Abstract:A highly regioselective iodosulfonylation of allenes in the presence of CuI and 1,10-phenanthroline has been developed for the synthesis of various useful ( E)-α-iodomethyl vinylsulfones in moderate to excellent yields. This practical reaction is fast, operationally simple, and in particular, proceeds under very mild conditions to afford the target products with high regio- and stereoselectivity. The selectivity was illustrated by a conceptual DFT analysis.

Abstract:Naturally occurring conjugated allenynes are of general interest owing to their potent and various biological activities. The 1,5-H transfer of alka-1,4-diyn-3-yl amines would be one of the most straightforward yet challenging approaches to such compounds since, in principle, two regioisomers may be formed involving two C-C triple bonds. Herein, a catalytic recipe of copper halides with mixed oxidation states, i.e., CuCl/CuBr2 , has been identified to address the issues of the side reaction of conjugate addition and the selectivity of 1,5-H transfer of alka-1,4-diyn-3-yl amines in EATA (enantioselective allenation of terminal alkynes) reaction involving 2-alkynals. This method provided various allenynes with excellent enantioselectivities, and was also applied to the first highly enantioselective total synthesis of natural product scorodonin. Mechanistic studies and DFT calculations elucidated the regioselectivity for the observed 1,5-H transfer.

Abstract:We describe a new method for the stereoselective synthesis of (Z)-1,2-dithio-1-alkenes via copper-catalyzed cross-coupling of (Z)-2-bromovinyl sulfides and thiols. The desired products are obtained in good to excellent yields. The double bond geometry of the starting (Z)-2-bromovinyl sulfides is retained under these reaction conditions.

Abstract:An organocatalytic, enantioselective domino Michael-aldol reaction of 2-mercaptobenzaldehydes with maleimides has been developed. The process, catalyzed by a bifunctional chiral amine thiourea via a hydrogen-bonding mediated activation mechanism using as low as 1 mol% catalyst loading, affords versatile succinimide-containing benzothiopyrans with the generation of 3 stereogenic centers in one single operation.

Abstract:1,2,3-Thiadiazoles are precursors of thioacylcarbenes and thioketenes and show various reactions with unsaturated compounds. The rhodium-catalyzed reactions of 1,2,3-thiadiazoles and alk-2-enenitriles, including alkyl, aryl, and heteroaryl acrylonitriles, have been investigated, showing chemospecific reactivity to generate 3-vinylisothiazole derivatives via the (3+2) transannulation between the cyano group of alk-2-enenitriles and generated thioacylcarbenes. (Z)-Cinnamonitriles are less active than (E)-cinnamonitriles due to the strong coordination of the (Z)-C=C bond and the Rh catalyst. The current reaction provides a method for the synthesis of 3-vinylisothiazole derivatives.

Abstract:A highly enantioselective synthesis of chiral β-hydroxy thioesters that uses a decarboxylative aldol reaction of malonic acid half thioesters and aldehydes catalyzed by a chloramphenicol base-derived bifunctional organocatalyst is reported. The resulting chiral β-hydroxy thioesters were obtained in high yields (up to 82%) with good to excellent enantioselectivities (up to 94% ee). The synthetic application of the methodology is illustrated by the asymmetric synthesis of the selective serotonin reuptake inhibitor dapoxetine.

Abstract:Using thianthrene S-oxide (TTSO) as a transient mediator, para-arylation and alkenylation of mono-substituted arenes have been demonstrated via a para-selective thianthrenation/Pd-catalyzed thio-Suzuki-Miyaura coupling sequence under mild conditions. This reaction features a broad substrate scope, and functional group and heterocycle tolerance. The versatility of this approach was further demonstrated by late-stage functionalization of complex bioactive scaffolds, and direct synthesis of some pharmaceuticals, including Tetriprofen, Ibuprofen, Bifonazole, and LJ570.

