Abstract:We report a high-speed, dispersion-encoded, full-range (DEFR) swept-source optical coherence tomography system for in vivo ocular imaging and biometry of small animals. The fast DEFR algorithm removes the depth ambiguity, gives access to objects located at the zero delay position, and doubles the sampling depth to 2x5.0 mm (at -101 to -71 dB sensitivity) in a single scan using 2048 samples/depth scan 0.43 nm line width of a light source operating at 1056 nm with 70 nm tuning range. The acquisition speed (frames of 512 depth scans in 18.3 ms) permits precise on-line monitoring during positioning and provides cross-sectional views of the mouse eye. Preliminary studies demonstrate high-throughput, reproducible assessment of multiple biometric features (e.g., day-to-day reproducibility of axial length measurement +/-5.3 microm) that is insensitive to eye motion sufficient for long-term monitoring.

Abstract:Optical coherence tomography (OCT) is a new imaging technology that uses an optical analog of ultrasound imaging for biological tissues. Image segmentation plays an important role in dealing with quantitative analysis of medical images. We have proposed a novel framework to deal with the low intensity problem, based on the labeled patches and Bayesian classification ( LPBC ) model. The proposed method includes training and testing phases. During the training phase, firstly, we manually select the sub-images of background and Region of Interest (ROI) from the training image, and then extract features by patches. Finally, we train the Bayesian model with the features. The segmentation threshold of each patch is computed by the learned Bayesian model. In addition, we have collected a new dataset of mouse eyes in vivo with OCT, named MEVOCT , which can be found at URL https://17861318579.github.io/LPBC. MEVOCT consists of 20 high-resolution images. The resolution of every image is 2048 × 2048 pixels. The experimental results demonstrate the effectiveness of the LPBC method on the new MEVOCT dataset. The ROI segmentation is of great importance for the distortion correction.

Automatic biometry of the anterior segment during accommodation imaged by optical coherence tomography

Abstract:Objective: To test accuracy and repeatability of a software algorithm that performs automatic biometry of the anterior segment of the human eye imaged with long scan depth optical coherence tomography (OCT). Methods: The ocular anterior segment imaging was performed with custom-built long scan depth OCT. An automatic software algorithm including boundary segmentation, image registration, and optical correction was developed for fast and reliable biometric measurements based on the OCT images. The boundary segmentation algorithm mainly used the gradient information of images and applied the shortest path search based on the dynamic programming to optimize the edge finding. The automatic algorithm was validated by comparison of the biometric dimensions between automatic and manual measurements and repeatability study. Results: Biometric dimensions of the anterior segment, including central corneal thickness, anterior chamber depth, pupil diameter, crystalline lens thickness, and radii of curvature of the anterior and posterior surfaces of lens, were obtained by the automatic algorithm successfully. There were no significant differences between the automatic and manual measurements for all biometric dimensions. The intraclass correlation coefficients (ICC) of agreement between automatic and manual measurements ranged from 0.85 to 0.98. The coefficients of repeatability and ICC for all automatic dimensions were satisfactory (1.1%-6.1% and 0.663-0.990, respectively). Conclusions: The high accuracy, good repeatability, and fast execution speed for automatic measurement of the anterior segment dimensions on the OCT images were demonstrated. The application of this automatic biometry is promising for investigating dynamic changes of human anterior segment during accommodation in real time.

