Abstract:Background The aim of this study was to establish a primary mouse gastric carcinoma cell line. Methods Gastric adenocarcinoma in the body region was induced in immunocompetent BALB/c mice using N-Methyl-N-nitrosourea and a 2% NaCl solution. Fresh gastric cancer tissue samples were cultured in 1640 medium supplemented with 10% fetal bovine serum for primary culture and subculture. Cellular morphology was assessed via light microscopy, and a cell growth curve was established. Genomic and proteomic analyses were conducted to characterize the molecular features of the cell lines. This cell line demonstrated a 100% success rate in forming subcutaneous tumors in BALB/c mice. By integrating proteomic profiles from clinical gastric cancer patients and the murine subcutaneous tumor model, several molecular targets suitable for preclinical investigation were identified. Trametinib, a MEK inhibitor, was employed as a model compound in our preclinical study. Results A novel gastric carcinoma cell line, designated MCC, was established from BALB/c mice. This cell line exhibited a doubling time of approximately 33 hours. Genomic and proteomic analyses identified mutations frequently observed in clinical gastric cancer patients, such as Kras, Egfr, and Ccnd3. Additionally, MCC overexpresses proteins, including SLC1A5, MCC6, and ITGA2, which are significantly upregulated in gastric cancer tissues compared to adjacent non-cancerous tissues. The MCC cell line demonstrated stable tumorigenicity in immunocompetent BALB/c mice, forming subcutaneous tumors that closely resemble the proteomic profile of clinical gastric cancer samples. This high concordance facilitated the identification of several potential therapeutic targets for gastric cancer. Preclinical studies with trametinib revealed that treatment effectively inhibited gastric cancer growth, likely mediated through the activation of immune cells, particularly neutrophils and T cells. Conclusions The MCC cell line is a valuable tool for gastric cancer research, enabling studies of cancer development, progression, and metastasis, and providing a robust platform for evaluating therapeutic strategies and drug efficacy. Its strong tumorigenicity and high correlation with clinical proteomic profiles underscore its relevance in translational research, aiding in the identification of novel therapeutic targets and molecular pathways for effective treatments.
Apoptosis of Implanted Tumor of Human Primary Gastric Cancer Cells in Nude Mice Induced by Resveratrol

Abstract:AIM: To investigate the molecular mechanism of the apoptosis of implanted tumor of human primary gastric cancer cells in nude mice induced by resveratrol. METHODS: Human primary gastric cancer cells were planted into nude mice to establish the cancer model. Resveratrol at different doses were injected near the carcinoma on the nude mice. After treatment, transmission electron microscope and TUNEL staining method were used to detect the apoptosis of implanted tumor cells. Immunohistochemical staining and RT-PCR were used to detect the expression of apoptosis-related genes bcl-2 and bax in implanted tumor. RESULTS: Resveratrol significantly inhibited carcinoma growth when it was injected near the carcinoma. The apoptotic cells in implanted tumor induced by resveratrol were detected by transmission electron microscope and TUNEL staining, immunohistochemical staining and RT-PCR showed resveratrol inhibited bcl-2 expression and increased bax expression in human primary gastric cancer cells. CONCLUSION: Resveratrol inhibits implanted tumor of human primary gastric cancer cells in nude mice through inducing apoptosis. This apoptosis may be mediated by down-regulation of bcl-2 expression and up-regulation of bax expression.
Influence of CO2 Pneumoperitoneum on Gastric Cancer Cells

