computation system and an image display system, wherein the optic computation system comprises a broadband light source, a signal generating device, a light intensity modulator, a chromatic dispersion device, a first coupler, a first circulator, a second circulator, a first focusing lens, a second focusing lens, a reflector, a scanning galvanometer and a second coupler, and the image display system comprises a balance detector, an envelope detector, a data acquisition card and a computer. The spectral optical coherence imaging system based on the optic computation realizes the real-time Fourier transform of the light frequency domain signals. Since the real-time Fourier transform can timely process the image data of the spectral optical coherence imaging system, the imaging speed is extremely high, and the spectral optical coherence imaging system based on the optic computation can be widely applied to the spectral optical coherence imaging.
Spectral Coding Based Single-Pixel NIR Spectroscopy in the Sub-Millisecond Regime

Abstract:In this contribution, we present a high-speed multiplex grating spectrometer based on a spectral coding approach that is founded on principles of compressive sensing. The spectrometer employs a single-pixel InGaAs detector to measure the signals encoded by an amplitude spatial light modulator (digital micromirror device, DMD). This approach leads to a speed advantage and multiplex sensitivity advantage atypical for standard dispersive systems. Exploiting the 18.2 kHz pattern rate of the DMD, we demonstrate 4.2 ms acquisition times for full spectra with a bandwidth of 450 nm (5250 cm-1 – 4300 cm-1; 1.9 μm – 2.33 μm). Due to the programmability of the DMD, spectral regions of interest can be chosen freely, thus reducing acquisition times further, down to the sub-millisecond regime. The adjustable resolving power of the system accessed by means of computer simulations is discussed, quantified for different measurement modes, and verified by comparison with a state-of-the-art Fourier-transform infrared spectrometer. We show measurements of characteristic polymer absorption bands in different operation regimes of the spectrometer. The theoretical multiplex advantage of 8 was experimentally verified by comparison of the noise behavior of the spectral coding approach and a standard line-scan approach.

Abstract:We present an ultrahigh-speed optical coherence tomography (OCT) based on an all-optical swept-source with an A-scan rate of 40 MHz. The inertia-free swept-source, which has its output power of 41.2 mW and tuning range of 40 nm and high scan linearity in wave-number with Pearson's correlation coefficients r of 0.9996, consists of a supercontinuum laser, an optical band-pass filter, a linearly chirped fiber Bragg grating, an erbium-doped fiber amplifier, and two buffer stages. With sensitivity of 87 dB, high-speed OCT imaging of biological tissue in vivo is also demonstrated. (C) The Authors.

Abstract:Monolithic Vernier tuned super-structure grating distributed Bragg reflector (SSG-DBR) lasers are expected to become one of the most promising sources for swept source optical coherence tomography (SS-OCT) with a long coherence length, reduced sensitivity roll-off, and potential capability for a very fast A-scan rate. However, previous implementations of the lasers suffer from four main problems: 1) frequencies deviate from the targeted values when scanned, 2) large amounts of noise appear associated with abrupt changes in injection currents, 3) optically aliased noise appears due to a long coherence length, and 4) the narrow wavelength coverage of a single chip limits resolution. We have developed a method of dynamical frequency tuning, a method of selective data sampling to eliminate current switching noise, an interferometer to reduce aliased noise, and an excess-noise-free connection of two serially scanned lasers to enhance resolution to solve these problems. An optical frequency comb SS-OCT system was achieved with a sensitivity of 124 dB and a dynamic range of 55-72 dB that depended on the depth at an A-scan rate of 3.1 kHz with a resolution of 15 μm by discretely scanning two SSG-DBR lasers, i.e., L-band (1.560-1.599 μm) and UL-band (1.598-1.640 μm). A few OCT images with excellent image penetration depth were obtained.

Abstract:Optical coherence tomography (OCT) is an emerging technology for high-resolution bio-medical imaging. The rapid scanning optical delay line (RSOD) has developed basically for depth scanning and dispersion compensation. If the pivot center of the galvamirror in RSOD is offset, phase modulation can be realized, providing a high carrier frequency required for heterodyne detection. However, the size of galvamirror should be increased, leading to lower response for its oscillation and hence a low scanning rate. In this paper we propose to apply a simisoidal waveform (resonant scan) instead of a triangle waveform (linear scan) to the galvamirror. Numerical analysis on phase modulation by RSOD and successive envelop demodulation under different driving waveform is conducted. We demonstrate that it is possible to improve OCT frame rate by resonant scan without compromising the signal-to-noise performance if complete and accurate envelope demodulation and appropriate signal processing are adopted.