Abstract:Fluorinated motifs are frequently encountered indrugs and agrochemicals. Incorporatingfluorine-containing motifsin drug candidates for lead optimization in pharmaceutical researchand development has emerged as a powerful tool. The constructionof molecules that feature a trifluoromethyl (CF3-) group on astereogenic carbon has accumulated broad research efforts. Unlike its well-explored, biologically active methyl counterpart,asymmetric construction of beta-trifluoromethylated alcohols bearing adjacent stereocenters still remains elusive. Throughretrosynthetic analysis, we posited that followed by sequential reduction of carbonyl, the initial construction of chiral alpha-trifluoromethylated ketones could render the desired product in a facile, one-pot fashion. Herein, we developed thefirst example ofnickel-catalyzed asymmtric reductive cross-coupling trifluoroalkylation of acyl chlorides for enantioselective synthesis of diverse alpha-trifluoromethylated ketones. The one-pot reduction of these alpha-trifluoromethylated ketones furnished corresponding alcohols bearing beta-CF3-substituted stereogenic carbons with excellent diastereoselectivity and complete enantioselective retention. High yields/enantioselectivity, mild conditions, and good functional group compatibility are shown in the system. Utilities of the method are alsoillustrated by applying asymmetric, late-stage trifluoroalkylation of biologically active complex molecules, revealing tremendouspotential for development of CF3-containing chiral drugs.

Abstract:An organocatalytic asymmetric enantioselective domino oxa-Michael–Mannich-[1,3]-amino rearrangement reaction of N-tosylsalicylimines with a wide range of α,β-unsaturated aldehydes utilizing diarylprolinol silyl ether catalysis is described. The catalytic reactions proceed with excellent enantioselectivity (up to 99% ee) to produce the corresponding chair N-tosylimines-chromenes with a yield of up to 99%, tolerating a range of functional groups. This methodology offers a new method with great potential to further extend the synthetic power and versatility of chiral aminocatalysis.

Abstract:The use of alkylthio electrophiles in synthesis has remained elusive because of the lack of a suitable reagent that is practical and of excellent enantioselectivity and appropriate reactivity. In this work we introduce a novel alkylthio reagent based on the 2-mercapto-5-methyl-1,3,4-thiadiazole (MMTD) fragment for direct alkylsulfenylation of ketones and aldehydes. It can be readily prepared by the oxidative coupling between thiadiazole and other alkylthio reagents and be combined with chiral primary aminocatalysis. This protocol provides facile access to diverse α-alkylthio quaternary carbon centers with good stereoselectivities.

Abstract:We have developed a highly selective one-pot method for the synthesis of (E)-vinyl sulfones and sulfoxides from thiols with terminal alkynes. The sulfones and sulfoxides could be obtained with excellent selectivity in good isolated yields. It is simple, efficient and environmentally benign, and metal-free. The mechanism for the formation of the (E)-vinyl sulfones was also proposed.

Abstract:A tertiary amino‐thiourea catalyzed asymmetric aldol reaction between 2‐isocyanatomalonate esters and isatins has been achieved. Optical active spirooxazolidinone oxindole derivatives could be obtained with excellent yields (80%–96%) and enantioselectivities (67%–99% ee) under mild conditions, providing an updated example to apply isatins as the electrophile in the asymmetric aldol reaction with 2‐isocyanatomalonate esters.

Abstract:Highly enantioselective synthesis of acyclic α-tertiary amines through asymmetric desymmetrization is reported. This approach is based on chiral phosphoric acid mediated, enantioselective, oxidative desymmetrization of 2-substituted 2-nitro-1,3-diolbenzylidine acetals in the presence of DMDO as an oxidant. The method allows for the formation of a wide variety of chiral 2-nitro-1,3-diols in high enantioselectivity, which could be transformed into optically pure, unnatural α-alkyl series. The synthetic utility of this method has been further demonstrated by the expedient construction of the core structure of natural products manzacidins enantioselectively.

Abstract:In this article, we present an efficient, one-pot regioselective approach towards the synthesis of 2-aryl/hetaryl-4-methyl-5-acylthiazoles obtained during the reaction of α-bromo-1,3-diketones generated in situ by triturating unsymmetrical 1,3-diketones with N-bromosuccinimide, with various thioamides under solvent-free conditions. This environmentally benign protocol showed large functional group tolerance, resulted in the exclusive formation of a single isomer, out of the two possible regioisomers in admirable yields. The structure of the isomeric thiazole was assigned unambiguously on the basis of multinuclear NMR [(1H-13C) HMBC, (1H-13C) HMQC, (1H-15N) HMBC] and X-ray crystallographic studies. The entire protocol is ecologically desirable as it employs no organic solvent.GRAPHICAL