Enhancing Sensitivity of High Resolution Optical Coherence Tomography Using an Optional Spectrally Encoded Extended Source (conference Presentation)

Abstract:High-resolution optical coherence tomography (OCT) is of critical importance to disease diagnosis because it is capable of providing detailed microstructural information of the biological tissues. However, a compromise usually has to be made between its spatial resolutions and sensitivity due to the suboptimal spectral response of the system components, such as the linear camera, the dispersion grating, and the focusing lenses, etc. In this study, we demonstrate an OCT system that achieves both high spatial resolutions and enhanced sensitivity through utilizing a spectrally encoded source. The system achieves a lateral resolution of 3.1 μm and an axial resolution of 2.3 μm in air; when with a simple dispersive prism placed in the infinity space of the sample arm optics, the illumination beam on the sample is transformed into a line source with a visual angle of 10.3 mrad. Such an extended source technique allows a ~4 times larger maximum permissible exposure (MPE) than its point source counterpart, which thus improves the system sensitivity by ~6dB. In addition, the dispersive prism can be conveniently switched to a reflector. Such flexibility helps increase the penetration depth of the system without increasing the complexity of the current point source devices. We conducted experiments to characterize the system’s imaging capability using the human fingertip in vivo and the swine eye optic never disc ex vivo. The higher penetration depth of such a system over the conventional point source OCT system is also demonstrated in these two tissues.

Abstract:Optical coherence tomography angiography (OCTA) is a non-contact, high-resolution biomedical imaging technology widely used in eye disease diagnosis. This paper introduces a novel mouse retinal near-infrared imaging system with a spectral domain OCTA operating at a 250 kHz A-scan rate. The system achieves a 2.3 μm axial resolution and a 1.2 mm, 5.7 dB imaging range. With a robust signal-to-noise ratio of 99.95 dB at 250 kHz A-scan, after optimizing the scanning parameters, the system captured clear retinal cross-sections, optical coherence tomography (OCT) scans, and OCTA images during the imaging process of the mouse retina.

Abstract:Objects: A novel computer method for three-dimensional in vivo imaging of human anterior eye segment is described with optical coherence tomography. Methods: A laser beam was projected at an oblique angle on the cornea and scanned to acquire 18 optical section images at 10-degree intervals across the corneal area. Tissue's edges were detected using Canny detector, and Track Points arithmetic was designed to extract the single-pixel edges, which were smoothed by curve fitness. According to the characters of rotating image, which appeared displaced because of the motion blur of patients, the classical hat-head method was improved to build 3D model for OCT images sequence. Three B-Spine interpolation and curve fitness would be used to obtain the outlines if they were lost in original OCT images. The optical section images were segmented to construct a series of anterior eye. Imaging was performed in normal subject and patients. Results: A series of digital 3D model of anterior eye was reconstructed, which allowed enhanced visualization of the micro-structures, including the section of anterior eye segment at any rotating degree, the contours of corneal thickness and curvature, and the depths and angles of anterior chamber. Conclusion: A method for in vivo three-dimensional anterior eye imaging was developed, which has potential as a tool for objective, noninvasive and real-time evaluation of diseases.

Abstract:Non-invasive imaging plays a crucial role in diagnosing and studying eye diseases. However, existing photoacoustic ophthalmoscopy (PAOM) techniques in mice have limitations due to handling restrictions, suboptimal optical properties, limited availability of light sources and permissible light fluence at the retina. This study introduces an innovative approach that utilizes Rose Bengal, a contrast agent, to enhance PAOM contrast. This enables visualization of deeper structures like the choroidal microvasculature and sclera in the mouse eye using visible light. The integration of near-infrared-II optical coherence tomography (NIR-II OCT) provides additional tissue contrast and insights into potential NIR-II PAOM capabilities. To optimize imaging, we developed a cost-effective 3D printable mouse eye phantom and a fully 3D printable tip/tilt mouse platform. This solution elevates PAOM to a user-friendly technology, which can be used to address pressing research questions concerning several ocular diseases such as myopia, glaucoma and/or age-related macular degeneration in the future.

