Abstract:AIM To prepare the monoclonal antibodies (mAbs) against Norovirus capsid protein for the development of a rapid assay of Norovirus and to investigate the pathogenesis of this virus. METHODS Sp2/0-Ag-14 myeloma cells were fused with spleen cells of BALB/c mice immunized with the recombinant protein of Norovirus NVgz01 (DQ369797), which was overexpressed in E.coli. The monoclonal antibodies against Norovirus capsid protein were screened by selective culture medium. The obtained Ig subtypes, titer and specificity of the mAbs were examined by ELISA and Western blot respectively. RESULTS After cell fusion and subcloning, four hybridoma cell lines which secreted monoclonal antibodies specifically against Norovirus capsid protein were obtained and named as N2C3, N7C2, N4B1 and N8A9. Indirect ELISA and Western blot assay showed that the four mAbs specifically recognized the Norovirus capsid protein expressed in E.coli. The native Norovirus capsid protein in the stool samples were also recognized by them. The Sandwich ELISA, a rapid detection assay of the expressed and native Norovirus capsid protein, demonstrated that N2C3 and N7C2 were matched successfully. CONCLUSION The mAbs against GII Norovirus capsid protein have been developed, which can be used for the development of detection assay and the basic research of Norovirus.

Characterization of a Norovirus-specific monoclonal antibody that exhibits wide spectrum binding activities

Abstract:Noroviruses (NoVs) are increasingly recognized as the leading cause of acute non-bacterial gastroenteritis worldwide. To screen for NoV-specific monoclonal antibodies (mAbs) with wide spectrum binding activities that could be used for the development of NoV-related detection reagents, we immunized mice with a combination of virus like particles (VLPs) derived from eight different genotypes (two from genogroup I and six from genogroup II), of which two (GI.7 and GII.2) were newly produced VLPs. Indirect enzyme-linked immunosorbent assay (ELISA) confirmed that two mAbs (8D8 and 10B11) bound to all eight major capsid proteins (VP1) with varied binding abilities. Epitope mapping using short peptides covering the N-terminal half of GII.3 VP1 indicated that the binding site of mAb 8D8 was located between amino acid 31 and 60. Multiple amino acid sequence alignment of VP1 suggested that this site harbors conservative sequences across all genogroups. Indirect and sandwich ELISA indicated that mAb 8D8 was unable bind intact VLPs. In summary, we successfully produced GI.7 and GII.2 VLPs using recombinant baculovirus expression system and a cross-reactive mAb by immunizing mice with eight different VLPs that might be useful in the studying and detecting NoVs.

Abstract:Ultra-sensitive, accurate, and specific diagnostic probes are urgently developed to overcome the limitations of conventional norovirus (NV) probes. Herein, we presented the fabrication of a dual-signal probe using multifunctionalized gold and magnetic nanoparticles hybrid composite (Fe3O4 @Au NPs) carriers for the specific detection of NV by magnetic relaxation-fluorescence bimodal sensing. The dual-specific probes were created by modifying Fe3O4 @Au NPs with antibody (Ab) and aptamer-FAM. The antigen-Ab immunoreaction caused aggregation of Fe3O4 @Au NPs, altering the magnetic relaxation signal; simultaneously, the aptamer-FAM that were originally adsorbed on the Au surface were desorbed and cleaved into the solution in the presence of NV, causing a change in the fluorescence signal. Ultrahigh detection sensitives of 0.38 and 3.45 pg/mL were obtained for the dual-modality detection platform, respectively. Furthermore, two quantitative methods obtained satisfactory spiked recoveries for NV-spiked oyster, scallop, strawberry, and tomato samples, consistent with the results of commercial ELISA kits. The results indicated that this dual-specificity assay not only was ultrasensitive but also markedly boosted analysis reliability and precision. Hence, the dual-mode biosensor exhibits high potential for NV detection in the field of food safety.