Abstract:Objective To investigate the influence of CO2 pneumoperitoneum on the adhesive and invasive ability of gastric cancer cells based on the expression of adhesive and invasive molecules. Methods With an artificial CO2 pneumoperitoneum model in vitro, human gastric cancer cells MKN-45, SGC-7901 and MKN-28 were exposed to 3 different CO2 gradients: 9 mm Hg, 15 mm Hg and control group (0 mm Hg). The expression of E-cadherin, ICAM-1, MMP-2 and VEGF-A were measured at 2 and 4 hours exposure by using RT-PCR, CytoMatrixTM kit and ECMatrixTM kit. The pretreated gastric cancer cells were injected into abdominal cavity of nude mice(2×106 cells per mouse). Five mice in each group were sacrificed 4 weeks later to record the number of tumor nodules in abdominal cavity. The remaining mice were kept for observation of survival time. Results The expression of E-cadherin (MKN-45: from 2.26 to 2.19, SGC-7901 :from 2.16 to 2.09、MKN-28 :from 2.06 to 1.99), ICAM-1 (MKN-45 : from 2.20 to 2.28、SGC-7901: from 2.10 to 2.18、MKN-28: from 2.00 to 2.08), MMP-2 (MKN-45:from 2.05 to 2.13、SGC-7901: from 1.95 to 2.03、MKN-28: from 1.85 to 1.93) and VEGF-A(MKN-45 : from 2.10 to 2.16、SGC-7901 :from 2.00 to 2.06、MKN-28: from 1.90 to 1.96) didn't change significantly with increasing pressure and time (P>0.05). The expression of adhesive and invasive molecules didn't change significantly between the experimental groups and the control group. There was no statistical significance of tumor metastasis in abdominal cavity of nude mice(MKN-45:from 22 to 23、SGC-7901 :from 20 to 22、MKN-28:from 21 to 22) and survival time(MKN-45 :from 23 to 21、SGC-7901 :from 22 to 21、MKN-28 :from 22 to 21) among all the groups. Conclusion Under low pressure and short time of CO2 exposure, the adhesive and invasive capacity of gastric cancer cells did not change significantly hence did not increase the possibility of neoplasm metastasis.

Abstract:Objective: To study the influence of carbon dioxide pneumoperitoneum (CP) on abdominal malignant tumor growth. Methods: Thirty nude rats were stratified into 3 groups( n =10). Group 1 was laparotomy, group 2 was CP, and group 3 was nitrogen pneumoperitoneum(NP). Thirty minutes before operation every rat was injected intraabdominally with 5×10 5 mammary adenocarcinoma cells (SHZ88 cell).One hour and 48 h after operation, morphologic alterations of the mesothelium was examined by transmission electron microscopy. Thirty days later, rats were killed to observe the difference of tumor growth in the 3 groups. Results: No significantly morphologic alterations of the mesothelium in group 1 were found. Group 2 and group 3 had the similar alterations such as mesothelial cell degeneration and broken connection between mesothelial cells and damaged underlying basal lamina.The average intraabdominal tumor weight in group 1 was (0.736 8±0.189 5) g, group 2 was (1.786 1±0.039 9) g, group 3 was (0.826 0±0.355 9) g.The average intraabdominal tumor weight of group 2 was significantly increased compared with that of group 3 and group 1( P 0.01).The average weight of group 3 abdominal tumor had no significant difference compared with that of group 1 ( P 0.05) Conclusion: CP significantly promotes abdominal tumor cells growth, the reason may be that carbon dioxide is not suitable for pneumoperitoneum.
Heated and Humidified Co2 Pneumoperitoneum Inhibits Tumour Cell Proliferation, Migration and Invasion in Colon Cancer

Abstract:Background: Peritoneal carcinomatosis (PC) arising from colorectal cancer is associated with poor prognosis and few treatment options are currently available. Laparoscopic CO2 insufflation stimulates the progression and metastatic potential of gastrointestinal carcinomas. However, heated and humidified CO2 pneumoperitoneum (HH-CO2) is a promising treatment for PC, although its effects and mechanism of action in human colon cancer cells remain unclear. This study evaluated the anti-tumour effects of HH-CO2 on human colon cancer in vitro. Methods: Cell viability was assessed using the WST-8 assay in two colon cancer cell lines. Apoptosis was assessed by Annexin V PI flow cytometry, and migration and invasion were examined using wound healing and Transwell (R) invasion assays. The expressions of Bcl-2, Bax, matrix metalloproteinase-2 (MMP-2), E-cadherin, ICAM-1, and CD44 were detected by western blotting. Results: HH-CO2 significantly inhibited cell proliferation, migration, invasion and adhesion. HH-CO2 induced apoptosis and significantly inhibited the expression of Bcl-2, MMP-2, ICAM-1 and CD44, and increased Bax and E-cadherin expression in colon cancer cells. Conclusions: HH-CO2 induces apoptosis and inhibits proliferation, migration, invasion and adhesion of human colon cancer cells. Our results suggest that HH-CO2 may serve as a potential candidate for the treatment and/or prevention of peritoneal carcinomatosis from colorectal cancer and warrant further in vivo investigation.

