Influencing Factors in Glucose Concentration Detection by Surface Plasmon Resonance with GGBP Amine Coupling

Abstract:The glucose concentration of human interstitial fluid is closely related to that of human blood.The minimally invasive blood glucose monitoring can be realized by measuring the glucose concentration of interstitial fluid.Considering the complexity of the interstitial fluid,the D-galactose/Dglucose binding protein(GGBP) which can specifically absorb glucose molecules can be used to modify the surface plasmon resonance(SPR) sensor with amine coupling method to realize glucose selective measurement.The measuring process is complicated,so the factors which influence the measurement accuracy need to be evaluated to obtain the optimized measurement condition.A SPR glucose concentration detection system based on a miniature integrated SPR sensor was set up,the factors affecting the measurement results were studied systemically by experiments,including protein purification,protein activity,baseline,flow velocity,mass transfer effect,temperature and bubble,and some solutions were proposed accordingly.The study optimizes the measurement process and provides important instruction for the standardization and development of miniaturization instrument.