Abstract:Human norovirus (HuNoV) is the most predominant viral agents of acute gastroenteritis. Point‐of‐care testing (POCT) based on lateral flow immunochromatography (LIFC) has become an important tool for rapid diagnosis of HuNoVs. However, low sensitivity and lack of quantitation are the bottlenecks of traditional LIFC. Thus, we established a rapid and accurate technique that combined immunomagnetic enrichment (IM) with LFIC to identify GII HuNoVs in fecal specimens. Before preparing immunofluorescent nanomagnetic microspheres and achieving the effect of HuNoV enrichment in IM and fluorescent signal in LFIC, amino‐functionalized magnetic beads (MBs) and carboxylated quantum dots (QDs) were coupled at a mass ratio of 4:10. Anti‐HuNoV monoclonal antibody was then conjugated with QDs‐MB. The limit of detection was 1.56 × 104 copies/mL, and the quantitative detection range was 1.56 × 104 copies/mL–1 × 106 copies/mL under optimal circumstances. The common HuNoV genotypes GII.2, GII.3, GII.4, and GII.17 can be detected, there was no cross‐reaction with various enteric viruses, including rotavirus, astrovirus, enterovirus, and sapovirus. A comparison between IM‐LFIC and RT‐qPCR for the detection of 87 fecal specimens showed a high level of agreement (kappa = 0.799). This suggested that the method is rapid and sensitive, making it a promising option for point‐of‐care testing in the future.

Abstract:Norovirus infection is the main cause of epidemic acute gastroenteritis outbreaks. As a result, norovirus merits careful attention. Acute gastroenteritis caused by norovirus is very harmful, necessitating effective prevention and monitoring. This study extracted norovirus using the Fe3O4 nanomagnetic bead method while establishing a rapid and sensitive quantitative norovirus detection method for emergency use. Norovirus RNA was extracted from norovirus-positive stool samples, norovirus-specific primers were designed, and SYBR Green I fluorescent dye was used to construct specific fluorescence quantitative reverse-transcription polymerase chain reaction (RT-PCR) method. Norovirus in the sample was successfully detected with the quantitative RT-PCR method established in this study, with a measured cycle threshold value of 28.73; the melting temperature value of the PCR product was approximately 83.5 °C. Additionally, by analyzing melting curve with quantitative PCR and evaluating the repeatability of the detection method, the performance of the reaction system in terms of specificity and repeatability was determined to be good. The fluorescence quantitative detection method established here can rapidly and effectively detect norovirus, and the method has good specificity and high repeatability. Currently, there is a trend of norovirus infection becoming increasingly severe. Thus, this rapid and sensitive fluorescence quantitative detection method will play a significant role in the monitoring and detection of norovirus and is thus worthy of promotion and application.

Abstract:We previously identified a Nanobody (termed Nano-85) that bound to a highly conserved region on the norovirus capsid. In this study, the Nanobody was biotinylated and gold conjugated for a lateral flow immunoassay (termed Nano-IC). We showed that the Nano-IC assay was capable of detecting at least four antigenically distinct GII genotypes, including the newly emerging GII.17. In the clinical setting, the Nano-IC assay had sensitivities equivalent to other commercially available lateral flow systems. The Nano-IC method was capable of producing results in ~5 min, which makes this method useful in settings that require rapid diagnosis, such as cruise ship outbreaks and elder care facilities. The Nano-IC assay has several advantages over antibody-based IC methods: for example, Nanobodies can be readily produced in large quantities, they are generally more stable than conventional antibodies, and the Nanobody binding sites can be easily obtained by X-ray crystallography.

Abstract:Background Human noroviruses (HuNoVs) are a major cause of nonbacterial gastroenteritis in all age groups worldwide. HuNoVs can be detected in vitro using molecular assays such as RT-PCR and RT-qPCR. However, these molecular-based techniques require special equipment, unique reagents, experienced personnel, and extended time to obtain results. Besides, the diversity of viral genotypes is high. Therefore, methods that are rapid, broad-range and effective in the detection of HuNoVs are desiderated for screening the feces or vomit of infected people during outbreaks. Results In this study, a colloidal-gold-based immunochromatographic assay (ICA) was developed for effective detection of HuNoVs in clinical samples. Monoclonal antibodies (MAbs) against the shell (S) domain in the major capsid protein of HuNoVs were used in the ICA. The limitations of detection for HuNoVs in clinical samples were 1.2 × 10 6 genomic copies per gram of stool sample (gc/g) and 4.4 × 10 5 gc/g for genogroup I and II (GI and GII) HuNoVs, respectively. A total of 122 clinical samples were tested for HuNoVs by ICA and compared against RT-qPCR. The relative sensitivity, specificity and agreement of ICA was 84.2% (95% CI: 83.6–84.8%), 100.0% (95% CI: 98.5–100.0%) and 87.7% (95% CI: 85.6–89.8%), respectively. No cross-reaction with other common enteric viruses or bacteria was observed. The ICA detected a broad range of genotypes, including GI.1, GI.3, GI.4, GI.6, GI.14, GII.2, GII.3, GII.4, GII.6, GII.13, and GII.17 HuNoVs. Conclusions This study demonstrates that ICA targeting the S domain of VP1 is a promising candidate for effectively identifying the different genotypes of HuNoVs in clinical samples with high sensitivity and specificity.