Abstract:The present study explored the inhibitory effect of hyperthermic CO2 pneumoperitoneum on the proliferation and migration of colon cancer cells, and its mechanism. Colon cancer cell line SW-480 was sealed into a urine collection bag to simulate pneumoperitoneum with 100% CO2 under a pressure of 14 mmHg. The growth and morphology of cells were observed under a microscope, the inhibition on cell proliferation was measured using WST-8 test, cell apoptosis and the cell cycle were monitored using fluorescence-activated cell sorting analysis, the migration of cells was tested using the scratch assay, and the expression of HSP-70, caspase-3, hypoxia-inducible factor-1 alpha (HIF-1 alpha) and matrix metalloproteinase-9 (MMP-9) proteins and genes was investigated using western blotting and reverse transcription polymerase chain reaction. Compared with the control group, there was no significant difference in the CO2 group (P>0.05), while the apoptosis and necrosis rates in the hyperthermo-CO2 group was significantly increased (P<0.05). Compared with the control group, the number of cells at G0/G1 phase significantly increased and the number of cells at S phase significantly decreased in the hyperthermo-CO2 group (P<0.05), indicating that hyperthermo-CO2 could arrest the cell cycle. It was suggested by the results of the scratch assay that cell migration ability enhanced in the CO2 group, but decreased in the hyperthermo-CO2 group compared with the control. CO2 pneumoperitoneum promoted cell migration by upregulating HIF-1 alpha and MMP-9 expression. However, the CO2 pneumoperitoneum with hyperthermia enhanced apoptosis and inhibited migration by upregulating the expression of HSP-70, HIF-1 alpha and caspase-3, but downregulating the expression of MMP-9.

Abstract:Objective To study the effects of different CO2 pressures on adhesive and invasive alility of MKN-28gastric carcinoma cells.Methods With an artificial pneumoperitoneum model in vitro,MKN-28 human gastric carcinoma cells were exposure to CO2 in 3 different pressure groups:9mmHg,15mmHg and control group.The adhesive ability of MKN-28 cells before and after exposed to CO2 was detected by CytoMatrixTMkit.The percentage of cells expressing E-cadherin and ICAM-1 among MKN-28 cells were counted by flowcytometry in vitro.All the MKN-28 gastric carcinoma cells were injected into abdominal cavity of nude mice(2×106cells/mouse) respectively.Five mice of each group were sacrificed 4 weeks later to record the number of tumor nodules in abdominal cavity.The remaining mice were kept to observe the survival time.Results In the 3 groups,all of the adhesive ability of MKN-28 cells(cd/m2).the percentage of cells expressing E-cadherin,and ICAM-1;the number of intra-abdominal tumor metastases.the survival time and the tumor metastasis in abdominal cavity of nude mice were no sigmificant difference call(P>0.05).Conclusions When the pressure is below is below 15 mmHg and for less thqn 4 h,different CO2 pressures and different time intervals do not affect the adhesive and invasive ability of MKN-28 cells in the abdominal cavity,and not increase the chancws of tumonr metastasis.