Abstract:We present a modulation transfer spectroscopy (MTS) configuration based on an acousto-optic modulator by using a variant of the typical double pass structure. One beam is modulated by using an acousto-optic modulator in opposite diffraction order to cancel the carrier frequency shift and produce a modulated pump beam. The line shape performance is investigated theoretically and experimentally. Laser frequency stabilization of the proposed configuration is demonstrated for the Cs-133 vertical bar 6(2)S(1/2),F = 4 >-> vertical bar 6(2)P(3/2),F ' = 5 > transition. The Allan deviations, which are measured by using beat note signals and the three-cornered hat method, are 3.6 x 10(-11) in an integration time of 100 s and approximately 4 x 10(-11) in a longer integration time.

Abstract:Dual-channel spectral domain optical coherence tomography (SD-OCT) is one of the effective methods for improving imaging depth and imaging speed. In this paper, we design a dual-channel SD-OCT system based on a single spectrometer that can operate in two modes: (1) Increasing imaging speed and (2) expanding imaging depth. An optical path offset is preintroduced between the two channels to separate the two-channel data. However, this offset increases the requirement for the spectral resolution of the spectrometer in mode (1), so compressive sensing (CS) technology is used herein to overcome this problem. Consequently, in mode (1), when the spectral resolution of the spectrometer is the same as that used in the single-channel system, we use a dual-channel SD-OCT system combined with CS technology to double the imaging speed. In mode (2), when the spectral resolution of the spectrometer is only half of that used in a single-channel system, the imaging depth can be nearly doubled. We demonstrate the feasibility and effectiveness of the method proposed in this work by imaging a mirror, a fish fin, a fish eye, and an onion.

Abstract:Current spectroscopic optical coherence tomography (OCT) methods rely on a posteriori numerical calculation. We present an alternative for accessing optically the spectroscopic information in OCT, i.e. without any post-processing, by using a grating based correlation and a wavelength demultiplexing system. Conventional A-scan and spectrally resolved A-scan are directly recorded on the image sensor. Furthermore, due to the grating based system, no correlation scan is necessary. In the frame of this paper we present the principle of the system as well as first experimental results.

Abstract:We propose an all-optical Fourier transformation system for real-time massive data processing in high speed optical coherence tomography (OCT). In the so-called optical computing OCT, fast Fourier transformation (FFT) of A-scan signal is optically processed in real time before being detected by photoelectric detector. Therefore, the processing time for interpolation and FFT in traditional Fourier domain OCT can be dramatically eliminated. A processing rate of 10 mega-A-scans/second was experimentally achieved, which is, to our knowledge, the highest speed for OCT imaging. Due to its fiber based all-optical configuration, this optical computing OCT system is ideal for ultrahigh speed volumetric OCT imaging in clinical application.

Abstract:An ultrahigh-speed, wide-field OCT system for the imaging of anterior, posterior, and ocular biometers is crucial for obtaining comprehensive ocular parameters and quantifying ocular pathology size. Here, we demonstrate a multi-parametric ophthalmic OCT system with a speed of up to 1 MHz for wide-field imaging of the retina and 50 kHz for anterior chamber and ocular biometric measurement. A spectrum correction algorithm is proposed to ensure the accurate pairing of adjacent A-lines and elevate the A-scan speed from 500 kHz to 1 MHz for retinal imaging. A registration method employing position feedback signals was introduced, reducing pixel offsets between forward and reverse galvanometer scanning by 2.3 times. Experimental validation on glass sheets and the human eye confirms feasibility and efficacy. Meanwhile, we propose a revised formula to determine the “true” fundus size using all-axial length parameters from different fields of view. The efficient algorithms and compact design enhance system compatibility with clinical requirements, showing promise for widespread commercialization.

Abstract:A spectral filtering method that can enhance the axial resolution and suppress sidelobes of the point spread function in Optical Coherence Tomography (OCT) system is demonstrated. A mechanically induced long-period fiber grating (MLPG) is used to tailor the spectral shape of a homemade Er-doped fiber amplified spontaneous emission (ASE) source. By using a MLPG, the high and narrow characteristic peak, centered at 1533 nm, of the ASE source was fairly well removed, which enabled us to use a broader spectral bandwidth of the source. The experimental results show that the proposed spectral filtering method can suppress the sidelobes in the axial point spread function up to 5.4 dB and thus enhance the axial resolution of an OCT system by four times. The OCT images of an extracted human premolar taken with and without the spectral filtering are presented and compared with each other.

Abstract:The parameters of the spectrometer were analyzed in order to optimize the performance of spectral-domain optical coherence tomography (SD-OCT). Because probing depth is proportional to spectral resolution, it can be increased with the choice of a higher resolution spectrometer, which implies greater pixel numbers and lower imaging speed, or by sacrificing part of the spectrum, which compromises the axial resolution and side lobes. With a dynamic range of 8 bits or more, the signal-to-noise ratio remains constant for different noise levels. The results were verified experimentally with in vivo retinal SD-OCT imaging.