Abstract:A one-step multiplex real-time reverse transcription-PCR (RT-PCR) assay was developed for one-tube and simultaneous detection of three genogroups of human norovirus, genogroup I, II and IV (GI, GII and GIV). The specificity and sensitivity of the assay were evaluated and 50 samples were tested by using this assay. The results showed that the multiplex assay had high sensitivity and specificity. The amplification efficiencies of the assay were 91.3%, 90.1%, 88.9% and the detection limits were up to 16.9, 6.3, 43.0 copies/reaction respectively for norovirus GI, GII and GIV detection. No cross-reaction with the other examined RNA viruses was observed, and the qualitative analysis of samples showed that the multiplex assay had a good consistency with its corresponding monoplex assays for the detection of norovirus GI, GII and GIV (Kappa values were 0.848, 0.876 and 0.812 respectively).

Abstract:ABSTRACT Noroviruses (NoVs) commonly cause acute gastroenteritis outbreaks. Broadly reactive diagnostic assays are essential for rapid detection of NoV infections. We previously generated a panel of broadly reactive monoclonal antibodies (MAbs). We characterized MAb reactivities by use of virus-like particles (VLPs) from 16 different NoV genotypes (6 from genogroup I [GI], 9 from GII, and 1 from GIV) coating a microtiter plate (direct enzyme-linked immunosorbent assay [ELISA]) and by Western blotting. MAbs were genotype specific or recognized multiple genotypes within a genogroup and between genogroups. We next applied surface plasmon resonance (SPR) analysis to measure MAb dissociation constants ( K d ) as a surrogate for binding affinity; a K d level of <10 nM was regarded as indicating strong binding. Some MAbs did not interact with the VLPs by SPR analysis. To further assess this lack of MAb-VLP interaction, the MAbs were evaluated for the ability to identify NoV VLPs in a capture ELISA. Those MAbs for which a K d could not be measured by SPR analysis also failed to capture the NoV VLPs; in contrast, those with a measurable K d gave a positive signal in the capture ELISA. Thus, some broadly cross-reactive epitopes in the VP1 protruding domain may be partially masked on intact particles. One MAb, NV23, was able to detect genogroup I, II, and IV VLPs from 16 genotypes tested by sandwich ELISA, and it successfully detected NoVs in stool samples positive by real-time reverse transcription-PCR when the threshold cycle ( C T ) value was <31. Biochemical analyses of MAb reactivity, including SPR analysis, identified NV23 as a broadly reactive ligand for application in norovirus diagnostic assays.
A norovirus detection architecture based on isothermal amplification and expanded genetic systems

Abstract:Noroviruses are the major cause of global viral gastroenteritis with short incubation times and small inoculums required for infection. This creates a need for a rapid molecular test for norovirus for early diagnosis, in the hope of preventing the spread of the disease. Non-chemists generally use off-the shelf reagents and natural DNA to create such tests, suffering from background noise that comes from adventitious DNA and RNA (collectively xNA) that is abundant in real biological samples, especially feces, a common location for norovirus. Here, we create an assay that combines artificially expanded genetic information systems (AEGIS, which adds nucleotides to the four in standard xNA, pairing orthogonally to A:T and G:C) with loop-mediated isothermal amplification (LAMP) to amplify norovirus RNA at constant temperatures, without the power or instrument requirements of PCR cycling. This assay was then validated using feces contaminated with murine norovirus (MNV). Treating stool samples with ammonia extracts the MNV RNA, which is then amplified in an AEGIS-RT-LAMP where AEGIS segments are incorporated both into an internal LAMP primer and into a molecular beacon stem, the second lowering background signaling noise. This is coupled with RNase H nicking during sample amplification, allowing detection of as few as 10 copies of noroviral RNA in a stool sample, generating a fluorescent signal visible to human eye, all in a closed reaction vessel.
Development of Recombinant Antibodies and Its Application in Immunomagnetic Separation-Based Rapid Detection of Vibrio Cholerae in Aquatic Environments