Abstract:Background: Biometric parameters play an important role in studies on myopia. This study aimed to explore the application of a commercially available two-dimensional visualized swept-source optical coherence tomography (OCT) system, for in vivo biometric measurement in multiple myopia model species. Methods: In this study, chickens, guinea pigs, and C57BL/6 mice underwent eye imaging with the commercially available OCT (CASIA2), and the original images were used to calculate the central corneal thickness, anterior chamber depth (ACD), lens thickness (LT), vitreous chamber depth (VCD), and axial length (AL). The retinal thickness and choroidal thickness were also calculated in chicken eyes. The repeatability of the biometric measurement outcomes was analyzed. Results: Excellent repeatable AL measurements were obtained for all three species, with an intraclass correlation coefficient (ICC) of >= 0.941 and a within-subject standard deviation of <= 0.055. Excellent repeatability was found in chicken eyes for ACD, LT, and VCD, with an ICC of >= 0.932; in guinea pig eyes for ACD and VCD, with an ICC of >= 0.934; and in mouse eyes for LT, with an ICC of >= 0.941. Conclusions: It is effective to use commercially available OCT to measure biometric parameters in chickens, guinea pigs, and C57BL/6 mice. This methodology could potentially increase the accuracy and efficiency of future myopia animal experiments.

Abstract:NaCl based solutions were applied as osmotic stress agents to alter the hydration state of the mouse eye. Full-eye responses to these osmotic challenges were monitored in vivo using a custom-built optical coherence tomography (OCT) with an extended imaging range of 12.38 mm. Dynamic changes in the mouse eye were quantified based on the OCT images using several parameters, including the central corneal thickness (CCT), the anterior chamber depth (ACD), the crystalline lens thickness (LT), the cornearetina distance (CRD), the iris curvature (IC), and the lens scattering intensity (LSI). Apparent but reversible changes in the morphology of almost all the ocular components and the light transparency of the lens are exhibited. Particularly, the ocular dehydration induced by the hypertonic challenges resulted in a closing of the iridocorneal angle and an opacification of the lens. Our results indicated that the ocular hydration is an important physiological process which might be correlated with various ocular disorders, such as dry eye, cataract, and angle-closure glaucoma, and would affect the biometry and imaging of the eye. OCT uniquely enables the comprehensive study of the dynamic full-eye responses to the ocular hydration in vivo.

Abstract:Current optical coherence tomography (OCT) technology, which is used for imaging the eye's anterior segment, has been established as a clinical gold standard for the diagnosis of corneal diseases. However, the cellular resolution level information that is critical for many clinical applications is still not available. The major technical challenges toward cellular resolution OCT imaging are the limited ranging depth and depth of focus (DOF). In this work, we present a novel ultrahigh resolution OCT system that achieves an isotropic spatial resolution of <2 µm in tissue. The proposed system could approximately double the ranging depth and extend the DOF using the dual-spectrometer design and the forward-model based digital refocusing method, respectively. We demonstrate that the novel system is capable of visualizing the full thickness of the pig cornea over the ranging depth of 3.5 mm and the border of the corneal endothelial cells 8 times Rayleigh range away from the focal plane. This technology has the potential to realize cellular resolution corneal imaging in vivo.

Abstract:Eye movements are commonly seen as an obstacle to high-resolution ophthalmic imaging. In this context we study the natural axial movements of the in vivo human eye and show that they can be used to modulate the optical phase and retrieve tomographic images via time-domain full-field optical coherence tomography (TD-FF-OCT). This approach opens a path to a simplified ophthalmic TD-FF-OCT device, operating without the usual piezo motor-camera synchronization. The device demonstrates in vivo human corneal images under the different image retrieval schemes (2-phase and 4-phase) and different exposure times (3.5 ms, 10 ms, 20 ms). Data on eye movements, acquired with a spectral-domain OCT with axial eye tracking (180 B-scans/s), are used to study the influence of ocular motion on the probability of capturing high-signal tomographic images without phase washout. The optimal combinations of camera acquisition speed and amplitude of piezo modulation are proposed and discussed.

