Abstract:The pathological presentation of Alzheimer disease (AD), the leading cause of senile dementia, involves regionalized neuronal death and an accumulation of intraneuronal and extracellular filaments termed neurofibrillary tangles and senile plaques, respectively. One of the β-amyloid peptides (Aβ), the Aβ1-42 form, is primarily responsible for neuronal damage and cell death that is the main component in the senile plaques. Over the past twenty years, the amyloid hypothesis has been strongly supported by a wealth of evidence, including data from genetic studies of Alzheimer disease. Amyloid cascade hypothesis states that the accumulation and deposition of fibrillar Aβ is the primary driver of neurodegeneration and cognitive decline leading to dementia. AD is a clinicopathological syndrome in which different gene defects can lead--directly or indirectly--to alter APP expression or proteolytic processing as such to change Aβ stability or aggregation. These result in a chronic imbalance between Aβ production and clearance. Gradual accumulation of aggregated Aβ initiates a complex, multistep cascade that includes gliosis, inflammatory changes, neuritic/synaptic change, tangles and transmitter loss. The evidence that links Aβ to the pathogenesis of AD is substantial, but the means by which these peptides exert their toxic effects, and where in neuronal cells they act, is far from clear. The up-to-date proceeding in the molecular mechanism of β-amyloid peptides is overviewed.

Abstract:Alzheimer's disease (AD) and type 2 diabetes (T2D) are two common protein aggregation diseases. Compelling evidence has shown a link between AD and T2D, which may derive from interspecies cross-sequence interactions between amyloid-β peptide (Aβ), associated with AD, and human islet amyloid polypeptide (hIAPP), associated with T2D. Herein, we present experimental and computational protocols and tools to study the aggregate structures and kinetics, conformational conversion, and molecular interactions of Aβ-hIAPP mixtures. These protocols could be generally applied to other cross-seeding behaviors of amyloid peptides.

Abstract:Alzheimer's disease (AD) is a primary neurodegenerative disorder mainly affecting aged people over 60 years old. It is characterized by extracellular senile plaques (SPs) and intracellular neurofibrillary tangles (NFTs) in patients' brains. The major component of SP is β-amyloid peptide (βA) with molecular weight of about 4 ku which is a neurotoxic derivative of β-amyloid precursor protein (APP) and can cause cell damage mainly through oxidative stress and being able to form calcium channels in lipid bilayers. The precursor, APP, whose function is not yet elucidated in detail but evidence exists that it may mediate cell adhesion, maintain synaptic plasticity and so on, can be processed in conventional secretary way by several putative secretases and alternatively in endosomal/ lysosomal way. Longer βA is more predisposed to aggregate to form SP than its shorer counterpart, so some APP mutants may cause familial AD through producing more longer βAs or increasing the production of shorter ones. The changes of some factors which are important for the metabolism of APP, for example, mutations of presenilins, may also cause AD by increased generation of βA.

Abstract:One of the neuropathologic hallmarks of Alzheimer’s disease (AD) is the presence of senile plaques which consist of β-amyloid peptide (Aβ) in the brain. Aβ is derived from β-amyloid precursor protein (APP) through sequential cleavages by the β-secretase and the γ-secretase. In addition to Aβ, γ-cleavage releases the intracellular domain of APP (AICD). However, although it is well-established that Aβ is the prime culprit for AD pathogenesis, the physio/pathological functions of AICD remain largely elusive. Here we review recent progress toward elucidating the functional roles of AICD, which include modulating intracellular trafficking/processing of APP, inducing apoptosis, and regulating gene expression at transcriptional level.

Abstract:Alzheimer's disease (AD), a common neurodegenerative disease in the elderly and the most prevalent cause of dementia, is characterized by progressive cognitive impairment. The prevalence of AD continues to increase worldwide, becoming a great healthcare challenge of the twenty-first century. In the more than 110 years since AD was discovered, many related pathogenic mechanisms have been proposed, and the most recognized hypotheses are the amyloid and tau hypotheses. However, almost all clinical trials targeting these mechanisms have not identified any effective methods to treat AD. Scientists are gradually moving away from the simple assumption, as proposed in the original amyloid hypothesis, to new theories of pathogenesis, including gamma oscillations, prion transmission, cerebral vasoconstriction, growth hormone secretagogue receptor 1α (GHSR1α)-mediated mechanism, and infection. To place these findings in context, we first reviewed the neuropathology of AD and further discussed new insights in the pathogenesis of AD.