Abstract:Cholera caused by Vibrio cholerae remains a major public health concern in many countries. The greatest obstacle to detection of V. cholerae contamination in drinking water or aquatic environments mainly relates to sample preparation steps, especially the enrichment step. In this study, immunomagnetic separation methods were developed based on sequence-defined recombinant antibodies (rAbs) against V. cholerae, then used for the specific and efficient enrichment of V. cholerae in water samples. Using the variable region genes of the anti-V. cholerae monoclonal antibodies (mAbs) 5F2, the full-length IgG rAbs (R5F2) were produced using mammalian human embryonic kidney 293T cells. Two antibodies, 5F2 and R5F2, were used to prepare immunomagnetic beads (IMBs), and their capture efficiencies (CEs) were evaluated. The results showed that 0.4 mg of 5F2-IMBs and R5F2-IMBs exhibited good CEs (96.0% and 75.9%, respectively) against V. cholerae within 40 min. The IMBs could still effectively capture V. cholerae in large-volume reaction systems (5 ml to 25 ml). The CEs of 5F2-IMBs and R5F2-IMBs ranged from 90.2% to 70.7% and 65.1% to 44.2%, respectively. Furthermore, 5F2-IMBs and R5F2-IMBs did not show significant cross-reactivity with other bacteria and exhibited high specificity. When R5F2-IMS was used in combination with quantitative real-time PCR, the detection limit was approximately 5 colony-forming units/25 ml after enrichment for 4 h. Our results suggest that the rAbs produced herein could provide useful alternatives to traditional hybridoma-based antibodies for accurate detection of V. cholerae in food safety and environmental monitoring.

Abstract:ABSTRACT Human norovirus (HuNoV) is recognized as the leading causative agent of foodborne outbreaks of epidemic gastroenteritis. Consequently, there is a high demand for developing point-of-care testing for HuNoV. We developed an origami microfluidic device that facilitates rapid detection of murine norovirus 1 (MNV-1), a surrogate for HuNoV, encompassing the entire process from sample preparation to result visualization. This process includes RNA absorption via a paper strip, RNA amplification using recombinase polymerase amplification (RPA), and a lateral flow assay for signal readout. The on-chip detection of MNV-1 was completed within 35 min, demonstrating 100% specificity to MNV-1 in our settings. The detection limit of this microfluidic device for MNV-1 was 200 PFU/mL, comparable to the in-tube RPA reaction. It also successfully detected MNV-1 in lettuce and raspberries at concentrations of 170 PFU/g and 230 PFU/g, respectively, without requiring extra concentration steps. This device demonstrates high compatibility with isothermal nucleic acid amplification and holds significant potential for detecting foodborne viruses in agri-food products in remote and resource-limited settings. IMPORTANCE HuNoV belongs to the family of Caliciviridae and is a leading cause of acute gastroenteritis that can be transmitted through contaminated foods. HuNoV causes around one out of five cases of acute gastroenteritis that lead to diarrhea and vomiting, placing a substantial burden on the healthcare system worldwide. HuNoV outbreaks can occur when food is contaminated at the source (e.g., wild mussels exposed to polluted water), on farms (e.g., during crop cultivation, harvesting, or livestock handling), during packaging, or at catered events. The research outcomes of this study expand the approaches of HuNoV testing, adding value to the framework for routine testing of food products. This microfluidic device can facilitate the monitoring of HuNoV outbreaks, reduce the economic loss of the agri-food industry, and enhance food safety.