Abstract:A full-field swept-source optical coherence tomography (FF-SS-OCT) for in vivo murine retinal imaging is demonstrated. The on-axis FF-SS-OCT system was built in a Mach-Zehnder interferometer configuration employing a tunable laser source with an adjustable sweep rate and sweep range in conjunction with a fast 2D-CMOS camera. A large field retinal (coherent) illumination was accomplished using an imaging interface comprised of a short-focal length imaging lens and a contact lens. The magnification between the camera and retina (spatial sampling) was appropriately chosen to record the microscopic structural features of the retina in the image. A pupil stop was employed in the detection path to reject unwanted backscattering from the mouse eye and other sources and limit aberrations distorting the retinal images. In vivo mouse retinal imaging was performed at a sweep rate of 150 Hz to acquire volumes unaffected by the system vibrations, which predominated at lower frequencies. Operating the FF-SS-OCT at this speed yielded an effective axial scan rate of 20 million A-scans/s and a field of view of 820 × 410 µm (24.12° × 12.06°). High-quality retinal B-scans and enface images of the retina were obtained with the SS-FF-OCT, revealing all major retinal layers and vascular plexuses.

Abstract:Optical coherence tomography (OCT), a promising noninvasive bioimaging technique, has become one of the most successful optical technologies implemented in medicine and clinical practice. Here we report a novel technique of depth-resolved transverse-plane motion tracking with configurable measurement features via optical coherence tomography, termed OCT-MT. Based on OCT circular scanning combined with speckle spatial oversampling, the OCT-MT technique can perform depth-resolved transverse-plane motion tracking. Benefitting from the optical interference and depth-resolved feature, the proposed OCT-MT can reduce the requirements on the input power of the irradiation signal and the surface reflectivity and roughness of the target, when performing motion tracking. Furthermore, OCT-MT can conduct such kind of motion tracking with configurable measurement ranges and resolutions by configuring A-line number per scanning circle, circular scanning radius, and A-line scanning time. The proposed OCT-MT technique may expand the ability of motion tracking for OCT in addition to imaging.

Abstract:Optical coherence tomography (OCT) is an emerging optical imaging technology that holds great potential in medical and biological applications. Apart from its conventional ophthalmic uses, it has found extensive applications in studying various brain activities and disorders in anesthetized/restricted rodents, with a particular focus on visualizing brain blood vessel morphology and function. However, developing a compact wearable OCT probe for studying the brain activity/disorders in freely moving rodents is challenging due to the requirements for stability and lightweight design. Here, we report a robust wearable OCT probe, which, to the best of our knowledge, is the first wearable OCT angiography probe capable of long-term monitoring of mouse brain blood flow. This wearable imaging probe has a maximum scanning speed of 76 kHz, with a 12 mu m axial resolution, 5.5 mu m lateral resolution, and a large field of view (FOV) of 4 mm x 4 mm. It offers easy assembly and stable imaging, enabling it to capture brain vessels in freely moving rodents. We tested this probe to monitor cerebral hemodynamics for up to 4 hours during the acute ischemic phase after photothrombotic stroke in mice, highlighting the reliability and long-term stability of our probe. This work contributes to the advancement of wearable biomedical imaging.

Abstract:Significance: Microcirculation and neurovascular coupling are important parameters to study in neurological and neuro-ophthalmic conditions. As the retina shares many similarities with the cerebral cortex and is optically accessible, a special focus is directed to assessing the chorioretinal structure, microvasculature, and hemodynamics of mice, a vital animal model for vision and neuroscience research. Aim: We aim to introduce an optical imaging tool enabling in vivo volumetric mouse retinal monitoring of vascular hemodynamics with high temporal resolution. Approach: We translated the spatio-temporal optical coherence tomography (STOC-T) technique into the field of small animal imaging by designing a new optical system that could compensate for the mouse eye refractive error. We also developed post-processing algorithms, notably for the assessment of (i) localized hemodynamics from the analysis of pulse wave-induced Doppler artifact modulation and (ii) retinal tissue displacement from phase-sensitive measurements. Results: We acquired high-quality, in vivo volumetric mouse retina images at a rate of 113 Hz over a lateral field of view of ∼ 500 μ m . We presented high-resolution en face images of the retinal and choroidal structure and microvasculature from various layers, after digital aberration correction. We were able to measure the pulse wave velocity in capillaries of the outer plexiform layer with a mean speed of 0.35 mm/s and identified venous and arterial pulsation frequency and phase delay. We quantified the modulation amplitudes of tissue displacement near major vessels (with peaks of 150 nm), potentially carrying information about the biomechanical properties of the retinal layers involved. Last, we identified the delays between retinal displacements due to the passing of venous and arterial pulse waves. Conclusions: The developed STOC-T system provides insights into the hemodynamics of the mouse retina and choroid that could be beneficial in the study of neurovascular coupling and vasculature and flow speed anomalies in neurological and neuro-ophthalmic conditions.
Noninvasive volumetric imaging and morphometry of the rodent retina with high-speed, ultrahigh-resolution optical coherence tomography