Abstract:Objective To investigate the influence of different pressures and duration of CO2 pneumoperitoneum on the adhesive and invasive ability of gastric cancer cells based on the expressions of adhesive and invasive molecules.Methods With an artificial CO2 pneumoperitoneum model in vitro,human gastric cancer cell lines including MKN-45,SGC-7901,and MKN-28 were exposed to CO2 in different environments:0 mm Hg(1 mm Hg=0.133 kPa),9 mm Hg(2 h,4 h),and 15 mm Hg(2 h,4 h).The expressions of mRNA of E-cadherin,intercellular adhesion molecule-1(ICAM-1),matrix metalloproteinase-2(MMP-2),and vascular endothelial growth factor-A(VEGF-A)in the different environments were measured by RT-PCR.The expressions of protein of E-cadherin and ICAM-1 in the environments of 0 mm Hg and 15 mm Hg(4 h)were measured by FCM.Results With the increase of duration or pressure,RT-PCR showed that there was a downward trend in the expression of E-cadherin mRNA as well as there were upward trends in the expressions of ICAM-1,MMP-2,and VEGF-A mRNA;FCM showed that there was a downward trend in the expression of E-cadherin protein while the expression of ICAM-1 protein showed the opposite change.But there were no obvious differences under different environment(P0.05).Conclusions Under low pressure(≤15 mm Hg)and short time(≤4 h)of CO2 pneumoperitoneum,the adhesive and invasive ability of gastric cancer cells could not be affected,which means that under this environment,CO2 pneumoperitoneum will not increase the possibility of neoplasm metastasis.
Antitumor Effects of Hyperthermic CO2 Pneumoperitoneum on Human Gastric Cancer Cells.

Abstract:AIM:To elucidate the effects of hyperthermic CO2 pneumoperitoneum on human gastric AGS cells.METHODS:Based on a newly devised in vitro study model, we evaluated the anti-cancer effects of HT-CO2 (42-44 degrees C for 2-4h) on human gastric cancer cells, and also the corresponding mechanisms.RESULTS:HT-CO2 (42-44 degrees C for 2-4h) severely inhibited cell proliferation as assessed by Cell Counting Kit-8 assay, while inducing apoptosis in a temperature- and time-dependent manner demonstrated by annexin-V/PI flow cytometry and morphological analysis (Hoechst/PI fluorescence). In addition, it was found that HT-CO2 (42-44 degrees C for 2-4h) promoted the up-regulation of Bax by western blotting. Significantly, it could also suppress gastric cancer cell invasion and metastasis by in vitro invasion and motility assay.CONCLUSION:In conclusion, HT-CO2 had an efficacious cytotoxic effect on gastric cancer cells through Bax-induced mitochondrial apoptotic signaling. Our studies indicate that it may serve as a potential therapy for peritoneal carcinomatosis of gastric cancer. Further investigations in vivo using animal models are now urgently needed.
Inhibition of Peritoneal Dissemination of Colon Cancer by Hyperthermic Co2 Insufflation: A Novel Approach to Prevent Intraperitoneal Tumor Spread