An ultrasensitive SiO2-encapsulated alloyed CdZnSeS quantum dot-molecular beacon nanobiosensor for norovirus

Abstract:Ultrasensitive, rapid and selective diagnostic probes are urgently needed to overcome the limitations of traditional probes for norovirus (NV). Here, we report the detection of NV genogroup II via nucleic acid hybridization technology using a quantum dot (QD)-conjugated molecular beacon (MB) probe. To boost the sensitivity of the MB assay system, an ultrasensitive QD fluorophore with unique optical properties was synthesized, characterized and exploited as a fluorescence signal generator. Alloyed thioglycolic (TGA)-capped CdZnSeS QDs with a high photoluminescence (PL) quantum yield (QY) value of 92% were synthesized, and a modified silanization method was employed to encapsulate the thiol-capped QDs in a silica layer. The resulting highly luminescent alloyed SiO2-coated CdZnSeS QDs had a remarkable PL QY value of 98%. Transmission electron microscopy and dynamic light scattering confirmed the monodispersity of the alloyed nanocrystals, and zeta potential analysis confirmed their colloidal stability. Powder X-ray diffraction and PL lifetime measurements confirmed the surface modification of the QDs. The alloyed TGA-capped and SiO2-coated CdZnSeS QD-conjugated MB bioprobes detected extremely low concentrations of NV RNA. Ultrasensitive detection of low concentrations of NV RNA with a limit of detection (LOD) of 8.2copies/mL in human serum and a LOD of 9.3 copies/mL in buffer was achieved using the SiO2-coated CdZnSeS QD-MB probes, an increase in sensitivity of 3-fold compared with the detection limit for NV RNA using TGA-capped CdZnSeS QD-MBs. The additional merits of our detection system are rapidity, specificity and improved sensitivity over conventional molecular test probes.

Abstract:Enzyme-linked immunosorbent assay (ELISA) provides a convenient way for the detection of viral pathogens. However, conventional ELISA performed on mirowell plates suffers from poor sensitivity, laborious coating and complicated blocking procedures. Herein, we designed a sensitive colorimetric immunoassay by taking advantages of the enrichment and isolation ability of magnetic beads (MBs) and the high loading capacity of gold nanoparticles (AuNPs) for detecting respiratory syncytial virus (RSV) as a pathogen model. RSV was selectively captured and preconcentrated from samples with antibodies functionalized MBs, followed by binding with antibodies labeled AuNPs, which carrying a large amount of alkaline phosphatase (ALP) molecules for colorimetric signal amplification by catalyzing the dephosphorylation of non-colored pNPP to generate colored product pNP. After optimizing the experimental conditions based on the principle of low nonspecific signal, low cost, and high sensitivity, the analytical sensitivity of the developed immunoassay can be improved to 0.27 pg/mL, which is over sevenfold higher than that of commercially available RSV ELISA kits (2 pg/mL). In addition, the total assay time was less than 2.5 h without any pretreatment, which is much more rapid than other reported assays. Therefore, the proposed immunoassay holds great promise for the fabrication of rapid, sensitive, and economic method for the viral pathogen detection.

Abstract:Graphical and Lay Summary Immunoassay has been developed to detect highly contagious viruses. However, high sensitivity is required for these immunoassays to be reliable for detecting infectious agents in low amount viral particles, such as norovirus. Here, we addressed this issue by incorporating bioconjugates to Ag@Fe3O4@Au NPs NPs as a magneto‐catalyst. We also addressed the issue in high impurity detection for norovirus immunoassay by a simple pre‐treatment. We obtained recovery of 99.95% for spiked feces and 6.97 NoV copy number/mL for clinically‐isolated norovirus. A new method has been developed to improve the detection of norovirus (NoV) in complex fecal samples using nanocatalyst‐based immunoassays. The method involves using multifunctional trimetallic nanoparticles, known as Ag@Fe3O4@Au NPs. These nanoparticles consist of a core of silver (Ag) and a shell of iron oxide (Fe3O4) decorated with isolated gold nanoparticles (Au NPs). The nanoparticles have enhanced catalytic activity, making them an ideal nanocatalyst for reducing 4‐nitrophenol (4‐NP, yellow) to 4‐aminophenol (4‐AP, colorless). The developed Ag@Fe3O4@Au NPs‐based immunoassay achieved a limit of detection (LOD) of 1.9 pg/mL for norovirus‐like particles (NoV‐LP) and 6.97 RNA copy number/mL for fecal NoV. In fecal sample analysis for NoV, a heat treatment at 65°C was necessary to prevent degradation of the target protein, ensuring sensitive detection. This work successfully combined multifunctional nanocatalysts for advanced immunoassays, which could contribute to developing nano‐biosensing platforms.

