Measurement of Glucose Concentration by Surface Plasmon Resonance with D-Galactose/D-Glucose Binding Protein

Abstract:A minimally invasive way for the detection of diabetes by measuring the glucose in interstitial fluid was investigated in this paper.The measurement system can estimate the blood glucose concentration through the interstitial fluid.D-galactose/D-glucose binding protein(GGBP)was expressed from the Escherichia coli which can specifically absorb glucose in the interstitial fluid.And then a bio-optical method of surface plasmon resonance(SPR)technology was introduced to build up glucose measurement system.After the modification of SPR sensor surface by immobilized GGBP on it,the sensor succeeded in distinguishing the concentration of glucose solution in various concentration ranges.Three kinds of mutated GGBP(mutated at E149C;mutated at E149C,A213S;and mutated at E149C,A213S and L238S)were used in the indirect detection.Results show that the sensor with GGBP E149C has the highest sensitivity and a good linearity when measuring the glucose concentration of 0.5 mg/L—5 mg/L.The result reveals a bright future for SPR in the minimally invasive diabetes testing and detection of other components in human body.