Abstract:BACKGROUND:The increasing use of laparoscopic surgery for advanced gastrointestinal cancer raises concerns about intra-peritoneal tumor spread. Prevention of peritoneal dissemination is extremely important but a preventive modality is lacking. The aim of this study was to examine a novel approach (hyperthermic CO2 insufflation, HT-CO2) for preventing peritoneal dissemination during laparoscopic surgery. METHODS:A peritoneal dissemination model was established in Balb/c nu/nu mice by intraperitoneal injection of human colon cancer cells (SW1116, 1×106). The mice (n = 48) were subsequently randomized into two groups and subjected to hyperthermic CO2 (43°C, >95% humidity, HT-CO2 group) or standard normothermic CO2 (21°C, <1% relative humidity, NT-CO2 group) insufflation for 3 hours. The mice were sacrificed 28 days later. The peritoneal dissemination was quantitatively analyzed by counting and weighing the peritoneal nodules. The port sites and ascites volume were measured. The peritoneal damage of HT-CO2 was histologically examined with light microscopy and scanning electron microscopy. Intra-abdominal adhesions were evaluated 4 weeks later. RESULTS:The number of peritoneal nodules in the HT-CO2 group was significantly less than that in the NT-CO2 group (10.21±3.72 vs. 67.12±5.49, P<0.01). The mean weight of metastatic tumors in the HT-CO2 group was significantly lower than that in the NT-CO2 group (0.31±0.10g vs. 2.16±0.31g, P<0.01). Massive ascites were found in the NT-CO2 group while significantly less ascites developed in HT-CO2- treated mice (8.26±0.31ml vs. 1.27±0.28ml, P<0.01). No port-site metastases were detected in the HT-CO2 group while the incidence of the NT-CO2 group was 12.5% (3/24). HT-CO2 subjection resulted in slight peritoneal damage; the peritoneum returned to normal within five days. No adhesions formed after HT-CO2 treatment. CONCLUSIONS:HT-CO2 can suppress peritoneal dissemination of colon cancer cells and only causes slight and transient peritoneal damage. HT-CO2 may serve as a promising adjuvant treatment for preventing peritoneal dissemination in laparoscopic resection of advanced colorectal cancer.
Effect of Carbon Dioxide Pneumoperitoneum on Human Renal Cell Carcinoma Proliferation and Metastasis in an Orthotropic Xenograft Nude Mouse Model.

Abstract:INTRODUCTION:This study aimed to explore the effect of carbon dioxide (CO2) pneumoperitoneum on tumor proliferation and metastasis in an orthotropic xenograft nude mice model of human renal cell carcinoma (RCC) and evaluate the safety of CO2 pneumoperitoneum laparoscopy for treating RCC. MATERIAL AND METHODS:RCC 786-0 cells were injected to establish an orthotropic xenograft model. Fifty nude mice were given orthotropic inoculations and randomized to five groups: group A (control); group B (CO2 pneumoperitoneum for 2 h); group C (CO2 pneumoperitoneum for 4 h); group D (CO2 pneumoperitoneum for 4 h and 24 h after waking); group E (CO2 pneumoperitoneum for 4 h and 48 h after waking). The proliferation status was observed in RCC specimens by immunohistochemical staining for Ki67. The protein levels of hypoxia-inducible factor-1α (HIF-1α) and vascular endothelial growth factor (VEGF) were examined by western blotting. RESULTS:All groups showed similar Ki67-positive staining in RCC samples (p > 0.05). The relative expression of HIF-1α and VEGF gradually increased in both group B and group C, as compared with group A, but only the difference between group C and group A reached statistical significance (p < 0.05). The protein levels of HIF-1α and VEGF decreased in both group D and group E, as compared with group B and group C; however, the differences between group D, group E, and group A did not reach statistical significance (p > 0.05). CONCLUSIONS:In an orthotropic xenograft nude mice model of RCC, CO2 pneumoperitoneum has no effect on expression of the cellular proliferation marker Ki67. However, CO2 pneumoperitoneum rapidly induces transient expression of HIF-1α and VEGF. Thus, CO2 pneumoperitoneum laparoscopy may be a safe method for treating RCC.
Development of Intraperitoneally Transplantated Human Ovarian Carcinoma Model with Immune Reconstruction in Severe Combined Immunodeficient Mice

