The effect of Ezhu (莪术)on metastasis-associated capacities of melanoma cells

Abstract:Objective: To study the effect of Ezhu, a kind of traditional Chinese medicine which can dissolve the stasis by activating the blood circulation, on the metastasis-associated capacities of B16-BL6 metastatic mouse melanoma cells. Methods: The proliferation, adhesion, invasion and migration capacity of B16-BL6 was evaluated respectively by cck-8 assay, adhesion assay and reconstituted basement membrane invasion and migration assay in vitro. Mouse spontaneous melanoma model was used to study the effect on metastasis in vivo. Results: At some high concentrations, the extract of Ezhu inhibited the proliferation of B16-BL6 cells significantly; It could inhibit the adhesion of B16-BL6 cells to the basement component Laminin, suppress B16-BL6 cells invading the reconstituted basement membrane and reduce the chemotactic migration capacity of B16-BL6 cells; It could suppress significantly the growth and metastasis of B16-BL6 mouse melanoma. Conclusion: The suppressive effect of Ezhu on the metastasis of B16-BL6 metastatic mouse melanoma cells may relate to the effect of Ezhu on inhibiting the B16-BL6 cells adhering to the extracellular matrix and reducing the invasion and migration capacity of B16-BL6 cells.
