Abstract:Male moths detect and find their mates using species-specific sex pheromones emitted by conspecific females. Olfaction plays a vital role in this behavior. Since the first discovery of an insect sex pheromone from the silkmoth Bombyx mori, great efforts have been spent on understanding the sensing of the pheromones in vivo. Much progress has been made in elucidating the molecular mechanisms that mediate chemoreception in insects in the past few decades. In this review, we focus on pheromone reception and detection in moths, from the molecular to the behavioral level. We trace the information pathway from the capture of pheromone by male antennae, binding and transportation to olfactory receptor neurons, receptor activation, signal transduction, molecule inactivation, through brain processing and behavioral response. We highlight the impact of recent studies and also provide our insights into pheromone processing.

Abstract:Insect pheromones are critical chemical signals that regulate intraspecific behavior and play a key role in the dynamic monitoring and control of pest populations. Historically, research on insect pheromones has primarily focused on lipid-based compounds. However, terpenes and terpenoids, which are widely occurring classes of bioactive compounds, also play significant roles in insect pheromone blends. Over 50 terpene and terpenoid-based pheromones have been identified in over 52 insect species, spanning various orders such as Coleoptera, Hymenoptera, Blattodea, Hemiptera, Diptera, and Lepidoptera. These compounds are associated with several types of pheromones, including female or male sex pheromones, aggregation pheromones, alarm pheromones, and aphrodisiac pheromones. Terpenes and terpenoids may act as either primary or secondary components of pheromone blends and influence a wide range of critical insect behaviors. They play essential roles in the physiological and ecological adaptation of insects to their environment. This review provides a comprehensive overview of current research on terpene and terpenoid-based pheromones in insects, examining their structures, types, and physiological and ecological functions. Additionally, we propose future research directions to guide the application of these pheromones in insect behavioral regulation and pest management, while advocating for their broader use in insect pest monitoring and control.

Abstract:Pheromones are chemical signals that are given off by creatures to communicate with other conspecifics. Because of their influence on animal behavior and relationships throughout the animal world, pheromones have fascinated scientists for decades. The newest findings and new perspectives on the intriguing field of pheromones are synthesized in this thorough review article. We investigate the many roles played by pheromones, from mating attraction and territorial marking to alarm signaling and social organization. The chemical diversity of pheromones is explored in depth in this paper, highlighting the variety of substances used by different animals as messengers. We go over the processes involved in pheromone detection and processing to help understand how chemosensory receptors and brain circuits interact in complex ways. Additionally, we discuss the role that pheromones play in ecology and evolution by promoting speciation and adaptation. This review emphasizes both the new research on pheromones in humans as well as their function in the animal world. We investigate the data supporting human pheromones and their possible impact on social and reproductive behaviors. The review aims to offer a detailed exploration and insight into the role and effects of pheromones in the natural world. This includes examining the chemical essence of pheromones and their functionality as communication signals across different species, investigating the specificity of these chemicals to certain species and how non-specific pheromones come about, and delving into the evolution of pheromones with a focus on the physiological and evolutionary mechanisms that drive communication. Additionally, the review seeks to understand the role pheromones play in pest management, specifically how species-specific pheromones can be utilized in eco-friendly pest control strategies. It will also explore the enigmatic impact pheromones have on the behavior and interactions within and among species. Drawing from a variety of sources, such as scientific research articles, studies on pheromone evolution, and articles on pheromone application in pest management, the review aspires to synthesize existing knowledge to provide a comprehensive overview of pheromones and their intriguing influence in nature. This study intends to not only summarize existing information but also encourage further research, shedding light on these cryptic chemical signals and their tremendous influence on the biology and behavior of species. It does this by offering a comprehensive perspective on the complex world of pheromones.

Abstract:Since the first identification of the silkworm moth sex pheromone in 1959, significant research has been reported on identifying and unravelling the sex pheromone mechanisms of hundreds of insect species. In the past two decades, the number of research studies on new insect pheromones, pheromone biosynthesis, mode of action, peripheral olfactory and neural mechanisms, and their practical applications in Integrated Pest Management has increased dramatically. An interdisciplinary approach that uses the advances and new techniques in analytical chemistry, chemical ecology, neurophysiology, genetics, and evolutionary and molecular biology has helped us to better understand the pheromone perception mechanisms and its practical application in agricultural pest management. In this review, we present the most recent developments in pheromone research and its application in the past two decades.