Abstract:Noroviruses are recognized worldwide as the principal cause of acute, non-bacterial gastroenteritis, resulting in 19-21 million cases of disease every year in the United States. Noroviruses have a very low infectious dose, a short incubation period, high resistance to traditional disinfection techniques and multiple modes of transmission, making early, point-of-care detection essential for controlling the spread of the disease. The traditional diagnostic tools, electron microscopy, RT-PCR and ELISA require sophisticated and expensive instrumentation, and are considered too laborious and slow to be useful during severe outbreaks. In this paper we describe the development of a new, rapid and sensitive lateral-flow assay using labeled phage particles for the detection of the prototypical norovirus GI.1 (Norwalk), with a limit of detection of 107 virus-like particles per mL, one hundred-fold lower than a conventional gold nanoparticle lateral-flow assay using the same antibody pair.
Production and application of monoclonal antibodies to Macrobrachium rosenbergii Nodavirus

Abstract:Whitish muscle diseases of freshwater giant prawn, Macrobrachium rosenbergii, is a new epidemic disease in the larvae and post-larvae of M. Rosenbergii in recent years. The clinical sign of the disease is a whitish or opacity appearance of the muscles in abdomen. Mortalities may reach more than 60%, sometimes even reach 100%. The pathogen of the disease is confirmed as Macrobrachium rosenbergii Nodavirus(MrNV), an icosahedral and non-enveloped virus sized from 25-26 nm with two linear ssRNA sized 3.0 kb and 1.3 kb respectively. MrNV was the first nodavirus founded in crustacean. To develop a rapid method for MrNV detection, monoclonal antibody against MrNV was prepared and used for virus detection. Briefly, mouse myeloma cells(SP2/0) were fused with spleen cells from BALB/c immunized with purified MrNV from diseased postlarvae(PL). Positive colonies were screened by indirect ELISA with the plates coated with purified MrNV and cloned 2-3 times with limited dilution method. Twelve positive hybridoma cell lines secreting monoclonal antibodies (Mabs) against MrNV were at last obtained and used for ascites preparation. The titres of positive ascites Mabs were ranged from 1∶10~5 to 1∶10~6 by indirect ELISA. These twelve Mabs were isotyped by Southern Biotechnology clonotyping system, and the results showed that six Mabs were IgG1, four IgG2a and two IgG2b, respectively. No cross reactions were found when the twelve Mabs were used for indirect ELISA with white spot syndrome virus(WSSV) and Taura syndrome virus(TSV). A triple antibody sandwich enzyme-linked immunosorbent assay(TAS-ELISA) was developed with the plates pre-coated with rabbit polyclonal antibodies against MrNV for virus capturing, followed by MrNV and samples incubation, Mabs and goat-anti-mouse-IgG antibodies labeled with HRP. Mab 2B5, characterized as Mab against 43 kD coat protein of MrNV by Western blot, was selected for TAS-ELISA. The results showed that TAS-ELISA can be used successfully for minimum virus detection with the sensitivity of 0.98 ng MrNV, or with the viral titer of 1∶40960 when typical diseased PL homogenization supernatants were used for TAS-ELISA. TAS-ELISA could be considered as a useful technique for MrNV detection as well as for epidemiological studies for its rapid and inexpensive cost with high specificity and sensitivity.

Abstract:Norovirus is an infectious disease that can cause non-bacterial gastroenteritis, which has a low infectious dose, rapid onset, and strong transmission ability; therefore, rapid and sensitive detection is essential to reduce the transmission of gastroenteritis. In the study, a norovirus GII loop-mediated isothermal amplification assay was developed and prepared into freeze-drying microspheres, and a closed-cassette-based, integrated, reagent-ambient storage, on-site instant detection platform for norovirus GII was constructed using a commercial, fully automated nucleic acid analyzer with integrated magnetic bearing based nuclear acid extraction and nucleic acid detection, with a sensitivity of 10 copies/μL, with no cross-reactivity with other 5 viruses. For 28 simulated samples, the integrated assay platform was consistent with the experimental results of reverse transcription-quantitative polymerase chain reaction (RT-qPCR) assays after conventional laboratory nucleic acid extraction. The entire process can be finished in about 1 h, which is ideal for immediate rapid detection.
