Abstract:In order to design the precipitation process using gas antisolvent (GAS) to prepare micron particles, the equilibrium solubility of puerarin in alcohol+CO2 solution was measured in the temperature range of 308.2~328.2℃ and under the pressure changing up to 14 MPa. The measured results show that the pressure inside the vessel is the key factor of the GAS process which has a great influence on the solubility of puerarin in the solution. The experimental results also show unexpectedly that the concentration of the puerarin in the alcohol solution increases somewhat with increasing pressure at first and it can be explained as follows. When CO2 is mixed with the alcohol solution, some alcohol solvent will disperse into the supercritical fluid, so it causes the solution to become denser. After that, as the pressure increases further from 4 MPa to 10MPa, the concentration of the solution decreases dramatically, this is caused by the GAS crystallization of the solute puerarin. However the antisolvent effect will reduce when more CO2 is injected with the pressure increasing over 10MPa. The experiment results imply that, besides the antisolvent function of CO2 which results in solubility decrease, the extraction of alcohol from solution into supercritical fluid, which causes the densification of the solution, would also promote the process of GAS crystallization.

Abstract:The solubility of puerarin in water, ethanol, and acetone solutions was measured over the temperature range of (288.2 to 328.2) K. The solubility of puerarin in water increases with increasing temperature, whereas the solubility of puerarin in ethanol and acetone decreases with increasing temperature. The solubility data were correlated with the Apelblat equation.

Abstract:The effects of extraction time, temperature, pressure and different concentration of ethanol and their interactions on the yields of amentoflavone, quercetin and ginkgetin extracted from Taxus chinensis by supercritical CO2 were investigated by using a central composite design (CCD). An CCD experimental design with four factors and five levels was used to optimize the extraction parameters. Ultra performance liquid chromatography (UPLC) was used to analyze the content of the tree components in the extracts. Experimental results show that the main effects of factors and their interactions are significant on the yields (p < 0.05). The optimal extraction conditions were established for the three compounds: yield of 4.47 mg/g for amentoflavone at 48 °C, 25 MPa, 2.02 h and 78.5% ethanol, 3.73 mg/g for quercetin at 46 °C, 24 MPa, 2.3 h, 82% ethanol and 3.47 mg/g for ginkgetin at 48 °C, 20 MPa, 2.38 h, 82% ethanol, respectively.

Abstract:An efficient and solvent-free procedure was developed to improve the solubility and bioavailability of the poorly water-soluble drug puerarin by mechanochemical technology. The stable inclusion complex of puerarin and 2-hydroxypropyl-β-cyclodextrin (HPCD) was prepared by a ball mill under the following conditions: equimolar ratio of puerarin to HPCD; rotational speed of 250 rpm; milling time of 90 min; steel balls of 22 mm diameter. The solid complex was characterized by scanning electron microscopy (SEM), differential scanning calorimetry (DSC), powder X-ray diffractometry (XRD), and fourier transformation-infrared spectroscopy (FT-IR). Mechanochemical action could result in enhanced molecular encapsulation, homogeneous distribution and amorphization of the drug. In comparison to puerarin, a 1.64-fold increase in absolute bioavailability was obtained. The solubility of the inclusion complex was 25.33-fold higher and the drug release amount reached 79.44% at 15 min, 2.76-fold higher.
Study on Enzymatic Extraction of Isoflavone from Pueraria (Pueraria lobata) Root Residue

Abstract:Pueraria lobata(Willd.)Ohwi is a kind of raw material to produce starch in China.The root residue of pueraria from(pueraria-starch) manufactory still contains some isoflavone which is a kind of phytoestrogen with important physiological effects.By(pretreating) pueraria root residue with cellulase or xylanase before ethanol extraction,the total yield of isoflavone could be efficiently(increased.) The optimal cellulase dosage was 10 FPIU/g pueraria root residue and the suitable cellulase treatment time was 12 h.Total yield of isoflavone was 1.36 times as that by normal ethanol extraction.For pretreatment of pueraria root residue with xylanase,the(optimal) xylanase dosage was 300 IU/g residue for 6 h.Total yield of isoflavone was 1.52 times as that by regular ethanol extraction.It was proved that cellulase and xylanase could cooperate with each other to enhance extraction efficiency.When pueraria root residue was pretreated with both cellulase(7.5 FPIU/g residue) and xylanase(300 IU/g residue) for 6 h,total yield of isoflavone reached(138) mg/10 g pueraria root residue,which was 1.64 times as that by regular ethanol extraction.