Abstract:Chirality is a critical topic in the medicinal and agrochemical fields. One quarter of all agrochemicals was chiral in 1996, and this proportion has increased remarkably with the introduction of new compounds over time. Despite scientists have made great efforts to probe the enantiomeric selectivity of chiral chemicals in the environment since early 1990s, the different behaviours of individual enantiomers in biologically mediated processes are still unclear. In the present review, we highlight state-of-the-knowledge on the stereoselective biotransformation and accumulation of chiral contaminants in organisms ranging from invertebrates to humans. Chiral insecticides, fungicides, and herbicides, polychlorinated biphenyls (PCBs), pharmaceuticals, flame retardants hexabromocyclododecane (HBCD), and perfluorooctane sulfonate (PFOS) are all included in the target compounds. Key findings included: a) Changes in the enantiomeric fractions in vitro and in vivo models revealed that enantioselectivity commonly occurs in biotransformation and bioaccumulation. b) Emerging contaminants have become more important in the field of enantioselectivity together with their metabolites in biological transformation process. c) Chiral signatures have also been regarded as powerful tools for tracking pollution sources when the contribution of precursor is unknown. Future studies are needed in order to understand not only preliminary enrichment results but also detailed molecular mechanisms in diverse models to comprehensively understand the behaviours of chiral compounds.

Abstract:Insect biogenic amines play important roles in mediating behavioral and physiological processes as neurotransmitters, neuromodulators and neurohormones. They activate specific G-protein coupled receptors on the cell surfaces to induce downstream signaling pathways. These biogenic amine receptors show different pharmacological properties from their vertebrate counterparts and therefore become potential targets for pest control. However, their physiological functions are not well understood. We summarize our recent progress on insect biogenic amine receptors, and emphasize the characterizations of novel receptors and their functions in behavior and immunity.

Abstract:Chiral surface is a critical mediator that significantly impacts interaction with biological systems on regulating cell behavior. To better understand how the properties of interfacial Chirality affect cell behavior and address the limitations of chiral materials for biomedical applications, in this review, we mainly focus on the recent developments of chiral bio-interfaces for the controllable and accurate guidance of chiral biomedical phenomena. In particular, we will discuss how cells or organisms sense and respond to the chiral stimulus, as well as the chirality mediating cell fate, tissue repair, and organism immune response will be reviewed. In addition, the biological applications of chirality, such as drug delivery, antibacterial, antivirus and antitumor activities, and biological signal detection, will also be reviewed. Finally, the challenges of chiral bio-interfaces for controlling biological response and the further application of interface chirality materials for biomedical will be discussed.

Abstract:Insects communicating with pheromones, be it sex- or aggregation pheromones, are confronted with an olfactory environment rich in a diversity of volatile organic compounds of which plants are the main releaser. Certain of these volatiles can represent behaviorally relevant information, such as indications about host- or non-host plants; others will provide essentially a rich odor background out of which the behaviorally relevant information needs to be extracted. In an attempt to disentangle mechanisms of pheromone communication in a rich olfactory environment, which might underlie interactions between intraspecific signals and a background, we will summarize recent literature on pheromone/plant volatile interactions. Starting from molecular mechanisms, describing the peripheral detection and central nervous integration of pheromone-plant volatile mixtures, we will end with behavioral output in response to such mixtures and its plasticity.

Abstract:Chirality exists extensively in nature. Synthetic chiral plant growth regulators and other pesticides usually behave enantioselectively in phyto-biochemical processes. Chiral plant growth regulators regulate the physiological processes of plants enantioselectively, and chiral pesticides cause enantioselective toxicities or ecotoxicities to plants. On the other hand, these chiral agrochemicals can be absorbed and enantioselectively metabolized by plants. This review summarizes the enantioselective effects of chiral plant growth regulators on plants and the phytotoxic and biotransformation effects of chiral herbicides and several persistent organic pollutants (POPs) on plants. Together, this information on the interactions between chiral agrochemicals and plants might shed light on studies on the chemical and biological behaviors of chiral chemicals, and direct research into the selection of plants, which can potentially decontaminate the environment.

Abstract:Plants of the Glochidion (Phyllanthaceae) genus are pollinated exclusively by host-specific Epicephala (Gracillariidae) moths. Floral scent has been thought to play key role in the obligate pollination mutualism between Glochidion plants and Epicephala moths, but few studies have been reported about chemical variation in floral volatiles of Glochidion species in China. Floral volatiles of male and female flowers of five Glochidion species in south China were collected by dynamic headspace absorption technique and then were chemically analyzed by using gas chromatography-mass spectrometry. A total of 69 compounds were identified from floral scents of five investigated species. Glochidion hirsutum and G. zeylanicum showed no qualitative differences in floral scent, whereas there were clear variations of floral scent among other species (G. eriocarpum, G. daltonii, and G. sphaerogynum) and also they distinctly differed from these two species. Male flowers emitted significantly more scent than female flowers. Glochidion plants exhibited qualitative and quantitative differences in floral scent between two sexes of flowers. The findings suggest that the volatile variation of floral scent among Glochidion species reflects adaptations to specific pollinators. Sexual dimorphism in floral scent has evolved to signal alternative rewards provided by each sex to Epicephala moths.
Can Pheromones Contribute to Phylogenetic Hypotheses? A Case Study of Chrysomelidae