Abstract:Purpose: To demonstrate high-speed, ultrahigh-resolution optical coherence tomography (OCT) for noninvasive, in vivo, three-dimensional imaging of the retina in rat and mouse models. Methods: A high-speed, ultrahigh-resolution OCT system using spectral, or Fourier domain, detection has been developed for small animal retinal imaging. Imaging is performed with a contact lens and postobjective scanning. An axial image resolution of 2.8 mum is achieved with a spectrally broadband superluminescent diode light source with a bandwidth of approximately 150 nm at approximately 900-nm center wavelength. Imaging can be performed at 24,000 axial scans per second, which is approximately 100 times faster than previous ultrahigh-resolution OCT systems. High-definition and three-dimensional retinal imaging is performed in vivo in mouse and rat models. Results: High-speed, ultrahigh-resolution OCT enabled high-definition, high transverse pixel density imaging of the murine retina and visualization of all major intraretinal layers. Raster scan protocols enabled three-dimensional volumetric imagingand comprehensive retinal segmentation algorithms allowed measurement of retinal layers. An OCT fundus image, akin to a fundus photograph was generated by axial summation of three-dimensional OCT data, thus enabling precise registration of OCT measurements to retinal fundus features. Conclusions: High-speed, ultrahigh-resolution OCT enables imaging of retinal architectural morphology in small animal models. OCT fundus images allow precise registration of OCT images and repeated measurements with respect to retinal fundus features. Three-dimensional OCT imaging enables visualization and quantification of retinal structure, which promises to allow repeated, noninvasive measurements to track disease progression, thereby reducing the need for killing the animal for histology. This capability can accelerate basic research studies in rats and mice and their translation into clinical patient care.
Wearable optical coherence tomography angiography probe for monitoring mouse brain in <i>vivo</i>

Abstract:Optical coherence tomography (OCT) has become a promising tool for studying anatomical and functional dynamics of the cerebral cortex, offering advantages such as label-free imaging, high resolution, and non-invasive optical biopsy. However, observing the brains of non-anesthetized and freely moving mice has been a long-standing challenge for OCT. In this study, we designed a wearable OCT probe to observe the vascular morphology of the mouse brain and track shortterm vascular changes after thrombosis. We utilized a microelectromechanical system (MEMS) scanning mirror for three-dimensional scanning. Compared to traditional OCT systems, this wearable imaging probe features miniaturization, low cost, portability, and stability, allowing for imaging of the mouse brain in a non-anesthetized and freely moving state. The entire probe weighs 8 g and achieves a lateral resolution of 5.5 mu m, a longitudinal resolution of 12 mu m, and an effective imaging area of 4 mm x 4 mm. We evaluated the performance of the probe through phantom experiments and imaging of the mouse brain's vascular network, and successfully monitored local vascular morphological changes in the mouse brain shortly after stroke under awake conditions. We believe that the wearable probe can be applied in various fields such as ophthalmology, dermatology, and dentistry, and due to its portability and non-invasiveness, the wearable OCT probe is expected to have wide clinical research applications.