Abstract:One of the main neuropathological lesions observed in brain autopsy of Alzheimer's disease (AD) patients is the extracellular senile plaques mainly composed of amyloid-beta (Aβ) peptide. Recently, treatment strategies have focused on modifying the formation, clearance, and accumulation of this potentially neurotoxic peptide. β- and γ-secretase are responsible for the cleavage of amyloid precursor protein (APP) and the generation of Aβ peptide. Treatments targeting these two critical secretases may therefore reduce Aβ peptide levels and positive impact on AD. Vaccination is also an advanced approach against Aβ. This review focuses on recent advances of our understanding of this key peptide, with emphasis on Aβ peptide synthesis, accumulation and neurotoxicity, and current therapies including vaccination and two critical secretase inhibitors. MicroRNAs (miRNAs) are a class of conserved endogenous small noncoding RNAs, known to regulate the expression of complementary messenger RNAs, involved in AD development. We therefore address the relationship of miRNAs in the brain and Aβ generation, as a novel therapeutic approach to the treatment of AD while also providing new insights on the etiology of this neurological disorder.

Abstract:Amyloid precursor protein (APP) processing is central in Alzheimer's disease (AD) pathogenesis. The healthy unaffected neurons suffer the transmission of amyloid protein from pathologically affected neurons, which may play an important role in the anatomical spread of the disease. Exosomes are appropriate candidates for transmission of amyloid species, because of their potential role as “intercellular transportation”. To address a role of secreted exosomes in neuronal homeostasis in AD, we harvested exosomes from the conditioned medium of HEK293-APP Swe/Ind cells. We have demonstrated that these exosomes contained APP and were capable of efficiently transferring APP to normal primary neurons. Moreover, these exosomes had dose-dependent detrimental effect on cultured neurons. Our results suggest a key mechanism underlying the spread of amyloid protein in the brain and the acceleration of pathology in AD; exosomes secretion serves to amplify and propagate Alzheimer's disease related pathology.

Abstract:Alzheimer’s disease(AD) is characterized by early neuropathological pathways-extracellular fibrillar β-amyloid(Aβ) deposits in the brain,intracellular neurofibrillary tangles(NFTs),possibly leading to synaptic dysfunction,neurodegeneration and cognitive/functional decline.The clinical diagnosis of probable AD based on standard diagnostic criteria does not take into account the long preclinical and prodromal course of AD.Diagnosis was based on structured history,neurological examination and neuropsychological assessment,with emphasis on qualitative as well as quantitative aspects of performance.AD-related pathophysiological changes can occur many years and even decades before the appearance of clinical dementia syndrome.Biomarkers that are related to the pathophysiology of AD may thus be helpful in detecting the preclinical stages of disease,and improve early and differential diagnosis,provided that the biomarkers were specific enough to avoid interfering with normal processes and accuracy of preclinical diagnosis of AD was increasingly important for therapeutic and prognostic investigations.

Abstract:β-amyloid protein (Aβ) is a transmembrane glyc.oprotein β-amyloid precursor protein hydrolysate.The excessive production and accumulation in the brain,deposition may cause neuronal calcium homeostasis,free radicals,activation of inflammatory factors,local brain inflammation and a series of reactions.With the hippocampus and cerebral cortex of the degeneration and death of neurons in the brain cholinergic and dopaminergic neurotransmitter in a progressive decrease,it eventually leads to induce learning and memory and cognitive dysfunction.In recent years,in the brain of patients with diffuse axonal injury and patients with Alzheimer disease the formation of large amounts of β-amyloid plaques was found,and these diseases brought a heavy burden to the society and family,so the study of β-amyloid has great significance.