Abstract:Pheromones mediate species-level communication in the search for mates, nesting, and feeding sites. Although the role of pheromones has long been discussed by various authors, their existence was not proven until the mid-twentieth century when the first sex pheromone was identified. From this finding, much has been speculated about whether this communication mechanism has acted as a regulatory agent in the process of speciation, competition, and sexual selection since it acts as an intraspecific barrier. Chrysomelidae is one of the major Phytophaga lineages, with approximately 40,000 species. Due to this immense diversity the internal relationships remain unstable when analyzed only with morphological data, consequently recent efforts have been directed to molecular analyses to establish clarity for the relationships and found their respective monophyly. Therefore, our goals are twofold 1) to synthesize the current literature on Chrysomelidae sex pheromones and 2) to test whether Chrysomelidae sex pheromones and their chemical structures could be used in phylogenetic analysis for the group. The results show that, although this is the first analysis in Chrysomelidae to use pheromones as a phylogenetic character, much can be observed in agreement with previous analyses, thus confirming that pheromones, when known in their entirety within lineages, can be used as characters in phylogenetic analyses, bringing elucidation to the relationships and evolution of organisms.
Functional Characterization of Odorant Receptors for Sex Pheromone (Z)-11-Hexadecenol in Orthaga achatina

Abstract:Pheromone receptor (PR)-mediated transduction of sex pheromones to electrophysiological signals is the basis for sex pheromone communication. Orthaga achatina, a serious pest of the camphor tree, uses a mixture of four components (Z11-16:OAc, Z11-16:OH, Z11-16:Ald, and Z3,Z6,Z9,Z12,Z15-23:H) as its sex pheromone. In this study, we identified five PR genes (OachPR1-5) by phylogenetic analysis. Further RT-PCR and qPCR experiments showed that PR1-3 were specifically expressed in male antennae, while PR4 was significantly female-biased in expression. Functional characterization using the XOE-TEVC assay demonstrated that PR1 and PR3 both responded strongly to Z11-16:OH, while PR1 and PR3 had a weak response to Z3,Z6,Z9,Z12,Z15-23:H and Z11-16:Ald, respectively. Finally, two key amino acid residues (N78 and R331) were confirmed to be essential for binding of PR3 with Z11-16:OH by molecular docking and site-directed mutagenesis. This study helps understand the sex pheromone recognition molecular mechanism of O. achatina.
A plant volatile alters the perception of sex pheromone blend ratios in a moth

Abstract:Mate finding in most moths is based on male perception of a female-emitted pheromone whose species specificity resides in component chemistry and proportions. Components are individually detected by specialized olfactory receptor neurons (ORNs) projecting into the macroglomerular complex (MGC) of the male brain. We asked how robust ratio recognition is when challenged by a plant volatile background. To test this, we investigated the perception of the pheromone blend in <i>Agrotis ipsilon,</i> a moth species whose females produce a blend of Z7-dodecenyl acetate (Z7-12:Ac), Z9-tetradecenyl acetate (Z9-14:Ac), and Z11-hexadecenyl acetate in a 4:1:4 ratio optimally attractive for males. First, we recorded the responses of specialist ORNs for Z7 and Z9 and showed that heptanal, a flower volatile, activated Z7 but not Z9 neurons. Then, we recorded intracellularly the responses of MGC neurons to various ratios and showed that heptanal altered ratio responses of pheromone-sensitive neurons. Finally, we analyzed the behavior of males in a wind tunnel and showed that their innate preference for the 4:1:4 blend was shifted in the presence of heptanal. Pheromone ratio recognition may thus be altered by background odorants. Therefore, the olfactory environment might be a selective force for the evolution of pheromone communication systems.
Enantioselectivity in environmental safety of current chiral insecticides.