Abstract:OBJECTIVE:To develop an intraperitoneally transplanted human ovarian carcinoma model in humanized severe combined immunodeficient (SCID) mice.METHODS:Twelve CB17SCID mice were randomly divided into 4 groups: group A intraperineally injected with phosphate-buffered saline, group B injected with human ovarian carcinoma cells SKOV3, group C injected with human peripheral blood lymphocyte (PBL) for immune reconstruction, and group D injected with PBL and SKOV3 cells. The behaviors of mice, tumor growth and morphology, human IgG in peripheral blood, cancer antigen 125 (CA125) immunohistochemical staining, tumor infiltrating lymphocyte (TIL), and status of graft versus host disease (GVHD) were detected.RESULTS:The survival period was > 28 days in all groups. The ratio of successful tumor transplantation was 3/3 in groups B and D. The tumor was widespread in peritoneal cavity, mainly in diaphragm, liver, and mesentery, with bloody ascites. The number and size of tumor were less in the humanized group. CA125 expression was positive in primary transplanted tumor cells and SKOV3 cells. IgG was detected in humanized groups (C and D). The level of human IgG was significantly higher in group D than in group C (P < 0.05). TIL infiltration was remarkable in group injected with PBL and SKOV3 cells and failed to be found in group injected with SKOV3 cells. No GVHD was found in all groups.CONCLUSION:An intraperitoneal transplanted human ovarian carcinoma model has been established in humanized SCID mice that simulates the biological behavior of human ovarian carcinoma disseminating intraperitoneally in patients with immune function and may function as an ideal animal model for preclinical research of ovarian carcinoma treatment.
Investigating the mechanisms of peritoneal metastasis in gastric adenocarcinoma using a novel ex vivo peritoneal explant model

Abstract:Gastric adenocarcinoma, commonly known as stomach cancer, has a predilection for metastasis to the peritoneum, which portends limited survival. The peritoneal metastatic cascade remains poorly understood, and existing models fail to recapitulate key elements of the interaction between cancer cells and the peritoneal layer. To explore the underlying cellular and molecular mechanisms of peritoneal metastasis, we developed an ex vivo human peritoneal explant model. Fresh peritoneal tissue samples were suspended, mesothelial layer down but without direct contact, above a monolayer of red-fluorescent dye stained AGS human gastric adenocarcinoma cells for 24 h, then washed thoroughly. Implantation of AGS cells within the explanted peritoneum and invasion beyond the mesothelial layer were examined serially using real-time confocal fluorescence microscopy. Histoarchitecture of the explanted peritoneum was preserved over 5 days ex vivo. Both implantation and invasion were suppressed by restoration of functional E-cadherin through stable transfection of AGS cells, demonstrating sensitivity of the model to molecular manipulation. Thus, our ex vivo human peritoneal explant model permits meaningful investigation of the pathways and mechanism that contribute to peritoneal metastasis. The model will facilitate screening of new therapies that target peritoneal dissemination of gastric, ovarian and colorectal cancer.

Abstract:The laparoscopic operation is playing a more and more important role in the diagnose and treatment of gynaecology malignancy,but the reports about implantation metastasis in puncture site and dissemination of the cancer after this operation are also more and more,which limited its further application in malignant tumor operation.The main researches about the relationship between laparoscopic operation and metastasis lie in the influences about the nature of CO_2,pressure of aeroperitoneum and gas flow rate on the organism immune function,the structure and stress of peritoneum and so on.The aeroperitoneum from CO_2 is related closely to cancer metastasis and dissemination,but its mechanism needs further study.

Abstract:Tumor cell growth is influenced by the cellular microenvironment including the presence of immune cells and blood vessels. Currently, no transplantable gastric cancer syngeneic animal models exist; therefore, we set out to establish a mouse gastric carcinoma cell line, which was named mouse gastric carcinoma cell line 3I (MGCC3I), from forestomach carcinoma developed in benzo[a]pyrene-treated ICR mice. MGCC3I cells showed epithelial-like morphology, multinuclear giant cell formation, and retained an intestinal phenotype, which are similar to human gastric cancer carcinoma cells. The expression of gastric cancer markers MUC1, MUC2, and MUC5AC, and oncogenes c-myc, c-met, cyclin E1, and cancer stem cell marker CD44 was determined in MGCC3I cells. MGCC3I cells formed poorly differentiated stomach tumors following orthotopic implantation into the stomachs of syngeneic ICR mice. Histone deacetylase inhibitors are recognized as a new class of anticancer drugs. The immunological therapeutic effects of the histone deacetylase inhibitors sodium butyrate and valproic acid were evaluated in this new animal tumor model. Sodium butyrate inhibited MGCC3I stomach tumor formation in animal models. Increased tumor infiltration by CD8 T cells and neutrophils was observed in mice treated with sodium butyrate or valproic acid. Depletion of CD8 T cells significantly attenuated tumor regression mediated by histone deacetylase inhibitors, which is correlated with enhancement of the MHC class I pathway in MGCC3I cells. Taken together, we have successfully established an orthotopic transplantable gastric tumor animal model and demonstrated its usefulness in revealing the role of CD8 T cells in the therapeutic effects of sodium butyrate.