In recent years, a large number of studies have been conducted on the APP processing pathways and the role of secretion. Abstract:Alzheimer’s disease（AD）,a neurodegenerative disease characterized by cognitive decline and behavioral impairment,occurs mostly in the aging population.In recent years,the morbidity and mortality rates of AD have been increasing,and the disease onset tends to occur at a younger age.The etiology of AD is multifactorial and complex,involving aberrant deposition of amyloid β（Aβ）,tau accumulation,apolipoprotein abnormalities,cerebrovascular dysfunction,metal dysregulation,oxidative stress and genetic factors.Amyloid plaques and neurofibrillary tangles are the main pathological hallmarks of AD.Aβ,the main constituent of amyloid plaques,is generally recognized as the initiating cause of AD.Currently,there are still no effective treatments for AD.Herein,this review summarizes and highlights the advances in AD pathogenesis and therapeutic strategies,in the hope of providing new insight into AD study and treatment.

Abstract:The accumulation of Aβ can cause a series of structural and functional changes at the synapse,and plays a key role in the process of Alzheimer disease(AD),while phosphorylated tau also plays a central role in AD neurodegeneration.Research on the relationship between them as well as other relevant factors such as PI3K,GSK and so on is increasing gradually.Probing the exact effect and the correlation between Aβ and tau has become more and more prominent for the treatment and condition assessment in AD.

Abstract:Alzheimer's disease (AD) is a global medical challenge. Studies have shown that neurotoxicity caused by pathological aggregation of β-amyloid (Aβ) is an important factor leading to AD. Therefore, inhibiting the pathological aggregation of Aβ is the key to treating AD. The recombinant human HspB5-ACD structural domain protein (AHspB5) prepared by our group in the previous period has been shown to have anti-amyloid aggregation effects, but its inability to penetrate biological membranes has limited its development. In this study, we prepared a recombinant fusion protein (T-AHspB5) of TAT and AHspB5. In vitro experiments showed that T-AHspB5 inhibited the formation of Aβ 1–42 protofibrils and had the ability to penetrate the blood-brain barrier; in cellular experiments, T-AHspB5 prevented Aβ 1–42 -induced oxidative stress damage, apoptosis, and inflammatory responses in neuronal cells, and its mechanism of action was related to microglia activation and mitochondria-dependent apoptotic pathway. In animal experiments, T-AHspB5 improved memory and cognitive dysfunction and inhibited pathological changes of AD in APP/PS1 mice. In conclusion, this paper is expected to reveal the intervention mechanism and biological effect of T-AHspB5 on pathological aggregation of Aβ 1–42 , provide a new pathway for the treatment of AD, and lay the foundation for the future development and application of T-AHspB5.

Abstract:Alzheimer's disease (AD) has become one of the leading causes of health problems in the elderly, and studying its causes and treatments remains a serious challenge for researchers worldwide. The two main pathological features of Alzheimer's disease are the extracellular deposition of β-amyloid (Aβ) to form senile plaques and the intracellular aggregation of hyperphosphorylated Tau protein to form neurofibrillary tangles (NFTs). Researchers have proposed several hypotheses to elucidate the pathogenesis of AD, but due to the complexity of the pathophysiologic factors involved in the development of AD, no effective drugs have been found to stop the progression of the disease. Currently, the mainstay drugs used to treat AD can only alleviate the patient's symptoms and do not have a therapeutic effect. As researchers explore interactions among diseases, much evidence suggests that there is a close link between periodontitis and AD, and that periodontal pathogenic bacteria can exacerbate Aβ deposition and Tau protein hyperphosphorylation through neuroinflammatory mechanisms, thereby advancing the pathogenesis of AD. This article reviews recent advances in the pathogenesis of AD, available therapeutic agents, the relevance of periodontitis to AD, and mechanisms of action.

Abstract:Deposition of β-amyloid (Aβ) in the hippocampus is one of the hallmarks of Alzheimer's disease (AD), thus decreasing the production of Aβ has become one of the strategies of AD therapy. Aβ was proposed to be cleared from the body through the vascular pathway, but now it has been hypothesized that a kind of protease called Aβ-degrading enzyme (ADE) can hydrolyze Aβ into polypeptides. The ADEs include neprilysin, insulin-degrading enzyme, plasmin, endothelin converting enzyme and angiotensin-converting enzyme. Here the biological characteristics of ADEs and their potential roles in the pathogenesis and treatment of AD are summarized.