Abstract:The solubilization amount of oxymatrine in ammonium carboxylate perfluoropolyether (PFPE-NH4) water-in-supercritical carbon dioxide microemulsions with a water-to-surfactant molar ratio of (10.2, 14.8, and 21.0) was determined at temperatures of (308.15, 313.15, 318.15, 323.15, and 328.15) K and a density of 0.822 g.mL(-1) using a static apparatus with a sampling part. The determined solubilization amount of oxymatrine was 2 orders of magnitude higher than the solubility of oxymatrine in supercritical carbon dioxide and about 1/6 to 1/5 of the solubility of oxymatrine in bulk water. It was indicated that the solubilization amount of oxymatrine increased with an increase of the water-to-surfactant molar ratio and temperature in all experimental conditions.

Abstract:The equilibrium solubilities of two dodecylpolyoxyethylene polyoxypropylene ether nonionic surfactants (A(EO)BmB(PO)(n)) in supercritical carbon dioxide were measured at temperatures ranging from (308 to 328) K and pressures from (13.6 to 32.0) MPa in a stainless steel variable-volume view cell. The experimental data were correlated using a semiempirical model. The calculated results show satisfactory agreement with the experimental data.

Abstract:The solubility of Troeger's base (TB) in SC-CO2/CH3OH was measured at 308K ill the pressure range of 8 to 13 MPa, and methanol concentration ranges from 0 to 20 mol%. The solubility increases dramatically with methanol concentration. Comparison of solubility in supercritical CO2 with and without 10 mol% CH3OH ill the pressure of 8 MPa at 308K implies that both curves are sleep in the lower pressure range but become smooth as the pressure increases.

Abstract:Puerarin is an isoflavonoid extracted from the roots of Pueraria lobata in the Pueraria thomsonii family. Puerarin displays a broad range of pharmacological activities and has been widely used in clinics. However, puerarin's low solubility and permeability result in poor gastrointestinal absorption and low bioavailability. Simply increasing the dose does not effectively increase bioavailability and might lead to toxicity and side effects. Considerable progress has been achieved in the recent years on puerarin structural biotransformation using microorganisms and free enzymes. Increasingly numerous new puerarin derivatives with high solubility and bioactivity have been developed. This paper reviews recent progress in the biotransformation of puerarin.

Abstract:3,4-Dihydroxybenzaldehyde (protocatechuic aldehyde), (E)-3-(3,4-dihydroxyphenyl)prop-2-enoic acid (caffeic acid), (2S,3R,4S,5R,6R)-6-(hydroxymethyl)oxane-2,3,4,5-tetrol (D-galactose), and (3R,4S,5R,6R)-2-[[(2R,3S,4S,5R)-6-[(2S,3S,4S,5R)-3,4-dihydroxy-2,5-bis(hydroxymethyl)oxolan-2-yl]oxy-3,4,5-trihydroxyoxan-2-yl]methoxy]-6-(hydroxymethyl)oxane-3,4,5-triol (D-raffinose) are substances contained in danshen. In this study, the solubilities of protocatechuic aldehyde, caffeic acid, D-galactose, and D-raffinose pentahydrate in the mixtures of ethanol and water from 273.2 K to 303.2 K were determined. The solubilities of protocatechuic aldehyde and caffeic acid in ethanol-water solutions are significantly higher than those in pure water. The solubilities of D-galactose and D-raffinose pentahydrate decrease as ethanol mass fraction increases. The solubilities of the four compounds increase as equilibrium temperature increases. Prediction values of D-galactose calculated using universal functional activity coefficient (UNIFAC) models agreed well with experimental data.

Abstract:Puerarin (daidzein-8-C-glucoside) is an isoflavone isolated from several leguminous plants of the genus Pueraria. Puerarin possesses several pharmacological properties; however, the poor solubility of puerarin limits its applications. To resolve this poor solubility, Deinococcus geothermalis amylosucrase (DgAS) was used to modify puerarin into more soluble derivatives. The results showed that DgAS could biotransform puerarin into a novel compound: puerarin-4′-O-α-glucoside. The biotransformation reaction was manipulated at different temperatures, pH values, sucrose concentrations, reaction times, and enzyme concentrations. The results showed that the optimal reaction condition was biotransformed by 200 μg/mL DgAS with 20% (w/v) sucrose at pH 6 and incubated at 40 °C for 48 h, and the optimal production yield was 35.1%. Puerarin-4′-O-α-glucoside showed 129-fold higher solubility than that of puerarin and, thus, could be further applied for pharmacological use in the future.

