A Study on Promoter Methylation of CRMP4 in Prostate Cancer

Abstract:Objectives To investigate CpG island methylation status of CRMP4 promoter in prostate cancer and the relationship between promoter methylation and CRPM4 expresssion. To design MSP primers of CRMP4 promoter to predict micrometastasis of prostate cancer. Methods PC3M cells were treated with 0.5, 5, 12.5 μM demethylating agent 5-aza-dC for 5 days, and then were harvested for protein extraction for Western blot analysis of expression of CRMP4. Bisulfite Sequencing PCR (BSP) was performed to determine the methylation patterns of CRMP4 promoter. According to the BSP results, the Methylation specific PCR (MSP) primer pairs for CRMP4 were designed to detect the methylation status of CRMP4 promoter. Results The treatment with demethylating agent 5-aza-dC in PC3M cells was effective for re-expression of CRMP4, and the upregulation were correlated with the concentration of the demethylating agent. BSP showed that the CpG sites in -848, -841, -690, -680, -678, -674, -671, -665, -660 and -658 of CpG island were highly or moderately methylated among the clones derived from DNA obtained from cell lines (PC3, PC3M and DU145) and metastatic prostate cancer patient (primary lesion and metastatic lymph nodes). These methylated CpG sites were continuous CpG sites in two regions. In contrast, the CpG sites in localized prostate cancer, BPH and normal prostate were mostly unmethylated except a few sporadic 5mCs in some of the clones. With the designed MSP primer pairs, methylation was observed in most of the metastatic prostate cancer, most of locally advanced PCa and all of prostate cancer cell lines. Conclusion Downregulation of CRMP4 in prostate cancer correlates with promoter methylation and the designed MSP primer pairs to detect CRMP4 promoter methylation have a poten- tial in predicting metastasis of prostate cancer