The influence of CO2 insufflation on the expression of adhesion molecules on colon cancer cell

Abstract:Objective To investigate the influence of CO2-insufflation on the expression of some adhesion molecules. Methods With an artificial pneumoperitoneum model in vitro, SWI 116 colon carcinoma cells were exposed to CO2-insufflation at different pressure （6, 9, 12 and 15 mmHg） for 1 hour. Control groups were exposed to room air. E-cadherin, ICAM-1, E-selectin, CD44 and CD44v6 were measured 0, 12, 24, 48 and 72 hours after CO2-insufflation using flowcytometry and the mRNA level of these adhesion molecules were detected by real-time PCR. Results In transcription level the expression of ICAM-1 and CD44v6 on SW1116 cell line increased significantly in 6mmHg CO2-insufflation group, in 9mmHg and 12mmHg group, E-cadherin, CD44 and CD44v6 increased significantly, and when the pressure was 15mmHg, CD44v6 increased （ P 〈 0. 05 ）, the expression of these adhesion molecules returned to or even less than the control level by 72 h（ P 〈0.05 ）. With the CO2-insufflation pressure ascending, the increasing expression of the E-cadherin, CD44v6 and ICAM-1 were inhibited in some time points （P 〈0. 05）. In translation level we got the similar result. Conclusion The expression of these adhesion molecules was affected temporarily and bi-directionally after the continuous CO2-insufflation. The higher pressure of CO2-insufflation plays an important role in inhibiting the expression of the adhesion molecules. Abstract:Objective To evaluate the anti-proliferative effect of hyperthermic CO2 pneumoperitoneum (HT-CO2) on colonic cancer cells,to define its mechanism of action,and to assess the feasibility as a new therapeutic procedure for peritoneal carcinomatosis in colon cancer. Methods Based on an in vitro HT-CO2 study model,the cell proliferation was determined with WST-8 assay after treatment by HT-CO2 at 43℃ for 24 hours. Apoptosis was assessed by Annexin-V/PI flow cytometry and transmission electron microscopy. Cell cycle was detected by PI flow cytometry. Results HT-CO2 produced in significant anti-proliferative effect on colonic cancer cells. HT-CO2 sensibly induced apoptosis and G1 phrase cell cycle arrest. Conclusions HT-CO2 can significantly inhibit the proliferation of colonic cancer cells,and the anti-proliferation is attributable to induction of apoptosis and G1 phrase cell cycle arrest. HT-CO2 may potentially serve as a therapeutic procedure for the management of peritoneal carcinomatosis caused by colon cancer.

Abstract:The effects and potential molecular mechanisms underlying carbon dioxide (CO2) pneumoperitoneum on gastric cancer cell apoptosis are not fully understood. In this study, we assessed the effects of CO2 pneumoperitoneum on the apoptosis of MKN-45 gastric cancer cells. Additionally, we investigated the role of HIF-1α in CO2 pneumoperitoneum- induced apoptosis of gastric cancer cells. MKN-45 cells were cultured in CO 2 or air pneumoperitoneum at 0, 12 and 15 mmHg pressures for 4 h. We observed a change in cells morphology and increasing apoptotic ratios in MKN-45 cells when they were put into a 15 mmHg CO2 pneumoperitoneum environment. However, there was no significant difference between the 0, 12 mmHg CO2 pneumoperitoneum and the control groups. Exposure to 15 mmHg CO2 pneumoperitoneum significantly enhanced the expression levels of HIF-1α and Bax, while it attenuated Bcl-2 expression levels. When we inhibited HIF-1α by small interfering RNA (siRNA), we found that the apoptotic ratio of MKN-45 cells decreased in 15 mmHg CO2 pneumoperitoneum. This treatment markedly elevated Bcl-2 levels and decreased Bax expression. These data suggest that CO2 pneumoperitoneum may accelerate the apoptosis of MKN-45 cells at higher pressures. HIF-1α is a crucial factor that affects gastric cancer cell apoptosis by downregulating the Bcl-2/Bax ratio. © 2008 Springer-Verlag.
Influence of laparoscopic CO_2 pneumoperitoneum on proliferation and apoptosis of RCC-949 cells in vitro