Abstract:Chiral pesticides currently constitute about 25% of all pesticides used, and this ratio is increasing as more complex structures are introduced. Chirality occurs widely in synthetic pyrethroids and organophosphates, which are the mainstay of modern insecticides. Despite the great public concerns associated with the use of insecticides, the environmental significance of chirality in currently used insecticides is poorly understood. In this study, we resolved enantiomers of a number of synthetic pyrethroid and organophosphate insecticides on chiral selective columns and evaluated the occurrence of enantioselectivity in aquatic toxicity and biodegradation. Dramatic differences between enantiomers were observed in their acute toxicity to the freshwater invertebrates Ceriodaphnia dubia and Daphnia magna, suggesting that the aquatic toxicity is primarily attributable to a specific enantiomer in the racemate. In field sediments, the (-)enantiomer of cis-bifenthrin or cis-permethrin was preferentially degraded, resulting in relative enrichment of the (+)enantiomer. Enantioselective degradation was also observed during incubation of sediments under laboratory conditions. Enantioselectivity in these processes is expected to result in ecotoxicological effects that cannot be predicted from our existing knowledge and must be considered in future risk assessment and regulatory decisions.
Influences of Microbial Symbionts on Chemoreception of Their Insect Hosts

Abstract:Chemical communication is widespread among insects and exploited to adjust their behavior, such as food and habitat seeking and preferences, recruitment, defense, and mate attraction. Recently, many studies have revealed that microbial symbionts could regulate host chemical communication by affecting the synthesis and perception of insect semiochemicals. In this paper, we review recent studies of the influence of microbial symbionts on insect chemoreception. Microbial symbionts may influence insect sensitivity to semiochemicals by regulating the synthesis of odorant-binding proteins or chemosensory proteins and olfactory or gustatory receptors and regulating host neurotransmission, thereby adjusting insect behavior. The manipulation of insect chemosensory behavior by microbial symbionts is conducive to their proliferation and dispersal and provides the impetus for insects to change their feeding habits and aggregation and dispersal behavior, which contributes to population differentiation in insects. Future research is necessary to reveal the material and information exchange between both partners to improve our comprehension of the evolution of chemoreception in insects. Manipulating insect chemoreception physiology by inoculating them with microbes could be utilized as a potential approach to managing insect populations.

Abstract:Olfaction is a complex recognition process that is critical for chemical communication in insects. Though some insect species are capable of discrimination between compounds that are structurally similar, little is understood about how this high level of discrimination arises. Some insects rely on discriminating between enantiomers of a compound, demonstrating an ability for highly selective recognition. The role of two major peripheral olfactory proteins in insect olfaction, i.e., odorant-binding proteins (OBPs) and odorant receptors (ORs) has been extensively studied. OBPs and ORs have variable discrimination capabilities, with some found to display highly specialized binding capability, whilst others exhibit promiscuous binding activity. A deeper understanding of how odorant-protein interactions induce a response in an insect relies on further analysis such as structural studies. In this review, we explore the potential role of OBPs and ORs in highly specific recognition, specifically enantiomeric discrimination. We summarize the state of research into OBP and OR function and focus on reported examples in the literature of clear enantiomeric discrimination by these proteins.

Abstract:The exoskeletons of insects are covered by complex mixtures of cuticular hydrocarbons (CHCs) which are involved in social and sexual communication. However, little is known about the relationship between the structures of CHCs and their behavioral activity. The key component of the contact sex pheromone of the parasitoid Lariophagus distinguendus is 3-methylheptacosane (3-MeC27), which is present in CHC profiles of both females and newly emerged males. The CHCs of females and young males elicit wing-fanning behavior in older males. However, as young males age, 3-MeC27 disappears from their CHC profiles and they no longer elicit wing-fanning responses from other males. We applied enantiopure 3-MeC27 and structurally related CHCs (with respect to chain length or methyl-branch position) to the cuticle of aged male dummies and recorded the wing-fanning behavior of responding males. Only the two enantiomers of 3-MeC27 restored the dummies' attractiveness. The addition of structurally related CHCs or various n-alkanes to bioactive dummies of young males and females significantly decreased wing-fanning by test males. Hence, L. distinguendus males respond specifically but not enantioselectively to 3-MeC27, and perceive the CHC profiles as a whole. Both removal (as is the case with 3-MeC27 in aging males) and addition of individual compounds may disrupt the behavioral response.

Abstract:Chirality is an intriguing and fundamental property of natural matter, which is especially crucial in supporting the processes of living systems. The selective interactions between natural chiral compounds are widespread at all levels in living entities and play a vital role in biochemical reactions. The cutting-edge advancements in synthetic chiral inorganic nanostructures have led to significant progress in their applications within biological systems. These developments have unraveled chirality-dependent interactions at the nanoscale and molecular scale, providing a better understanding of intricate process of chiral selection in biological systems and demonstrating the potential of chiral inorganic nanostructures for life science applications. Herein, we summarize recent progress in understanding the chirality origin of inorganic chiral nanoparticles and the development of wet-chemical synthesis. We also discuss the captivating interaction between chiral inorganic nanostructures and biological entities at various scales.
