Effect of Treatment with Yiqi Baidu Recipe on Expression of the Tumor Metastatic Suppressor Gene in Mouse Melanoma Model

Abstract:Objective In order to explore the possible mechanism of traditional Chinese medicine (TCM) in treatment of metastatic melanoma,we have investigated the effect of treatment with Yiqi Baidu recipe on the expression of the tumor metastatic suppressor gene in mouse melanoma model with lung metastasis. Methods 60 C57BL/6 mice were divided in to three groups,each had 20 mice,to establish metastatic mouse melanoma model 156g YiQi Baidu recipe and 20mg/kg CTX were given by stomach tub for 3 days,the control mice were given saline,and all for 20 days. Yiqi Baidu recipe(56g TCM/kg) group,cyclophosphamide (CTX20mg/kg×3day) group,and saline control group,at the day after the end of treatment,they were executed by over bleeding from femoral artery,we used presser bits to count the number of metastatic tumors in the lungs. Both the expression of both nm23 and E-cadherin in tumor tissues by immunohistochemistry and the protein level of S100b by ELISA were examined after the treatment with Yiqi Baidu recipe. Results The number of metastatic tumors in CTX group were 59±22,79±20 in Yiqi Baidu recipe group,and 108±26 in saline group. After the treatment with Yiqi Baidu recipe,there was a significant reduction in the number of metastatic tumors in the lungs of the treatment group as compared to that of the group of saline controls(t=3.89,P0.05). After the treatment with Yiqi Baidu recipe and CTX,there were significant expressions of the tumor metastatic suppressor gene (nm23) in mouse melanoma model with lung metastasis with statistical significance in both groups,(χ2TCM=10.3,P0.05; χ2CTX=9.16,P0.05),while there was no statistical significance in nm23 expression level compared with CTX and Yiqi Baidu recipe Groups,whereas there was actually no expression of E-cadherinin in all tissues. After the treatment with Yiqi Baidu recipe and CTX,there were reductions of serum level of S100B in the groups with lung metastasis,although there was no statistical significance as compared to the group of saline controls(P0.05). Conclusion Data obtained have suggested that the treatment with Yiqi Baidu recipe suppress the metastatic activity in the experimental animal model of melanomas,in which molecular mechanism of regulating gene expression of nm23 may be involved.