Abstract:Objective To investigate the influence of laparoscopic CO2 pneumoperitoneum on the proliferation and apoptosis of renal cell carcinoma (RCC-949) cells in vitro, so as to assess the safety of laparoscopic surgery for renal tumors. Methods Human clear cell renal cell carcinoma (RCC-949) cells were cultured for 1 hour, 2 hours and 4 hours in a simulated laparoscopic CO2 pneumoperitoneum environment at 21 ℃, 99% CO2, 1% O2 and 15 mmHg (1 mmHg=0.133 kPa). The proliferation and apoptosis of RCC-949 cells cultured in the CO2 pneumoperitoneum environment were examined by terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay and methyl thiazolyl tetrazolium (MTT) assay, respectively. Results The optical density (OD) values of cell proliferation in the control group, hypoxia 1 h group, hypoxia 2 h group and hypoxia 4 h group were 0.348±0.051, 0.372±0.082, 0.389±0.110 and 0.378±0.114, respectively. Meanwhile, the apoptosis rates in the control group, hypoxia 1 h group, hypoxia 2 h group and hypoxia 4 h group were (3.2±0.5)%, (3.4±0.3)%, (3.8±0.4)% and (4.3±0.5)%, respectively. There were no significant differences in OD values or the apoptosis rates among all the groups (all P>0.05). Conclusion The proliferation and apoptosis rates of RCC-949 cells are not significantly changed within 4 hours of culture in the simulated laparoscopic CO2 pneumoperitoneum. Thus, we speculate that laparoscopic CO2 pneumoperitoneum does not promote tumor metastasis and implantation of RCC-949 cells, and laparoscopic surgery is safe and effective for renal tumors.
Hyperthermic CO2 Pneumoperitoneum Induces Apoptosis in Human Colon Cancer Cells Through Bax-associated Mitochondrial Pathway.

Abstract:Peritoneal carcinomatosis of colorectal cancer is common and associated with poor prognosis, which poses a serious challenge and satisfactory treatments are urgently needed. Hyperthermic CO2 pneumoperitoneum (HT-CO2) is a new strategy. This study was designed to determine the potential of HT-CO2 against colorectal cancer cells. Based on an in vitro HT-CO2 study model, the anti-tumor efficacy of HT-CO2 (42-44 degrees C for 2-4 h) on human colon cancer COLO 205 cells was evaluated and the mechanisms of actions were analyzed. We found that HT-CO2 (43-44 degrees C for 2-4 h) significantly decreased cell viability as determined by WST-8 assay, and the cytotoxicity was attributable to HT-CO2-induced hyperthermia and extracellular acidification. Apoptosis was the major form of cell killing as demonstrated by Annexin-V/PI flow cytometry and morphological analysis (Hoechst/PI fluorescence microscopy and transmission electron microscopy). Further Western blot analysis and flow cytometric analysis of mitochondrial membrane potential showed that Bax-associated mitochondrial apoptotic pathway played critical role in the induction of apoptosis. We conclude that HT-CO2 has significant cytotoxic effect on colon cancer cells through induction of Bax-associated mitochondrial apoptosis, and the cytocidal effect is attributable to HT-CO2-induced hyperthermia and extracellular acidifications. Our data suggest that HT-CO2 may serve as a potential candidate for treating and/or preventing peritoneal carcinomatosis of colorectal cancer and further investigations both in vitro as well as in vivo in animal models are needed.