Abstract:Puerarin is an isoflavone extracted from Chinese plant, Pueraria lobata (Wild.) Ohwi. It has been reported to have comprehensive pharmacological action in treatment of diabetes and cardiovascular diseases. The purpose of this study was to link the scattered effects of puerarin and to find the common mechanisms underlying. We investigated the effect of puerarin on the pivotal common pathogenic factors of metabolic syndrome, which includes obesity, Type II diabetes and cardiovascular diseases. Recently, a large body of evidence indicates that there is a complicated interplay among insulin resistance, adipocytes and endothelial dysfunction that links the abnormalities of metabolic syndrome. Results of present study showed that puerarin could potentiate insulin-induced preadipocyte differentiation, promote glucose-uptake of adipocytes that have been induced insulin resistance by high glucose, and prevent TNF-a-induced apoptosis and viability loss of endothelial cells. Furthermore, we found that these effects are probably due to promote PPARgamma expression and partly through inhibiting abnormal TNF-a-induced intracellular-free Ca(2+) accumulation of endothelial cells. Overall, our synthetical study links the comprehensive pharmacological actions of puerarin to the recognized common pathogenesis of metabolic syndrome, and provides a new insight into the mechanism of puerarin effect.
Studies on Isoflavone Production from Pueraria Root Residue under Solid-state Fermentation by Aspergillus Niger ZU-01

Abstract:Pueraria root residue from pueraria starch manufactory contains definite amount of isoflavone which is a kind of phytoestrogen with important physiological effects. Pueraria root residue was used as the main material for solid-state fermentation by Aspergillus niger ZU- 01 strain. Xylanase and endo-β-glucanase(CMCase) produced from solid state fermentation could hydrolyze the cell wall′s fibre of pueraria root residue, thus improving the yield of isolavone. Optimized culture medium of solid-state fermentation was composed of 70 % pueraria root residue and 30 % wheat bran, with 70 % water content. After 120 h fermentation at the temperature of 30 ℃, the yield of total isoflavone reached 1.82 times as that by normal ethanol extraction. HPLC showed that the content of daidzein decreased while the content of isoflavone glucoside increased after fermentation. A 70 % higher content of puerarin which reached 3.97 mg/g pueraria root residue was gained comparing to that of normal ethanol extraction.

The solubility of imperatorin in the above solvents decreased in the order ethyl acetate > methanol > ethanol > n-hexane and petroleum ether. The solubility data were well correlated with a modified Apelblat equation.

Abstract:Glucose via acylation and bromization pathway gives acetobromo-α-D-glucose.The etherification of puerarin to 7-hydroxyl-4′-(2,3,4,6-tetraacetyl-β-D-glucopyranosyloxy) puerarin with acetobromo-α-D-glucose was carried out in acetone at the presence of potassium carbonate.The isolated yield was 61.4%(based on puerarin).Finally,7-hydroxyl-4′-(β-D-glucopyranosyloxy)puerarin was obtained via deacetylation in anhydrous methanol at the presence of potassium carbonate with the isolated yield of 90.5%.The structures of intermediates and final product were characterized by 1HNMR spectra and MS.Compared with puerarin,the water-solubility of 7-hydroxyl-4′-(β-D-glucopyranosyloxy) puerarin was increased by 14.2 times.

Abstract:Radix Puerariae (RP), a traditional Chinese medicinal herb, has been used for a variety of disease prevention and treatment purposes. Three isoflavones, puerarin, daidzin and daidzein, isolated from RP are responsible for its broad therapeutic effects. In the present study, we demonstrate the application of three extraction methods, traditional, pressurized solvent extraction (PSE) and ultrasonic techniques, for preparing ethanolic RP extract. A comparison of the three extraction methods showed preferential higher yields of the three major isoflavones when the ultrasonic technique was applied. The extraction yield became higher as the mean size of RP particles decreased while the total accumulated power varied from 20 to 80MJ at the solid-to-solvent ratios of 1:5 and 1:10 (g/ml). The relationship among accumulated energies, extraction yields and mean particle sizes under different extraction ratios using ultrasound was also discussed. This study proves that using the ultrasonic method should be the most economic way for enhancing the extraction yield of isoflavones-containing herbal extract in a short time with a reduced amount of solvent at a lower temperature.
